톡소포자충 배양시 약제투여에 따른 Toxoplasma P30 유전자의 발현

To know the drugs effect on gene expression and its regulating mechanism by RT-PCR and southern blot hybridization assay based on detection of the P30 gene encoding the major surface antigen, P30 of Toxoplasma gondii and the DNA synthesis in T. gondii infected HeLa cells. The experiment was performed and the results were as follows. 1. The thymidine incooperation in DNA of HeLa cell and T. gondii infected HeLa cell reached peak level at 36 hours and suddenly decreased at 48 hours. The uracil incooperation in DNA of T. gondii reached peak level at 48 hours. 2. The decrease of P30 gene expression began from 24 hours after pyrimethamine treated and 36 hours after methotrexate treated in T. gondii. The results were some what different. Pyrimethamine was more sensitive and acted earlier to T. gondii than Methotrexate did. The decrease of P30 gene expression of T. gondii was regulated these drugs in dose dependent manner. 3. The effects of dibutyryl cyclic AMP on P30 gene expression increased peak at 24 hours and on contrary decreased at 60 hours after treatment in dose dependent manner, but at 10mM high concentration of dibutyryl cAMP, P30 gene expression was supressed. 4. Actinomycine D supressed the P30 gene expression of T. gondii in HeLa cell. As the results, we presume pyrimethamine acts more powerful and earlier to supresses P30 gene expression than methotrexate, and it is regulated at transcription level. The results of RT-PCR were in agreement with that of southern blot hybridization.

Purpose: This study researched the comparison of oral care methods between the wet gauze and the water gargling for nasal surgery patients. It executed to find out more effective method to relieve thirst. Method: We divided two groups; the wet gauze and the cold water garaling. We assessed patient's oral cavity state and sebjective oral dryness for three times; firstly before the intervention, secondly 15 minutes after 1st intervention and thirdly 15 minutes afrer 2nd intervention. Result: As the intervention, there was no difference on oral condition subhectvely, but there was a difference on thirsty reduction objectively. As the aral dryness has relieved through both methods. It showed the repeated gargling helps more to relieve oral dryness effectively than the wet gauge. At PACU, post nasal surgery patient's oral dryness was decreased with gauze more than two times, patient's oral dryness was relieved. Whereas the dryness was relieved after applying the cold water gargling for just one time. Conclusion: The water gargling is more effective intervention than the wet gauze. Especially, oral dryness was improved in the section of objetive oral assessment index.

The dream is one of unique mental functions during a sleep. Dream is a cue that shows us the unconscious mind. The Swiss psychiatrist C. A multi-residue matrix solid-phase dispersion extraction and the liquid chromatographic determination of enrofloxacin, ciprofloxacin, norfloxacin and ofloxacin in eggs is described. Blank and fluoroquinolones fortified egg samples (0.5 g) containing oxalic acid 0.05 g and sodium sulfate (anhydrose) 0.5 g were blended with octadecylsilyl (C_(18), 40 m, 18% load, endcapped, 2 g) derived from silica. After homogenization, C_(18)/egg matrix was transferred to column made of 10 ml plastic syringe and filter paper, and compressed 4.0 4.5 ml volume. A column was washed with 8 ml of hexane followed by 8 ml of ethylacetate and dried under N2 gas. Fluoroquinolones were eluated with 8 ml of methanol under gravity. The eluate contained fluoroquinolones, was free from interferences when analyzed by high performance liquid chromatography with fluorescence detection. Correlation coefficients for the fluoroquinolones standard curves (linear regression analysis, n = 5) ranged from 0.99918 (± 0.0035) to 0.99985 (± 0.0002). Aveage recoveries of fortified samples were between 89.4 and 103.5%. The method comparared favorably with the classical method in terms of the sample size, analysis time and overalls costs.
산란계에 투여된 플루오로퀴놀론계 합성항균제의 계란내 이행 잔류 연구

In the past few years the use of four fluoroquinolones such as of ofloxacin(OF), norfloxacin(NORE), ciprofloxacin(CIP), enrofloxacin(ENR) in veterinary practice has increased. The aim of this study was to determine the elimination of fluoroquinolones into eggs, by intramuscular injection of laying hens(2.5mg of ENR/kg, 5mg of CIP/kg, 5mg of NOR/kg, 5mg of OF/kg). The treatment of fluoroquinolones was carried out in 3 stages with 9 laying hens (Isabrown), with mean weight 1.8kg, which were housed in individual cages. Four fluoroquinolones were administered to 9 laying hens through intramuscular injection at 1 day, 2 days, 5 days trial respectively. Sample (Albumen, Yolk, Whole egg) were analysed by high performance liquid chromatography (HPLC) with a fluorescence programmable detector and matrix solid phase despersion (MSPD) extraction. Residues of fluoroquinolones were much higher in the yolk than in the albumen. Detectable residues persisted for 12 days, 14 days, 16 days, respectively, in albumen and yolk after the injection was discontinued. The increase of the fluoroquinolones concentration in yolk was very rapid and reached a plateau after 3 days of dosing. In albumen, the fluoroquinolones concentration increased during intramuscular injection. As soon as the injection was discontinued, residues decreased in the albumen and the yolk.

The relationship of bone mineral density and type of physical exercise was studied in 36 Postmenoposal Women(age 48-57). The exercise group consisted of Weight training([ =9), Aerobic dance(N=8), Swimming(N=9) and the control group consisted of 10 women. Bone mineral density was measured Lumber(L2-L4), Femur neck, Ward's triangle bone density. The results statstically analyzed on these data were summarized as follows: first, Lumber (L2-L4) bone density, Femoral neck bone density, Ward's triangle bone density in Weight training group was significantly higher than in Swimming group and Control group. Secondly, Lumber(L2-L4) bone density, Femoral neck bone density, Ward's triangle bone density in Aerobic dance group was significantly higher than in Swimming group and control group. Thirdly, No significant difference were round for Lumber(L2-L4) bone density, Femoral neck bone density in Weight training group and Aerobic dance group.

Prolapse of the fallopian tube into the vaginal vault after hysterectomy is a rare complication. Since most of cases may resolve before detection or remain undiagnosed, the exact incidence is still unknown. Though most common sign of tubal prolapse is tender friable tissue in vaginal cuff, with the simptoms of lower abdominal pain, dysparenunia, vaginal discharge, or postcoital spotting. The definitive diagnosis should be confirmed histologically Recently, we experienced a case of fallopian tube prolapse after abdominal hysterectomy in a 41-year-old woman and report with a brief review of the literature.

The leiomyomas of the vulva are rare benign tumors the incidence is 0.024%~0.55% in women. There are no relationship between leiomyomas of the vulva and uterus. But there are some reports that leiomyomas of the vulva and uterus simultaneously. The leiomyomas of the vulva present without symptoms and excision with biopsy is treatment of choice. Tumors of the gynecological apparatus may present without symptoms, or they may mimic other common gynecological conditions. Because of this, any vulva mass must be considered cautiously. We reported this case with a brief review of the concerned literatures.

In order to know the residual pattern of pesticides and predict to the degradation period until below MRL, we experimented diazinon, iprodinon and chlorfenapyr for shallot. They were the most detected pesticide in shallot by NAQS (National Agricultural product Quality management Service) survey. In this experiment, we sprayed those pesticides 15days before harvest and analyzed 0, 1, 2, 3, 5, 7, 10, 14 day sample to establish logical equation and to calculate DT_(50). During the cultivating period, the residue amount of diazinon was changed from 1.02 mg/kg (0 day) to 0.01 mg/kg (14 day), DT_(50) was 2.19 days, and iprodione was changed from 1.45 mg/kg (0 day) to 0.14 mg/kg (14 day), DT_(50) was 4.15 days, and chlorfenapyr was changed from 1.5 mg/kg (0 day) to 0.01 mg/kg (14 day), DT_(50) was 1.97 days. The DT_(50) of double amount in those pesticides, diazinon was 2.24 days, iprodione was 4.82 days, and chlorfenapyr was 2.24 days, respectively.
