        Expression of Wnt Target Genes in Solid Pseudopapillary Tumor of the Pancreas: A Pilot Study

        OBJECTIVES:: Solid pseudopapillary tumor (SPT) of the pancreas is very rare. This study was performed to analyze the expression of Wnt signal target genes (matrix metalloproteinase-7 [MMP-7], cyclin-D1, and c-myc) and Ki-67 in resected SPTs to determine their clinicopathologic characteristics according to their expression. METHODS:: From January 1995 to December 2005, 23 patients underwent pancreatic resections for SPT of the pancreas. Among 23 formalin-fixed, paraffin-embedded tissues, 12 were evaluated as a pilot study. Immunohistochemistry was performed using various detection and antigen retrieval methods to detect MMP-7, cyclin-D1, c-myc, and Ki-67. The expression of Wnt target genes was correlated with clinicopathologic features of the patients. RESULTS:: Solid pseudopapillary tumors of the pancreas always showed cytoplasmic/nuclear accumulation of &bgr;-catenin, frequent expression of cyclin-D1, and low proliferation index. MMP-7, cyclin-D1, c-myc, and Ki-67 were not correlated with microscopic features suggesting malignant potential (P > 0.05). Tumor size was closely related to microscopic features of malignant potential and apparently has an inverse relationship with the expression of cyclin-D1 and Ki-67 (P < 0.05). CONCLUSIONS:: Low proliferative index and associated MMP-7 expression may cause an unpredictable clinical course in this tumor. Subtle changes in the intracellular environment, not pathologic (morphologic) changes, may elucidate the unpredictable clinical course of this tumor.

        Background: Frequency electrical stimulation is clinically being used in variable skin therapeutic conditions such as skin rejuvenation and hair disorder. There have been several clinical studies demonstrating the positive effect of electrical stimuli on hair regrowth. However, its exact mechanism is yet to be clarified. Objectives: The objective of this study is to investigate effects of different electrical parameter settings on hair growth by revealing the changes of dermal papilla cell in vitro and at microscopic level in animal tissue. Methods: Cultured dermal papilla cells (DPCs) and dorsal skin of rabbit were electrically stimulated with different parameter settings at alternating-current to find the optimal condition for hair growth. Cell viability and proliferation were measured by MTT. In addition, Ki67, proliferation marker, expression was measured by immunofluorescence. Hair growth-related gene expression in DPCs and the skin of rabbit were measured by RT-PCR. Results: At certain electrical settings, DPCs responded well and their proliferation was successfully induced. Wnt/β -catenin, Ki67, p-ERK and p-AKT expressions in DPCs increased at certain frequency settings. Dermal thickness and hair related genes (PDGF, VEGF, SOX9 and KGF) expressions in the skin of rabbit also significantly increased. Conclusion: These data suggest that electrical stimulations at certain electrical settings, may give more effective therapeutic outcomes for hair growth.

        A 60-year-old man was admitted to our hospital complaining of fever with chillness and anarexia for 12 weeks. Physical examination revealed supraclavicular lymphadenopathy and left inguinal lymphadenopathy. Abdominal CT scan showed the evidence of maked lymphadenopathy on retraperitoneal, external iliac, obturator, left inguinal, and celiac lymph nodes. A pathologic diagnosis of Ki-1(+) lymphoma was made by the biopsed supraclavicular lymph node because the node consisted of large cells with pleomorphic nuclei, scanty cytoplasm, large basophilic nucleoli, atypical vacuoli and large cells were positive for leukocyte common antigen, Ber-H2, marker, and B-cell marker.

        To examine proper part and medium in in-vitro culture shoot formation of in-vitro Crisium japonicum var. ussurinese KITAMURA, the test was carried out. The results are as follow: The wsa good in differentiation of the plant on the medium was good in MS+BA 10.0+Ki 10.0㎎/ℓ. In the results of formation and roots formation, proper medium was MS+Ki 0.1/ℓ. The case of MS+Ki 0.1㎎/ℓ was much no of shoots, ratio of roots, no of roots, also was good on growth.

        A Study on Joke Misinterpretation in L2 Settings: Viewed from the Benign-Violation Theory

This paper examines cases in which Korean college students might misinterpret superiority, incongruity, and sarcastic English jokes. In parallel the writer tries to bring to light possible background reasons for their misinterpretation from the perspective of the Benign-Violation Theory’ concepts of norm violation, commitment, and psychological distance. Arguably Korean college students are likely to misinterpret English humor codes influenced by their ingrained belief system of strict moral standard as well as insufficient exposure to particular genres of English jokes.

      • 노화가 인체 중간엽 줄기세포로부터 조골세포로의 증식 및 분화에 미치는 영향

        연구배경: 일반적으로 골다공증과 연관된 위험인자로는 연령, 폐경, 약물, 불충분한 칼슘섭취, 만성질환 및 운동부족 등이 있는데, 특히 노화가 진행할수록 골밀도가 감소하는 것은 잘 알려져 있다. 노화와 관련하여 진행되는 골소실은 조골세포 및 전구조골세포의 기능적 결핍에 의한 골형성의 감소가 주요한 요인으로 여겨지고 있다. 그 동안 연령이 조골모 세포의 양과 조골모 세포로부터 성숙조골세포로의 분화 및 증식에 미치는 영향에 대한 일부 보고들이 있었으나 아직 일치된 견해는 없는 형편이다. 방법: 다양한 연령의 사람으로부터 골수를 채취, 중간엽 줄기세포가 포함된 단핵세포를 분리한 후 조골세포로 분화하기 좋은 조건하에서 배양하였다. 대상군은다시 젊은군과 노령군으로 구분하여 다양한 변수를 비교 분석하였다. 일차배양에서는 CFU-F를 계수하여 골수내 중간엽 줄기세포의 수를 추산하였고, 칼슘측정을 통하여 기질의 무기화 정도를 비교하였다. 계대배양후 이차배양에서는 시기별로 알카리성 포스파타제 활성도를 측정하고 오스테오칼신 mRNA의 발현을 관찰하여 젊은군과 노령군 사이의 증식능 차이를 비교하였다. 또한 이차배양 시기별로 MTT 측정을 하여 양군간에 증식능 차이가 있는지 알아보았다. 결과: 1. 일차배양 15일째에 평균 CFU-F의 수는 젊은군에서 유의하게 많았다(젊은군 148.3±28.9, 노령군 54.3±9.1, p=0.02). CFU-F의 평균면적은 젊은군에서 넓은 경향을 보였으나 통계적으로 유의하지는 않았다. 2. 일차배양 17일 경과 후 양군간에 기질 칼슘 침착정도는 유의한 차이를 보이지 않았다(젊은군 103.6±50.6, 노령군: 114.0±56.5, p=NS). 3. 이차배양 10일째에 젊은군에서 알카리성 포스파타제 활성도가 고령군에 비해 유의하게 높았다(젊은군: 935.5±115.0 U/mg, 노령군: 578.4±115.7U/mg,p.0.05). 고령군에서는 시간 경과에 따른 변화가 미약했으며 전반적으로 알카리성 포스파타제의 활성도가 젊은군에 비해 낮았다. 4. 이차배양도중 오스테오칼신 mRNA의 발현은 배양시기별로 젊은군에 비해 고령군에서 더 낮은 경향을 관찰할 수 있었다. 5. 이차배양 10일과 15일에 젊은군에서 노령군보다세포증식이 유의하게 증가된 양상을 보였다(10앓 젊은군 0.73±0.05, 노령군 0.58±0.04, p=0.05, 15일; 젊은군 0.80±0.05, 노령군 0.70±0.03, p=0.05).결론: 이상의 연구에서 저자들은 노령군에서 젊은군보다 골수 내 중간엽줄기세포의 수가 적고, 노령군에서 유래한 전구조골세포의 성숙조골세포로의 증식 및 분화가 젊은군 보다 감소해 있는 것을 관찰할 수 있었다. Background: Osteoblasts originate from osteoprogenitor cells in bone marrow stroma, termed mesenchymal stem cells (MSCs) or bone marrow stromal cells. Each MSC forms colonies (colony forming units-fibroblasts [CFL-Fs]) when cultured ex vivo. There are some reports about the age-related changes of the number and osteogenic potential of osteoprogenitor cells, but any relationship has not been clearly established in humans. In this study, we counted MSCs using CFU-Fs count and examined the proliferative capacity and differentiation potential of osteoprogenitor cells. Finally, we analyzed how these parameters varied with donor age. Methods: Bone marrow was obtained from the iliac crest of young (n=6, 27.2±8.6 years old) and old (n= 10, 57.4k6.7 years old) healthy donors. Mononuclear cells, including MSCs, were isolated and cultured in osteogenic medium. In primary culture, we compared the colony-forming efficiency of MSCs between the two groups and determined the matrix calcification. When primary culture showed near confluence, the cells were subcultured. Alkaline phosphatase activity, osteocalcin expression by RT-PCR and proliferative potential by MTT assay were examined by the time course of secondary culture. Results: At the 15th day of primary culture, the mean number of CFU-Fs was significantly higher in the younger donors (young: 148.3±28.9, old: 54.3±9.1, p=0.02) and the mean size of CFL-Fs was also larger in the younger donors than the older donors. However, matrix calcification was not different between the two groups (young: 103.6±50.6, old: 114.0±56.5, p=NS). In secondary culture, alkaline phosphatase activities were significantly lower in the older donors. The younger donors showed peak alkaline phosphatase activity at day 10, while the older donors didn't showed a remarkable peak (young: 935.5±115.OU/mg, old: 578.4±115.7U/mg, p<0.05). Total cell number as a proliferative index increased progressively during the secondary culture and a significantly greater cell number was noted in the younger donors. Osteocalcin expression was generally upregulated in the younger donors, but this was not statistically significant. Conclusion: Our study shows that the number of osteoprogenitor cells is decreased during aging and that the proliferative capacity and differentiation potential of osteoprogenitor cells seem to be reduced during aging (J Kor SOC Endocrinol 18:296-305, 2003).

        A study of 118 autopsy cases associated with medical care in 1990 was present according to the medical department, the mode of treatment and the cause of death. The Pediatrics showed the highest rate (25.4%), followed by the Obstetrics & Gynecology (22.9%), and Internal medicine (20.2%). According to the mode of treatment, medical accidents during operation and anesthesia were 29 cases (24.6%) with highest rate in Obstetrics & Gynecology (13 cases), 38 cases (32.2%) were during medication and/or injection with highest rate in Internal Medicine (18 cases), and 51 cases (43.2%) were during general medical practice with highest rate in pediatrics (19 cases). The most prevalent cause of death was the disease of respiratory system (34.9%, 30 cases) and the diseases of cardiovascular and urogenital system accounted for 31.4% and 11.6% respectively. The main cause of death in Internal medicine was the disease of cardiovascular system (11/24 cases), the respiratory system in pediatrics (15/30 cases) and the disease of urogenital system in Obstetrics & Gynecology (9/27 cases. ) Asphyxial deaths were 7 cases and the death due to adverse drug reaction accounted for 10 cases.

        인도메타신의 투여가 치아 맹출 시 기질금속단백분해 효소의 분포에 미치는 영향

        이 연구는 치낭과 그 주위 조직 세포에서 matrix metalloproteinase (MMP)-3과 -9의 발현에 대한 인도메타신의 영향을 관찰하여 인도메타신이 치아의 맹출에 미치는 영향의 일단을 규명하기 위해 시행되었다. 생후 10-12주된 10마리의 개를 실험군 8마리와 대조군 2마리로 나누고 실험군은 인도메타신을 체중에 대하여 통상적 복용량인 인도메타신 2㎎/㎏/day을 7일간 및 14일간 투여한 군과 과량의 8㎎/㎏/day을 7일간 및 14일간 투여한 군으로 나누고 대조군은 빈 캅셀을 placebo로 투여한 후 희생하고 맹출 중인 영구치 치배를 적출하여 조직 처리하고 H-E 염색 및 MMP-3과 -9에 대한 면역염색 시행 후 광학현미경으로 검경하였다. 관찰결과 대조군에서는 파골세포, 조골세포, 치주인대 세포, 법랑모세포 및 상아모세포에서 모두 MMP-3과 -9의 발현이 뚜렷하게 관찰되었다. 대조군에 비해 인도메타신 투여군에서 파골세포, 조골세포, 치주인대 세포는 MMP-3과 -9의 발현이 억제된 소견이 관찰되었으며 인도메타신의 투여기간이 길수록 투여량이 많을수록 더 뚜렷하게 관찰되었다. 법랑모세포와 상아모세포는 대조군과 실험군의 MMP-3과 -9의 발현의 차이가 관찰되지 않았다. 이상의 결과에 의하면 prostaglandin (PG) 생합성 억제제인 인도메타신은 치낭의 파골세포, 조골세포 및 치주인대 세포에서 MMP-3과 -9의 발현을 억제하였으며 이는 인도메타신 투여로 치아 맹출이 억제될 수 있음을 시사한다. Tooth eruption requires remodeling of surrounding tissues. This study was aimed to investigate the effect of indomethacin on the dental follicle and paradental tissues during tooth eruption by observing the distribution and expression of MMP by the immunohistochemical method. Ten mongrel dogs of ten to twelve weeks old were divided into 5 groups: four experimental groups administered indomethacin 2㎎/㎏/day 8㎎/㎏/day and orally 2 times a day for 14 days and 7 days respectively, and the control group was administered a placebo. Permanent teeth before eruption and their surrounding tissues were selected and excised. H & E staining and immunohistochemical stainings of MMP-3 and -9 were performed and examined under the light microscope. Osteoclasts, osteoblasts, periodontal ligament cells, ameloblasts and odontoblasts of the control group all expressed MMP-3 and -9. In the experimental group, osteoclasts, osteoblasts and periodontal ligament cells showed reduced expression of MMP-3 and -9. Magnitude of MMP reduction in the experimental group showed a time and dose of indomethacin administration dependent manner. These results show that indomethacin inhibited MMP-3 and -9 expression in the dental follicle and surrounding tissues and suggest that when indomethacin is administered for long periods, tooth eruption could be delayed.

        대기 플라즈마 용사 공정으로 제조한 텅스텐 구리 복합재료

        A free-standing tungsten/copper composite plate, which may be used as heat sink material for electronic packaging, was produced with air plasma spray by using spherically agglomerated W/Cu composite powder. The model for particle vaporization process was studied to predict the particle behavior in the high temperature of plasma jet. At the surface of the deposit layer, copper became oxidized to cuprite (Cu₂O). The degree of oxidation decreased towards the interface between deposit and substrate. Almost no Cu₂O was detected at the inner layer after grinding off 70 ㎛ from the surface. Most pores were found in the vicinity of tungsten-rich region. The microstructures of W/ Cu deposits were also discussed.
