Vascular Protective Role of Samul-Tang in HUVECs: Involvement of Nrf2/HO-1 and NO

<P>Samul-Tang (Si-Wu-Tang, SMT), composed of four medicinal herbs, is a well-known herbal formula treating hematological disorder or gynecologic disease. However, vascular protective effects of SMT and its molecular mechanisms on the vascular endothelium, known as the central spot of vascular inflammatory process, are not reported. The aim of this study was to investigate vascular protective effects of SMT water extract in human umbilical vein endothelial cells (HUVECs). Water extract of SMT was prepared and identified by HPLC-PDA analysis. Expression of cell adhesion molecules (CAMs) and heme oxygenase-1 (HO-1) and translocation of nuclear factor-kappa B (NF-<I>κ</I>B) and nuclear factor-erythroid 2-related factor 2 (Nrf2) were determined by western blot. Nuclear localization of NF-<I>κ</I>B and Nrf2 was visualized by immunofluorescence and DNA binding activity of NF-<I>κ</I>B was measured. ROS production, HL-60 monocyte adhesion, and intracellular nitric oxide (NO) were also measured using a fluorescent indicator. SMT suppressed NF-<I>κ</I>B translocation and activation as well as expression of CAMs, monocyte adhesion, and ROS production induced by TNF-<I>α</I> in HUVECs. SMT treated HUVECs showed upregulation of HO-1 and NO which are responsible for vascular protective action. Our study suggests that SMT, a traditionally used herbal formula, protects the vascular endothelium from inflammation and might be used as a promising vascular protective drug.</P>

In this study, To investigate the antioxidant activities of Rhaponticum uniflorum extracts (RUE), we assessed scavenging activities on DPPH, ABTS radicals, and reducing power. The DPPH and ABTS radical scavenging activities (SC50) of RUE were 96.97±1.19 μg/mL and 31.26±0.13 μ g/mL, respectively, and the reducing power significantly increased dose-dependent manner. To study the antioxidant effecters of RUE in H₂O₂ induced human keratinocytes, expression of Nuclear factor erythroid-2-related factor 2 (Nrf2) and heme oxygenase-1 (HO-1) were determined by Western blot and qRT-PCR. RUE induced the expression of HO-1, a critical target enzyme of Nrf2, in a concentration-dependent manner. The expression of HO- 1 mRNA was also induced in H₂O₂-treated HaCaT cells but no significant difference in the level of HO-1 mRNA was observed. Taken together, these results suggest that RUE may be useful as a potential ingredient for anti-aging products.

We have analyzed the genetic relationship among the mouse strains using RAPD-PCR. Polymorphic RAPD markers amplified from various primers were used to analyze the genetic relationship among seven mouse strains, CBA, C57BL/6, BALB/C, NOD, A/wy, DBA2, and AKR. The genetic similarity coefficients were estimated using the RAPD markers by UPGMA method. The genetic similarity coefficient between CBA and NOD was 0.474. This value was lower than any other genetic similarity coefficient among the strains tested. And CBA-specific RAPD band patterns were observed in 5 primers from 6 polymorphic primers used. This study makes possible phylogenetic identification among the inbred mouse strains at the molecular level using RAPD-PCR techniques.
기업전략에 따른 균형성과표의 가중치 설계변화 : A사의 사례를 중심으로

기업의 성과관리는 기업의 전략실천과 최고경영층의 기대를 조직 내에 소통시키고 기업의 목표달성을 도울 뿐만 아니라, 구성원들의 성과평가를 통해 보상과 개발 등이 이루어지게 함으로써 조직원들의 기업에 대한 집중과 몰입을 유도한다. 이로써 기업은 조직원들로 하여금 기업운영의 우선순위를 명확히 인지시키고, 한정된 기업자원의 효과적인 집중과 안배를 가져올 수 있을 것이다. 이는 전략실천을 위한 핵심성과지표의 선택과 기업전략의 우선순위가 명확히 반영된 가중치 설계를 통해 조직원들의 전략에 대한 이해 및 측정지표에 대한 공감을 불러일으킴으로써 전략실천을 보다 용이하게 할 것이다. 따라서 본 연구는 전략실천의 경영관리도구로서 널리 활용되고 있는 균형성과표 (Balanced Scorecard: 이하 BSC)의 효과적인 활용방안의 일환으로 전략에 따른 가중치설계변화가 기업의 전략목표대비 성과달성도에 미치는 영향을 검토하고자 한다. BSC를 전략실행을 위한 경영관리의 틀로서 인식하고, 기업전략에 따른 BSC의 가중치 설계변화의 추이를 비교 분석하여 가중치 설계변화의 근거를 살펴보고, 나아가 전략 및 경영환경의 변화, 사업계획, 재무계획, 보상계획 등 기업 고유의 원인이 가중치 설계에 미치는 직 · 간접 원인을 살펴보고자 한다. This study describes the Weights design analysis of the balanced scorecard depending on strategic aligning with corporate strategy and balanced scorecard. Corporate performance management communicates corporate strategy and CEO opinions to organization. Besides it helps a corporate goal achievement and evaluates members in organization. So performance management is very important issue. The results of this case study are summarized as follows. First, in spite of the difference of strategies, companies lay weight on financial perspective rather than other perspectives. From a point of view that the most important factor in designing weighting is the top management's intention, this result proves that the top management still make their decision based on short-term financial performance. Second, company A doesn't complete a systematic and scientific framework in designing weighting. The chief of department merely designs weighting and then he/she allocates voluntarily and intuitively weight to perspective measures. Third, weighting design according to the change of strategy can occur the difference of the level for performance achievement.
한국인 만성 긴장형두통 환자에서 바이오피드백을 활용한 자율적 이완훈련의 효과

Objectives : We aimed to evaluate the efficacy of biofeedback-assisted autogenic training for chronic tension-type headache, and to determine the relationship among the changes in electromyography (EMG) activity, headache activity, and mood states according to the psychophysiological treatment. Methods : Chronic tension-type headache patients aged from 20 to 40 years (n=35) were randomized to the treatment group receiving biofeedback-assisted autogenic training (8 sessions) or the monitoring-only control group. EMG activities, headache index, and various psychological variables were examined. Results : We found greater treatment response rate (≥50% reduction in headache index) in patients with biofeedback-assisted autogenic training than in the monitoring group (61% vs. 18% ; χ²=6.882, df=1, p=0.01). There were no significant changes in the mean values of the pretreatment EMG activities across the sessions m either group (all p’s>0.1). Mood states including anxiety and depression improved over time in the both groups, with the improvements being more prominent in the treatment group. Moreover, the reduction in depression level predicted treatment outcome in terms of headache index (95% confidence interval : 0.272-0.966, p=0.039). Conclusion : These results show that biofeedback-assisted autogenic training is effective for the treatment of chronic tension-type headache in a Korean population. Changes in mood states may be closely associated with the clinical outcome in the treatment of chronic tension-type headache using biofeedback-assisted autogenic training.
