The structural relationship of 16 asiatic acid (AA) derivatives, including AA and asiaticoside (AS) to cytotoxicity and anti-hepatofibrotic activity in HSC-T6 cells, were investigated. Cytotoxicities of AA derivatives varied from 5.5 mM to over 2000 mM of IC50 depending on AA functional group modifications. Substituting the hydroxyl group at the C(2) to NºC and substituting bulky groups for dihydroxyl groups at (3), (23) of the A-ring increased the cytotoxicity, but keto group at C(11) and benzoyl ester at C(2) were greatly reduced it. Modification of the carboxylic acid group at C28 also reduced the cytotoxicity. The collagen synthesis determined by hydroxyproline content in the cells was inhibited from a maximum of 48% (Zlx-i-85 and 87) to 15% (AS) by AA derivatives. The anti-hepatofibrotic effect of these compounds might be due to the reduced expression of prolyl 4-hydroxylase a and b subunits and TIMP2. However, the inhibition of collagen by asiaticoside derivatives did not show any structural-activity relationship.

Green tea and black tea are commonly drunk in the Asian and Western countries. It is known that the major tea catechins, (-)-epicatechin, (-)-epigallocatechin, (-)-epigallocatechin gallate, and (-)-epicatechin gallate are epimerized by fermentation processing. In this study, the effects of the extracts of the non-fermented and fermented teas on hepatic fibrosis were examined by means of the in vitro collagen secretion and collagen promoter assay. As a result, the treatment with green tea extract was found to reduce the secretion of collagen into the media, whereas black tea extract increased it, in the human hepatic stellate cell line.
헛개나무 열매 열수추출물 투여에 의한 생쥐의 지구력 운동 수행 능력 향상 및 피로개선 효과

The aim of this study was to evaluate the effect of hot water extract of peduncle obtained from Hovenia dulcis Thunb (HD) which is commercially developed for the protective effect on the alcoholic hepatotoxicity, on the endurance capacity for weight loaded forced swimming mice. The swimming times to exhaustion in mice fed 100 and 200 mg/kg HD for 2weeks were prolonged 3.6 and 3.7 fold, and for 4 weeks 1.9 and 2.7 fold compared with each vehicle control (42.8±20.5 min and 67.7±47.8 min, for 2 and 4 weeks), respectively. Blood biochemical parameters for ALT, AST, creatinine and BUN were not significantly different between from HD fed or control mice. Although HD fed mice swam over 2 fold longer time than vehicle control mice at 4 weeks, blood biomarkers of physical fatigue such as glucose, triglyceride and free fatty acid, lactate were not significant different and even tended to ameliorate. Hepatic lipid peroxidation and SOD activity did not significantly change in HD fed- and vehicle control exhausted swimming mice at 2 or 4 weeks. However, catalase activity in HD-fed mice was significantly increased in a dose-dependent manner compared with vehicle control mice. The present study indicates that HD improved physical fatigue and exercise performance in mice. Therefore, it has a potential for the pharmacological effect of anti-fatigue.
오리나무 열수 추출물이 실험적으로 유발된 흰쥐의 위염 및 위궤양에 미치는 영향

Alnus japonica Steud (A. japonica) have long been used in the traditional medicine for gastric disorder, hepatitis and fatty liver in Korea. Antiulcer effects of A. japonica hot water extract (AJ ext) were evaluated by in vitro antibacterial activity against H. pylori, by the inhibitory action against the in vitro gastric H^+/K^+ -ATPase and using rat models of gastric mucosal damage and gastric ulcer induced by HCl-ethanol, indomethacin, and restraint and water-immersion stress. For the determination of antibacterial activity of AJ ext against H. pylori, the activity of urease which released from H. pylori was measured in culture. AJ ext showed weak antibacterial activity against H. pylori with the growth inhibitions of 37% and 61% by adding final concentrations of 500 and 1000 µg/ml culture, respectively at 24 h. To observe the inhibitory activity of AJ ext against the H^+/K^+ -ATPase in hog gastric membrane vesicle, IC50 value of AJ ext was 806.3 µg/ml. Pretreatment of AJ ext (200, 500mg/kg, p.o.) prevented in a dose-dependent manner the acute gastritis in HCl-ethanol model and the formation of gastric ulcer in indomethacin model and restraint and water-immersion stress model. These results suggest that the AJ ext can be used for prevention and treatment of gastric mucosal damage and ulcers induced by various stress.

Rhus verniciflua stokes (RVS) is known to promote blood circulation by preventing blood stasis, although the active ingredients and the underlying mechanism are unclear. Platelets are the primary cells that regulate circulation and contribute to the development of diverse cardiovascular diseases by aggregation and thrombosis. The study assessed the antiplatelet activity of RVS and sought to identify the active constituents. Pretreatment of washed platelets with RVS heartwood extract blunted the aggregatory response of platelets to collagen. In the subfractions, fisetin, butein, and sulfuretin were identified as effective inhibitors of platelet aggregation by collagen, thrombin, and adenosine-5'-diphosphate. Antiplatelet activities of all three compounds were concentration dependent, and fisetin had longer in vitro duration of action compared with butein or sulfuretin. Extracellular signal-regulated kinase (ERK) mitogen-activated protein kinase activation by collagen was prevented by fisetin, whereas butein and sulfuretin failed to inhibit ERK and p38 activation was not affected by any of the compounds. Rats orally administered 100 mg/(kg·day - 1) fisetin for 7 days were resistant to arterial thrombosis, although total extract of RVS heartwood exhibited little effect at a dose of 1000 mg/(kg·day - 1). RVS heartwood may have cardiovascular protective activity by inhibiting platelet aggregation. The active constituents are fisetin, butein, and sulfuretin, and fisetin is orally effective against thrombosis.
랫드를 이용한 헛개나무과병 열수추출물 투여가 운동에 의한 피로회복과 운동수행능력에 미치는 기전 규명

The present study investigated the effects of Hovenia Dulcis (HD) fruit extract powder on the improvement of physical activity, especially exercise capacity. Forty mice were divided into 4 groups including normal controls, negative controls, 100 (HD-100) and 200 (HD-200) mg/kg HD fruit extract powder groups for 5-times exercises using treadmill. Nor- mal control did not performed treadmill running but others did 5-times for 10 days. HD fruit extract powders were admin- istrated orally one-times per day for 10 days before treadmill exercise and normal and negative controls were fed with excipient water. After 5-times exercise, blood biochemical analysis showed that aspartate aminotransferase (AST), lactate dehydrogenase (LDH) and creatine phosphokinase (CK) activities and blood lactate concentration were statistically increased in negative controls than in normal controls. They were decreased in HD fruit extract powder groups, compared with negative controls. These results were considered as the effects of HD fruit extract powders on reduced tissue damages during exercise. Other measured indices did not reveal remarkable differences. All together, these results suggest that HD fruit extract powders may enhance the exercise performance by recovering the exercise-fatigue via blood lactate con- centration by reducing blood LDH activity and via reduction of blood CK and AST activity.

Hovenia dulcis which is distributed in Korea, China and Japan is known to show hepatoprotctive effect and reduce the acute alcohol toxicity. In this study, the hepatoprotective effect against the chemically induced experimental liver injury models and lowering effect of blood alcohol level in animal models acutely administered alcohol by the peduncle extracts of Hovenia dulcis var. koreana were investigated. HdfHW-1, and HdfM-1 which are the extracts of fruit peduncles and young branches with hot water or 70% methanol and followed with 100% methanol, were significantly reduced the $CCl_4$ or D-galactosamine/LPS induced damage in sliced liver. The hot water or methanol extracts of fruit peduncle protected dose-dependently against $CCl_4$ induced toxicity in primary hepatocyte culture and particularly, the amount of LDH release was reduced to the control level at 500 $\mu\textrm{g}$/ml of hot water extracts. HdfHW-1 also decreased the hepatotoxicity induced by $CCl_4$ in rats. The active components of HdfHW-1 seemed to be high molecular weights because 0.2 M NaCl HdfHW-1 fraction was the most effective among NaCl fractions of HdfHW-1 eluted with various concentrations of NaCl on DEAE 650C column chromatography. HdfM and HdfHW were significantly reduced the levels of blood alcohol in rats and mice administered 40% alcohol. These results indicated that the hot water or methanol extracts of fruit peduncle of Hovenia dulcis var. koreana have hepatoprotective effect and may be reduce alcohol toxicity.
