Effects of zinc oxide nanoparticle dispersants on cytotoxicity and cellular uptake

Dispersion critically affects the physicochemical properties of nanoparticles and their interactions with biological systems. In this study, the effects of different zinc oxide nanoparticle (ZnO-NP) dispersants, that is, bovine serum albumin, citrate, carboxymethyl cellulose, fetal bovine serum, and cell culture medium, were investigated with respect to cytotoxicity and cellular uptake. Parallel comparative studies were also conducted with <TEX>$Zn^{2+}$</TEX> ions. The results demonstrated that ZnO-NPs dispersed in citrate exhibited the greatest cytotoxicity to human lung cells, probably related to their high cellular uptake via the citrate internalization mechanism, whereas, the energy-dependent endocytosis pathway of ZnO-NP internalization in cells was unaffected by dispersant type. These results emphasize that dispersant choice is important when evaluating the toxicity of nanoparticles and that results should be interpreted with caution.

In recent days, there is a growing attention toward well-being, health, and functional cosmetics. In this regard, the Jeju lava seawater and red ginseng have been widely applied as food and cosmetic in many industrial products. In the current research, a mixture of Jeju lava seawater and red ginseng extract has been confirmed to have an appropriate concentration for processing RAW264.7, HaCaT and THP-1 cells, 0.625% for RAW264.7, 3.125% for HaCaT. For THP-1 cell, the mixed sample at the concentration of less than 10% did not affect the cell viability. When the test was conducted based on this, the mixture of Jeju lava seawater and red ginseng extract significantly inhibited the expression of NO increase by LPS, and IL-8 increase by TNF-α and UVA. IL-8, IL-6 induced by UVB also showed significant inhibitory efficacy in a concentration-dependent manner. The result of suppression of the expressions of CD54 and CD86 cell surface proteins involved in the stimulation response in immune cells induced by stimulation with the chemical sensitizer DNCB, which is the cause of the immune response, suggest that the mixed sample is effective in anti-stimulation. This result shows that the mixture of Jeju lava seawater and red ginseng extract is a potential substance for skin inflammation relief and anti-irritation.

Sulforaphane [1-isothiocyanato-4-(methylsulfinyl)-butane] is an isothiocyanate found in some cruciferous vegetables, especially broccoli. Sulforaphane has been shown to display anti-cancer properties against various cancer cell lines. Matrix metalloproteinase-9 (MMP-9), which degrades the extracellular matrix (ECM), plays an important role in cancer cell invasion. In this study, we investigated the effect of sulforaphane on 12-O-tetradecanoyl phorbol-13-acetate (TPA)-induced MMP-9 expression and cell invasion in MCF-7 cells. TPA-induced MMP-9 expression and cell invasion were decreased by sulforaphane treatment. TPA substantially increased NF-B and AP-1 DNA binding activity. Pre-treatment with sulforaphane inhibited TPA-stimulated NF-B binding activity, but not AP-1 binding activity. In addition, we found that sulforaphane suppressed NF-B activation, by inhibiting phosphorylation of I?B in TPA-treated MCF-7 cells. In this study, we demonstrated that the inhibition of TPA-induced MMP-9 expression and cell invasion by sulforaphane was mediated by the suppression of the NF-B pathway in MCF-7 cells.

Herpes simplex virus (HSV) is a common pathogen, that causes a broad spectrum of diseases, ranging from minor skin infections to severe encephalitis and widespread infections. Acute retinal necrosis (ARN), one of the most serious manifestations of HSV infection, is defined as a rapidly progressing necrotizing retinopathy that presents discrete areas of circumferential retinal necrosis, along with signs of uveitis, vitreitis, and retinal vasculitis. We encountered a case of a female infant, born at 33 weeks of gestation with a body weight at birth of 2,080 g, who had ARN and encephalomalacia due to HSV infection. ARN associated with HSV infection should be suspected when nonspecific retinal exudates are observed in neonates, especially preterm infants.

Newcastle disease (ND) is an infectious poultry disease that caused high mortality and reduced egg production. NDVs are regularly present in the domestic duck population. And ducks play a possible role in the maintenance and transmission of NDVs. While we were monitoring the Avian Influenza, NDVs were isolated from field samples by accident. So we analysed the biological and genetic characteristics of these viruses. Lentogenic NDVs were isolated from two farms among twenty breeder duck farms. The ages of ducks were 39 weeks old in the ‘A’ farm and 3∼72 weeks old in the ‘B’ farm. And they were not inoculated with the NDVs vaccine. In the biological characteristics, the both viruses which separated from the farm ‘A’ and ‘B’ were thermostable. The amino acid sequence of a site from 112 to 119 in the fusion (F) protein was ‘GKQGRLIG’ which has monobasic motif in the samples of both farms. And this means the separated NDVs are lentogenic. Phylogenetic analysis was performed by en-tire nucleotide sequence of F protein. The virus strains from the A farm (MN095239) and the B farm (MN095240) belonged to class II genotype I. Using the analysis of whole F protein nucleic acid se-quence, the MN095239 (GenBank) had homology with Ulster strain about 99.95% and the MN095239 (GenBank) had homology with KR/CK/KU_LBM255/09 strain about 99.89%. NDV surveillance is needed to investigate epidemiological relationship of domestic breeder duck isolates in Korea.

<P>The present study aimed to investigate the comparative evaluation of pharmacological efficacy between sulfasalazine alone and sulfasalazine in combination with herbal medicine on dextran sodium sulfate- (DSS-) induced UC in mice. Balb/c mice received 5% DSS in drinking water for 7 days to induce colitis. Animals were divided into five groups (<I>n</I> = 9): Group I (normal group), Group II (DSS control group), Group III (DSS + sulfasalazine (30 mg/kg)), Group IV (DSS + sulfasalazine (60 mg/kg)), and Group V (DSS + sulfasalazine (30 mg/kg) + Cinnamomi Cortex and Bupleuri Radix mixture (30 mg/kg) (SCB)). Colonic pathological changes were analyzed using hematoxyline/eosin staining. The antioxidant, inflammatory, and apoptotic protein levels were determined using western blotting. SCB supplementation, as well as sulfasalazine, suppressed colonic length and mucosal inflammatory infiltration. In addition, SCB treatment significantly reduced the expression of proinflammatory signaling molecules through suppression of both mitogen-activated protein kinases (MAPK) and nuclear factor-kappa B (NF-<I>κ</I>B) signaling pathways and prevented the apoptosis of the colon. Moreover, SCB administration significantly led to the upregulation of antioxidant enzymes including SOD and catalase. Taken together, SCB treatment might offer a better treatment for human UC than sulfasalazine alone or may be useful as an alternative therapeutic strategy against UC, without any evidence of side effects.</P>
