Plastic 포매 조직표본 제작에서 Microwave를 이용한 조직고정에 관한 연구

Microwave를 이용하여 고정한 조직을 plastic 포매하여, 얻은 절편으로 조직화학 염색인 Hematoxylin-Eosin(H-E), periodic acid-Shiff(PAS), reticulum, Masson's trichrome(M·T)염색을 하고 통상적으로 이용되는 포르말린 고정법을 통한 조직표본에서 동일한 염색을 하여 세포의 형태학적 관찰과 염색성을 비교하고자 본 연구를 시도하여 다음과 같은 결론을 얻었다. 1.Microwave조사 시간이 짧고 최종온도가 낮을 수록 조직의 형태학적 소견은 전반적인 세포구조가 파괴되어 불량하고 염생성도 약하였다. 2.Microwave를 이용한 고정에서 최적온도는 장기에 따라 서로 다르며 간장조직은 조사시간 136초, 최종온도59±2℃신장조직은 조사시간 175초, 최종온도 69±2℃에서 세포의 구조가 가장 잘 보존되었고 염색성도 양호하였다. 3.장기별 최적조건에서 PAS, reticulum, Masson's trichrome염색시 결과는 일반적인 포르말린 고정을 한 조직에서 나타난 결과와 일치하였다. 이상의 결과로서 microwave를 이용한 고정방법은 장기별 최적조건이 서로 다르고 고정시간이 짧다는 장점과 다양한 조직화학적 염색에 있어서도 양호한 염색성을 보였으며 전반적으로 형태학적 구조가 잘 보존되었다. This study was undertaken to evaluate the effect of rapid heat fixation on specimens using microwave irradiation. Small portions of the liver and the kidney specimen were fixed by microwave irradiation and subsequently processed by plastic embedding for light microscopical study. The control specimens were fixed by 10% neutral buffered formalin and processed by same method as the experimental ones. Both were stained with hematoxylin and eosin, periodic acid-Schiff, reticulum and Masson's trichrome. The results were as follows: 1.The tissue structures were destroyed and staining property of cells was poor by microwave irradiation at low temperature. 2.The optimal condition of fixation was different in each tissues. For the liver it was 136 seconds microwave irradiation at 59±2℃, for the kidney it was 175 seconds microwave irradiation at 69±2℃. 3. When the specimens were fixed on optimal condition by microwave irradiation, the quality of fixation was as good as that of formalin fixation. Above results suggest that the fixation time can be shortened by microwave fixation.
PRELIMINARY SAFETY STUDY OF ENGINEERING-SCALE PYROPROCESS FACILITY

Pyroprocess technology has been considered as a fuel cycle option to solve the spent fuel accumulation problems in Korea. The Korea Atomic Energy Research Institute has been studying pyroprocess technology, and the conceptual design of an engineering-scale pyroprocess facility, called the Advanced Fuel Cycle (AFC) facility, has been performed on the basis of a 10tHM throughput per year. In this paper, the concept of the AFC facility was introduced, and its safety evaluations were performed. For the safety evaluations, anticipated accident events were selected, and environmental safety analyses were conducted for the safety of the public and workers. In addition, basic radiation shielding safety analyses and criticality safety analyses were conducted. These preliminary safety studies will be used to specify the concept of safety systems for pyroprocess facilities, and to establish safety design policies and advance more definite safety designs.
Excessive <i>O</i>-GlcNAcylation of proteins suppresses spontaneous cardiogenesis in ES cells

<P><B>Abstract</B></P><P>Increased modification of proteins with <I>O</I>-linked <I>N</I>-acetylglucosamine (<I>O</I>-GlcNAc) has been implicated in the development of diabetic cardiomyopathy. We used the well-characterized ES cells (Nkx2.5GFP knock-in ES cells), to investigate the role of <I>O</I>-GlcNAcylation in cardiomyocyte development. <I>O</I>-GlcNAcylation decreased in differentiating ES cells, as did the expression of <I>O</I>-GlcNAc transferase. Increasing <I>O</I>-GlcNAcylation with glucosamine or by inhibiting <I>N</I>-acetylglucosaminidase (streptozotocin or PUGNAc) decreased the number of cardiomyocyte precursors and cardiac-specific gene expression. On the other hand, decreasing <I>O</I>-GlcNAcylation with an inhibitor of glutamine fructose-6-phosphate amidotransferase (6-diazo-5-oxo-norleucine) increased cardiomyocyte precursors. These results suggest that excessive <I>O</I>-GlcNAcylation impairs cardiac cell differentiation in ES cells.</P>
TPC - W 성능 평가에서의 데이타베이스 시스템 성능 인자 튜닝

대량의 데이타를 관리하는 현대 데이타베이스 시스템 환경에서는 데이타베이스 시스템 튜닝에 대한 중요성이 증가하고 있다. 특히, 데이타베이스 시스템 성능 인자(performance parameters)를 시스템 부하에 따라 적절하게 튜닝하여야 한다. 본 논문에서는 TPC-W 환경에서 데이타베이스 시스템의 단일 성능 인자 튜닝 전략 2가지(처리량 평가 방법, 응답시간 평가 방법)를 제시한다. 효과성을 입증하기 위하여 제시한 튜닝 전략은 두 개의 상용 데이타베이스 시스템에 적용하였다. 실험 결과는 제시된 튜닝 전략이 성능 향상에 기여함을 보인다. There have been an emerging interests in the importance of database tuning techniques under modern database environments in which very large-scale data should be managed. In particular, database performance parameters should be tuned to reflect system loads appropriately. This paper presents two parameter tuning strategies, namely throughput-based and response-time-based, which tune each performance parameter accordingly. The proposed techniques are applied to two commercial database systems in the TPC-W benchmark to see the effectiveness of those methods. The results show that they can help improve system performance considerably.

Objective : The purpose of this study is to determine the effects of the methods of anesthesia on fetal acidemia following elective cesarean delivery among uncomplicated healthy parturients at term, with a single fetus and is to see the correlation of tha
