        Design, synthesis, and biological evaluation of chalcones for anticancer properties targeting glycogen synthase kinase 3 beta

        Chalcones compounds have been investigated to exhibit anticancer activity through various physiological modes of action. In order to develop chalcone compounds with novel anticancer-related modes of action, diverse chalcone compounds were designed and synthesized. Variously substituted poly-methoxy chalcone compounds 1 – 17 were prepared, and their structures were identified using high-resolution mass spectrometry (HR/MS) and nuclear magnetic resonance (NMR) experiments. Long-term survival clonogenic assay was applied to evaluate their anti-cancer abilities and revealed that their GI50 values ranged between 1.33 and 172.20 μM. When MCF-7SC cells were treated with various concentrations of compound 14 , reduced cell viability and induced apoptosis in MCF-7SC cells were observed in a dose-dependent manner. Wound healing assay demonstrated that compound 14 prevented the MCF7-SC migrated cells at non-lethal concentrations after 12 and 24 h of exposure. The efficiency of compound 14 on the levels of Epithelial-mesenchymal transition (EMT) markers was accessed by the western blot analysis. For the concrete understanding of anticancer properties at the molecular level, in vitro kinase assays on 12 cancer related proteins were carried out. Glycogen synthase kinase 3 beta (GSK3β) was most effectively inhibited by compound 14 with 89% inhibitory activity at 10 µM against GSK3β. The binding mode of compound 14 with GSK3β was reinforced through in silico experiments, which demonstrated compound 14 binds with GSK3β at binding affinity ranged between − 7.5 kcal/mol and − 6.8 kcal/mol. SwissADME analysis provided the druggability and leadlikeness of compound 14 , which unveiled drug development possibilities of chalcone compound 14 .

        문헌 연구를 통한 한국 춤 동작의 생체 역학적 분석 방법론 연구 : 신체 분절 협응 관점을 중심으로

        인체의 움직임을 분석하기 위해서는 '어떠한 목적과 관점에서 동작을 분석할 것인가'를 고려해야 한다. 본 연구는 한국춤 동작 특성에 따른 동작분석을 하기 위해 생체역학 이론 고찰과 문헌 연구를 바탕으로 신체 분절 협응의 관점에서 방법론을 제시하고 설명하는데 목적이 있다. 춤에서 움직임은 서양과 동양을 막론하고 그 원리는 같다. 하지만 동작의 형태에 따라 춤의 성격이 달라지며 각 나라의 독특한 양식의 춤이 형성된다. 한국 춤은 가슴에서 움직임의 충동이 시작되어 상완, 전완, 손으로 순차적 흐름이 나타난다. 이러한 움직임을 생체 역학적으로 운동 전이현상이라고 하며 순차적 운동 전이는 분절 간 및 분절 내의 유기적 조화를 이루어 부드러운 곡선의 형태를 만들어 낸다. 이 연구에서는 한국춤 동작의 특성 중 신체의 한 단위에서 다른 단위로 흐르는 중심 충동을 통해서 이어지는 계속적인 흐름을 증명하는 방법으로 생체역학의 영상분석 방법을 설정하고 신체 분절 협응 관점으로 해석하고자 하였다. 한국 춤의 오랜 수련을 통해 획득된 협응 패턴을 알기 위해서는 숙련도에 따른 비교 분석이 한국 춤의 동작 특성을 밝힐 수 있는 중요한 변수가 될 것이다. 또한 한국 춤이 신체 움직임의 완전한 협응에서 이루어지는 대표적인 예술 춤이라는 것을 규명할 수 있을 것이다. 이를 위해 영상 분석으로 획득한 운동학적 변인들을 이용해 각도-각도 도면을 작성하고 질적 분석을 할 수 있도록 유형별 해석(Winstein & Garfinkel, 1989)을 제시하였다. 하지만 관절각도 자체가 협응을 의미하지는 않으므로 관절각도 간 상관계수를 분절간 및 분절 내 협응 변수로 이용해 정량화된 자료를 양적 분석 할 것을 선행 연구를 통해 보고하였다. 본 연구는 생체역학의 이론 및 협응 관점의 선행연구 조사를 통해 신체 분절 협응 관점에서 한국 춤 동작분석 가능성을 제안하였다. In order to analyze the movement of human body, we need to consider 'with what purpose and perspective, the analyze shall be done.' This study aims to suggest and explain the methods on analyzing movement of Korean dance from the perspective of inter-segmental coordination based on biomechanics theory and documentary research. Dancing, whether it is oriental or western, has the same principle. However, depending on the way of movement itself, the characteristics of any dance will be changed, providing uniqueness to each country's folk dance. For instance, Korean dancing starts from the movement of chest to flow into brachial, wrist curl and hands. This movement is biomechanically called transition phenomena of movement and sequence transition of movement makes a soft round shape by creating organic organization between segmentation. In order to prove the continuous flow of movement from one body part to the other while dancing Korean dance, I introduced video analysis on biomechanics and tried to interpret the results from the segmental coordination point of view. Accumulated and diverse competitive analysis will be a decisive factor for finding out segmentation coordination pattern acquired by long-time practice of Korean dance and for reaffirming that Korean dance is a representative art dance created from a perfect coordination of segmentation. To this end, I will provide angle-angle diagram drawing applying the kinematics variables obtained through video analysis and different academic theory (Winstein & Garfinkel, 1989) to boost the quality aspects of analysis. However, as the joint angle itself does not mean coordination, I suggested the necessity of quantitative analysis on coefficient of correlation between joint angle by using inter and within segmentation coordination as variables in the previous study. This study suggests the possibility of analyzing Korean dance movement from the perspectives of inter-segmental coordination based on the biomechanics theory and documentary research of coordination.

        Fabrication of Poly(methyl methacrylate) Micro-Droplets Using a Complex Electric Field on a Rolling Collector

        Electrospraying was used to fabricate micro-sized droplets of various polymers using strong electric fields. For the mass production of poly (methylmethacrylate) droplets and more uniform-sized droplets than the normal electrospray process, an AC electric field was applied to the rolling collector in a normal electrospray process to provide an additional external force. The droplets were placed in an additional dispersion on the rolling collector due to the AC field conditions, resulting in the formation of smaller and more uniform-sized droplets.

        Synthetic polyphenol compounds inhibit β-catenin/Tcf signaling: Structure-activity relationship

        Activated b-catenin/T-cell factor signaling has been shown to be involved in human carcinogenesis. Here,the inhibitory effects of various synthetic naphthoflavones and napthochalcones on b-catenin/Tcfsignaling in b-catenin overexpressing-cells were investigated and compared. The naphthoflavones witha b-angular pattern (5,6-benzoflavone series) resulted in a more potent inhibitory activity than thosewith an a-angular pattern (7,8-benzoflavone series), indicating that the direction of the naphthyl ring isan important determinant of inhibitory activity. This result is in accordance with the napthochalconederivatives. Moreover, in silico docking results supported our in vitro biological results.

        Copper(II) Complexes Containing N,N′-Bidentate N-(Pyridin-2-ylmethyl)aniline and Its Derivatives: Synthesis, Structure and Magnetic Property

        The reaction between [CuCl2 . 2H2O] and N,N′-bidentate N-(pyridin-2-ylmethyl)aniline (La ) and its derivatives (Lb and Lc ) in methanol yielded new Cu(II) complexes; namely polynuclear [CuLaCl2 ]n , dinuclear [CuLb (μ-Cl)Cl]2 , and mononuclear [CuLcCl2 ], respectively. Monomeric [CuLcCl2 ] showed a distorted square planar geometry around copper metal-based on X-ray crystallography. The polymeric [CuLaCl2 ]n and dimeric [CuLb (μ-Cl)Cl]2 had a chloro-bridged five-coordinated and a distorted square pyramidal geometry around the Cu(II) center. The Cu∙∙∙Cu′ distance was 3.915(1) Å in [CuLaCl2 ]n and 3.928(1) Å in [CuLb (μ-Cl)Cl]2 . Interestingly, magnetic susceptibility (χ) measurements of complexes [CuLaCl2 ]n and [CuLb (μ-Cl)Cl]2 exhibited a weak antiferromagnetic interaction.

        한강지류 하천 주변 토양의 Phthalate ester 오염 분석

        Phthalate ester는 플라스틱의 물리적 성질, 특히 유연성을 향상시키시 위하여 플라스틱 제품에 첨가하는 가소재이며 내분비계 장애를 일으키는 것으로 추정되는 물질이다. 본 연구에서는 우리나라 한강지류 하천 주변 토양의 pathalate esters의 오염 정도를 gas chromatography-mass spectrometry(GC-MS) 방법으로 분석하였다. 그 결과 예상 오염 지역인 안양천과 서호천, 그리고 예상 청정지역인 가평천과 남한강 지역에서 채취한 토양시료 모두에서 bis(2-ethylhexyl) phthalate(DEHP)가 일정 수준 검출되었다. 그 오염 수준은 안양천 주변 토양이 113ppb로 가장 높았으며, 서호천 주변 토양이 64ppb,그리고 가평천과 남한강 주변 토양이 약 50ppb이었다. 따라서 한강 지류 하천 토양의 DEHP오염은 국내 하천 저질의 DEHP오염 수준이 최고 2.04 ppm임을 감안할때 상대적으로 낮은 수준임을 알 수 있었다. Phthalate esters are used as additives in plastics to improve mechanical properties, particularly flexibility.The contamination of soil samples by phthalate esters around four tributaries to the Han river was analyzed based on gas chromatography-mass spectrometry (GC-MS). All soil samples collected from the four tributatries, which include Anyangcheon and Seohocheon(expected polluted areas), and Gapyeongcheon and Namhan riveer (expected clean areas), were found to be contaminated by bis(2-ethylhexyl) phthalate(DEHP). Soil around Anyangcheon was the most contaminated(113 ppb), about twofold higher than the ones around Seohocheon(64 ppb),Gapyeongcheon and Namhan river(50 ppb). Based on the fact that DEHP contamination in sediments of thd rivef tributaries around the country is as high as 2.04 ppm, it was concluded that DEHP contamintion of the soils around the tributaries to the Han river is relatively low.

        Nearshore Current Pattern and Rip Current Occurrence at Jungmun Beach, Jeju by Numerical Computation

        A nearshore current or a wave-induced current is an important phenomenon in a nearshore zone, which is composed of longshore, cross-shore, and rip currents. The nearshore current is closely related to the occurrence of coastal accidents by beachgoers. A considerable number of coastal accidents by beachgoers involving the rip current have been reported at Jungmun Beach. However, in studies and observations of the nearshore current of Jungmun Beach, understanding of the rip current pattern remains unclear. In this study, a scientific approach is taken to understand the nearshore current and the rip current patterns at Jungmun Beach by numerical computation for year of 2015. From results of numerical computation, the occurrence and spatial characteristics of the rip current, and the similarities between the rip current and incident wave conditions are analyzed. The primary results of this study reveal that the rip currents are frequently generated at Jungmun Beach, especially in the western parts of the beach, and that the rip currents often occur with a wave breaking height of around 0.5 ~ 0.7 m, a wave period of around 6 ~ 8 seconds, and a breaking angle of around 0 ~ 15 degrees.
