        유방암 세포(EMT6) 이식 마우스에서 녹차폴리페놀 음용이 시스플라틴의 암 조직 성장 억제에 미치는 영향

        본 연구는 녹차폴리페놀이 시스플라틴의 항암작용과 신장독성에 미치는 영향을 유방암 세포(EMT6)와 암세포 이식 마우스를 이용하여 in vitro와 in vivo 실험으로 관찰하였다. 배양한 EMT6 세포에서 녹차폴리페놀은 시스플라틴에 의한 세포 독성을 증가시켰다. 마우스에 EMT6 세포를 주사하여 유발된 종양의 크기가 시스플라틴군(CP)보다 시스플라틴+녹차폴리페놀군(CP+GTP)에서 유의하게 작았고, 종양조직 p53와 caspase-3 활성화가 시스플라틴군(CP)보다 시스플라틴+녹차폴리페놀군(CP+GTP)에서 유의하게 높았으며, 신장 GGT와 AP 활성은 시스플라틴군(CP)보다 시스플라틴+녹차폴리페놀군(CP+GTP)에서 유의하게 높았고, 신장 조직학적 소견에서 신세뇨관 확장과 괴사가 시스플라틴군(CP)보다 시스플라틴+녹차폴리페놀군(CP+GTP)에서 유의하게 낮았다. 이상의 결과 녹차폴리페놀은 EMT6 유방암 세포를 이용한 in vitro 및 in vivo 실험에서 시스플라틴의 항암작용을 증강시키면서 신장에 대한 독성 부작용은 감소시키는 효과가 있는 것으로 추측된다. 녹차폴리페놀의 시스플라틴 항암작용 증강과 신장 독성 억제 및 감소 효과는 시스플라틴에 의한 암 치료 시 화학요법제의 보조제로서 이용가치가 있는 것으로 생각되며, 항암화학요법제에 대한 보조제로의 개발을 위해서는 대규모 동물실험을 통한 효과 입증 및 부작용에 대한 실험과 임상연구가 뒷받침되어야 할 것으로 사료된다. The aim of this study is to evaluate the effects of green tea polyphenol (GTP) on anticancer treatment with cisplatin (CP), using both an in vitro cell culture model and an in vivo mouse model of established breast tumor. Mouse breast cancer cells (EMT6) were treated with or without GTP and CP followed by determination of the cell viability using an MTT assay. The relative cell viability of CP treated EMT6 cells was 96% at a 20 μg/mL concentration of cisplatin; however, in combination with GTP (50 μg/mL), the cell viability decreased to 20% at the same concentration of CP (20 μg/mL). For the in vivo study, EMT6 cells were inoculated into Balb/c mice for the establishment of a tumor-bearing mice model. The tumor-bearing mice were treated with CP (5 mg/kg. i.p.) with or without dietary GTP (0.2% drinking water). Tumor growth was monitored by a measurement of tumor size using a digital caliper, and nephrotoxicity was determined by enzymatic and histological examinations. The levels of p53 and caspase-3 in tumor tissues were examined by a Western blot. In tumor-bearing mice treated with GTP plus CP, the increment of tumor volume showed a significant reduction, compared with CP or GTP alone. The levels of p53 and cleaved caspase-3 (caspase-3/p17) in tumor tissues of tumor-bearing mice were increased by CP and GTP compared to CP alone. In CP treated tumor-bearing mice, γ-glutamyltranspeptidase (GGT) and alkaline phosphatase (AP) activities were decreased, and marked tubular necrosis and dilatation were observed in the kidney. CP-induced enzymatic and histopathological changes in the kidney of tumor-bearing mice were reduced by combinations of GTP with CP. The results of these experiments demonstrated that dietary GTP has a potentiating effect on CP anti-tumor activity and a protective effect against CP-induced renal dysfunction. Therefore, GTP may be used as a modulator in anticancer treatment with CP.

        Response of low-temperature steel beams subjected to single and repeated lateral impacts

        This paper presents the experimental and numerical investigation results of the response of lowtemperature steel (LT-FH32 grade steel) beams under repeated impacts at room temperature and a single impact at a sub-zero temperature. After conducting tensile tests at room and sub-zero, repeated impact tests were conducted on two clamped single-beam models at room temperature, and singleimpact tests of two other clamped single-beam models were conducted at 50 C. The single and repeated impact tests were conducted by releasing a knife-edge striker using a drop testing machine. The permanent deflection of the model measured after each impact gradually increased with increasing number of impacts. Under the reduced temperature, the permanent deflection of the models slightly decreased. The numerical analyses were also performed to predict the damage response of the tested single-beam models. A comparison of the numerical prediction with those of experiments showed quite reasonable agreement.

      • Direct strength evaluation of the structural strength of a 500 cbm LNG bunkering ship

        The present paper describes a general procedure of the structural safety assessment for the independent type C tank of LNG bunkering ship. This strength assessment procedure consists of two main scheme, global Finite Element Analysis (FEA) model primarily for hull structure assessment and detailed LNG Tank structures FEA model including the cylindrical tank itself and saddle-support structures. Two kinds of mechanism are used, fixed and slides constraints in fore and rear of the saddlesupport structures that result in a variation of the reaction forces. Finite Element (FE) analyses have been performed and verified by the strength acceptance criteria to evaluate the safety adequacy of yielding and buckling of the hull and supporting structures. The detail of FE model for an LNG type C tank and its saddle supports was made, which includes the structural members such as cylindrical tank shell, ring stiffeners, swash bulkhead, and saddle supports. Subsequently, the FE buckling analysis of the Type C tank has been performed under external pressure following International Gas Containment (IGC) code requirements. Meanwhile, the assessment is also performed for yielding and buckling strength evaluation of the cylindrical LNG tank according to the PD 5500 unfired fusion welded pressure vessels code. Finally, a complete procedure for assessing the structural strength of 500 CBM LNG cargo tank, saddle support and hull structures have been provided.

      • 加熱 不活化한 B型 肝炎 Vaccine의 小亞에서의 臨床效果에 관한 硏究

        A different type of hepatitis B vaccine was prepared by purification of HBsAg from a pool of plasma containing surface antigen. After purification, the HBsAg was inactivated by double-heat treatment, 2 minutes 40 secs at 102℃ and 10 hours at 65℃. The heat-inactivated HBsAg was absorbed to aluminum phosphate. One dose of vaccine containing 3ug HBsAg absorbed to 0.6mg AIPO₄. The final product met the guideline recommended by WHO and KNIH(Korean National Institute of Health). After establishing the safety and immunogenicity of vaccine in chimpanzee and human volunteers, fifty children were administered at the intervals of one month with three intramuscular injections of a dose of vaccine containing 1.5ug or 3ug of HBsAg. Thirty five out of thirty seven children injected by a dose of vaccine containing 1.5ug of HBsAg and twelve out of thirteen children injected by a dose of vaccine containing 3ug of HBsAg have formed anti-HBs in two months after last vaccination.

        아동복지시설의 고객지향성에 관한 연구

        본 연구는 우리나라 아동복지시설의 고객지향성 수준을 파악하고, 고객지향성에 영향을 미치는 변인들을 시설의 인적, 물적 및 조직구조적 요인을 중심으로 파악하여 아동복지시설이 보다 고객의 욕구에 부응하는 방향으로 서비스의 질을 개선하기 위한 기초자료를 제공하고, 이에 따른 실천적 함의를 모색하는데 그 목적이 있다. 이를 위해 ‘연세대학교 사회통합배제연구단’에서 전국의 사회복지기관의 실무책임자들을 대상으로 2006년 5월 17일부터 9월 18일까지 수집한 데이터 중 아동복지시설(아동양육시설 140개소, 종합상담센터 56개소, 아동보호전문기관 23개소) 219개소를 분석대상으로 하였다. 연구결과는 다음과 같다. 먼저 아동복지시설의 인적, 물적, 조직구조적 요소별 고객지향성 수준은 평균 이상의 수준을 보였으며, 고객지향성의 수준차이를 인적, 물적, 조직구조적 변인의 하위 차원에 따라 분석해본 결과 인적 요소에서는 시설 내 정규직 종사자수와 정원대비 자격증 소지자 비율이, 물적 요소 중에서는 기관의 총 수입과 인건비 지출 비율이, 조직구조적 요소 중에서는 설립년도와 기관 유형에 따라 고객지향성이 유의미한 차이를 보였다. 그리고 고객 지향성에 영향을 미치는 요인을 인적, 물적, 조직구조적 요소차원으로 구분하여 분석한 결과 고급전문가 비율, 정부보조금 비율, 설립년도가 유의미한 영향을 미치는 변수로 나타났다. 이러한 연구결과를 바탕으로 아동복지시설의 고객지향성을 증진시킬 수 있는 사회복지적 함의를 논하였다. This study aims to identify predictive factors for customer orientation in child welfare agencies using human, financial and organizational resources as predictive factors and to explore implications for improvement of child welfare services in terms of customer orientation. For the purpose of the study, this study used a secondary data analysis method based on Yonsei Social Inclusion-Exclusion Study. Data were collected from May 17, 2006 to September 18, 2006 and participants were administrators in 219 child welfare agencies (Child orphanages, 140; Adolescents counseling centers, 56; Child Protection agencies, 23). Results are as following: First, the level of customer orientation in child welfare agencies was just above the average range. When differences in the level of customer orientation were examined according to human, financial and organizational resource characteristics, percentage rates of full-time workers and professional licences, overall income level and the percentage rate of workers salary, year of establishment and the type of agency showed statistically significant differences. Further, the percentage rates of professionals, subsidy level of government, year of agency establishment were identified as predictive factors for customer orientation in child welfare agencies. Based on the results, implication for child welfare services were discussed.

        가축분 퇴비와 어분 액비 시용이 유기농 오이 생육 및 토양환경에 미치는 영향

        유기농 오이의 안정생산을 위한 양분관리 방법으로 가축분 퇴비와 어분액비 시용이 오이 생육과 토양환 경에 미치는 영향을 검토하기 위하여 본 연구를 수행하였다. 시험은 가축분 퇴비 100% (LC100), 가축분 퇴비 50% + 어분액비 50% (LC50 + LF50), 가축분 퇴비 50% (LC50), 화학비료 (NPK), 그리고 무비 (NF) 등 5처리로 하여 처리구별 토양 화학성, 토양미생물 군집 변화, 그리고 오이 생육 및 수량을 조사하였다. 그 결과, 토양 화학성의 경우에는LC50 + LF50 처리는 LC100 처리와는 EC 함량을 제외하고는 통계적인 유의차가 없었으며 화학비료 처리와는 pH를 제외하고 통계적인 유의차를 나타냈다. 토양 미생물 군집의 경우에는 미생물 밀도는 처리에 따른 통계적 유의차가 없었으며 Microbial biomass C 함량은 NF, NPK 처리에 비해 가축분 퇴비, 액비와 같은 유기물 시용구에서 높게 나타났다. 각 처리에 의한 오이 생육을 비교한 결과, 오이 초장과 생체중은 LC100, LC50 + LF50, 그리고 NPK 처리간에는 통계적인 유의차가 없었으나 NF와 LC50 처리와는 유의적인 차이를 나타냈다. 오이 수량은 NPK 처리가 7,397 kg/10a으로 가장 많았지만 LC50 + LF50, 그리고 LC100 처리와는 통계적인 유의차가 없었다. 이러한 결과로부터 유기농 오이 재배에서 가축분 퇴비와 어분액비 시용으로 양분관리가 가능할 것으로 판단되었다. This study was carried out to investigate the effects of livestock manure compost and fish-meal liquid fertilizer on the growth of cucumber and the soil properties for the stable production of organic cucumber. Cucumber was transplanted in greenhouse on the 6 th of April in 2017, and this experiment contained five treatments: livestock manure compost 100% (LC 100%), livestock manure compost 50% + fish-meal liquid fertilizer 50% (LC50 + LF50), livestock manure compost 50% (LC50), chemical fertilizer (NPK), and no fertilizer (NF). As a result, it was shown that soil chemical properties of LC50 + LF50 plot is not different from that of LC100 plot except for the EC content, but soil chemical properties of LC50 + LF50 plot is statistically significantly different from that of NPK plot except for pH. As a result of evaluating the functional diversity of soil microbial communities using Biolog system, the substrate richness (S) and the diversity index (H) were the highest in LC50 + LF50 plot. As a result of comparing the cucumber growth and yield, it was found that there was no statistically significant difference between the plant height and the fresh weight of LC100, LC50 + LF50, and NPK plot, but the plant height and the fresh weight of LC100, LC50 + LF50, and NPK plot were different from that of LC50 and NF plot. The yield of cucumber was the highest in NPK plot r(7,397 kg/10a), but there was no statistically significant difference in the yield of cucumber between NPK plot and LC100, LC50 + LF50 plot. The above-described results suggested that the livestock manure compost and fish meal liquid fertilizer can be used for organic cucumber production under greenhouse condition.

        Effects of Long-Term Fertilization on Microbial Diversity in Upland Soils Estimated by Biolog Ecoplate and DGGE

        Organic amendment practices can influence diversity and activities of soil microorganisms. There is a need to investigate this impact compared with other types of materials. This study was carried out to evaluate the long term effects of chemical and organic fertilizer on soil microbial community in upland field. During the last 11 years green manure, rice straw compost, rapeseed cake, pig mature compost, NPK, and NPK + pig mature compost were treated in upland soil. Organic fertilizer treatment found with high bacterial colony forming units (CFUs) as compared to chemical and without fertilizer treatment. There was no significant difference in the actinomycetes and fungal population. The average well color development (AWCD) value was the highest in green manure and, the lowest in without fertilizer treatment. Analyses based on the denaturing gradient gel electrophoresis (DGGE) profile showed that rice straw compost and pig mature compost had a similar banding pattern while rapeseed cake, NPK, NPK + pig mature compost and without fertilizer treatment were clustered in another cluster and clearly distinguished from green manure treatment. Bacterial diversity can be highly increased by the application of organic fertilizer while chemical fertilizer had less impact. It can be concluded that green manure had a beneficial impact on soil microbial flora, while, the use of chemical fertilizer could affect the soil bacterial communities adversely.

        유기농 오이재배를 위한 어분액비 공급이 토양특성 및 오이 수량에 미치는 영향

        본 연구는 유기농 오이 재배를 위하여 어분액비를 웃거름으로 공급할 경우 오이 생육과 토양환경에 미치는 영향을 검토하여 양분공급원으로 활용하고자 수행하였다. 시험구 처리는 무시비, 액비 무처리, 액비처리농도에 따라 100, 50, 25 mg/L 처리구, 화학비료 처리 등 6처리를 두었다. 주요 조사항목은 어분액비처리에 의한 오이 생육, 수량, 토양 화학성, 그리고 토양미생물 군집 변화를 조사하였다. 어분액비 처리에 의한 토양화학성 변화를 살펴본 결과, 어분액비 100 mg/L 처리는 다른 처리구와 비교하여 pH와 치환성 양이온 칼륨, 마그네슘 함량이 뚜렷하게 낮아지는 결과를 나타냈다. 또한 Biolog을 이용하여 토양미생물 군집의 기능적 다양성을 분석한 결과, 토양미생물 군집의 기질 이용도는 액비처리, 화학비료구, 무비구가 각각 1.57∼1.63, 1.5, 1.3으로 액비처리와 화학비료구가 무비구에 비해 높았지만 액비 처리 수준별 차이는 나타나지 않았다. 액비처리에 의한 오이 생육을 비교한 결과, 액비처리 농도별 오이 초장과 과실의 생체중 및 건물중은 통계적 유의차는 없었으며 잎과 줄기의 생체중과 건물중은 어분액비 100 mg/L 처리 시 통계적 유의차를 나타냈다. 오이 수량은 액비처리 수준별 차이는 없었으며 어분액비 처리와 화학비료 처리간에 비교하더라도 차이를 나타내지 않았다. 따라서 어분액비와 화학비료 처리에 대한 오이의 생육과 수량을 비교한 결과, 수량과 생육에 큰 차이를 나타내지 않아 어분액비는 유기농업에서 웃거름으로 활용 가능 할 것으로 판단되었다. This study was carried out to evaluate the application effects of fish meal liquid fertilizer on soil characteristics and growth of cucumber for organic cultivation. Cucumber in greenhouse was transplanted on March 31th in 2016, and the experimental treatments involve six treatments: No fertilizer, 0, 25, 50, and 100 mg/L N application by fish meal liquid fertilizer and chemical fertilizer. In the results of soil chemical property, application of 100 mg/L of fish meal liquid fertilizer showed a significant differences in pH, K, and Mg contents. The soil microbial community varied in relation to the fish meal liquid fertilizer treatments. Microbial biomass was lower in the chemical fertilizer than in the liquid fertilizer treatment. Result of principal component analysis obtained from Ecoplate showed that fish meal liquid fertilizer treatments, no liquid fertilizer, chemical fertilizer, and no fertilizer were divided into distinct groups, with the no fertilizer treatment located furthest from the other treatments. There were no significant differences in plant height of cucumber between the fish meal liquid fertilizer treatments and chemical fertilizer treatments. Also, the cucumber yield did not vary significantly between the concentrations of liquid fertilizers, and there were also no significant differences in the yield among the fish meal liquid and chemical fertilizer treatments. In conclusion, it is suggested that the application of fish meal liquid fertilizer can be used as a additional fertilizer for cucumber production with organic culture in greenhouse.
