      • Clinicopathological Characteristics of Triple Negative Breast Cancer at a Tertiary Care Hospital in India

        Background: Triple-negative breast cancer (TNBC), characterized by the lack of expression of estrogen receptor, progesterone receptor and human epidermal growth factor receptor-2, is typically associated with a poor prognosis. The majority of TNBCs show the expression of basal markers on gene expression profiling and most authors accept TNBC as basal-like (BL) breast cancer. However, a smaller fraction lacks a BL phenotype despite being TNBC. The literature is silent on non-basal-like (NBL) type of TNBC. The present study was aimed at defining behavioral differences between BL and NBL phenotypes. Objectives: i) Identify the TNBCs and categorize them into BL and NBL breast cancer. ii) Examine the behavioral differences between two subtypes. iii) Observe the pattern of treatment failure among TNBCs. Materials and Methods: All TNBC cases during January 2009-December 2010 were retrieved. The subjects fitting the inclusion criteria of study were differentiated into BL and NBL phenotypes using surrogate immunohistochemistry with three basal markers $34{\beta}E12$, c-Kit and EGFR as per the algorithm defined by Nielsen et al. The detailed data of subjects were collated from clinical records. The comparison of clinicopathological features between two subgroups was done using statistical analyses. The pattern of treatment failure along with its association with prognostic factors was assessed. Results: TNBC constituted 18% of breast cancer cases considered in the study. The BL and NBL subtypes accounted for 81% and 19% respectively of the TNBC group. No statistically significant association was seen between prognostic parameters and two phenotypes. Among patients with treatment failure, 19% were with BL and 15% were with NBL phenotype. The mean disease free survival (DFS) in groups BL and NBL was 30.0 and 37.9 months respectively, while mean overall survival (OS) was 31.93 and 38.5 months respectively. Treatment failure was significantly associated with stage (p=.023) among prognostic factors. Conclusions: Disease stage at presentation is an important prognostic factor influencing the treatment failure and survival among TNBCs. Increasing tumor size is related to lymph node positivity. BL tumors have a more aggressive clinical course than that of NBL as shown by shorter DFS and OS, despite having no statistically significant difference between prognostic parameters. New therapeutic alternatives should be explored for patients with this subtype of breast cancer.

        Isolation and Characterization of Water Soluble Bioactive Substances from Marine Photosynthetic Microalga Tetraselmis Species

        Isolation, quantification and characterization of bioactive compounds from microalgae Tetraselmis species have been performed to elucidate the potential use as a bioactive material. Microalga are a promising source of lipids for biofuel production and represent a diverse group of microscopic photosynthetic organisms and are able to convert solar energy into biomasses. The imposition of oxidative stresses by various environmental factors leads to the production of reactive oxygen species (ROS) in plant cells, including algal cell. Antioxidant activity of water soluble bioactive substances (WSBS) was determined by DPPH, ABTS, FRAP and similar SOD radical-scavenging assay. The content analysis of lipid, sugar, polyphenol and protein was performed to evaluate the potent usefulness of WSBS from the whole cells cultivated under the optimum conditions for the production of biodiesel. The yield of WSBS was determined to be 17% based on the total carbohydrate assay. The relative activity of WSBS toward FRAP and DPPH reagent was observed ranging from 13 to 55%, showing the significant anti-oxidative properties with wide range of temperatures from room temperature to 80℃. The outcomes of this study support utilization of WSBS from microalgae Tetraselmis species as a potential source of naturally occurring antioxidants.

        Poly(NIPAAm-co-SPO) random copolymer was synthesizedvia RAFT polymerization. For the preparation of Janus particles, micron sizedroplets containing the random copolymer were then prepared by using droplet microfluidicsand studied their self-assembly behavior under different conditions. Janusmorphology was developed by phase separation and photo-crosslinking reactionunder UV irradiation. Characteristics of Janus particle preparation andcorresponding self-assembly behavior will be discussed in the presentation (NRFgrant: 2014R1A1A4A01007436).

        Biochemical Properties of Water Soluble Polysaccharides from Photosynthetic Marine Microalgae Tetraselmis Species

        Water soluble polysaccharides (WSPs) were isolated from defatted microalgae Tetraselmis sp. KCTC 12432 BP (T.S1) and KCTC 12236 BP (T.S2), and purified by a combination of ethanol precipitation, immobilized metal affinity chromatography and size-exclusion column chromatography. Biochemical properties of WSPs obtained from the supernatant (S 1 and S 2) and purification steps (EP 1 and EP 2) have been elucidated for their potential use as a biologically active material. Oxidative effect on efficiency from the hydrolytic fenton reaction (FS 1 and FS 2) was also employed to assess the WSPs production and biological activity from T.S1 and T.S2, respectively. Antioxidant activity of the purified WSPs from EP 2 toward 1,1-diphenyl-2-picryl-hy-drazyl (DPPH), 2,2’-Azinobis (3-ethylbenzothiazoline-6-sulfonic acid) di-ammonium salt (ABTS), and ferric reducing antioxidant power (FRAP) of radical-scavenging showed significant differences depending on the monosaccharide compositions of WSPs. On the other hand, a significant specific tyrosinase inhibitory activity was observed from the WSPs purified from EP 1 (10 μmol/g/min) indicating the potential use as a whitening source. The outcomes of our study support the utilization of WSPs from these microalgal species as a potential source of naturally occurring whitening and antioxidants to ease oxidative stresses. According to our knowledge related to microalgal studies, until now this is the first study which demonstrates the significance between primary structure and potential biological activities of WSPs purified from the defatted microalgae Tetraselmis species.

        Tool Wear, Chip formation and Workpiece surface issues in CBN Hard Turning: A Review

        Steel parts that carry critical loads in everything from automotive drive trains and jet engines to industrial bearings and metal-forming machinery are normally produced by a series of processes, including time-consuming and costly grinding and polishing operations. Due to the advent of super-hard materials such as polycrystalline cubic boron nitride (PCBN) cutting tools and improved machine tool designs, hard turning has become an attractive alternative to grinding for steel parts. The potential of hard turning to eliminate the costs associated with additional finishing processes in conventional machining is appealing to industry. The objective of this paper,is to survey the recent research progress in hard turning with CBN tools in regard of tool wear, surface issues and chip formation. A significant pool of CBN turning studies has been surveyed in an attempt to achieve better understanding of tool wear, chip formation, surface finish, white layer formation, micro-hardness variation and residual stress on the basis of varying CBN content, binder, tool edge geometry, cooling methods and cutting parameters. Further important modeling techniques based on finite element, soft computing and other mathematical approaches used in CBN turning are reviewed. In conclusion, a summary of the CBN turning and modeling techniques is outlined and the scope of future work is presented.

        Ovarian Torsion in a Postmenopausal Woman: A Case Report and Review of Literature

        Postmenopausal ovarian masses are not uncommon. Most of them are malignant due to inflammation and adhesion with the surrounding structures; these masses are fixed and torsion is not a usual phenomenon. We present a 73-year-old postmenopausal woman, parity five and five living issues with an acute abdomen. The examination revealed a 28–30-week size abdominal pelvic mass, which was confirmed to be ovarian in origin, with characteristics that indicated torsion on ultrasound Doppler. She underwent an emergency laparotomy with a total abdominal hysterectomy and bilateral salpingo-ophrectomy. Histopathology confirmed the diagnosis of serous cystadenoma. Ovarian torsion is a crucial differential diagnosis in postmenopausal adnexal masses presenting with acute abdomen. A high index of suspicion is required to diagnose and promptly manage women to improve the outcome.

        Stimuli-responsive polymers are very fascinating materials. Among all kinds of stimuli responsive polymers, light responsive polymers deserve particular attention because of easy control of their intensity, spatial localization, and wavelength changes. In this work, light-stimuli block copolymers containing spironaphthoxazine (SPO), which is one of lightresponsive material, were synthesized by reversible-addition fragmentation chain transfer (RAFT) polymerization. Corresponding vesicles were prepared from the synthesized block copolymers and their behavior were investigated under the UV light (NRF grant no.: 2014R1A1A4A 01007436).

        Protective Effect of Naringin, a Citrus Flavonoid, Against Colchicine-Induced Cognitive Dysfunction and Oxidative Damage in Rats

        Alzheimer's disease is a neurodegenerative disorder. Central administration of colchicine is well known to cause cognitive impairment and oxidative damage, which simulates sporadic dementia of the Alzheimer type in humans. The present study has been designed to investigate the protective effects of naringin against the colchicine-induced cognitive impairment and oxidative damage in rats. Colchicine (15μg/5μL), administered intracerebroventricularly, resulted in poor memory retention in both the Morris water maze and elevated plus maze task paradigms and caused marked oxidative damage. It also caused a significant decrease in acetylcholinesterase activity. Naringin (40 and 80mg/kg, p.o.) treatment was given daily for a period of 25 days beginning 4 days prior to colchicine administration. Chronic treatment with naringin caused significant improvement in the cognitive performance and attenuated oxidative damage, as evidenced by lowering of malondialdehyde level and nitrite concentration and restoration of superoxide dismutase, catalase, glutathione S-transferase, and reduced glutathione levels, and acetylcholinesterase activity compared to control. The present study highlights the therapeutic potential of naringin against colchicine-induced cognitive impairment and associated oxidative damage.
