Background: Superior colliculus is a part of midbrain, and participates in the visual reflexes, It receives afferent fibers from optic nerve, visual cortex, and spinotectal tract. After optic deprivation, the microscopic structure of the superior colliculus changed. Calcium-binding proteins (CBPs) Play an important role in the neuronal protection, differentiation and reorganization of the central nervous system, Objectives and Methods: The effects of neonatal retinal deafferentation on a CBPs, calbindm D-28k (CB), Parvalbumin (PB) and calretimn (CR), and the existence of colocalization between the CBPs were examined immunohistochemically in the rat superior colliculus. Results: On the experimental (contralateral to enucleation) side of superior colliculus, the number of CB-immunoreactive (IR) cells was reduced (77.4% compared to control), but not fibers. The number of PB-IR neurons and fibers was also reduced on the experimental side (88.5% compared to control), In the other hand, the CR-IR cells were dramatically increased (642% compared to control), but CR-IR fibers were markedly decreased on the experimental side. The colocalization between CB-CR and PV-CR was rarely observed in the superior colliculus Conclusion: These results suggest that the changes of retinotectal projection may alter the expressional pattern of CBPs in different manners; relatively stable in CB- and PV-IR neurons and plastic in CR-IR neurons.
Determination of Cobalt(III) Ion Using a Nafion-Ethylenediamine Modified Glassy Carbon Electrode

Determination of cobalt(III) ion with a perfluorinated sulfonated polymer-ethylenediamine (nafion-en) modified glassy carbon electrode is studied. It is based on the chemical reactivity of an immobilized layer, nafion-en, to yield complex [Co(en)3]3+. The reduction peak potential by differential pulse voltammetry (DPV) is observed at - 0.437±0.047 V (vs. Ag/ AgCl). The linear calibration curve is obtained in cobalt(III) ion concentration range 1.0×10-8~1.0× 10-3 M (5.893×10-12~ 5.893×10-5 g/mL).

In our previous study, we preliminarily demonstrated that Celluclast and Neutrase extracts exhibited the strongest $H_2O_2$-scavenging activities among five carbohydrases (Viscozyme, Celluclast, Termamyl, Ultraflo and AMG) and five proteases (Kojizyme, Alcalse, Flavourzyme Protamex and Neutrase) extracts. Thus, Celluclast and Neutrase extracts were selected for use in further experiments and were separated into four different molecular weight fractions (<5, 5-10, 10-30 and >30 kDa). Among them, the 5-10 kDa fraction showed the highest $H_2O_2$-scavenging activity. The 5-10 kDa fraction also strongly enhanced cell viability against $H_2O_2$-induced oxidative damage. Furthermore, the fraction reduced the proportion of apoptotic cells induced by $H_2O_2$, as demonstrated by decreased sub-G1 hypodiploid cells and decreased apoptotic body formation by flow cytometry. These results indicated that the 5-10 kDa fraction of the Celluclast and Neutrase extracts from S. coreanum exhibited strong antioxidant activity over $H_2O_2$-mediated cell damage in vitro.

The aim of the current study was to clarify the melanogenesis inhibitory effects of PA and its molecular mechanisms in murine melanoma cells (B16F10). We first predicted the 3D structure of tyrosinase and used a molecular docking algorithm to simulate binding between tyrosinase and PA. These molecular modeling studies calculated a binding energy of -527.42 kcal/mol and indicated that PA interacts with Cu400 and 401, Val283, and His263. Furthermore, PA significantly decreased α-MSH-induced intracellular tyrosinase activity and melanin content in a dose-dependent manner. PA also inhibited key melanogenic proteins such as tyrosinase, tyrosinase-related protein 1 (TRP-1), and TRP-2 in α-MSH-stimulated B16F10 cells. In addition, PA decreased MITF expression levels by inhibiting phosphorylation of cAMP response element-binding protein (CREB) and cAMP-dependent protein kinase A (PKA). These results demonstrate that PA can effectively suppress melanin synthesis in melanoma cells. Taken together, our results show that PA could serve as a potential inhibitor of melanogenesis, and hence could be explored as a possible skin-lightening agent.

BACGROUND/OBJECTIVES: In this randomized, placebo-controlled, double-blind study, we evaluated the antihypertensive effects of enzymatic hydrolysate from Styela clava flesh tissue in patients with type 2 diabetes mellitus (T2DM) and hypertension. SUBJECTS/METHODS: S. clava flesh tissue hydrolysate (SFTH) (n = 34) and placebo (n = 22) were randomly allocated to the study subjects. Each subject ingested two test capsules (500 mg) containing powdered SFTH (SFTH group) or placebo capsules (placebo group) during four weeks. RESULTS: In the SFTH group, systolic and diastolic blood pressure decreased significantly 4 weeks after ingestion by 9.9 mmHg (P < 0.01) and 7.8 mmHg (P < 0.01), respectively. In addition, the SFTH group exhibited a significant decrease in hemoglobin $A_{1c}$ with a tendency toward improvement in homeostasis model assessment of insulin resistance, triglyceride, apolipoprotein B and plasma insulin levels after 4 weeks. No adverse effects were observed in other indexes, including biochemical and hematological parameters in both groups. CONCLUSION: The results of our study suggested that SFTH exerts a regulatory, antihypertensive effect in patients with T2DM and hypertension.
