Isolation of Paenibacillus pinesoli sp. nov. from Forest Soil in Gyeonggi-Do, Korea

Using a new culture method for unculturable soil bacteria,strain NB5T was isolated from forest soil at Kyonggi University,and characterized taxonomically on the basis of 16SrRNA gene sequence as well as phenotypic and chemotaxonomiccharacteristics. The novel strain was a Gram- and catalase-positive, rod-shaped bacterium, which grew in the pHrange 6.0–9.5 (optimum, 6.5–9.5) and at temperatures between15°C and 45°C (optimum, 25–40°C). Growth was possibleat 0–5% NaCl (optimum, 0% to 3%) in nutrient, Luria-Bertani, and trypticase soy broths (TSB), as well as R2A medium(with optimal growth in TSB). A phylogenetic analysisof the 16S rRNA gene sequence showed that the novel strainwas affiliated with the genus Paenibacillus and had 96.8%and 96.5% similarity to P. nanensis MX2-3T and P. agaridevoransDSM 1355T, respectively. The predominant menaquinonein NB5T was MK-7; the major fatty acids were anteiso-C15:0 and iso-C16:0; and the DNA G+C content was 54.5mol%. We propose this strain as a novel species of the genusPaenibacillus, and suggest the name Paenibacillus pinesolisp. nov. (type strain, KACC 17472T =KEMB 9005-025T =JCM19203T).
U2OS 골육종 세포주의 세포자멸사에서 MMP억제제와 Doxorubicin 작용의 비교연구

목적: wild-type p53 단백질을 가지고 있는 U2OS 인간 골육종 세포주의 세포자멸사에서 MMP억제제와 doxorubicin의 작용을 비교하고자 하였다. 대상 및 방법: 배양한U2OS 세포주에 Doxorubicin, MMP억제제(MMPI III)를 따로 투여한 것과 두 약물을 동시에 투여한 것을 비교하였다. 또한 Doxorubicin의 작용이 Fas/FasL 경로를 통해 유발되는지 알아보기 위해 Fas 중화항체를 doxorubicin과 함께 투여하였다. 약물을 처리하여 배양한 세포에서 발생한 세포자멸사와 세포괴사를 확인하기 위해서 유세포 검사를 시행하였다. 결과: Doxorubicin을 처리한 세포가 처리하지 않은 세포보다 약물 농도에서 더 많은 세포괴사(p=0.000)를 보였다. 이에 반해 MMPI III를 처리한 세포는 약물 농도와 반응시간에 따른 세포자멸사 및 세포괴사에서 차이를 보이지 않았다. 두 약물을 동시에 투여한 군은 그렇지 않은 군에 비해 세포 자멸사 및 세포괴사에서 차이가 없었다. Doxorubicin에 Fas 중화항체를 추가한 군과 doxorubicin 단독 투여 군 사이에 세포자멸사 및 세포괴사에서 차이가 없었다. 결론: Doxorubicin과 같이 MMP 억제제를 골육종 치료에 사용하려면 wild-type p53 뿐만 아니라 wild-type p14를 가지고 있는 골육종 세포에 대한 검사가 필요할 것으로 사료된다. Purpose: The purpose of this study was to compare the proapoptotic effects of matrix metal-loproteinase inhibitor (MMPI) and doxorubicin on wild-type p53 osteosarcoma cell line, socalled U2OS cell line. Materials and Methods: U2OS cells were treated with MMP inhibitor III (MMPI III) and doxorubicin, either respectively or simultaneously. In cells treated with doxorubicin, Fas-neutralizing antibody so called ZB4 was additionally treated to examine whether the doxorubicin played a role through the Fas/FasL pathway. Cells were analysed regarding to apoptosis and cell death by flow cytometry. Results: U2OS cells incubated with doxorubicin showed significant amount of cell death in dose-dependent manner. However, those incubated with MMPI III mostly remained viable state. In addition, there is no relationship between two drugs. Cells treated with doxorubicin and ZB4 at the same time did not show down regulation of apoptosis through inhibition of Fas/FasL pathway. Conclusion: It is important to re-examine MMP inhibitor's effect on other osteosarcoma cell line with wild-type p14 as well as wild-type p53 to evaluate its proapoptotic effect.
過脂肪食餌가 흰쥐 腸間膜 비만세포에 미치는 형태학적인 硏究

This experiment was carried out to study the morphology of the mesenteric mast cell of the rat. We used relatively 40 mature, health rats less than 300g^(m) in weights, divided into 4 groups, controlled, carbohydrate, protein and fat, respectively, breeding for 100 days and killed. After that we examined and observed the relative emergence numbers, morphology, staining and distributions of the mast cells in the mesentery and so the following results were obtained. 1. Relative emergence number from the mesenteric mast cells of the controlled group was 75.1±0.6, and most of them contained rich, fine mature granules and seen that mature granules in some of the mast cells were escaped out of the cell. The morphology of mast cell was relatively round or oval. 2. Relative emergence numbers from the merenteric mast cells of the carbohydrate group and protein group were 45.6±1.7, 41.4±1.2. in mature, fine granules were fully contained in the cytoplasm of the carbohydrate group, and the protein group was relatively rare in the cytoplasm and sometimes seen mature granuler escaped from the cell. 3. Relative emergence number of the fat group was 39.5±0.7 and the morphology of the mast cells was relatively oval or collapsed as a whole. Granules in the cytoplasm were in matare rare.

The adrenal medulla using immunohistochemistry few somatostatin immunoreactive nerve fibers were demonstrated in adrenal medulla, In addition, a large population of chromattin cells in the dog adrenal medulla displayed intense somatostatin-like immunoreactivity especially in the peripheral and perivascular region. About 10~20% of the adrenaline cells were the somatostatin-immunoreactive. The present study confirms the somatostatin is localized in the dog adrenal chromattin cell, and it is suggested that somatostatin may have a paracrine or endocrine role in the dog adrenal gland.
