First Report of Xenoroussoella triseptata Isolated from Soil in Korea

A fungal strain, designated KNUF-20-NI009, was isolated from soil collected from Gunsansi, Jeollabuk-do, Korea. The isolate showed cultural features typical of the genus Xenoroussoella. Colonies cultivated on malt extract agar were olivaceous-brown to pale olivaceous-white at the margins, with undersides of dark olivaceous to olivaceous-brown and a white margin. The conidia, with a size range of 2.7-5.1×1.6-3.3 µm (x=3.6×2.6 µm, ̅ n=50), were globoid to ellipsoid in shape, hyaline when immature, becoming light brown to golden-brown when mature, and characterized by 1 or 2 guttules. Multi-locus sequence analysis based on a combined dataset of internal transcribed spacer regions (ITS), large subunit rDNA (LSU), small subunit rDNA (SSU), translation elongation factor 1-alpha (TEF1α), and RNA polymerase II largest subunit (RPB2) sequences revealed KNUF-20-NI009 to be a strain of Xenoroussoella triseptata. This is the first report of this species in Korea.
Spirosoma harenae sp. nov., a Bacterium Isolated from a Sandy Beach

<P>A Gram-stain-negative, non-motile, non-spore-forming, rod-shaped, aerobic bacterium, designated 15J8-9(T), was isolated from a sandy beach in Jeju Island, South Korea. The isolate was able to grow between 10 and 30 A degrees C, pH 5-8, and in presence of 0-1% (w/v) NaCl. Based on 16S rRNA gene phylogenetic analysis, the novel strain was closely related to members of the genus Spirosoma (96.1-90.9% similarities) and showed highest sequence similarity to Spirosoma panaciterrae DSM 21099(T) (96.1%). The G + C content of the genomic DNA of strain 15J8-9(T) was 45.1 mol%. The isolate contained menaquinone MK-7 as the predominant respiratory quinone, phosphatidylethanolamine as the major polar lipid, and summed feature 3 (C-16:1 omega 6c/C-16:1 omega 7c; 28.0%), C-16:1 omega 5c (23.4%), iso-C-15:0 (13.5%), and C-16:0 (11.5%) as the major fatty acids that supported the affiliation of strain 15J8-9(T) to the genus Spirosoma. The isolate could be differentiated clearly from recognized Spirosoma species on the basis of several phenotypic, genotypic and chemotaxonomic features. Therefore, strain 15J8-9(T) is considered to represent a novel species of the genus the genus Spirosoma, for which the name Spirosoma harenae sp. nov. is proposed. The type strain is 15J8-9(T) (= KCTC 52030(T) = JCM 31993(T)).</P>

A Gram-negative, aerobic, rod shaped, non-spore-forming bacterial strain, designated Dae08^T, was isolated from sediment of the stream near Daechung dam in South Korea, and was characterized in order to determine its taxonomic position, using a polyphasic approach. Comparative 16S rRNA gene sequence analysis showed that strain Dae08^T belongs to the family Xanthomonadaceae of the Gammaproteobacteria, and is related to Lysobacter brunescens ATCC 29482^T (97.3%). The phylogenetic distances from any other species with validly published names within the genus Lysobacter were greater than 3.7%. The G+C contents of the genomic DNA of strain Dae08^T was 69.3 mol%. The detection of a quinone system with Q-8 as the predominant compound and a fatty acid profile with iso-C_15:0, iso-C_17:1 ω9c, iso-C_17:0, iso-C_16:0, and iso-C_11:0 3-OH as the major acids supported the affiliation of strain Dae08^T to the genus Lysobacter. DNA-DNA relatedness between strain Dae08^T and its phylogenetically closest neighbour was 28%. On the basis of its phenotypic properties and phylogenetic distinctiveness, strain Dae08^T (= KCTC 12600^T) should be classified in the genus Lysobacter as the novel species, for which the name Lysobacter daecheongensis sp. nov. is proposed.
Hymenobacter knuensis sp. nov., Isolated From River Water

<P>A Gram-stain-negative, non-motile, non-spore-forming, rod-shaped, aerobic bacterial strain, designated 16F7C-2(T), was isolated from the Han River, South Korea, and was characterized taxonomically using a polyphasic approach. Comparative 16S rRNA gene sequence analysis showed that strain 16F7C-2(T) belonged to the family Cytophagaceae in the phylum Bacteroidetes and was most closely related to Hymenobacter algoricola VUG-A23a(T) (98.3%) and Hymenobacter fastidiosus VUG-A124(T) (97.7%). The G + C content of the genomic DNA of strain 16F7C-2(T) was 63.4 mol%. The detection of menaquinone MK-7 as the predominant respiratory quinone; a fatty acid profile with summed feature 3 (C-16:1 omega 7c/C-16:1 omega 6c; 19.5%), summed feature 4 (C-17:1 iso I/C-17:1 anteiso B; 15.9%), C-15:0 iso (12.6%), C-16:1 omega 5c (10.5%), and C-15:0 anteiso (10.4%) as the major components; and a polar lipid profile with phosphatidylethanolamine as the major component also supported the affiliation of strain 16F7C-2(T) to the genus Hymenobacter. The DNA-DNA relatedness between strain 16F7C-2(T) and H. algoricola JCM 17214(T) and H. fastidiosus JCM17224(T) were 45.2 +/- 5.8 and 40.3 +/- 2.9%, respectively, clearly showing that the isolate constitutes a new genospecies. Strain 16F7C-2(T) could be clearly differentiated from its closest neighbors on the basis of its phenotypic, genotypic, and chemotaxonomic features. Therefore, strain 16F7C-2(T) represents a novel species of the genus Hymenobacter, for which the name Hymenobacter knuensis sp. nov. is proposed. The type strain is 16F7C-2(T) (=KCTC 52538(T) = JCM 31814(T)).</P>
Hymenobacter daeguensis sp. nov. isolated from river water

The G+C content of the genomic DNAof strain 16F3Y-2T was 62.9 mol%. The isolate contained MK-7as the predominant respiratory quinone, and summed feature3 (C16:1 ω7c/C16:1 ω6c; 35.5%), C15:0 iso (16.9%), C16:1 ω5c(10.9%), and C15:0 anteiso (9.9%) as major fatty acids. The majorpolar lipid was phosphatidylethanolamine. Phenotypicand chemotaxonomic data supported the affiliation of strain16F3Y-2T with the genus Hymenobacter. However, strain16F3Y-2T exhibited relatively low levels of DNA-DNA relatednesswith 'H. terrae' KCTC 32554 (44.1%) and H. soli KCTC12607T (24.3%), clearly indicating that the isolate constitutesa new genospecies. Strain 16F3Y-2T could be differentiatedfrom its phylogenetic neighbors on the basis of several phenotypic,genotypic, and chemotaxonomic features. Therefore,strain 16F3Y-2T represents a novel species in the genus Hymenobacter,for which the name Hymenobacter daeguensissp. nov. is proposed. The type strain is 16F3Y-2T (=KCTC52537T =JCM 31654T).
Hymenobacter jeollabukensis sp. nov., isolated from soil

A Gram-stain-negative, non-motile, rod-shaped, aerobic bacterial strain, designated <TEX>$1-3-3-8^T$</TEX>, was isolated from soil and characterized taxonomically using a polyphasic approach. Comparative 16S rRNA gene sequence analysis showed that strain <TEX>$1-3-3-8^T$</TEX> belongs to the family Cytophagaceae of phylum Bacteroidetes and is most closely related to Hymenobacter paludis <TEX>$KBP-30^T$</TEX> (96.8% similarity), Hymenobacter ocellatus <TEX>$Myx2105^T$</TEX> (96.8%), Hymenobacter coalescens <TEX>$WW84^T$</TEX> (95.6%), and Hymenobacter deserti <TEX>$ZLB-3^T$</TEX> (95.4%). The G + C content of the genomic DNA of strain <TEX>$1-3-3-8^T$</TEX> was 63.6 mol%. The isolate contained <TEX>$C_{15:0}$</TEX> iso (28.4%), summed feature 4 (<TEX>$C_{17:1}$</TEX> anteiso B/<TEX>$C_{17:1}$</TEX> iso I; 18.9%), and <TEX>$C_{15:0}$</TEX> anteiso (17.6%) as major fatty acids, MK-7 as the predominant respiratory quinone, and sym-homospermidine as the predominant polyamine. The major polar lipids were phosphatidylethanolamine and an unidentified lipid. The phenotypic and chemotaxonomic data supported the affiliation of strain <TEX>$1-</TEX><TEX>3-3-8^T$</TEX> with the genus Hymenobacter. The DNA-DNA relatedness between strain <TEX>$1-3-3-8^T$</TEX> and H. paludis KCTC <TEX>$32237^T$</TEX> and H. ocellatus DSM <TEX>$11117^T$</TEX> were 24.5 and 27.4% respectively, clearly showing that the isolate is not related to them at the species level. Overall, the novel strain could be differentiated from its phylogenetic neighbors on the basis of several phenotypic, genotypic, and chemotaxonomic features. Therefore, strain <TEX>$1-3-3-8^T$</TEX> represents a novel species of the genus Hymenobacter, for which the name Hymenobacter jeollabukensis sp. nov. has been proposed. The type strain is <TEX>$1-3-3-8^T$</TEX> (<TEX>$=KCTC\;52741^T=JCM\;32192^T$</TEX>).
