        국내 식품 중 구이, 찜, 볶음, 조림에 존재하는 수용성 비타민 B<sub>1</sub>, B<sub>2</sub> 그리고 B<sub>3</sub> 함량 조사

        A conventional Korean meal typically includes various roasted, steamed, stir-fried, and braised foods. For this study, we investigated the contents of water soluble vitamins, $B_1$ (thiamin), $B_2$ (riboflavin) and $B_3$ (niacin) in various roasted, steamed, stir-fried, and braised foods. Method validation for analytical data in this study showed a high linearity ($r^2$>0.999), and the limit of detection and quantification were 0.001-0.067 and $0.002-0.203{\mu}g/mL$, respectively. For accuracy and precision, analytical values using standard reference materials were in the certified ranges. Roasted foods contained 0.039-1.057 mg/100 g of thiamin, 0.058-0.686 mg/100 g of riboflavin and 0.021-21.772 mg/100 g of niacin. Steamed foods contained 0.049-1.066 mg/100 g of thiamin, 0.025-0.548 mg/100 g of riboflavin and 0.134-21.509 mg/100 g of niacin. Stir-fried foods contained 0.114-0.388 mg/100 g of thiamin, 0.014-1.258 mg/100 g of riboflavin and 0.015-2.319 mg/100 g of niacin. Braised foods contained 0.112-1.656 mg/100 g of thiamin, 0.024-0.298 mg/100 g of riboflavin and 0.322-2.157 mg/100 g of niacin. The data on water-soluble vitamins in this study can be used for a nutritional database of conventional Korean meals. 본 연구는 국내 식생활의 기본적인 구성식단으로 알려진 구이, 찜, 볶음, 조림에 존재하는 다양한 영양성분 중 수용성 비타민인 $B_1$ (thiamin), $B_2$ (riboflavin), 그리고 $B_3$ (niacin)의 함량을 확인하였다. 실험분석 타당성 검증을 통해서 높은 직선성($r^2$>0.997)을 확인하였고, 검출한계의 경우 $0.001-0.067{\mu}g/mL$ 그리고 정량한계의 경우 $0.002-0.203{\mu}g/mL$를 확인하였다. 또한 실험값에 대한 정밀도와 반복성에 대한 검증을 위해서 standard reference materials를 통해 실험값과 표준값과의 오차가 신뢰도 이내에 존재한다는 결과를 확인하였다. 본 연구에서 제시된 구이식품의 경우 thiamin은 0.039-1.057 mg/100 g의 범위에 존재하였고, riboflavin은 0.058-0.686 mg/100 g의 범위에 존재했으며, niacin의 경우 0.021-21.772 mg/100 g 의 범위로 나타났다. 찜 종류의 경우 thiamin은 0.049-1.066 mg/100 g의 범위에 존재하였고, riboflavin은 0.025-0.548 mg/100 g의 범위에 존재했으며, niacin의 경우 0.134-21.509 mg/100 g의 범위로 나타났다. 볶음의 경우 thiamin은 0.114-0.388 mg/100 g의 범위에 존재하였고, riboflavin은 0.014-1.258 mg/100 g의 범위에 존재했으며, niacin의 경우 0.015-2.319 mg/100 g의 범위로 나타났다. 조림의 경우 thiamin은 0.112-1.656 mg/100 g의 범위에 존재하였고, riboflavin은 0.024-0.298 mg/100 g의 범위에 존재했으며, niacin의 경우 0.322-2.157 mg/100 g의 범위로 나타났다. 본 연구에서 제시된 구이, 찜, 볶음, 그리고 조림 식품에 대한 수용성 비타민 함량에 대한 영양성분 데이터베이스 구축 연구가 국민 식생활의 중요한 기초자료로 이용될 것으로 확신한다.

        재생 중인 흰쥐 간의 형태학적 변화 및 PCNA 발현에 미치는 rrhGM-CSF의 영향

        Granulocyte macrophage-colony stimulating factor (GM-CSF)는 과립구 및 대식세포뿐만 아니라 상피세포의 증식과 분화를 자극하는 당단백질이며, 최근 생산된 벼세포 유래 재조합 GMCSF(rrhGM-CSF)는 감염원으로부터 안전하고 당사슬이 매우 풍부하여 물질의 안정성 혹은 효과의 지속성을 높여 주는 것으로 알려졌다. 본 실험에서는 간 재생 능력이 우수한 흰쥐를 실험모델로 하여 간의 78%를 제거한 후, 간 재생을 유도하는 과정에서 rrhGM-CSF를 처리하고, 시간 경과에 따라 형태변화의 차이와 더불어 단백질 발현 분석법과 면역조직화학법을 이용하여 PCNA 발현에 미치는 효과에 대해서 알아보고자 하였다. rrhGMCSF는 간 재생 속도를 뚜렷이 증가시키지는 못하였으나, 대조군에 비해 실험군에서는 재생 초기에 간 세포판의 붕괴와 재구성 시기를 다소 앞당기는 것으로 관찰되었다. 증식 중인 세포에서 증가하는 것으로 알려져 있는 핵단백질인 proliferating cell nuclear antigen (PCNA)의 간 조직에서의 분포와 발현 정도를 보면 부분간절제 후 12시간과 24시간에서는 PCNA 단백질이 두 그룹에서 조금씩 발현되다가 간 절제 3일과 5일이 경과한 실험군에서 단백질이 높게 발현되었다. 간 재생이 진행될수록 간 조직 전체에서 고르게 PCNA 양성반응이 나타났으며, 대조군 보다 실험군에서 반응성이 더 뚜렷한 것으로 나타났다. 이러한 결과들로 보아 부분 간절제 후 간 재생을 유도하는 과정에서 rrhGM-CSF가 세포분열을 촉진시키는 인자들 중의 하나로 작용하여 간 재생에 효과를 나타낼 수 있을 것으로 사료된다. Liver regeneration is a result of highly coordinated proliferation of hepatocytes and nonparenchymal liver cells. Partial hepatectomy (PH) is the most often used stimulus to study liver regeneration because, compared with other methods that use hepatic toxins, it is not associated with the tissue injury and inflammation, and the initiation of the regenerative stimulus is precisely defined. Granulocyte macrophage-colony stimulating factor (GM-CSF), which is a cytokine able to regulate the proliferation and differentiation of epithelial cells, was first identified as the most potent mitogen for bone marrow. Particularly, rrhGM-CSF, which is highly glycosylated and sustained longer than any other types of GM-CSF in the blood circulation, was specifically produced from rice cell culture. In this experiment, effects of rrhGM-CSF administration were evaluated in the regenerating liver after 78% PH of rats. Morphological changes induced by PH were characterized by destroyed hepatocyte plate around the central vein and enlarged nuclear cytoplasmic ratio and increased hepatocytes with two nuclei. And then, proliferation of liver cells (parenchymal and nonparenchymal) and rearrangement of plates and lobules seemed to be carried out during liver regeneration. These alterations in the experimental group preceded those of the control. Since proliferating cell nuclear antigen (PCNA) is known to be a nuclear protein maximally elevated in the S phase of proliferating cells, the protein was used as a marker of liver regeneration after PH in rats. PCNA levels by western blot analysis and immunohistology were compared between the two groups. PCNA protein expression of two groups at 12 hr and 24 hr after injury showed similar pattern. The protein expression showed the peak at 3 days in both groups, however, the protein level of the experimental group was higher than that of the control. On immunohistochemical observations, the reaction product of PCNA was localized at the nuclei of proliferating cells and the positive reaction in experimental group at 3 days was clearly stronger than that in control group. The results by Western blotting and immunohistology for PCNA showed similar pattern in terms of the protein levels. In conclusion, rrhGM-CSF administration during liver regeneration after 78% PH accelerated breakdown and restoration of the hepatic plate and expression of PCNA. These results suggest that rrhGM-CSF might play an important role during liver regeneration in rats.

        피부 미생물총의 생장과 구성에 대한 화장품과 그 방부제의 영향

        화장품 및 화장품 함유 방부제에 대한 병원미생물과 사람의 피부서식세균들의 증식에 미치는 효과를 조사하였다. 파라벤, 1,2-헥산디올, 페녹시 에탄올과 같은 방부제를 함유한 토너, 에멀젼, 크림, 유아용크림은 포도상구균, 대장균, 녹농균에 대해 강한 방부효과를 보였으며, 파라벤, 1,2-헥산디올, 페녹시 에탄올과 같은 방부제들은 포도상구균, 대장균, 녹농균과 같은 병원균 및 Staphilococcus epidermidis, Shigella flexneri, Enterobacter aerogenes 등과 같은 일부 피부서식세균에 대한 강한 항균효과를 보였다. 사람의 피부에 페녹시에탄올 함유 크림을 도포하였을 때, 피부서식 미생물들의 구성이 바뀌었다. 문 수준에서는 Proteobacteria가 증가하였고, 종 수준에서는 4P004125_s가 증가하였고, Propionibacterium humerusii 감소하였다. 이 결과들로부터 파라벤, 1,2-헥산디올, 페녹시 에탄올과 같은 방부제는 병원균에 대한 항균효과를 가짐과 동시에 페녹시 에탄올을 함유한 화장품은 피부세균군집에 변화를 줄 수 있음을 시사하고 있다. We investigated the growth-inhibitory activities of cosmetics and their preservatives against pathogens and resident skin bacteria. Of the tested cosmetics, preservatives such as parabens, 1,2-hexanediol, phenoxyethanol-contained toner, emulsion, cream and baby cream exhibited potent antibacterial effects against Staphylococcus aureus, Escherichia coli and Pseudomonas aeruginosa. Parabens, 1,2-hexanediol and phenoxyethanol inhibited the growth of pathogens, as well as skin-resident bacteria such as Staphilococcus epidermidis, Shigella flexneri, Enterobacter aerogenes and so on. The application of a basic cream containing phenoxyethanol to human skin was shown to disturb the skin microbiota: at the phylum level, Proteobacteria increased and at species level, 4P004125_s increased and Propionibacterium humerusii decreased. Based on these findings, parabens, 1,2-hexanediol and phenoxyethanol have antimicrobial activity and cosmetics containing phenoxyethanol may disturb skin microbiota.

      • 잇솔질 방법에 따른 구강미생물의 변화

        In order to establish the most effective toothbrushing method, five women in their twenties who maintain a healthy oral condition brushed their teeth using a rolling tooth brushing method, scrub tooth brushing method, and tooth brushing using a sonic vibration electric toothbrush. Samples were taken from the tooth surface to compare the changes in Streptococcus mutans , Lactobacillus casei , general oral microorganisms, and dental plaque. Results were as follows. 1. S. mutans and L. casei on the tooth surface were most abundant following employment of the scrub tooth brushing method; lowest after using the rolling tooth brushing method; and low after brushing with a sonic vibration electric toothbrush. Therefore, the most effective tooth brushing method for the removal of S. mutans and L. casei was confirmed as the roll method. 2. As a result of comparing the changes of oral microorganisms on the surface of the teeth, the tooth brushing method which can be said to most effectively remove general oral microorganisms is the rolling tooth brushing method. Use of a sonic vibration electric toothbrush also had a large effect on removing general oral microorganisms. 3. Using the O’Leary index, the most effective tooth brushing method in reducing dental plaque is the rolling tooth brushing method. The scrub tooth brushing method was found to be less effective in removing dental plaque than the rolling tooth brushing method and tooth brushing method using a sonic vibration electric toothbrush.

        당뇨가 유발된 햄스터 창상치유에 미치는 벼세포 유래 GM-CSF의 효과

        상처치유는 growth factor와 cytokine들의 복합적인 작용에 의해 일어나는 과정이다. GM-CSF는 호중구와 대식세포를 분화시키고 각질세포를 증식시키는 다면발현성 사이토카인으로 여러 세포에서 재조합되어 사용되고 있다. (주)핸슨바이오텍에서 개발한 rice cell 유래의 재조합 rrhGM-CSF는 다른 세포에서 유래된 GM-CSF들보다 탄수화물 측쇄가 많은 것이 특징이다. rrhGM-CSF의 창상치유효과를 알아보기 위하여 햄스터에 만성 당뇨를 유도하여 상처치유가 어려운 동물모델을 만든 후, 등쪽에 인위적으로 피부 전 층을 제거하여 창상을 유발하였다. 실험군에는 rrhGM-CSF가 포함된 연고를 적용하고, 대조군에는 base cream만을 적용하여 재상피화되는 과정과 피부조직의 구조 및 PCNA 반응을 통한 세포증식 정도를 비교하였다. 육안 및 조직학적 관찰 결과, rrhGM-CSF를 적용한 그룹의 재상피화 속도가 더 빨랐으며, PCNA 면역조직화학적 조사에서도 증식하는 각질세포의 수가 대조군보다 상대적으로 많은 것으로 나타남으로써, rrhGM-CSF에 의한 피부상처 치유 효과를 관찰할 수 있었다. 이러한 결과들로 미루어 보아 벼세포 유래의 rrhGM-CSF는 국소적인 피부 창상 치유에 효과적인 것으로 보이며, rrhGM-CSF의 풍부한 당화 사슬은 이 단백질이 상처부위에 안정된 상태로 기능을 나타냄으로써 효과를 오랫동안 지속시킬 수 있었던 것으로 사료된다. GM-CSF is a multipotent growth factor, which also plays an important role during the process of wound healing. rrhGM-CSF was specifically produced from rice cell culture in our laboratory (Hanson Biotech Co., Ltd, Daejeon). The rrhGMCSF contains more oligosaccharide side chains than any other types of GM-CSF. This work was taken to evaluate the influence on wound healing of rrhGM-CSF in male golden hamsters. Full thickness skin defects of 9 mm in diameter were made in the back of hamsters, and 100 ${\mu}L$ ointment containing rrhGM-CSF 50 ${\mu}g/mL$ was applied. Control groups were given ointment without rrhGM-CSF. The wound sizes were relatively reduced and skin was well regenerated in the experimental group compared with the control group. Structurally, reepithelialization and architecture of the skin following injury were well accomplished in the experimental group. And also, positive reaction of PCNA of the skin following injury was more prominent in rrhGM-CSF containing ointment treatment group. Since this type of GM-CSF has highly glycosylated side chains, the effectiveness might be retain longer and stable, regarding acceleration of wound healing in the animal model. The present study has important implications for further development of the therapeutic manipulation of wound healing using rrhGM-CSF.

      • Isolation and Identification of a Bacterial Agent Causing Soft Rot in Chinese Cabbage

        The Chinese cabbage (Brassica rapa subsp. pekinensis), part of the Brassicaceae family, is a biennial plant that has long been cultivated in China and East Asia. However, various plant diseases cause considerable damage to Chinese cabbage yields during cultivation. In this study, diseased Chinese cabbages that showed typical symptoms of bacterial soft rot were collected from different fields in Poheon, Gyeonggi-do, South Korea from September to October in 2019. The causal agent was isolated from the cabbage samples and subsequently re-inoculated into healthy cabbage tissues. The re-inoculated healthy cabbages showed the same symptoms as the diseased cabbages, from which the pathogen was originally isolated. The isolated pathogen was identified as a member of the genus Pseudomonas using 16S rRNA analysis. This result may suggest that a Pseudomonas species isolated from the cabbage can be the causal agent of soft rot that are currently spread in Pocheon, Korea. Further research for accurate identification of the bacterial species should be needed.

        볶음땅콩가루, 새싹 땅콩가루를 첨가한 쌀파이의 품질 특성

        The purpose of this study is to make rice pie, a bakery item, to enhance use of rice flour and analyze quality characteristics of rice pie by adding roasted peanut flour and peanut sprout flour. Water content of rice pie adding roasted peanut flour or peanut sprout flour is lower than that of wheat pie. The pH of rice pie is lower than that of wheat pie, and pH becomes higher when roasted peanut flour or peanut sprout flour is added. Density of dough is lower in rice pie than in wheat pie, and when roasted peanut flour or peanut sprout flour is added, rice pie reveals higher density. Loss rate is higher in rice pie than in wheat pie, and when roasted peanut flour or peanut sprout flour is added, rice pie reveals lower loss rate. When roasted peanut flour or peanut sprout flour is added to rice pie, its brightness (L) and yellowness (b) decrease, while redness (a) increases. Hardness is higher in pie made of rice powder than in wheat pie, and when roasted peanut flour or peanut sprout flour is added, hardness becomes lower. Regarding composition of fatty acids in rice pie, saturated fatty acids observed are myristic acids, palmitic acids, and stearic acids. Monounsaturated fatty acids found are mainly oleic acids. Polyunsaturated fatty acids detected are linoleic acids, and are found more in pie made of rice powder, than in wheat pie.
