        한강시민공원의 이용자 활동 특성 분석 및 개선방안 연구

        본 연구는 한강시민공원을 이용하는 시민들의 특성 및 이용행태를 분석하고 초기 조성방향과 실질적인 이용행태 간의 차이를 확인하여 향후 공원계획과 관리에 대한 시사점을 제시하는 것을 목적으로 둔다. 대상지별 분석 결과를 정리하면 첫째, 여의도한강공원은 주변 지역에 업무, 상업지구가 분포하고 문화 및 축제 프로그램이 많다는 특성을 가지고 있다. 그러나 이와 상관없이 인근 주민이나 업무, 상업 지구에 종사하는 사람들이 휴식이나 산책, 가벼운 운동 등을 위해 한강시민공원을 일상적으로 이용하는 것으로 분석되었다. 둘째, 반포한강공원은 주변이 주거지역이라는 특성을 가지고 있으며 한강르네상스 사업으로 시설이 조성되어있다. 그러나 시설 이용을 목적으로 하기보다 자전거 이용 중에 휴식을 취하기 위해 공원을 찾거나 휴식 및 산책 또는 가벼운 운동을 하는 행태가 나타났다. 본 연구는 이러한 결과를 바탕으로 한강시민공원의 실질적인 이용행태를 알 수 있으며 생활권 공원으로서의 한강공원의 활용도가 높았음을 확인하였다는 측면에서 의의를 가질 수 있다. Han-Gang Park, Seoul?s representative and iconic public places is dedicated to the role of the theme park. The purpose of this study is to compared the current user behavior and purpose of the composition in past, and derived the implications for planning and management for Han-Gang Park. The results of analysis target area shows that, firstly, Yeoido Han-Gang Park is surrounded by commercial and business district. That area has the many cultural, festival programs. But, without specific purpose, Yeoido Han-Gang Park has been used for resting or walking, and light exercise. And vicinity residents, office worker, or commercial area worker were using regularly. In case of Banpo, environs have a property of residential area and Banpos ? facilities were made by Han-Gang Renaissance Project. But most people use the park to take a rest in the way riding a bicycle rather than use facilities. On these results, we can recognize the practical usage behavior. And this study has significance that Han-Gang Park appeared to show higher utilization level of the citizens as a neighborhood space.

      • 한국인 좌심실 비대증 환자들에서 파브리병 선별검사의 의의

        목적: 파브리병(Fabry disease)은 alpha-galactosidase A의 결핍으로 인하여 리소좀에 globotriaosylceramide(Gb3)가 축적되어 여러 장기에 이상을 일으키는 질병이다. 본 연구에서는 파브리병의 만성 합병증 중 심장 질환을 주로 보이는 환자들, 그 중에서도 좌심실 비대증을 보이는 한국인 환자들을 대상으로 파브리병의 빈도를 알아보고자 하였다. 방법: 좌심실비대증을 진단받은 환자 257명을 연구대상으로 선정하였고, 남성이 172명(평균 56세, 범위 30-81세), 여성이 84명(평균 66세, 범위 45-85세)이었다. 파브리병 선별을 위하여 고성능액체크로마토그래피-탠덤질량분석기를 이용하여 소변 Gb3 농도를 측정하였다. 확진은 형광분석법에 의한 말초혈액의 alpha-galactosidase A 활성도와 염기서열분석법에 의한 GLA 유전자 돌연변이 유무를 검사하여 이루어졌다. 결과: 소변 Gb3 검사에서 cutoff (25 ug/mmoL creatinine)를 초과하는 환자는 4명이었지만, 최종적으로 추가 검사를 통해 진단된 파브리병 환자는 여성 환자 한 명이었다(1/257명, 0.4%). 확진된 환자는 54.3 ug/mmoL creatinine의 Gb3 농도와 15.5 nmole/hr/mg protein (참고범위, $55.2{\pm}12.7nmole/hr/mg$ protein)의 alpha-galactosidase A 활성도를 보였다. GLA 유전자에서는 c.796G>A (p.D266N) 돌연변이가 이형접합체로 관찰되었다. 추가로 시행한 가족검사에서 환자의 딸은 아직 파브리병의 증상을 보이지 않았지만, 엄마와 같은 GLA 돌연변이(c.796G>A)를 가지고 있었으며, alpha-galactosidaseA 활성도는 42.5 nmole/hr/mg protein, 소변 Gb3 농도는 25.5 ug/mmoL creatinine을 나타냈다. 결론: 한국인 좌심실 비대증을 가진 환자들에서 파브리병의 유병율은 0.4%였다. 유병율이 낮아 보임에도 불구하고, 파브리병 진단 전 환자와 가족 구성원을 발견할 수 있는 장점 덕분에 선별검사의 의의가 있는 것으로 사료된다. Objectives: Fabry disease (FD) is a lysosomal storage disease caused by the inappropriate accumulation of globotriaosylceramide (Gb3) in tissues due to a deficiency in the enzyme ${\alpha}$-galactosidase A. Hypertrophic cardiomyopathy is one of the chronic complications of FD. We tried to evaluate the prevalence of Fabry disease in the Korean patients with left ventricular hypertrophy (LVH). Methods: A total of 257 patients with LVH were recruited and they were 172 males (mean 56 years, range 30-81 years) and 84 females (mean 66 years, range 45-85 years). Urinary Gb3 was used to screen FD by high performance liquid chromatography-tandem mass spectrometry. Confirmatory tests were done by alpha-galactosidaseA activity using fluorometric assay and by GLA mutation analysis using sequencing. Results: Four patients were screening positive by urinary Gb3 analysis (cutoff, 25 ug/mmol creatinine). But, one female patient was diagnosed with FD confirmed by enzyme analysis in leukocytes as well as by genetic analysis (1/257 patients, 0.4%). She showed 54.3 ug/mmoL creatinine of Gb3 and 15.5 nmole/hr/mg protein (reference range, $55.2{\pm}12.7nmole/hr/mg$ protein) of alphagalactosidase A activity. And she had a heterozygous GLA mutation of c.796G>A (p.D266N). Her daughter was found to be a carrier for FD confirmed by GLA mutation analysis. Asymptomatic carrier showed 25.5ug/mmol creatinine of Gb3 and 42.5 nmole/hr/mg protein (reference range, $55.2{\pm}12.7nmole/hr/mg$ protein) of alpha-galactosidase A activity. Conclusions: The prevalence of FD in Koran patients with LVH was detected as 0.4%. Although the prevalence seems to be low, screening studies are of great importance for detecting hidden cases as well as for identifying other effected family members.

        <P>We found that beta-lapachone (beta-lap), a novel bioreductive drug, caused rapid apoptosis and clonogenic cell death in A549 human lung epithelial cancer cells in vitro in a dose-dependent manner. The clonogenic cell death caused by beta-lap could be significantly inhibited by dicoumarol, an inhibitor of NAD(P)H:quinone oxido-reductase (NQO1), and also by siRNA for NQO1, demonstrating that NQO1-induced bioreduction of beta-lap is an essential step in beta-lap-induced cell death. Irradiation of A549 cells with 4 Gy caused a long-lasting upregulation of NQO1, thereby increasing NQO1-mediated beta-lap-induced cell deaths. Although the direct cause of beta-lap-induced apoptosis is not yet clear, beta-lap treatment reduced the expression of p53 and NF-kappaB, whereas it increased cytochrome C release, caspase-3 activity, and gammaH2AX foci formation. Importantly, beta-lap treatment immediately after irradiation enhanced radiation-induced cell death, indicating that beta-lap sensitizes cancer cells to radiation, in addition to directly killing some of the cells. The growth of A549 tumors induced in immunocompromised mice could be markedly suppressed by local radiation therapy when followed by beta-lap treatment. This is the first study to demonstrate that combined radiotherapy and beta-lap treatment can have a significant effect on human tumor xenografts.</P>

        자궁 내 감염 관련 유전자 및 산화 스트레스 관련 유전자와 조기분만의 연관성

        Objective: To investigate the association between preterm birth and cytokine genes (IFN-γ, interleukin (IL)-10) in intrauterine infection and enzyme gene (CYP1A1) in oxidative stress response. Methods: This study involved a case-control study conducted at Ewha Womans University Hospital in Seoul, Korea. Subjects with preterm deliveries (<37 weeks of gestation) and normal controls with term deliveries (≥37 weeks of gestation) were selected from gravidas who had undergone prenatal examinations in the hospital and were followed until infant delivery. The weight, height, and blood samples of each participants were obtained according to standard protocols. We included subjects who gave birth to a singleton infant and had a gestational age between 24 and 42 weeks. Mutiple births, stillbirths, and congenital anomalies were excluded. Finally, 164 gravidas with preterm births and 305 normal controls with term deliveries were enrolled in the present study. Results: Preterm delivery group and term delivery group had significant difference in gestational age and neonatal body weight (P<0.0001). There were no statistically significant association between preterm birth and IFN-γ, IL-10, CYP1A1 genes (P>0.05). Conclusion: In this study, IFN-γ (874A/T), IL-10 (1082A/G), IL-10 (819C/T), IL-10 (592A/C) and CYP1A1 (T6235C), CYP1A1 (Ile462val [A/G]) genes had no significant association with preterm birth.

      • Calcium Mobilization through L-type Channels in Hepatic Stellate Cell Is Essential for TGF-b-mediated CTGF

        Aims: In hepatic fibrogenesis hepatic stellate cell (HSC) is a major cell type responsible for producing a major profibrogenic cytokine TGF-b, and connective tissue growth factor (CTGF), a major fibrogenic mediator in several organs. The multi-functional nature of TGF-b signaling in hepatic fibrogenesis is still elusive. At the previous The Liver Week(2015) we reported that Pyk2 is essential for TGF-b-mediated, Smad-independent CTGF induction. Pyk2 is known to be calcium- sensitive, and TGF-b was reported to increase intracellular calcium level. Therefore, we investigated the relation between intracellular calcium levels and pro-fibrogenic TGF-b/Pyk2 axis in hepatic stellate cell. Methods: Immortalized human stellate cell line, LX-2, has been cultured. After TGF-b treatment, expression of CTGF and a-SMA were assessed with RT-PCR and western blot. Pharmacological inhibitor and siRNA-mediated knockdown were used to modulate the activities and expression levels of protein. Intracelllular calcium mobilization was measured with Fura-4/AM. Activation of Pyk2 was addressed in western blot using different phosphorylation site-specific antibodies. Results: CTGF expression was up-regulated within 1hr in TGF-b stimulated LX-2. This up-regulation was greatly suppressed by siRNA-mediated knockdown and pharmacological inhibitor of Pyk2. TGF-b treatment increased phosphorylation of Pyk2 on tyrosine 402, 579/580, and 881. Consistent with the previous reports, TGF-b increased intracellular calcium concentration in Fura-4-preloaded LX-2. CTGF induction by TGF-b was blocked in dose dependent manner by pre-treatment with BAPTA-AM, an intracellular calcium chelator, while A23187, a calcium ionopore, increased CTGF induction even in the absence of TGF-b, suggesting that increase of intracellular calcium level is enough to induce CTGF expression. In addition, A23187 increased phosphorylation of Pyk2, and CTGF induction by A23187 is also greatly reduced by siRNA of Pyk2. Pre-treament of LX-2 with Nifedipine (an L-type calcium channel blocker) suppressed CTGF induction by TGF-b in dose-dependent manner, while FPL64176 (L-type channel activator) increased CTGF expression without TGF-b. The CTGF up-regulations by TGF-b, A23187, and FPL64176 were all suppressed by siRNA-mediated knockdown and pharmacological inhibitors of Pyk2. Conclusions: In hepatic stellate cell, TGF-b increases the intracellular calcium level through L-type calcium channel, leading to downstream Pyk2 signaling for CTGF induction.

        Clostridium difficile에 의한 설사의 예후인자

        배경 : Clostridium difficile에 의한 설사(Clostridium difficile associated diarrhea; CDAD)는 임상 경과가 다양하다. 최근에 유럽과 북미에서 C. difficile 감염의 발생률이 증가하고 고전적 치료에 잘 반응하지 않으며 이환률이 증가하였고, 이러한 원인이 새로운 균주의 탄생에 기인한다고 보고되었다. CDAD의 예후에 영향을 미치는 세균성 요인과 숙주 요인을 확인하기 위해 본 연구를 진행하였다. 재료 및 방법 : 2002년 8월부터 2003년 12월까지 CDAD가 진단된 20세 이상인 환자들을 대상으로 후향적 Cohort 연구를 하였다. 세균성 요인을 확인하기 위해 cdt A, cdtB, tcd A, tcd A rep 그리고 tcd B 유전자(binary toxin)를 확인하였다. 설사가 치료 시작 후 11일 이상 지속되거나, 2달 이내에 재발하거나, 수술 혹은 다른 시술이 필요한 경우, 사망한 경우 예후가 좋지 않은 것으로 정의하였다. 결과 : 총 115예에서 toxin을 생성하는 C. difficile가 동정되었으며, Toxin A와 toxin B 모두 양성인 균이 91예, toxin B만 양성인 균이 24예였다. Toxin A 생성 여부가 예후에 영향을 미치지 않았지만, 제산제를 사용한 환자에서 그렇지 않은 환자에 비해 toxin B만 양성인 균이 많이 동정되었다(P<0.05). 예후가 좋지 않은 경우는 39예(33.9%)였고 76예(66.1%)에서 예후는 양호하였다. 단변량 분석에서 70세 이상의 고령, 남성, 증상 발현 후 사용한 항생제의 개수 사용, 증상 발현 후 carbapenem, aminoglycoside, glycopeptide 사용, 당뇨 및 뇌졸중 병력이 있는 경우, 그리고 높은 Charlson index가 불량한 예후 인자로 확인되었다. 그러나 독립적인 예후 인자를 조사했을 때에는70세 이상의 고령(odds ratio=3.378, P=0.009), 증상 발현후 carbapenem 사용(odds ratio 7.210, P<0.001)이 예후에 중요한 영향을 미치는 요소로 확인되었다. 결론 : 70세 이상의 고령과 증상 발현 후 carbapenem 사용이 CDAD 독립적인 불량한 예후인자이다. Background : Clostridium difficile associated diarrhea (CDAD) has a wide range of clinical manifestations. The prognostic factors of CDAD are not fully understood. Materialsand Methods : A retrospective cohort study of 115 patients with CDAD from Aug. 2002 to Dec. 2003 was conducted to evaluate prognostic factors of CDAD. Bacteriologic factors were determined by detecting the binary toxin gene, tcd A, tcd A rep and tcd B gene. Poor prognosis was defined as diarrhea more than 10 days even with classic treatment, recurrence, death, and moribund discharge. Results : Approximately 79% of isolated strains were toxin A+/B+ strains and 21% were toxin A-/B+ strains. There was no difference in prognosis between toxin A+ and toxin A- strains. 39 (33.9%) cases showed poor prognosis and 76 (66.1%) cases showed good prognosis. Univariate analyses revealed that the poor prognostic factors were old age over 70 years old, male, the number of antibiotics used after onset of symptom, the administration of carbapenems, aminoglycosides, glycopeptides after onset of symptom, history of DM and stroke, and high Charlson comorbidity index. Multiple logistic regression analysis identified old age over 70 years old (odds ratio=3.378, P=0.009) and the administration of carbapenems after onset of symptom (odds ratio 7.210, P<0.001) as the independent poor prognostic factors. Conclusion : Old age over 70 and the administration of carbapenems after onset of symptom were the poor prognostic factors for CDAD caused by none-binary toxin producing strains.

        중합효소연쇄반응을 통해 확진되고 Atovaquone과 Azithromycin으로 치료한 Human Babesiosis 1예

        저자들은 국내에서 처음으로 중합효소연쇄반응 검사를 통하여 B. microti에 의한 인체 바베시오시를 진단하였으며, atovaquone과 azithromycin 병합요법을 사용하여 치유한 1예를 경험하였기에 문헌고찰과 함께 보고하는 바이다. Human babesiosis is a tick-borne infectious disease caused by Babesia species. The clinical diagnosis is difficult because of nonspecific symptoms like flu. Rapid diagnosis of human babesiosis is microscopic examination in peripheral blood smear (Giemsa-stain) which reveals characteristic forms of an intracellular quadruplet parasite. But differentiation between Babesia microti and Plasmodium species can be quite difficult because of the morphologic similarity. We experienced a case of human babesiosis. The patient was a 62-year old Korean male who had been in New Jersey, U.S.A for 2 months. We initially diagnosed as malaria infection because the peripheral blood smear revealed intracellular single ring form organism. But the patient was not improved significantly by the treatment with chloroquine regimen. Finally we confirmed human babesiosis by polymerase chain reaction for Babesia microti. We treated the patient successfully with a regimen of atovaquone and azithromycin which has fewer adverse reactions than a regimen of clindamycin and quinine.

Plant Sci. 94: 73-83. Pectin is one of the main components of plant cell walls, and its biosynthesis is controlled by pectin methylesterase (PME). Pectin methylesterase inhibitors (PMEIs) are key regulators of PME. We report here the cloning and characterization of a novel Solanum lycopersicum L. PMEI gene, SlPMEI. RT-PCR studies of leaf, seed, fruit, flower, and flower organs confirmed that SlPMEI is expressed specifically in pollen. Promoter analysis of SlPMEI revealed pollen-specific cis-acting elements (pollen lat52 and g10). In addition, SlPMEI is expressed independently of abiotic stress, pathogen exposure, and growth stage in tomato, and a histochemical analysis of promoter activity revealed pollen-specific expression in both Arabidopsis and tomato. Under the microscope, we observed pollen-specific GUS expression in the stamen of transgenic tomato plant. These results indicate that the promoter of SlPMEI has strong pollen-specific activity, and could therefore be useful for development of industrially and agronomically important transgenic plants. </P>

        The Role of Plant Extracts in Alleviating Particulate Matter-induced Inflammation in Barrier-interrupted Skin

        Background: The adverse effects of particulate matter (PM) on the skin have been concerning recently. Objective: We investigated PM induced inflammation on barrier-interrupted skin and relieving effects of plant extracts on PM-induced skin irritation. Methods: A total of 20 participants were enrolled for the induction of skin irritation. Sodium lauryl sulfate (SLS) patch was affixed to the forearm for 24 hours. After patch removal, total suspended particulates (TSPs) were applied. After inducing skin irritation using SLS and TSP, cream containing 0.1% mixture of plant extracts (Citrus sunki, Sceptridium ternatum (Botrychium ternatum), and Korthalsella japonica) was applied. The a* value, erythema index (EI), and transepidermal water loss (TEWL) were measured. Results: The a* value was significantly higher at the site where TSPs were applied than that at the site where TSPs were not applied at 1 and 2 days after SLS patch removal. The a* value, EI, and TEWL were significantly low in the cream-applied area 7 days after the introduction of the SLS patch and TSPs. Conclusion: PM induced skin irritation in a previously disrupted skin barrier. Topical application of plant extracts alleviated skin irritation symptoms caused by dust exposure.         최근에 식물추출물을 이용하는 병해충관리용 유기농업자재의 생산이 증가하면서 유기농업자재의 제조 개발이 활발하게 이루어지고 있다. 황련(Coptis)은 항균 효과, 제초 효과, 고추역병 방제, 배추좀나방 방제, 오이횐가루병 등 다양한 살충, 살균 효과가 있어 유기농업자재의 원료로 사용되고 있다. 이에 따라 황련 추출물로 이루어진 유기농업자재의 품질관리 및 효과적 사용을 위해 유효성분 확인 및 함량 분석법 개발에 대한 농업인, 농자재 생산사의 요구가 계속되고 있어 주성분 설정 및 분석법 개발이 필요하다. 본 시험에서는 황련의 주성분을 berberine, coptisine, palmatine으로 설정하고 정량분석을 위한 전처리법을 확립하고 제품 중 주성분의 함량을 분석하였다. 시료는 한 약재 황련을 주정과 ethanol로 추출하여 농축한 추출과 황련 추출물을 함유한 병해충관리용 유기 농업자재 시판제품 4종을 대상으로 하였다. 주성분의 함량을 HLB cartridge 60mg 3cc를 이용하여 정제하여 syringe filter(0.45um, PTFE)를 이용하여 거른 후 HPLC-UVD로 분석하였다. Column은 Luna 5u C18 (250*4.60mm 5micron), UV검줄파장은 346nm, 이동상은 용매A 0.1% Acetic acid in DW, 용매B 0.1% Acetic acid in ACN을 사용하여 gradient system으로 분석하였다. 그 결과 berberine, coptisine, palmatine의 표준품 검량선은 0.2~10 mg kg<sup>-1</sup> 범위에서 0.999이상의 직선성 (R2)을 보였다. Coptisine의 회수율은 89.7~102.1%, Palmatine 87.1-94.1%, Berberine 86.3~97.5% 로 확인할 수 있었다. 황련의 EtOH 추출액의 함량은 3개 주성분의 합으로 3.6~8.95%가 나왔고, 황련이 포함된 유기농업자재 제품의 함량 분석 결과는 제품 A~D에서는 0.037~0.582%이었다. 이 실험에서 확립된 분석법은 황련 추출물과 황련 함유 유기농업자재 중 berberine, coptisine, palmatine의 함량분석에 활용할 수 있을 것으로 기대된다.
