        Effects of Streptozotocin-Induced Type 1 Diabetes on Cell Proliferation and Neuronal Differentiation in the Dentate Gyrus : Correlation with Memory Impairment

        We examined the effects of steptozotocin (STZ)-induced type 1 diabetes on cell proliferation and neuroblasts in the subgranular zone of the hippocampal dentate gyrus (SZDG) of male Wistar rats. Change in memory function was also investigated using the passive avoidance test. In the SZDG, Ki67 (a marker for cell proliferation) positive nuclei were significantly decreased at 2 and 3 weeks and slightly decreased at 4 weeks after STZ treatment. Doublecortin (DCX, a marker for neuronal differentiation)-immunoreactive (+) neuroblasts with tertiary dendrites were significantly decreased in the STZ-treated group compared to those in the vehicle-treated group. However, DCX+ neuroblasts without tertiary dendrites were abundant at 4 weeks after STZ treatment. In addition, retention latency time in STZ-treated group was similar to that of vehicle-treated group at 2 and 3 weeks after STZ treatment. However, the retention latency time was significantly decreased at 4 weeks after STZ treatment. These results suggest that STZ significantly reduced cell proliferation and neuroblasts at 2~3 weeks after STZ treatment, but not at 4 weeks after STZ treatment although memory impairment was detected at 4 weeks after STZ treatment. The gradual reduction of DCX+ neuroblasts with tertiary dendrites may be associated with the impairment of hippocampus-related memory function.

        In this report, we present a case of the male pseudohermaphrodite in a beagle dog. The animal was subjected for examination due to enlargement of clitoris that was shown as reddish finger-like structure protruding from the vulva. Visual examination of the internal genitalia revealed that the animal possessed both testes and uterus. Based on the findings of physical examination and exploratory laparotomy, it was tentatively diagnosed as male pseudohermaphrodite. Internal genitalia and enlarged clitoris were removed for the prevention of pyometra and Sertoli cell tumor.

      • 비강 및 비인두에 발생한 도립유두종과 편평상피암종의 p53단백 및 세포증식능에 관한 연구

        The inverted papilloma and squamous cell carcinoma are common neoplasia in the sinonasal cavity and nasopharynx, but the incidence of these tumors are very low and the study on the oncogenesis or biological activity of the tumor cells are not well known. This study was designed to evaluate the oncogenic roles of the p53 gene and the proliferative activity of the tumor cells in the inflammatory polyp, inverted papilloma and squamous cell carcinoma. The experiment was carried by the immunohistochemical stains about the p53 protein, PCNA and Ki-67, histochemical stain about the AgNORs. and flow cytometric analysis about the DNA ploidy using the formalin fixed paraffin embedded tissues. The frequency of the expression of p53 protein was 0%(0/16 cases) in the inflammatory polyps, 18.8% (3/16 cases) in the inverted papillomas, and 87.5%(14/16 cases) in the squamous cell carcinomas. The labelling index(%) of the PCNA and Ki-67 was 5.3% and 3.3% in the inflammatoy polyps, 29.6% and 25.2% in the inverted papillomas, and 51.9% and 36.8% in the squamous cell carcinoma. The expression of the PCNA and Ki-67 was distributed in the periphery of the tumor islands of the inverted papilloma and was distributed in the both of center and periphery of the tumor islands of the squamous cell carcinoma. The number of nuclear AgNORs was increased in the order of inflammatory polyps (0.96), inverted papillomas(1.34) and squamous cell carcinoma(2.09). The frequency of the DNA aneuploidy was 0%(0/16 cases) in the inflammatory polyps, 18.8%(3/16 cases) in the inverted papillomas. and 12.5%(2/16 cases) in the squamous cell carcinomas. Above results indicates that the changes of the p53 gene and proliferative activity of the tumor cells are involved on the oncogenesis and the biological activity of the inverted papilloma or squamous cell carcinoma in the nasopharynx and sinonasa cavity.

      • Effects of Cu,Zn-superoxide dismutase on cell proliferation and neuroblast differentiation in the mouse dentate gyrus.

        <P>Oxidative stress is one of the most important factors in reducing adult hippocampal neurogenesis in the adult brain. In this study, we observed the effects of Cu,Zn-superoxide dismutase (SOD1) on lipid peroxidation, cell proliferation, and neuroblast differentiation in the mouse dentate gyrus using malondialdehyde (MDA), Ki67, and doublecortin (DCX), respectively. We constructed an expression vector, PEP-1, fused PEP-1 with SOD1, and generated PEP-1-SOD1 fusion protein. We administered PEP-1 and 100 or 500 관g PEP-1-SOD1 intraperitoneally once a day for 3 weeks and sacrificed at 30 min after the last administrations. PEP-1 administration did not change the MDA levels compared to those in the vehicle-treated group, while PEP-1-SOD1 treatment significantly reduced MDA levels compared to the vehicle-treated group. In the PEP-1-treated group, the number of Ki67-positive nuclei was similar to that in the vehicle-treated group. In the 100 관g PEP-1-SOD1-treated group, the number of Ki67-positive nuclei was slightly decreased; however, in the 500 관g PEP-1-SOD1-treated group, Ki67-positive nuclei were decreased to 78.5% of the vehicle-treated group. The number of DCX-positive neuroblasts in the PEP-1-treated group was similar to that in the vehicle-treated group. However, the arborization of DCX-positive neuroblasts was significantly decreased in both the 100 and 500 관g PEP-1-SOD1-treated groups compared to that in the vehicle-treated group. The number of DCX-positive neuroblasts with tertiary dendrites was markedly decreased in the 500 관g PEP-1-SOD1-treated group. These results suggest that a SOD1 supplement to healthy mice may not be necessary to modulate cell proliferation and neuroblast differentiation in the dentate gyrus.</P>

      • 10주간 댄스스포츠 프로그램 수행이 비만여성의 신체구성 및 체력 변화에 미치는 영향

        This study investigated the changes of body composition and physical fitness between before and after dancesports program of 10 weeks, for the suggestion of the theoretical basis for effective prevention and adminstration of obesity with dancesports. Subjects were consisted of 20-aged female students(n=12), and were divided to obese(n=6) and normal weight group(n=6). Body weight, percentage of body fat, BMI, skinfold thickness(8 sites), and body circumferences were measured for analysis of body composition, and physical fitness were measured by the back muscular strength, sit-up, side step test, and sit and reach. Obese group was decreased body weight and %fat after 10 weeks dance sport program but normal weight group was no change of body weight. Obese group showed a significant decrease of skinfold thickness in subecupular, abdomen, suprailiac, thigh, and axillary after 10 weeks dancesports program The decrease of body fat and skinfold thickness after 10 weeks dancesports program showed different tendency depend on different level of obesity and body part. Therefore hypothesis on change of body composition after dancesports program could be accepted in these results. Physical fitness of muscular strength and flexibility showed the improvement after 10 weeks dancesports program. ln these results, dancesports program could be suggested the effective exercise program for obesity treatment.

        In this study, Mechanical property, thermal property, watervapor, and oxygen permeability of biodegradable Soy protein/Pectin-montmorillonite nanocomposite, prepared with Na^(+)-MMT, have been studied. X-ray diffraction and TEM experiments an polymer/silicate composite samples demonstrated that the intercalation of polymer chains leads to increase in the spacing between silicate layers. In addition, it were found that the intercalated Soy protein/Pectin/montmorillonite nanocomposite were prepared. It was found that the addition of only a small of clay was enough to improve the thermal stability and mechanical property of Soy protein/Pectin/montmorillonite nanocomposite. watervapor of Soy protein/Pectin/montmorillonite nanocomposite was generally lower than that of Soy protein/Pectin. oxygen permeability of Soy protein/Pectin/montmorillonite nanocomposite was generally lower than that of Soy protein/Pectin.

        Streptococcus의 유당분해에 대한 자일리톨의 효과

        자일리톨은 탄소 5개가 있는 탄수화물로 치아우식증을 억제할 목적으로 사용되는 자당 대체물이다. 본 논문은 구강 중요 세균인 Streptococcus mutans, Streptococcus oralis, Streptococcus salivarius에서 유당분해에 대한 자일리톨의 작용을 관찰하고자 하였다. 세균에 대한 유당과 자일리톨의 병합효과를 보기 위하여 분광광도계를 이용한 흡광도를 측정하였고 생균수 검사를 실시하였다. Streptococcus mutans 또는 β-galactosidase에 의한 유당분해에 대한 자일리톨 효과를 보기 위하여 thin layer chromatography와 lactose-PTS activity test를 실시하여 다음과 같은 결과를 얻었다. 1. 유당과 자일리톨을 병합 첨가한 배지에 Streptococcus mutans를 배양하면 배양액 흡광도가 배양 8시간에 증가하지 않다가 배양 24시간에 증가하였다. 배양 8시간에서의 생균수도 유당 단독보다 유당과 자일리톨 병합 첨가 시 적게 나타났다. 2. 유당과 자일리톨을 병합 첨가한 배지에 Streptococcus oralis, Streptococcus salivarius를 배양하면 배양액 흡광도가 배양 8시간에 증가하지 않다가 배양 24시간에 증가하였다. 3. 배지에 유당 단독으로 첨가할 때보다도 유당과 자일리톨을 병합 첨가한 배지에 Streptococcus mutans를 8시간 배양하면 thin layer chromatography상 남아있는 유당이 많았으나, 배양 24시간에는 모든 유당이 분해되었다. 4. 배지에 유당 단독으로 첨가할 때보다도 유당과 자일리톨을 병합 첨가한 배지에서 Streptococcus mutans의 lactose-PTS 활성도가 낮았다. 5. 배지에 유당 단독으로 첨가할 때보다도 유당과 자일리톨을 병합 첨가한 배지에 β-galactosidase 사용시 thin layer chromatography상 유당이 많았다. 이상의 결과는 자일리톨이 Streptococcus의 유당 이용을 억제함을 시사하였다. Xylitol is a 5-carbons carbohydrate, which can be replaced with sucrose for preventing dental caries. To study the effect of xylitol on the fermentation of lactose in bacteria, the important oral bacteria such as Streptococcus (S.) mutans, S. oralis and S. salivarius were studied. The optical density using spectophotometer and the cell concentration were assessed to evaluate the combined effect of lactose and xylitol against the bacteria. Thin layer chromatography and lactose-PTS activity test were performed to evaluate the effect of xylitol on the fermentation of lactose in S. mutans and by β-galactosidase with the following results. 1. The optical density of Streptococcus mutans culture was not increased for 8 hours-incubation in the media added with lactose and xylitol, but was increased at 24 hours-incubation. The number of viable cells at 8 hours-incubation was smaller in the media containing lactose and xylitol in comparison with lactose only. 2. The optical densities of Streptococcus oralis culture and Streptococcus salivarius culture were not increased for 8 hours-incubation in the media added with lactose and xylitol, but were increased at 24 hours-incubation. 3. When Streptococcus mutans was incubated for 8 hours in the media added with lactose and xylitol. the amount of remained lactose was larger compared with the media added with lactose only. But all lactose was fermented in both media after 24 hours-incubation. 4. When Streptococcus mutans was incubated in the media added with lactose and xylitol, the activity of lactose-PTS was higher compared with the media added with lactose only. 5. When β-galactosidase was incubated in the media added with lactose and xylitol, the amount of remained lactose was larger compared with the media added with lactose only. These results indicated that xylitol inhibited the fermentation of lactose by Streptococcus.

      • 그루지아 민가의 전통 발효유에서 분리한 유산균의 이화학적 특성

        그루지아의 전통 발효유로부터 유산 생성과 항균성이 우수한 3주의 균주를 최종 분리, 선발하고 각각 Lactobacillus bulgaricus (L bulga G82), Lactobacillus acidophilus (L. acid G86), 그리고, Streptococcus thermophilus (S. thermo G90)로 동정하였다. 이들 유산균을 starter로 요구르트를 제조하여 이화학적 특성을 조사한 결과, pH는 4.0∼4.5의 범위를 보였으며, pH 4.0으로 L. acid G86 균주가 가장 낮게 나타났다. 또한 0.1N HCl에 대한 완충능은 4.50∼5.12ml, 0.1N NaOHdp 대한 완충능은 3.74∼4.98ml의 범위를 보였고, 3 균주 중 L acid G86 균주가 산-염기에 대하여 가장 높은 완충능을 나타내었다. 산도는 0.95∼1.16%, 점도는 1,984∼2,232 cps 그리고 생균수는 1.4x109∼3.0x109으로 조사 되었다. in vitro에서의 콜레스테롤 저하 정도는 L. acid G86 균주가 약 35%로 가장 높은 저하 정도를 보였고, L. bulga G82와 S. thermo G90 균주는 각각 약 32%, 24%이었다. Three strains of inhibitory lactic acid bacteria against pathogenic microorganism were isolated from traditional fermented milk of Georgia, and than, identified to be Lactobacillus bulgaricus (L bulga G82), Lactobacillus acidophilus (L acid G86) and Streptococcus thermophilus (S. thermo G90), respectively. These strains were used to starter for yogurts. So, we made yogurts, and evaluated their phyco-chemical properties. The range of pH value was 4.0∼4.5, and L. acid G86 strain was the lower pH value for 4,0. The buffer capacity against 0.1N HCI and 0.1N NaOH were 4,50~5.12ml and 3.74~4.98ml. The acidity, viscosity and viable cell counts of yogurt were 0.95∼1/16%, 1.984∼2,232 cps and 1.4x109∼3.0x109, respectively. Among the three strains, L. acid G86 strain was have the best ability of lowering cholesterol level in vitro, the value were about 35%. The L. bulga G82 and S. thermo G90 were around 32% and 24%, respectively.

        Utilizing the electrospinning technique nanofiber-based biodegradable scaffolds made of PLGA was suggested. Under various conditions, their diameters and porosity as well as mechanical strength were evaluated. In addition to those, cell(chondrocyte) proliferation and formation of extracelluar matrices were also investigated along with the conventional membrane type PLGA scaffolds for the potential use in tissue engineering. As conclusions, this type of scaffold showed a potential of application to tissue engineering in view of mechanical stability as well as cellular responses.

        The cable-stayed bridges are composed of cables, towers and girders. when the cable-stayed bridges are loaded, the whole structure display very complex behavior due to excessive cable deflection. Eventhough the material in the members of a cable-stayed bridge structure behaviors in a linear elastic manner. the overall load-displacement relationship for the structure will be nonlinear. The nonlinear behavior is a result of: due to the sag caused by their own dead weight and the nonlinear axial and bending force-deformation relationships for the bend members. In the construction stage analysis, some technical approaches and structure safety are needed to stimulate numerically the complicated changes of structural system during construction. In this study static and construction stage analysis is performed and compared. Static and construction stage analysis on unsymmetric Cable-stayed bridge is performed and linear and nonlinear analysis is performed to evaluate the structure characteristics. After Static analysis of final construction stage was performed, construction stage analysis was performed.
