Quantitative Traits Loci (QTL) Mapping of Pleurotus eryngii

Pleurotus eryngii, an edible white-rot fungus, is cultivated widespread in Eurasia, northern Africa and China. It occupies the second position in the world mushroom market. Despites the importance, the small numbers of studies have been done. To promote the availability of P. eryngii, it is important to know the genome and organization of the fungus. In this study, the whole genome sequence of P5 monokaryon from P. eryngii KNR2312 strain was sequenced using Next Generation Sequencing (NGS) strategy. Identified 222 SSR markers based on newly known genome information and various type of markers used to construct the map consisted of 12 linkage groups (LGs) spanning 1047.8 cM. Using composite interval mapping, 71 quantitative trait loci (QTL)s were identified on 12 linkage groups (LG) for nine traits such as yield, quality, cap color and earliness in four different populations. Clusters of more than five QTLs for various traits were identified on three genomic regions on LG 1, 7, and 9. The largest cluster was identified on LG 1 in the range from 65.4 to 110.4 cM. We performed the genes prediction analysis responsible for yield in the LG1 genomic region with highest logarithm of the odd (LOD) scores The identified genes were involved in biomass degradation and synthesis and signal transduction. However, the region was wide to identifie the genes crucial role in important trait, we need to narrow down. To improve the resolution of QTL mapping, we enlarge the populations crossing with additional 205 monokaryons. Find mapping of QTLs will increases the accuracy and efficiency of interpret the genomic region and enhance the usefulness of genomic information. Abelmoschus manihot jinhuakui petal hot water extract(AJPH) was administered to ovariectomized female rats and the experimental groups were as follows: sham-operated rats (SHAM), ovariectomized rats (OVX-CON), ovariectomized rats supplemented with estradiol 10 μg/kg (OVX-E2) and ovariectomized rats supplemented with AJPH 50 or 200 mg/kg (OVX-AJPH50, OVX-AJPH200). Serum ALP activity was significantly increased in the OVX-CON group than in the SHAM group, on the other hand the OVX-AJPH200 group significantly decreased than the OVX-CON group, and showed a similar level to the SHAM group. The content of NTx and CTx in the serum was significantly increased in the OVX-CON group than in the SHAM group. Both OVX-E2 and AJPH groups decreased significantly compared to the OVX-CON group and decreased to the same level as the SHAM group. The collagen content in the bone and cartilage tissues was significantly decreased in the OVX-CON group with ovariectomy compared with the SHAM group, and increased in the OVX-E2 group and the AJPH groups. Therefore, the AJPH is expected to have a positive effect on bone by suppressing bone loss due to ovariectomy.

<P>We have prepared functionalized nanoporous thin films from a polystyrene-block-polyethylene oxide block copolymer, which was made cleavable due to the intervening disulfide bond. The cleavage reaction of the disulfide bond leaves behind free thiol groups inside the nanopores of polystyrene thin film. This nanoporous thin film can be used as a template for generating gold nanoring structures. This strategy can provide a facile method to form a highly ordered array of biopolymer or metal-polymer composite structures.</P>

Molecular markers were crucial role in understanding and using genomic information for marker-assisted breeding, mapping genes of interest and cloning genes. Among the molecular markers, Simple Sequence Repeat (SSR) has became increasingly important marker due to their co-dominant and high polymorphic nature and abundant distribution throughout the genome. Simple sequence repeats (SSR), also called “microsatellites” consist of tandemly repeated short DNA sequence motifs and have applied in various marker-based studies. SSRs were isolated and characterized from “Heuktari” and “Miso”, which are one of the major oyster mushroom cultivars in Korea by the genome sequencing and bioinformatic analysis. The genome sizes of “Heuktari” were 40.8 Mb and “Miso” were estimated to be 40.3 Mb, which were larger than the other P.ostreatus species (PC9 and PC10) and less than P.eryngii (KNR2312 P5). A total of 949 and 968 SSRs were found in the “Heuktari” and “Miso” genomes, respectively. A comparative analysis of five mushrooms including P.ostreatus var. florida (PC9 and PC15), P.eryngii, revealed that SSR numbers from “Heuktari” and “Miso” were the highest among them. Studied all mushrooms showed similar pattern of SSR distributions. Tri-, hexa- and octanucleotide motifs accounted for the top three fractions of all SSRs. <P><B>Abstract</B></P> <P> <I>Pleurotus ostreatus</I> is one of the most important edible mushrooms. Many cultivars have been bred to meet consumer needs. The identification of cultivars based on the morphological characteristics is restricted because fruiting bodies are frequently capricious due to environmental conditions; accordingly, sequence-based methods are required. A total of 546 simple sequence repeat primers derived from the <I>P. ostreatus</I> genome were screened, and one primer, JHH_SSR-184, was found to show polymorphisms on the major cultivars in Korea. The sequences of the polymorphic loci showed variable-number tandem repeat loci-like features enabling cultivar specificity. Thus, these loci might be applicable to discriminate <I>P. ostreatus</I> cultivars.</P> <P><B>Highlights</B></P> <P> <UL> <LI> The SSR marker discriminated the Korean major <I>Pleurotus ostreatus</I> cultivars. </LI> <LI> The cultivar specific sequences showed variable number tandem repeat loci-like feature. </LI> <LI> Nine loci with variable copy number of the repeat conferred the cultivar specificity. </LI> </UL> </P>
홍국색소의 항산화 활성 및 조골세포 분화에 미치는 영향

본 연구에서는 홍국색소의 항산화 활성 및 조골세포의 분화에 미치는 영향을 검토하였다. 홍국색소의 항산화 활성은 DPPH radical 소거능, ABTS radical 소거능 및 SOD 유사 활성을 측정하여 평가하였다. 홍국색소의 DPPH radical 및 ABTS radical 소거 활성은 농도 의존적으로 증가하였으며, 특히 1,000 μg/ml 농도에서 각각 94% 및 99%의 최대 활성을 나타내었다. 또한 SOD 유사 활성을 검토한 결과에서, 홍국색소 1,000 μg/ml 농도에서 62%의 최대 활성을 나타내었다. 우선 홍국색소 1~1,000 μg/ml의 농도에서 세포독성을 검토하기 위해 MTT assay를 실시한 결과, 1~100 μg/ml 농도의 범위에서 세포 독성은 나타나지 않았다. ALP 활성은 1~100 μg/ml 농도에서 농도의존적으로 증가하였으며, 100 μg/ml의 농도에서 최대 124%의 활성을 나타내었다. 또한, 석회화 형성능 시험에서도 대조군 대비 모든 농도에서 유의적으로 증가하는 경향이 나타났으며, 100 μg/ml의 농도에서 대조군 대비 1.5배의 석회화 형성 결과가 나타났다. 이러한 결과를 통해 홍국색소는 항산화 활성이 우수하며, 조골세포 분화에 긍정적인 영향을 줌으로써 골다공증 예방 소재로서의 개발 가능성을 가진 부가가치가 높은 천연물 소재로 활용 가능할 것으로 생각된다. The purpose of this study was to investigate antioxidant activity and cell differentiation effects of Monascus purpureus pigment on osteoblast-like MC3T3-E1 cell. In order to examine the antioxidant activities of Monascus purpureus pigment, DPPH radical scavenging, ABTS radical scavenging and SODlike activities were investigated. DPPH radical and ABTS radical scavenging activities of Monascus purpureus pigment were increased in a dose-dependent manner, and maximum activity were 94% and 99% at a concentration of 1,000 μg/ml, respectively. Additionally, SOD-like activity of Monascus purpureus pigment showed 62% at a concentration of 1,000 μg/ml. MC3T3-E1 cells did not show cytotoxicity in the concentration range of Monascus purpureus pigment 1~100 μg/ml. The ALP activity was increased by addition of Monascus purpureus pigment, and the maximum activity was 124% as compared with control. In addition, nodule formation, a late differentiation factor for bone formation, was increased by adding Monascus purpureus pigment compared to control. These results suggest that Monascus purpureus pigment is expected to be a natural source for developing functional materials to prevent bone-related diseases by osteoblast differentiation.
2023 IRA reports and case study for avocado fruit from Philippines

Division of the risk management, APQA(Animal and Plant Quarantine Agency) is conducting the IRA(Import Risk Assessment) to prevent the introduction of pests when importing foreign agricultural products. In the 2023 IRA, 55 cases of 19 items from 23 countries was analyzed, and fresh avocado(variety: HASS) from the Philippines was concluded in September 2023 and can be imported in accordance with appropriate phytosanitary measures. The main issue in the IRA of fresh avocado fruits from the Philippines was whether HASS variety avocados were host plants of Bactrocera dorsalis. As a result of the risk analysis, B. dorsalis were included in the pest list that needs to be managed by special risk management. As a risk management for B. dorsalis, one trap using methyl-eugenol is installed per 20 ha once a week. After surveying, if the population is more than 2/trap/day, control measures such as adding traps and spraying pesticides are necessary. As equivalence, regionalization, and transparency have been strengthened with the recent entry into force of the mega FTA, a higher level of scientific basis is required. Accordingly, division of the risk management, APQA is working harder to improve its expertise, and we need your cooperation and attention of the IRAs.
