국내 식품으로부터 분리한 Listeria Species의 RAPD 분석

Randomly Amplified Polymorphic DNA(RAPD) 기술을 이용하여 국내 식품으로부터 분리한 Listeria sp. 분리균주에 대한 DNA polymorphism을 분석하고 유연관계를 비교하며, 유용 marker를 개발할 목적으로 10가지 10-mer primer를 이용하여 PCR을 수행한 결과 5개의 primer(OPA-01, OP-26-01, OP-26-02, OPB-01, OP-26-10)가 선별되었고, 76개의 DNA 단편이 증폭되었다. 이 중 OPA-01과 OP-26-10 primer에 의한 약 1.5 kb와 0.7 kb의 증폭 band는 모든 Listeria 분리균에서 관찰할 수 있었으나, 이 증폭된 DNA 단편은 Listeria sp.에만 특이적인 것은 아니었다. NTSYS 프로그램을 이용해서 Listeria sp. 분리균간의 유전적 유연관계를 알아본 결과 7개의 cluster로 나뉘어졌고 유사도는 대체로 0.54~0.93사이었으며, 특히, No.3과 No. 20은 93%로 가장 높은 유사도를 나타내었고, No. 7과 No. 24 또는 No. 7과 No. 45는 54%로 가장 낮은 유사도를 나타내었다. 이러한 결과는 RAPD 기술을 이용하여 쉽게 Listeria sp.를 subspecies로 분류할 수 있음을 시사하였다. This study was carried out for comparing Listeria strains developing genetic markers for Listeria strains using Listeria sp. genetic markers using Randomly Amplified Polymorphic DNA (RAPD) analysis method. Five of 10 RAPD primers (OPA-01, OP-26-01, OP-26-02, OPB-01, OP-26-10) showed the distinctive polymorphism among Listeria sp. isolated from domestic foods. RAPD-PCR with five arbitrary primers produced 76 DNA polymorphism. Among them, OPA-01 and OP-26-10 primers produced about 1.5 kb and 0.7 kb amplified DNA fragments for all the Listeria sp., but these amplified DNA fragment did not correspond to specific Listeria genus. The genetic relationships of Listeria sp. using NTSYS program were grouped into 7 clusters and showed 0.54 to 0.93 similarity among strains. Especially, No. 3 and No. 20 isolates showed genetically most similar relationship by 0.94, and No. 7 and No. 24, or No. 7 and No. 45 isolates showed the least similarity by 0.54. From these results, RAPD analysis method seemed to be successfully applied the classification and genetic analysis for Listeria sp. isolates.
인체 폐암종에서 Intercellular Adhesion Molecule - 1 의 발현에 관한 연구

Background: Intercellular adhesion molecule-1 (ICAM-1) is a 90 kD surface glycoprotein, that serves as cell-cell and cell-substratum adhesion molecules and helps to regulate cellular morphology, differentiation, and proliferation. The adhesion molecules likely play important roles in maintaining the normal structure and function of the lung, as well as participating in the development and spread of malignant lung diseases. ICAM-1 system among many cell adhesion molecules is importantly issuing in the pathogenesis of the tumor cell invasion or metastasis of lung cancer. Methods: By using IgG, monoclonal antibody for ICAM-1, we investigated the expression of ICAM-1 immunohistochemically in the formalin-fixed, paraffin-embedded tissue sections of the normal and lung cancers. Results: In the normal lung tissue sections, the expression of ICAM-1 was not discernible. Up-regulation of the ICAM-1 expression was shown on the surface of tumor cells of lung cancers in 11 cases of 13 primary lung cancers (84.6%). ICAM-1 was strongly positive depending on cell types of lung cancer, adenocarcinoma, squamous ce11 carcinoma, small cell carcinoma in the order of degree of expression but in the large cell carcinoma, the expression of ICAM-1 was not noted. Conclusion: It was concluded from these findings that up-regulation of the ICAM-1 expression may be related to pathogenesis of lung cancer.
HlyA 유전자 Primer를 이용한 PCR에 의한 식품으로부터 Listeria monocytogenes의 신속 검출 방법

본 연구는 식품에 존재하는 L. monocytogenes균의 신속한 검출방법을 조사하기 위하여 hlyA 유전자 primer를 사용하여 PCR 기법으로 행하였으며 primer의 특이성과 PCR의 민감성, L. monocytogenes균의 검출을 위한 최적조건 및 우유 및 소고기의 적용시험을 각각 조사하였다. PCR 특이성 실험에서는 L. monocytogenes 20주에 대한 PCR 분석결과 모두 713 bp 크기의 PCR products를 확인할 수 있었고, Listeria spp. 및 다른 세균에서는 동일한 poducts가 증폭되지 않으므로써 hlyA based primer의 L. monocytogenes에 대한 PCR 특이성이 관찰되었다. PCR 분석법의 민감도는 L. monocytogenes ATCC 19111의 1 pg DNA와 2.4×10⁴ cell 수준에서 target DNA가 증폭되었다. Tailing의 제거와 민감도를 높이기 위하여 PCR cycle 수에 따른 최적조건을 조사한 결과 20~30cycle 정도의 PCR clcyle 수가 좋은 것으로 나타났으며 민감도는 20 cycle PCR amplification을 행한 후 한번더 10~15 cycle로 처리했을 때 훨씬 증가하였다. 한편, 우유(10 mL)와 쇠고기 절편(10 g)에 0~10^7 CFU/mL 또는 g 수준으로 L. monocytogenes를 신속하게 검출하기 위하여 PCR을 적용한 결과, 우유시료에서는 2번 PCR을 반복함으로써 2 cells까지 검출할 수 있었고, 쇠고기 절편은 LEB로 35℃에서 24시간 증균하여 PCR함으로써 2.6×10² cell까지 검출할 수 있었다. The study was conducted to develop a rapid method for the detection of Listeria monocytogenes in foods via polymerase chain reaction (PCR) technique using hemolysin gene (hlyA) primers. Specificity and sensitivity of PCR, optimal conditions for PCR and application of hlyA gene primers for the detection of L. monocytogenes from milk and beef were investigeted. Each of the 20 L. monocytogenes strains gave a single 713 bp band, but other Listeria spp. and other bacteria did not show any bands. As few as 1 pg of L. monocytogenes DNA or 2.4×10⁴ L. monocytogenes cells could be detected with hlyA gene primers. PCR product was most improved at 20~30 cycle in terms of removal of tailing and sensitivity. Also, the sensitivity was significantly improved by the further 10~15 cycle after 20 cycle PCR amplication. Milk (10 mL) and beef (10 g) samples were inoculated with L. monocytogenes at the concentrations ranging from 0 to 10^7 CFU/mL or g to determine the best sensitivity of PCR for the rapid detection of L. monocytogenes. PCR assay could detect 2 cells in milk with repeating PCR amplication and 2.6×10² cells in beef sample after 24 hr enrichment growth at 35℃ in LEB.
성인의 T - lymphoblastic Lymphoma 의 임상상과 Modified LSA2 - L2 화학요법의 치료성적

N/A Adult T-lymphoblastic lymphoma was first described by Barcos and Lukes in 1974. It is famous for early systemic metastasis and should be treated systematically as soon as possible. We diagnosed 8 adult T-lymphoblasic lymphomas by means of clinical manifestation, tissue biopsy and their immunophenotyping from May 1986 to September 1988. Seven cases were treated with an aggressive regimen, the modified LSA2-L2 protocol. The results were as follows: 1) The median age in the group was 24 years (range 17 -35). 2) At diagnosis, all patients had mediastinal involvement, and 5 patients had bone marrow involvement. Three patients demonstrated malignant cells in the peripheral blood. 3) Six patients were positive for Tdt stain and 4 patients were CD7 (+). 4) Complete clinical response was attained in all patients with induction therapy including cytoxan, vincristine, and adriamycin. 5) Two complete responders who did not take the prescribed chemotherapeutic medication and CNS prophylaxis have had relapses in the CNS and testis. In summary, adult T-lymphoblastie lymphoma is a very aggressive and systemic disease. Systemic chemotherapy with CNS prophylaxis, such as the modified LSA2-L2 protocol, should be recommended.

N/A Behcet's disease is known as a multisystemic disease caused by nonspecific vasculitis of obscure origin. Among the systemic manifestations, pulmonary involvement is know to be very rare and only a few cases have been documented. The most important features of pulmonary lesions in Behcet's disease are the fleeting nature of the pulmonary infiltrate and recurrent hemoptysis, which is often massive and fatal. We report a case of Behcet's disease manifested as multiple pulmonary nodules which was caused by pulmonary infarction as a result of pulmonary arteritis.

The simultaneous association of a gastric carcinoid tumor with adenocarcinoma as a synchronous dual malignancy of the stomach in a same patient is extremely rare. The carcinoid syndrome is an endocrine manifestation of neoplastic enterochromaffin cells. The humoral consequences of entero- chromaffin carcinoid tumors include facial flushing, diarrhea, bronchoconstriction, and so on. Thus classical functional carcinoid syndrome, however, is surprisingly infrequent in gastric lesions. Therefore, the precise diagnosis of carcinoid tumor of the stomach is occasionally made by the histopathologic demonstration of argyrophilic granules in tumor cell cytoplasm. We report, hereby, two cases of the coincidence of carcinoid tumor and adenocarcinoma of the stomach, and review the literatures.

In the normal the expression of ICAM-1 was not discernible. Up-regulation of the ICAM-1 expression was showed on the surface of tumor cells of lung cancers in 11 cases of 13 primary lung cancers (84.6%).
