음양곽과 $\alpha$ -MSH에 의한 B16 Melanoma 세포의 멜라닌화와 Retinoic Acid의 억제 효과

음양곽 물추출액과 $\alpha$-melanocyte stimulating hormone ($\alpha$-MSH)에 의한 B16 melanoma 세포의 멜라닌화를 비교 조사하였으며, retinoic acid (RA)에 의한 억제효과도 연구하였다. B16 melanoma 세포(1×$10^5$개 정도)를 $\alpha$-MSH로 처리하고 72시간 배양한 결과 $\alpha$-MSH 처리농도에 비례하여 tyrosinase 활성도와 melanin 생성이 증가되었으며, 8 ng/mL 처리시 tyrosinase 활성도는 350%, melanin 함량은 290% 이상이었다. 음양곽 물추출액으로 처리한 경우에는, 100 ${\mu}g$/mL 처리시 tyrosinase 활성도는 200%, melanin 함량은 180% 이상이었다. 위 조건에 RA를 가하면, $\alpha$-MSH의 경우 tyrosinase 활성도는 350%에서 210%로, melanin 함량은 290%에서 250%로 억제되었다. 음양곽 물추출물의 경우 tyrosinase 활성도는 200%에서 100%로, melanin 함량은 180%에서 120%로 억제되었다. 이 결과는 음양곽 물추출액이 cAMP 신호전달 경로를 통하여 B16 melanoma 세포의 멜라닌화를 촉진하며, RA는 그것을 억제하는 것으로 해석된다. Melanization of B16 melanoma cells was comparatively studied by the aqueous extract of Epimedium koreanum Nakai(EK) and $\alpha$-MSH. In addition, inhibitory effects of RA was investigated. B16 melanoma cells(about 1${\times}10_5$) have been shown an increase in tyrosinase activity and melanin contents in proportion to concentration of $\alpha$-MSH when treated with $\alpha$-MSH and incubated for 72 hrs. They indicated a 350% increase in tyrosinase activity and a 290% increase in melanin contents at 8 ng/mL. In case of EK, they have been shown a 200% increase in tyrosinase activity and a 180% increase in melanin contents at 100 ${\mu}g$/mL. In addition of RA to the above condition, they have been shown an inhibition from 350% to 210% in tyrosinase activity and from 290% to 250% in melanin contents in $\alpha$-MSH, and inhibition from 200% to 100% in tyrosinase activity and from 180% to 120% in melanin contents in EK. From the above results, it is suggested that EK promotes melanization of B16 melanoma cells through cAMP pathway, whereas RA inhibits it.

Antifungal activities of the extracts from 26 medicinal plants were investigated utilizing paper-disk diffusion method and (1,3)β-glucan synthase inhibitory assay. (1,3)β-glucan synthase is considered as valuable target in the development of antifungal agents. Among the screened extracts, the ethyl acetate extract of Equisetum arvense, the ethyl acetate extract of Polygonum aviculare, the butanol extract of Crataegus pinnatifida and the n-hexane extract of Saussurea lappa showed significant antifungal activities on Candida albacans in both disk diffusion and enzyme assays.

Quercetin is one of the bioflavonoid compounds and has multiple biological effects such as antioxidant and effective anti-inflammatory agent. Melanin has an important role in protecting human skin from the damaging effects of ultra-violet (UV) radiation. We studied the effect of quercetin on proliferation of B16/F10 melanoma cells. After 48h treatment of cells with quercetin, the cells exhibited a dose- dependent inhibition in their proliferation without apoptosis. Therefore, the decrease in cell numbers may be due to cell growth arrest, not due to cell death by cytotoxiciy. We also investigated the effect of quercetin on melanogenesis of this cells. B16/F10 melanoma cells were grown for 48h in the presence of 0.01∼50μg/ml Quercetin and the total melanin contents were measured. Quercetin stimulated melanization of the cells in low concentrations (0.01-20μg/ml), whereas it inhibited melanization in high concentrations (30-50 μg/ml). It was observed that quercetin differently regulates melanogenesis of B16/F10 melanoma cells dependent on its concentrations.

Caesalpinia sappan has lung been comnlonly used as emmenagogue, analgesic, and cure for thrombosis in the Oriental medicine. The main constituent of C. sappan Sprain as well as a remedy for thrombusis in the Oriental medicine. The main constituent of C. sappan is brazileln, which is an antioxidative substance that has a flavonoid structure. In this study, we examined the effect of butanol extract of C. sappan on proliferation and melanogenesis in B16/F10 melanoma cells. After 48h treatment of cells with various concentrationls of butanol extract, the cells exhibited a dose-dependent inhibition in their proliferation without apotosis. There fore, the growth retardation by the extract may be due tho the cell arrest, not due tho the cell death induced by cytotoxicity. We also estimated total melanin contents as a final product and activity of tyrosinase, a key enzyme, in melanogenesis of B16/F10 melanoma cells. Our result showed that tole me]anin contents and tyroslnase activity were decreased in butanol extract-treated cell in a dose depndent manner compared to control group. In conclusion, it was observed tllat butanol extract of C. sappan inhibited melanization of these cells and therefore butanol extract could be devoir)ped as skin whitening components of cosmetics.
느릅나무의 에틸 아세테이트 추출물에 의한 Melanin생성 효과

Melanogenesis is a physiological process resulted in the synthesis of melanin pigments, which has a role in protecting skin from the damaging effect of ultra-violet (UV)radiation. The main aim of the present study was to examine the effect of Ulmus davidiana var. japonica (UL) on melanogenesis. Cells were cultured in the presence of various concentrations of Ulmus davidiana var. japonica For 48 h, and there were estimated total melanin contents as a final product and activity tyrosinase, a key enzyme, in melanogenesis. Among the four solvent extracts tested, EtOAc extract mostly increased tyrosinase activity And EtOAc extract increased the melanin contents and tyrosinase activity in a dose-dependent manner, Especially it was observed that 100mcg/ml/ EtOAc extract promotes melanin secretion in B16/F10 melanoma cells by 140% at 48 h treatment and activity of increased by 180% in the presence of same concentration. In conclusion, as for EtOAc extract treatment, there was no effect on the viability of B16/F10 cell, only to stimulate melanization.
