인체 대장암 및 자궁경부암에서 PCR-SSCP법을 이용한 Ki-ras 암유전자의 점돌연변이에 관한 연구

인체 대장암 20례와 자궁경부암 10례의 암조직을 대상으로 c-Ki-ras 유전자 codon 12와 13에서의 점돌연변이를 알아보고자 polymerase chain reaction(PCR)과 single-strand conformation polymorphism(SSCP) 검사법을 시행하였다. 대장암의 경우 20례중 9례(45%)에서, 자궁경부암의 경우 10례중 1례(10%)에서 양성으로 나왔다. 대장암에서는 codon 12에서 GGT가 TGT로의 치환이 4례로서 가장 많았으며, AGT, CGT로의 치환이 각 1례였고 codon 13에서는 GGC가 GAC로의 치환이 2례, TGC로의 치환이 1례인 것으로 나타났다. 자궁경부암의 경우 codon 12에서 GGT가 AGT로의 치환이 1례인 것으로 나타났다. In an attempt to clarify the role of genetic alteration in the genesis of human colorectal and cervical cancers, tissue specimens from 20 patients with colorectal cancer and 10 patients with cervical cancer were examined for the presence of point mutation in K-ras2 exon 1 by single strand conformation polymorphism analysis of PCR product. Exon I of c-Ki-ras2 was amplified by polymerase chain reaction(PCR) and comparison was made between the normal and mutated genes by nondenaturing polyacrylamide gel electrophoresis(PAGE) of PCR product and nucleotide sequence analysis using asymmetric PCR with direct sequencing. Genomic DNA from white blood cells were used as normal control and those from A427 cell line were used as mutated control. 1.PCR product from A427 cell line showed a distinct migration shift pattern compared to the normal control in PAGE and the direct sequencing indicated that nucleotide sequence of codon 12 was mutated from GGT to GAT. In addition, there was a loss of normal allele in A427 cell line. 2.PCR product from 9 cases(45%) out of a 20 colon cancer patients showed migration shifts in PAGE, and all of these 9 patients invariably demonstrated mutation of ras gene, either in codon 12(6 cases:30%) or 13(3 cases:15%). The base substitutions in codon 12 were: from GGT to AGT(1), CGT(1), or TGT(4). Base changes in codon 13 were from GGC to TGC(1) or GAC(2). 3.PCR product from 1 cases(10%) out of a 10 cervical cancer patients showed migration shrifts in PAGE, Invariably demonstrated mutation of ras gene in codon 12. The base substitutions was from GGT to AGT(1) TQE incidence of point mutation of c-Ki-ras in colon cancer was high, however, it was low in cervical cancer, which showed the relation of ruts gene mutation with colon cancer. PCR -SSCP analysis is a simple, rapid and efficient method of detection of point mutation, especially when dealing with multiple samples.

The recently recognized immunodeficiency syndrome of idiopathic CD4+ T-lymphocytopenia was defined by the Centers for Disease Control and Prevention (CDC) to include depressed numbers of circulating CD4+ T-lymphocytes (<300 cells/㎣ or <20% of total T cells) on more than one occasion, no laboratory evidence of infection with human immunodeficiency virus type 1(HIV-1) or type 2 (HIV-2), and the absence of any defined immunodeficiency or therapy associated with depressed levels of CD4+ T cells. We had experienced one case of Idiopathic CD4+ T-lymphocytopenia without HIV infection, combined with Kaposi`s sarcoma and Pneumocystis carinii pneumonia. She had visited to emergency room due to moderate dyspnea and fever for about three months. On physical examination, multiple purple colored mass lesions were detected on lower back, both upper and lower extremities and right tonsillar area. Skin biopsy on mass of right lower extremity revealed Kaposi`s sarcoma. On HRCT of the chest, patch bilateral, symmetric reticulonodular infiltration were founded. So, we performed sputum cytology repeatedly, we identified pneumocystis carinii cysts. Idiopathic CD4+ T-lymphocytopenia is rare and represents various clinical and immunologic states. It is perhaps more important to work harder to track the epidemiologic feature of this syndrome closely, to define the associated immune deficits carefully, and to search broadly for the cause. Further epidemiologic, immunologic, and virologic studies will aid in the characterization of this newly recognized condition.

This paper propose of noise and restoring image of low fingerprint quality as a pre-processing stage. Noises which were not eliminated during the procedure of fingerprint image enhancement become a reason for false minutiae extraction. According to this. the fall of the recognitive ration makes the performance of the recognitive system reduced. So ratio occurring false of minutiae can be reduced and cognitive performance can improved by restoring the image which have the type of the enhanced ridge after eliminating noise which is detected in input procedure.
판매종업원의 역할행동에 영향을 미치는 업무열의요인에 관한 연구

이 연구는 기존연구에서 업무와 판매종업원 자신 간의 지속적인 인지적 감정적 상태인 업무열의에 대한 판매종업원의 자기효능감과 업무가치감의 효과와 업무열의의 역할행동효과에 관한 연구가 희박함을 파악하였다. 따라서 판매종업원의 역할행동의 영향요인으로 업무열의개념을 도입하고 업무열의의 영향요인으로 자기효능감 및 업무가치감과 기존연구에서 식별된 감독자지원을 설정하여, 이들 간의 영향관계를 이론적으로 검토하였다. 그리고 전문 자동차판매종업원을 대상으로 실증연구하여 다음과 같은 결과를 얻었다. 역할 내 행동에 대하여 자기효능감과 업무가치감이 업무열의를 매개로 간접적으로 긍정적 영향을 미치지만, 역할 외 행동에 대하여 자기효능감, 업무가치감 그리고 감독자 지원이 모두 업무열의를 매개로 간접적으로 영향을 미치지 못하였다. 그런데 자기효능감이 역할 외 행동에 대하여 직접적으로 긍정적 영향을 주었다. 그러나 업무가치감, 감독자지원은 역할 외 행동에 대하여 직접적으로도 긍정적 영향을 주지 못하였다. 따라서 감독자는 판매종업원의 역할행동을 촉진하기 위한 업무열의수준을 유도하기 위하여 판매종업원의 업무지식수준을 향상시키고 종업원들 간의 업무교류를 향상시키는 관리를 하여 자기효능감수준울 높여야 하고, 또 전문가로서 역량의 증진, 승진 등의 개인적 목표달성, 즉 개인적 비전실현에 도움이 되는 관리를 하여 업무가치감의 수준을 높게 해야 할 것이다. In the current research, self-efficacy as personal resource variable, work values as personal vision variable and supervisory support as environmental variable were presumed to affect work engagement. And this study made efforts to explore if work engagement might have impact on the in-role behavior that is formally required of employees by the job description and impact on the extra-role behavior that is neither prescribed formally in the job description nor is directly related to work performance but is voluntary, not forcible, acts of employees which could contribute to organizational achievements. The results of this study that used car salespersons as subjects for empirical study are as follows. First, the work engagement of car salespersons had a positive impact upon both in- and extra-role behaviors. Second, it was found that work values, self-efficacy of car salespersons and supervisory support had positive influence on the work engagement. Third, work values, self-efficacy had more positive influence on the work engagement than supervisory support did. Finally, in the full model including direct paths from all of the exogenous variables to both in-role and extra-role behavior significant mediating roles of work engagement were explored in case of self-efficacy and work values only under the condition of in-role behavior. And self-efficacy also had direct impacts on both in-role behavior and extra-role behavior. This study by thus, suggests that work values and self-efficacy of the car salespersons should be managed to strengthen work engagement to lead to the in-role behaviors which can make the growth of a corporation.
