<P><B>OBJECTIVE</B></P><P>Patients with diabetic erectile dysfunction often have severe endothelial dysfunction and respond poorly to oral phosphodiesterase-5 inhibitors. We examined the effectiveness of the potent angiopoietin-1 (Ang1) variant, cartilage oligomeric matrix protein (COMP)-Ang1, in promoting cavernous endothelial regeneration and restoring erectile function in diabetic animals.</P><P><B>RESEARCH DESIGN AND METHODS</B></P><P>Four groups of mice were used: controls; streptozotocin (STZ)-induced diabetic mice; STZ-induced diabetic mice treated with repeated intracavernous injections of PBS; and STZ-induced diabetic mice treated with COMP-Ang1 protein (days −3 and 0). Two and 4 weeks after treatment, we measured erectile function by electrical stimulation of the cavernous nerve. The penis was harvested for histologic examinations, Western blot analysis, and cGMP quantification. We also performed a vascular permeability test.</P><P><B>RESULTS</B></P><P>Local delivery of the COMP-Ang1 protein significantly increased cavernous endothelial proliferation, endothelial nitric oxide (NO) synthase (NOS) phosphorylation, and cGMP expression compared with that in the untreated or PBS-treated STZ-induced diabetic group. The changes in the group that received COMP-Ang1 restored erectile function up to 4 weeks after treatment. Endothelial protective effects, such as marked decreases in the expression of p47<SUP>phox</SUP> and inducible NOS, in the generation of superoxide anion and nitrotyrosine, and in the number of apoptotic cells in the corpus cavernosum tissue, were noted in COMP-Ang1–treated STZ-induced diabetic mice. An intracavernous injection of COMP-Ang1 completely restored endothelial cell-cell junction proteins and decreased cavernous endothelial permeability. COMP-Ang1–induced promotion of cavernous angiogenesis and erectile function was abolished by the NOS inhibitor, <I>N</I>-nitro-L-arginine methyl ester, but not by the NADPH oxidase inhibitor, apocynin.</P><P><B>CONCLUSIONS</B></P><P>These findings support the concept of cavernous endothelial regeneration by use of the recombinant Ang1 protein as a curative therapy for diabetic erectile dysfunction.</P>
Promoted Growth of Maize by the Phosphate Solubilizing Bacteria Isolated from North-east China

A strain of phosphate solubilizing bacterium was isolated from rhizosphere and identified as Burkholderia sp. by 16S-rRNA gene sequence analyses. The bacterium was found to release gluconic acid and the solubilization of hydroxyapatite in the liquid medium by a significant drop in pH to 3.7 from an initial pH 7.0. The soluble-P concentration continuously increased during the incubation periods and the total amount of soluble P released in culture filtrate was detected at 990 mg L<SUP>-1</SUP> after 10 days of inoculation. Most promoted maize growth was found in the standard NPK (240-120-120 kg ha<SUP>-1</SUP>) soil inoculation with Burkholderia sp. (Twenty milliliters/ plant, 106 CFU) and also in the absence of Burkholderia sp. inoculation, the soil amended with only 2/3 levels of P gave significant higher plant yield compared to 1/3 levels of P or without P supplementation.

Purpose: This study aims to analyze the clinical-pathological characteristics of multifocal and multicentric breast cancer (MMBC) in Chinese women. Methods: Sixty-seven cases with MMBC were randomly collected and reviewed at seven hospitals in representative districts of China during 1999 to 2008. Results: The incidence of MMBC in breast cancer in China was 1.75%. Compared to those with unifocal breast cancer, women with MMBC were more likely to have larger tumor size, lymph node metastasis (59.70% vs. 45.62%) and stage III to IV (46.26% vs. 21.10%). The peak age at onset of MMBC was 40 to 49 years old and has been gradually increasing during 1999 to 2008. Most of the MMBC women were treated with surgery and adjuvant therapy. Conclusion: In China, the incidence of MMBC in breast cancer is significantly lower than that in Western countries. Compared to unifocal breast cancer, MMBC is biologically more aggressive. Most MMBC women underwent mastectomy, instead of breast conservation surgery.

DBOP specifies palea development by suppressing the expansion of the margin of palea in rice

Palea and lemma are grass specific floral organs, and their identities to the typical floral organs in eudicots remains unclear. In the present study, we identified and characterized a defective body of palea (dbop) from ethyl methyl sulfonate mutagenesis lines of Oryza sativa ssp. japonica cv. Nipponbare. The dbop mutant exhibited abnormal palea without the body of palea. By map-based cloning, the DBOP was narrowed down to a 68.9 kb interval on the long arm of chromosome 9. Sequencing result showed that there was G to A substitution and C to A substitution at the 245th and 383rd nucleotide of LOC_Os09g24480 coding sequence region, respectively. DBOP was found allelic to the RETARDED PALEA1 (REP1) gene which encoded a TCP family transcription factor. DBOP/REP1 might specify palea development by suppressing the expansion of the margin of palea.

Purpose: Transforming growth factor-β1 (TGF-β1) has been implicated in cavernous fibrosis due to a variety of causes of erectile dysfunction (ED), such as diabetes mellitus and post-radical prostatectomy. To examine the role of the TGF-β signaling pathway in cavernous fibrosis, we established a rat model of cavernous fibrosis by using adenovirus expressing TGF-β1 (ad-TGF-β1). Materials and Methods: Four-month-old male Sprague-Dawley rats received intracavernous injection of ad-TGF-β1 (1×108, 1×109, or 1×1010 virus particles [vp] in 100 μl of PBS) and the penis was harvested for histologic examination at 10, 20, or 30 days after injection (n=4 per group and per time point). Based on the initial findings, the animals were divided into three groups (n=6 per group): Group 1, age-matched control; Group 2, intracavernous injection of ad-LacZ (1×1010 vp/100 μl); and Group 3, intracavernous injection of ad-TGF-β1 (1×1010 vp/100 μl). At 30 days after injection, erectile function was evaluated during electrical stimulation of the cavernous nerve. The penis was then harvested and stained with Masson’s trichrome and antibody to smooth muscle α-actin. Results: Masson’s trichrome staining revealed that intracavernous delivery of ad-TGF-β1 sufficiently induced cavernous fibrosis in a dose-dependent manner. The fibrotic scars persisted up to 30 days after injection at the highest dosage (1×1010 vp/100 μl), whereas no histologic evidence of cavernous fibrosis was found in the control rats or the ad-LacZ-injected rats. The rats receiving ad-TGF-β1 showed a higher cavernous collagen content and less smooth muscle content than the control rats or ad-LacZ-injected rats. Erectile function was significantly decreased in rats receiving ad-TGF-β1 compared with that in controls or rats receiving ad-LacZ. Conclusions: This model induced by ad-TGF-β1 may play an important role in understanding the pathophysiologic mechanisms of cavernous fibrosis-associated TGF-β signaling and the development of new therapeutics targeting this pathway.
