      • Glucagon 및 Cycloheximide 投與에 의한 흰쥐 肝細胞 lysosome의 變化에 관한 超微形態學的 硏究

        저자는 체중 30∼35gm 정도의 ICR계 생쥐에 체중 100gm당 100㎍의 glucagon을 그리고 체중 100gm당 0.15mg의 cycloheximide를 각각 단독으로 또 이들을 같이 복강내에 병합 투여하여, 이때 간세포에 일어나는 형태학적 변화와 아울러 lysosome막의 유래의 일단을 알아보기 위한 실험성적을 요약하면 다음과 같다. 광학현미경적으로는 중심정맥 주위의 간세포이 종창과 한국성 지발변화가 나타났었는데, 그 정도는 glucagon 단독투여군과 cycloheximide와 glucagon을 병합투여한 군에서는 다같이 중등도로 일어났었다. 전자현미경적으로는 glucagon 단독투여 군에서는 간세포에 있어서의 lysosome의 현저한 증가, 특히 autophagic vacuole의 증가, 세포핵의 비대 및 핵소체수의 증가, 그리고 kupffer세포의 탐식 작용의 증가등이고 cycloheximide 단독투여군에서는 경한 lysosome의 증가, RER의 확장, 단열, ribosome의 탈락, free ribosome의 증가, Golgi complex의 위축, SER의 감소, mitochondria의 종창 및 공포화와 lipid droplet의 증가 등이다. 그리고 glucagon과 cycloheximide 병합투여군에서는 상기한 두가지 단독투여군의 소견과 여기에 더하여 peroxisome의 증가가 일어났으며, 이들 변화중 특히 autophagic vacuole의 증가는 glucagon 단독투여군보다 좀더 현저한 경향이었다. 이상의 연구성적으로 보아, glucagon은 간세포에 있어서 autophagy를 증가시킬 뿐 아니라, 핵에도 작용하여 그 비대를 일으키며, 이때 cycloheximide를 투여하여 단백합성을 억제하여도 autophagy를 저해하지는 않는다고 믿어진다. 따라서 autophagic vacuole의 한계막은 새롭게 신생된다기 보다는 세포내의 기존의 막, 예를 들면 SER, RER, Golgi막등 막성 성분에서 유래될 가능성이 지금으로서는 더 많은 것으로 생각된다. Glucagon은 autophagic vacuole을 증가시킬 뿐 아니라, cycloheximide를 투여한 후 glucagon을 투여한 경우에는 autophagic vacuole의 증가와 더불어 peroxisome도 증가시킬 것으로 추정된다. In order to investigate the morphologic changes and origin of membrane of lysosomes, the author administered glucagon, 100㎍ per 100gm of body weight, cycloheximide, 0.15㎎ per 100 gm of body weight and both to the healthy male ICR mice intraperitoncally. Each hepatic tissue was observed by light microscope and electron microscope. The results were summarized as follows. Light microscopically, swelling of heaptocytes and focal fatty change appeared around the-central vein. The severity of these changes was mild in the group administered glucagon and was moderate in groups administered cycloheximide alone, both cycloheximide and glucagon. Electron microscopically, in the group administered glucagon, marked increase of lysosomes, particularly autophagic vaculoles, enlargement of nuclei with increase of nuclcoli in thc hepatocytes and increase of phagocytosis in kupffer cells were appeared. In the group administered cycloheximide, slight increase of lysosomes, dilatation and fragmentation of RER, shedding of ribosomes, increase of free ribosomes, atrophy of Golgi complex, decrease of SER, swelling and vacuolizatlon of mitochondria, and increase of lipid droplets occured. In thr group aiministered both cycloheximide and glucagon, above findings in each group and additional increase of peroxisomes appeared. In these changes, increase of autophagic varuoles was more prominent in the group administered glucagon. From the above results, it is believed that glucagon induces enlargement of nuclei as increase of autophagy in the hepatocytes. At this time, in spite of suppression of protein synthesis by administration of cycloheximide, the autophage was not inhibited. Therefore it is more possible that the limiting membrane of autophagic vacuoles originates from preexisting membrane rather than front de novo synthesis. when glucagon was administered after administration of cycloheximide, peroxisomes as well as autophagic vacuoles were increased.

        Verrucous mycosis fungoides is very rare variant of cutaneous T cell lymphoma that manifests verrucous crusted lesion and may be clinically misdiagnosed as a wart, deep fungal infection or atypical mycobacterial infection. If it is located acrally, it might then be better classified as mycosis fungoides palmaris et plantaris or pagetoid reticulosis. We report a case of solitary verrucous mycosis fungoides on the left knee in 25-year-old woman successfully treated ultraviolet A-1 phototherapy.

      • 세포질내 정자주입법(ICSI)에 있어서 정자흡입 및 난자내 주입방법에 관한 연구

        Study on Method of Sperm Aspiration and Injection into an Oocyte in Intracytoplasmic Sperm Injection(ICSI) Immobilization of spermatozoa prior to intracytoplasmic sperm iniection(ICSI) sometimes results in crooked tail and this makes it difficult to aspirate sperm into an injection pipette tail first. Head-first sperm aspiration into an injection pipette avoid this problem due to the bigger size of the sperm head. The effect of head or tail-first sperm injection into an oocyte on fertilization cleavage, percentage of grade I embryos and development to blastocyst stage in ICSI program has been studied. A single living immobilized spermatozoa from oligoasthenozoospermic patient was injected into an oocyte head-first or tail-first according to the treatment. Eighteen hours after microinjection, oocytes ware inspected for survival and fertilization Fertilized oocytes with two pronuclei were cultured in 30μl drop of mHTF supplemented with 10% heat-inactivated follicular fluid(FF) at 37℃. On day 2. embryo transfer was performed with cleaved embryos. The remaining 2-8 cell stage embryos were co-cultured with BRL cells in mHTF + 10% FF for 72 hours and the developmental stage was observed. The data were analyzed by Analysis of Variance. A total of 164 oocytes from 36 cycles were assigned to earth treatment and ICSI was performed(88 head-first, tail-first). The rates of normal fertilization were 81.8% and 76.3% for head-first and tail-first, respectively. Of the fertilized oocytes, the percentage of cleaved embryos and the percentage of grade 1 embryo among cleaved embryos were 88.9% and 68.8%, 93.1% and 74.1% for head-first and tail-first, respectively. Of the 2-8 cell embryos cultured, 44.4%(16/36) and 50.0%(10/20) for head first and tail first, respectively developed to blastocyst stage. There were no differences in fertilization, cleavage, rates of grade 1 embryos, and development to blastocyst stage. In conclusion, head-first or tail-first sperm injection into an oocyte in ICSI program does not affect fertilization and subsequent embryo development to blastocyst stage in vitro.

      • 위암세포에 의한 종양침윤 림프구의 면역반응 억제기전에 관한 연구

        Tumor-infiltrating lymphocytes ('1°ILs) interact most closely with tumor cells and thus are more likely to reflect tumor host interactions accurately. But it is unknown whether such T cells are nonspecific inflammatory cells or a subset of specific host immune responses. In this study, there was no clear correlation between the infiltration of T lymphocytes in stomach cancer and the overexpression of c-ErbB-2 or increasing class I MHC expression on tumor cells. A positive correlation was seen between the presence of TILs in the tumor and tumors with diploidy by flow cytometric DNA analysis. The proliferative responses of Ills stimulated with IL-2, anti-CD3 mAb, or both were examined. When compared to normal mucosal-associated lymphoid tissue lymphocytes, the proliferative response of TILs to high dose IL-2 was minimal. A similarly poor response to anti-CD3 mAb plus IL-2 was also observed. The freshly isolated TILs exhibit reduced ability to proliferate in response to IL-2, anti-CD3 mAb or both. The microenvironment of the tumor suppresses the proliferative capacity of the TILs. The mechanism of this suppression remains unknown. It could be mediated by suppressor cells, by soluble substances within the tumor, or both. To examine this question, supernatants of stomach cancer cells (SNSNU-1) were tested for the presence of immunosuppressive factors. Human peripheral blood T-cells and tumor-draining lymph node lymphocytes (TDLNL) were incubated for 3 days with SNSNU-1 and then assessed for proliferative responses to PMA, anti-CD28 mAb, or both and for the inducibility to express IFN- r or IL-4 mRNA to PMA. Peripheral blood T-cells pretreated with SNSNU-1 were unable to proliferate in response to PMA, anti-CD28 mAb or both. SNSNU-1 also produces inhibitory activities of TDLNL proliferative response to PMA or anti-CD28 mAb and PMA (49%, 52%, respectively). In contrast, culture supernatants obtained from HEp-2, K562 or Daudi showed normal proliferative responsiveness of peripheral blood T-cells and TDLNL by PMA, anti-CD28 mAb or both.

      • 우리나라에서 분리되는 Haemophilus influenzae의 9종 항균제에 대한 시험관내 감수성

        To assess the occurrence of antimicrobial resistance among clinical isolates of H. influenzae in Korea, a total of 238 isolates were tested for their in vitro susceptibitlity to ampicillin and cefaclor by disc diffusion and 105 isolates were tested against nine antimicrobial agents-by the agar dilution method recommended by NCCLS. The antimicrobial agents sued for testing were ampicillin (Sigma A-9518, U.S.S.), cefaclor (Daewoong Lilly, Korea), cephalexin (Sigma C-4895, U.S.A.), cephradine (Sigma C-8395, U.S.A.), cephalothin (Sigma C-4520, U.S.A.), cefadroxil (Sigma C-7020, U.S.A.), erythromycin (Sigma E-6376, U.S.A.), teracycline (Sigma T-3383, U.S.A.) and cefatrizine (Dong-A Pharmaceutical Co., Korea). The isolation rate of H. influenzae from various clinical samples was 18.9%. Using the disc diffusion test, the resisant rates to ampicillin and cefaclor were shown to be 21.9% and 3.4%, respectively. Among the 52 ampicillin-resistant strains, 42 were susceptible to cefaclor (80.8%), and only two strains wre resistant. The MIS_(s) of ampicillin ranged from <0.125 ㎍/㎖ to >128 ㎍/㎖, and its MIC_(90) was 1 ㎍/㎖. Ampicillin showed the lowst MIC_(90) among the tested antivicrobial agents. The MIC_(90) of the tested cephalosporins ranged from 9.2㎍/㎖ to > 128. Among them, cefaclor showed the lowest MIC_(90) and its MIC_(90) was 9.2 ㎍/㎖. The MIC_(90) of tetracycline and erythromycin were 2.5 ㎍/㎖ and 14.4 ㎍/㎖, respectively. By the MIC, the resistant for ampicillin, cefalor, cephalexin, cephracine, cephalothin, cefadroxil, erythromycin, tetracycline and cefatrizine were 9.5%, 1.9%, 61.9%, 76.2%, 3.8%, 68.6%, 65.7%, 13.3% and 5.7%, respectively. There was no evidence that the MIC_(s) of cefaclor for the ampicillin-resistant strains differed significantly from those of cefaclor obtained with the ampicillin-susceptible strains. Except two islates, all of the multiple-esistant strains of H. influenzae were suceptible to cefaclor.

      • n-CdS_(0.46)Se_(0.54)/p-Cu_92-x)S_(0.46)Se_(0.54) 이종접합 태양전지의 제작과 그 특성에 관한 연구

        승화방법에 의해 CdS_0.46Se_0.54 단결정을 성장하여 결정구조를 조사하고, Van der Pauw 방법으로 Hall effect를 측정하여 carrier density의 온도 의존성과 mobility의 온도 의존성을 조사하였다. 성장된 CdS_0.46Se_0.54 단결정을 치환반응하여 n-CdS_0.46Se_0.54/p-Cu_2-xS_0.46Se_0.54 이종접합 태양전지를 제작하였다. Spectral response, 전류-전압특성 및 전력변환 효율을 조사하여 그 결과로부터 개방전압은 0.48V, 단락 전류 밀도는 21mA/㎠, fill factor와 전력변환효율은 각각 0.75와 9.5%를 얻었다. CdS_0.46Se_0.54 single crystal was grown by a sublimation method. The crystal structure and the temperature dependence of carrier density and mobility of CdS_0.46Se_0.54 single crystal were studied. Heterojunction solar cells on n-CdS_0.46Se_0.54/p-Cu_2-xS_0.46Se_0.54 were fabricated by the substitution reaction. The spectral response, the J-U characteristics and the conversion efficiency of the n-CdS_0.46Se_0.54/p-Cu_2-xS_0.46Se_0.54 heterojunction solar cells were studied. The open-cricuit voltage, short-circuit density, fill factor and conversion efficiency of n-CdS_0.46Se_0.54/p-Cu_2-xS_0.46Se_0.54 heterojunction solar cells under 80mW/㎠ illumination were found to be 0.48V, 21mA/㎠, 0.75 and 9.5%, respectively.
