<P>APIP, Apaf-1 interacting protein, has been known to inhibit two main types of programmed cell death, apoptosis and pyroptosis, and was recently found to be associated with cancers and inflammatory diseases. Distinct from its inhibitory role in cell death, APIP was also shown to act as a 5-methylthioribulose-1-phosphate dehydratase, or MtnB, in the methionine salvage pathway. Here we report the structural and enzymatic characterization of human APIP as an MtnB enzyme with a <I>K</I><SUB><I>m</I></SUB> of 9.32 μM and a <I>V</I><SUB><I>max</I></SUB> of 1.39 μmol min<SUP>−1</SUP> mg<SUP>−1</SUP>. The crystal structure was determined at 2.0-Å resolution, revealing an overall fold similar to members of the zinc-dependent class II aldolase family. APIP/MtnB exists as a tetramer in solution and exhibits an assembly with <I>C4</I> symmetry in the crystal lattice. The pocket-shaped active site is located at the end of a long cleft between two adjacent subunits. We propose an enzymatic reaction mechanism involving Glu139* as a catalytic acid/base, as supported by enzymatic assay, substrate-docking study, and sequence conservation analysis. We explored the relationship between two distinct functions of APIP/MtnB, cell death inhibition, and methionine salvage, by measuring the ability of enzymatic mutants to inhibit cell death, and determined that APIP/MtnB functions as a cell death inhibitor independently of its MtnB enzyme activity for apoptosis induced by either hypoxia or etoposide, but dependently for caspase-1-induced pyroptosis. Our results establish the structural and biochemical groundwork for future mechanistic studies of the role of APIP/MtnB in modulating cell death and inflammation and in the development of related diseases.</P>

The controller of an automotive braking system requires complex parameter calibration, and current parameter calibration relies on expensive real vehicle testing or simulation methods. Simulation takes less time, but the parameters of the braking system are very large and the search range is also very wide, which takes a lot of time to optimize. Through the AMDEAS method proposed in this study, the important control parameters can be quickly identified through the optimization system and the user can tune the major parameters. In addition to finding optimal solution, AMDEAS can also classify parameters by their importance.

Surveillance is one of the major applications in wireless sensor network areas, and it is important to detect, classify and localize the targets. In this paper, we divide the research into two sections: (1) detecting and classifying the targets and (2) localizing them. To detect and classify multiple moving targets, we use acoustic and seismic sensors, and we analyze raw data from the sensors in both time and frequency domains. In this process, we must decide which features are useful for the classification to improve the performance and make it work in real time. Thus, we exploit Weibull likelihood and short-time Fourier transform (STFT) to extract the features as a sampling method. Then, we implement a support vector machine (SVM) and a neural network to classify the type of targets based on those features. Using the suggested algorithms, the proposed classifiers provide more accurate performance than the method that analyzes the raw data from only the frequency or time domain. For localization, Gaussian Process Regression (GPR) is used to estimate the relative location that corresponds to the received signal strength indication (RSSI) data. We also demonstrate the simultaneous localization with the process of detection and classification in real time. Finally, experimental results validate the suggested algorithm.

Although indoor positioning technologies have been extensively researched, many indoor positioning methods are limited by the need for actual measurement data to be collected within a specified environment. To address issues, this paper proposes an indoor positioning simulation to implement an indoor environment and measure object position. In Addition, it presents the architecture and method of simulation through simple situations.
수요응답교통서비스가 이용자의 활동성에 미치는 효과: 지역 간 차이 실증분석

본 논문은 국토교통부의 도시형 교통모델 시범사업(공공형 택시) 자료를 이용해 교통모델 사업의 농촌지역 교통서비스 개선 효과를 회귀분석을 통해 실증적으로 분석한다. 도시형 교통모델 시범사업을 통한 외출횟수와 방문지의 수는 지역 간 상이한 변화를 보이며, 이중차분을 통한 분석의 결과는 시지역에 비해 군지역에서 시범사업을 통한 외출횟수와 방문지의 수 증가가 통계적으로 유의하게 큼을 제시한다. 특히, 군지역에서 사업시행으로 인한 외출횟수와 방문지 수의 증가가 시지역에 비해 각각 0.52회와 0.39곳만큼 큰 것으로 나타났다. 이러한 분석결과는 대부분의 선행연구들이 사업시행 지역에 한정하여 분석함으로써 경기변동에 의한 변화를 통제하지 못하는 한계를 극복한다. 교통약자의 교통편의 증진을 위한 정책은 형평성 측면에서 그 당위성이 제시되었으나 본 논문의 실증분석결과는 (예산의) 효율성 측면에서도 이러한 정책의 타당성이 확보될 수 있음을 함축한다. This paper empirically analyzes the effect of improving transportation services in rural areas of the transportation model project through regression analysis using the data of the urban transportation model pilot project (public taxi) of the Ministry of Land, Infrastructure and Transport. The number of going out and the number of places visited through the urban transport model pilot project show different changes between regions. The result of the analysis through double difference suggests that the increase in the number of going out and the number of places visited through the pilot project is statistically significantly greater in the rural area than in the urban area. In particular, it is analyzed that through the project the number of going out and the number of places visited have become greater in rural areas than those in urban areas by 0.52 times and 0.39 places, respectively. These results overcome the limit of most previous studies where the changes caused by cyclical fluctuation are not controlled for since their analyses are restricted to the areas with the project. The policy for the promotion of transportation for the transportation disadvantaged was justified in terms of equity, but the results of the empirical analysis of this paper imply that the validity of these policies can be secured in terms of efficiency (budget) as well.
Effects of Levetiracetam Monotherapy on the Cognitive Function of Epilepsy Patients

<P>Abstract</P><P><B><I>Aims:</I></B> To estimate the effect of levetiracetam (LEV) on cognitive function and mood status in newly diagnosed epilepsy patients. <B><I>Methods:</I></B> Fifty-five drug-naïve epilepsy patients (M:F ratio = 31:24; mean age = 30.9 years) were included. All patients underwent two neuropsychological (NP) tests, one before receiving LEV and then another 12.9 ± 5.0 months after starting LEV monotherapy. We evaluated general cognitive function, verbal/visual attention and memory, linguistic and visuospatial functions, frontal lobe function, and mood status. Repeated-measures regression and generalized estimating equation models were applied to assess the effects of all the confounding variables such as seizure control, average LEV dose, duration of epilepsy, inter-test interval, and subtype of epilepsy syndrome. <B><I>Results:</I></B> LEV monotherapy over 1 year revealed significant improvements in the following domains of NP tests with the correction of possible confounding variables: verbal and visual attention, psychomotor speed, mental flexibility, executive function, verbal fluency and word generation. No NP domains showed any significant decrease. <B><I>Conclusion:</I></B> Our study suggested that LEV monotherapy had no harmful effect on cognitive function in drug-naïve patients with epilepsy.</P><P>Copyright © 2013 S.
