        Allergenicity and Stability of 6 New Korean Bony Fish Extracts

        Purpose Diagnostic tests for allergen sensitization should reflect real exposure. We made 6 new bony fish extracts, which are consumed popularly in Korea, and evaluated their allergenicity and stability. Methods We manufactured fish extracts from codfish, mackerel, common eel, flounder, cutlass, and catfish. Protein and parvalbumin (PV) were evaluated by Bradford assay, 2-site enzyme-linked immunosorbent assay, sodium dodecyl sulphate–polyacrylamide gel electrophoresis (SDS-PAGE), and anti-PV immunoblotting. The immunoglobulin E (IgE) reactivities of the extracts were evaluated with ImmunoCAP and IgE immunoblotting using sera from 24 Korean fish allergy patients, 5 asymptomatic sensitizers, and 11 non-atopic subjects. Stability of the extracts stored in 4 different buffers were evaluated for up to a year. Results The protein concentrations of commercial SPT fish extracts varied with up to a 7.5-fold difference. SDS-PAGE showed marked differences in the PV concentrations of commercial SPT reagents. Specific IgE measurements for the following investigatory fish extracts—iCodfish, iMackerel, and iEel—were concordant with that of their corresponding Phadia ImmunoCAP measurements. ImmunoCAP results showed marked IgE cross-reactivity among the fish species, and the overall sensitivity of ImmunoCAP with the investigatory fish extracts for identification of culprit fish species was 85.7%. The protein and PV concentrations in the investigatory extracts were highly stable in saline with 0.3% phenol–50% glycerol at 4°C for up to a year. Conclusions The commercial SPT fish extracts exhibited considerable variation in terms of allergenicity, which may impact on diagnostic accuracy. Our new fish extracts have sufficient allergenicity and stability and may be adequate to various clinical applications.

        <P>The cell wall skeleton of Mycobacterium bovis Bacillus Calmette-Guerin (BCG/CWS) is an effective antitumor immunotherapy agent. Here, we demonstrate that BCG/CWS has a radiosensitizing effect on colon cancer cells through the induction of autophagic cell death. Exposure of HCT116 colon cancer cells to BCG/CWS before ionizing radiation (IR) resulted in increased cell death in a caspase-independent manner. Treatment with BCG/CWS plus IR resulted in the induction of autophagy in colon cancer cells. Either the autophagy inhibitor 3-methyladenine or knockdown of beclin 1 or Atg7 significantly reduced tumor cell death induced by BCG/CWS plus IR, whereas the caspase inhibitor z-VAD-fmk failed to do so. BCG/CWS plus IR-mediated autophagy and cell death was mediated predominantly by the generation of reactive oxygen species (ROS). The c-Jun NH(2)-terminal kinase pathway functioned upstream of ROS generation in the induction of autophagy and cell death in HCT116 cells after co-treatment with BCG/CWS and IR. Furthermore, toll-like receptor (TLR) 2, and in part, TLR4, were responsible for BCG/CWS-induced radiosensitization. In vivo studies revealed that BCG/CWS-mediated radiosensitization of HCT116 xenograft growth is accompanied predominantly by autophagy. Our data suggest that BCG/CWS in combination with IR is a promising therapeutic strategy for enhancing radiation therapy in colon cancer cells through the induction of autophagy.</P>

        Comparison Of Antibacterial Activity Of 10 Essential Oils And Oxacillin Against Staphylococcus Epidermidis

        다양한 연구가 에센셜 오일과 피부 상재균에 관한 초점을 맞추고 있지만, 에센셜 오일의 항균 특성과 미생 물에 대한 효능 비교 연구는 거의 없다. 이 연구에서 우리는 옥사실린을 대조군으로 사용하여 표피포도상구균에 대한 10가지 에센셜 오일의 항균 활성을 테스트했다. 두 가지 다른 농도에서 에센셜 오일과 표피포도상구균을 함유한 용액의 활성을 측정하기 위해 광학 밀도(OD)를 사용했다. 옥사실린에 비해 에센셜오일 3종(팔마로사, 레몬 그라스, 멜리사 트루)이 항균활성은 더 높았으나 농도는 항균활성에 미미한 영향을 미치는 것으로 나타났다. Although various studies have focused on the relationship between essential oils (EOs) and skin flora, there are only few comparative studies on the antibacterial properties of EOs and their efficacy against common microorganisms. In this study, we tested the antibacterial activity of 10 different EOs against Staphylococcus epidermidis(S. epidermidis) with oxacillin as control. Optical density was used to measure the activity of solutions containing the EOs and S. epidermidis at two different concentrations. Three EOs (palmarosa, lemongrass, and Mellissa True) had higher antibacterial activity than oxacillin, but their concentrations had negligible effect on antibacterial activity.

        This study was carried out to investigate the prevalence of Ascaris suum infection of slaughtered pigs in Chungju and Eumseong area from January to June in 2007. The percentage of pigs and farms detected milk spot, adult worm, and adult worm in detected milk spot were 4.2% and 23.8%, 0.2% and 9.3%, and 4.9% and 39%, respectively. The prevalence of A suum infection from April to June 2007 was lower than that of January to March 2007. There is not enough of difference in farms` infection rate based on their breeding type.

        학교 밖 청소년의 학업 중단 과정 탐색 및학업 중단 인식 연구 -강북 지역 학교 밖 청소년을 중심으로-

        본 연구는 학교 밖 청소년의 학업 중단 과정을 탐색하고 학업 중단에 관한 인식 변화를 조사하기 위해 실시한 것이다. 이를 위해 2016년 7월초부터 9월말까지 강북 지역의 청소년 16명을 대상으로 반구조화된 질문지를 바탕으로 한 면담을 실시, 분석해 다음과 같은 결과를 얻었다. 첫째, 면담에 참여한 학교 밖 청소년들은 대부분 현재 실시되고 있는 학업 중단 예방 관련 제도의 적용을 받지 못한 채 비교적 간단 하게 학업 중단을 결정한 것으로 조사되었다. 둘째, 학업을 중단한 청소년들이 대부분 후회하며 학업 중단을 부정적으로 인식할 것이라는 일반적인 생각과 달리 면담 대상 청소년 중 2/3가 학업 중단 결정에 만족하고 학업 중단을 긍정적으로 인식하고 있었다. 셋째, 학업 중단 결정을 후회하는 청소년들은 학교생활 부적응 경험의 아픔 으로 인해 학교 복귀에 대해 양가감정을 가지고 있었고, 학업 중단에 만족하는 청소 년들은 모두 학교 복귀를 거부했다. 넷째, 학업 중단에 만족하고 현재 자신의 삶을 긍정적으로 인식하고 있는 청소년들의 70%가 사회적 인식, 현실적 어려움 등을 이유로 주변에 학업 중단을 권유하지 않겠다고 응답했다. 이러한 연구 결과는 학업 중단 예방과 학교 밖 청소년에 대한 지원정책의 다양화가 이루어져야 함을 시사한다. The purpose of this study is to investigate the drop-out process and analyse the perception of out-of-school adolescents of their regret or satisfaction on their drop-out decisions. We hope to provide policy implications for out-of-school youths. For study, 16 of out-of-school adolescents in gangbuk area were interviewed. The research results are like those; First, out-of-school adolescents relatively processed drop-out in short time without application of prevention system on drop-out. Second, unlike the myth that the almost of out-of-school adolescents might be regret for their drop-out decisions, considerable numbers of adolescents have positive perception. Third, adolescents of the regret group, who had experienced maladjustment in the school life, hesitated to return to school. On the other hand, adolescents of satisfaction group would not like to return to school. Fourth, adolescents of satisfaction group, who had experienced social barrier, didn t recommend drop-out. These findings suggest that the policies of prevention to drop-out and intervention for out-of-school adolescents need to be diversified.

        Nitric Oxide Synthesis is Modulated by 1,25-Dihydroxyvitamin D3 and Interferon-${\gamma}$ in Human Macrophages after Mycobacterial Infection

        Background: Little information is available the role of Nitric Oxide (NO) in host defenses during human tuberculosis (TB) infection. We investigated the modulating factor(s) affecting NO synthase (iNOS) induction in human macrophages. Methods: Both iNOS mRNA and protein that regulate the growth of mycobacteria were determined using reverase transcriptase-polymerase chain reaction and western blot analysis. The upstream signaling pathways were further investigated using iNOS specific inhibitors. Results: Here we show that combined treatment with 1,25-dihydroxyvitamin D3 (1,25-D3) and Interferon (IFN)-${\gamma}$ synergistically enhanced NO synthesis and iNOS expression induced by Mycobacterium tuberculosis (MTB) or by its purified protein derivatives in human monocyte-derived macrophages. Both the nuclear factor-${\kappa}B$ and MEK1-ERK1/2 pathways were indispensable in the induction of iNOS expression, as shown in toll like receptor 2 stimulation. Further, the combined treatment with 1,25-D3 and IFN-${\gamma}$ was more potent than either agent alone in the inhibition of intracellular MTB growth. Notably, this enhanced effect was not explained by increased expression of cathelicidin, a known antimycobacterial effector of 1,25-D3. Conclusion: These data support a key role of NO in host defenses against TB and identify novel modulating factors for iNOS induction in human macrophages.
