Identification and Analysis of the Novel pGAPDH-w Gene Differentially Expressed in Wild Ginseng

Objective: Panax ginseng is one of the most medicinally used herbal medicines in the world. Wild ginseng is widely accepted to be more active than cultivated ginseng in chemoprevention. However, little has actually been reported on the differences between wild ginseng and cultivated ginseng. Method: To identify wild ginseng-specific genes, we used suppressive subtraction hybridization. Results: We report that one of the clones isolated in this screen was the GAPDH (glyceraldehyde 3-phosphate dehydrogenase) gene (designated pGAPDH-w). DNA BLAST sequence analysis revealed that this pGAPDH-w gene contained novel sequences of 94 bp. RT-PCR results showed that the expression of the pGAPDH-w gene was significantly up-regulated in the wild ginseng as compared with the cultivated ginseng. Conclusion: The pGAPDH-w gene may be one of the important markers of wild ginseng.

Although wild ginseng is widely accepted to be more active than cultivated ginseng in chemoprevention, little has actually been reported on the difference between wild ginseng and cultivated ginseng. Using suppressive subtraction hybridization, we cloned the p-psbB gene as a candidate target gene for a wild ginseng-specific gene. Here, we report that one of the clones isolated in this screen was the chloroplast p-psbB gene, a chlorophyll a-binding inner antenna protein in the photosystem II complex, located in the lipid matrix of the thylakoid membrane. Real-time results showed that the expression of the p-psbB gene was significantly up-regulated in wild ginseng as compared to cultivated ginseng. Thus, the p-psbB gene may be one of the important markers of wild ginseng.

Thus, to identify and analyze those differences, we used suppressive subtraction hybridization (SSH) sequences with microarrays, realtime polymerase chain reaction (PCR), and reverse transcription PCRs (RT-PCRs). One of the clones isolated in this research was the chloroplast rpoC1 gene, a ${\beta}$subunit of RNA polymerase. Real-time RT-PCR results showed that the expression of the rpoC1 gene was significantly upregulated in wild ginseng as compared to cultivated ginseng, so, we conclude that the rpoC1 gene may be one of the important markers of wild ginseng.
O형 고무 밴드 결찰법을 이용한 Mallory-Weiss 증후군의 치료

Background/Aims: Mallory-Weiss syndrome refers to a laceration or lacerations in the region of the gastroesophageal junction due to vomiting, retching, or coughing induced by several causes, and preceding hematemesis. In the last several years, endoscopic therapies have dramatically changed the need for emergency surgery in patients with upper gastrointestinal bleeding. There is only little information however, regarding the indication criteria and the efficacy of endoscopic therapies in severe upper gastrointestinal bleeding, due to Mallory-Weiss syndrome. This study was designed to assess the usefulness and the indications of endoscopic O-ring band ligation. Methods: Thirty patients with Mallory- Weiss syndrome who were experiencing a related hemorrhage were studied. Among these, 5 patients with active bleeding or a visible vessel revealed during an endoscopic examination were treated with O-ring band ligation. Patients with blood clots or linear tears received only conservative treatment. Result : 1) It was determined that Mallory-Weiss syndrome was the cause of bleeding n 6.1% of patients with upper gastrointestinal bleeding. 2) active bleeding or a visible vessel seen in an endoscopic examination was found in 5 patients (16.7%); definitive heomostasis was obtained in all cases with O-ring band ligation. 3) The remaining 25 patients were successfully teated with conservative therapy. 4) Patients undergoing O-ring band ligation displayed a significantly lower initial diastolic blood pressure and hemoglobin levels, and also showed significantly more numbers of melena and needed more transfusions than patients with conservative therapy. Conclusions: Severe bleeding due to Mallory-Weiss syndrome can be successfully treated by O-ring band ligation. Moreover, active bleeding or visible vessel detected in n endoscopic examination can be considered an indication of O-ring band ligation in upper gastrointestinal bleeding due to Mallory-Weiss syndrome.

The experiment described in this paper focused on determining the effects of high-temperature(20℃) treatment of Satsuma mandarins, prior to their storage, on their color development and quality. High-temperature treatment promoted fruit skin color during the storage, decreased the weight loss regardless of their color at the time of their harvest and reduced the decay occurrence until the middle of March, but there was no difference between treatments after the end of March. There was a tendency of higher soluble solid and acid levels in the fruit when high-temperature treatment was applied but this difference was not significant.
