        Different distribution patterns of microorganisms between aquaculture pond sediment and water

        Aquatic microorganisms in the sediment and water column are closely related; however, their distribution patterns between these two habitats still remain largely unknown. In this study, we compared sediment and water microeukaryotic and bacterial microorganisms in aquaculture ponds from different areas in China, and analyzed the influencing environmental factors as well as the inter-taxa relationships. We found that bacteria were significantly more abundant than fungi in both sediment and water, and the bacterial richness and diversity in sediment were higher than in water in all the sampling areas, but no significant differences were found between the two habitats for microeukaryotes. Bacterial taxa could be clearly separated through cluster analysis between the sediment and water, while eukaryotic taxa at all classification levels could not. Spirochaetea, Deltaproteobacteria, Nitrospirae, Ignavibacteriae, Firmicutes, Chloroflexi, and Lentimicrobiaceae were more abundantly distributed in sediment, while Betaproteobacteria, Alphaproteobacter, Cyanobacteria, Roseiflexaceae, Dinghuibacter, Cryomorphaceae, and Actinobacteria were more abundant in water samples. For eukaryotes, only Cryptomonadales were found to be distributed differently between the two habitats. Microorganisms in sediment were mainly correlated with enzymes related to organic matter decomposition, while water temperature, pH, dissolved oxygen, and nutrient levels all showed significant correlation with the microbial communities in pond water. Intensive interspecific relationships were also found among eukaryotes and bacteria. Together, our results indicated that eukaryotic microorganisms are distributed less differently between sediment and water in aquaculture ponds compared to bacteria. This study provides valuable data for evaluating microbial distributions in aquatic environments, which may also be of practical use in aquaculture pond management.

        중국 온라인쇼핑 체험마케팅이 태도, 구매의도, 구전의도에 미치는 영향

        Purpose: This study was conducted to study the effect of online shopping mall experience marketing in China on consumer behavior. Through this study, we tried to find out which factors of experience marketing affect consumer attitude, purchase intention, and conversational intention. In addition, by analyzing the effects of consumer attitude and purchase intention in forming conversational intention in experience marketing, this study aims to present a direction for the establishment of an online shopping mall experience marketing strategy in China. Research design, data, and methodology: For the purpose of this study, an online survey was conducted on consumers with experience in using online shopping malls in China, and 246 valid samples were finally analyzed. Descriptive statistics frequency analysis, factor analysis, and reliability analysis were performed using the SPSS 25.0 statistical package program, and regression analysis and mediating effect analysis were performed to verify the hypothesis. Results: As a result of the study, it was confirmed that the five experience marketing factors, sense, feel, recognition, behavior, and relationship, had a positive effect on consumer attitude, and that behavior, sene, and recognition experience had a greater effect. In addition, it was found that behavior experience had the greatest influence on purchase intention and recognition experience on conversational intention. Finally, it was confirmed that consumer attitude and purchase intention exert a mediating effect in the influence between recognition experience and conversational intention. Conclusions: If experience marketing factors can be combined with each other to provide various marketing solutions, it is meaningful to secure very strong strategic means for companies and marketers. To this end, companies need to reinforce experience marketing solutions first, and in particular, they need to provide a variety of behavior experiences, sense experiences, and recognition experiences.

        Political Uncertainty and Finance: A Survey

        An emerging stream of literature investigates the impact of political uncertainty on financial markets. In this survey, we review this line of literature from four perspectives, namely, asset prices, corporate policies, financial intermediaries, and economy and households, suggesting that political uncertainty generally increases market friction and as a result changes corporate behavior and adversely affects the economy. At the end of the survey, we discuss a few future directions worth being explored in view of the relationship between political uncertainty and finance.

        Dysfunction of Shh signaling activates autophagy to inhibit trophoblast motility in recurrent miscarriage

        In early pregnancy, the placenta anchors the conceptus and supports embryonic development and survival. This study aimed to investigate the underlying functions of Shh signaling in recurrent miscarriage (RM), a serious disorder of pregnancy. In the present study, Shh and Gli2 were mainly observed in cytotrophoblasts (CTBs), Ptch was mainly observed in syncytiotrophoblasts (STBs), and Smo and Gli3 were expressed in both CTBs and STBs. Shh signaling was significantly impaired in human placenta tissue from recurrent miscarriage patients compared to that of gestational age-matched normal controls. VEGF-A and CD31 protein levels were also significantly decreased in recurrent miscarriage patients. Furthermore, inhibition of Shh signaling impaired the motility of JAR cells by regulating the expression of Gli2 and Gli3. Intriguingly, inhibition of Shh signaling also triggered autophagy and autolysosome accumulation. Additionally, knockdown of BECN1 reversed Gant61-induced motility inhibition. In conclusion, our results showed that dysfunction of Shh signaling activated autophagy to inhibit trophoblast motility, which suggests the Shh pathway and autophagy as potential targets for RM therapy.

        Deep oxidative desulfurization of model fuel using ozone generated by dielectric barrier discharge plasma combined with ionic liquid extraction

        The dielectric barrier discharge (DBD) is often used to prepare ozone. In this study, a novel room temperature oxidative desulfurization method involving ozone oxidation produced in the DBD reactor combined with ionic liquid (IL) [BMIM]CH3COO ([BMIM]Ac) extraction was developed. The method was suitable for the deep removal of sulfur (S)-containing compounds from model fuel. By this desulfurization technology, 4,6-dimethyldibenzothiophene (4,6-DMDBT), dibenzothiophene (DBT), benzothiophene (BT) and thiophene (TS) were efficiently removed. Normally, the removal of TS and BT from fuel is highly difficult. However, using the proposed method of this study without any catalyst, the removal rate of TS and BT reached 99.9%. When TiO2/MCM-41 was used as a catalyst, the S-removal of DBT and 4,6-DMDBT increased to 98.6 and 95.2%, respectively. The sulfur removal activity of the four sulfur compounds decreased in the order of TS > BT >> DBT > 4,6-DMDBT. 2013 The Korean Society of Industrial and Engineering Chemistry.         <P><B>Abstract</B></P><P>The InN rods were grown by metal–organic chemical vapor deposition with a density of 1.4×10<SUP>9</SUP>cm<SUP>−2</SUP>. Optical properties of InN rods have been systematically investigated by means of temperature dependent photoluminescence (PL) and power dependent PL. Four peaks appear in the PL spectra and the origination was analyzed. The lowest energy peak P1 (0.665eV) is attributed to transitions of conduction band electrons to the photo-holes captured by deep acceptor; P2 (0.717eV) is the direct band-to-band transition peak of InN; main peak P3 (0.759eV) results from the recombination of degenerate electrons with photo-holes near the top of the valence band (Burstein–Moss effects); the high energy shoulder P4 (0.787eV) was by the co-effect of quantum confinement and the Burstein–Moss effects due to the small size distribution of InN wetting layers.</P>

        SMC3 contributes to heart development by regulating super-enhancer associated genes

        Abnormal cardiac development has been observed in individuals with Cornelia de Lange syndrome (CdLS) due to mutations in genes encoding members of the cohesin complex. However, the precise role of cohesin in heart development remains elusive. In this study, we aimed to elucidate the indispensable role of SMC3, a component of the cohesin complex, in cardiac development and its underlying mechanism. Our investigation revealed that CdLS patients with SMC3 mutations have high rates of congenital heart disease (CHD). We utilized heart-specific Smc3-knockout (SMC3-cKO) mice, which exhibit varying degrees of outflow tract (OFT) abnormalities, to further explore this relationship. Additionally, we identified 16 rare SMC3 variants with potential pathogenicity in individuals with isolated CHD. By employing single-nucleus RNA sequencing and chromosome conformation capture high-throughput genome-wide translocation sequencing, we revealed that Smc3 deletion downregulates the expression of key genes, including Ets2, in OFT cardiac muscle cells by specifically decreasing interactions between super-enhancers (SEs) and promoters. Notably, Ets2-SE-null mice also exhibit delayed OFT development in the heart. Our research revealed a novel role for SMC3 in heart development via the regulation of SE-associated genes, suggesting its potential relevance as a CHD-related gene and providing crucial insights into the molecular basis of cardiac development.

        Transcriptional profiling analysis of OsDT11-mediated ABA-dependent signal pathway for drought tolerance in rice

        Small cysteine-rich peptides (CRPs) are important in plant responses to biotic and abiotic stresses. OsDT11, encoding an 88 amino acid CRP-type protein, has been reported to enhance tolerance to drought in rice (Oryza sativa L.) without negatively afecting other agronomic traits. However, the molecular mechanisms of OsDT11-mediated drought tolerance are still unclear. Here, we performed RNA-Seq analysis to compare the transcriptome profles between wild-type (WT) and OsDT11-overexpressing (OE) rice lines under drought stress or under control (non-drought) conditions. A total of 1570 and 1421 diferentially expressed genes were identifed in the OE lines and the WT under drought treatment, respectively, compared to non-drought conditions. Gene ontology analysis indicated that the 430 up-regulated genes in common to both OE and the WT lines were induced for functions related to responses to water deprivation and to abscisic acid (ABA). More than half of these genes had higher expression in the OE than in the WT under drought stress. In the OE, but not in the WT, 294 genes were specifcally up-regulated under drought stress and were functionally enriched in starch and sucrose biosyn�thetic processes and in response to stress. This implies that OsDT11 not only triggers strongly response to drought stress, but also alters several metabolic processes to enhance drought tolerance. Gene expression profling suggests that OsDT11 confers drought tolerance by mediating an enhanced response to drought stress in an ABA-dependent signaling pathways.

      • Culture of a Whole Porcine Liver Ex Situ without Red Blood Cells

        Aims: Liver transplantation is an effective approach to end-stage liver disease. Shortage of donor liver and increased waiting time for liver transplantation necessitate the development of an organ culture system by which livers can be cultured and maintained ex situ for a prolonged period of time. The aim of this work is to test whether cell culture condition in vitro could be used to culture whole livers ex situ without the use of erythrocytes. Methods: Eight castrated male land race/farm young porcine livers were exposed to 30 min warm ischemia and 30 min cold perfusion. Livers were isolated and connected to an ex situ liver culture system using a standard culture medium RPMI 1640 supplied with 10% of fetal calf serum and sufficient dissolved oxygen under a normothermic condition for 6 hours. Metabolic biomarkers, bile and urea production, hepatic cell viability, and histology analysis of biopsies were performed and analyzed. Results: Dissociated porcine hepatic cells survived and grew in vitro under the standard RPMI 1640 culture medium. When the same RPMI 1640 medium supplemented with 10% of FCS and sufficient oxygen was used to culture livers ex situ, over 98% of liver cells were viable for at least 6 hours during ex situ whole organ culture based on the results from biochemical assays. Conclusions: Our data demonstrate that the liver culture system established in this work can be used to culture whole livers ex situ in the absence of erythrocytes.
