        Molecular Screening of Small Biopsy Samples Using Next-Generation Sequencing in Korean Patients with Advanced Non-small Cell Lung Cancer: Korean Lung Cancer Consortium (KLCC-13-01)

        Background: Non-small cell lung cancer (NSCLC) is a common type of cancer with poor prognosis. As individual cancers exhibit unique mutation patterns, identifying and characterizing gene mutations in NSCLC might help predict patient outcomes and guide treatment. The aim of this study was to evaluate the clinical adequacy of molecular testing using next-generation sequencing (NGS) for small biopsy samples and characterize the mutational landscape of Korean patients with advanced NSCLC. Methods: DNA was extracted from small biopsy samples of 162 patients with advanced NSCLC. Targeted NGS of genomic alterations was conducted using Ion AmpliSeq Cancer Hotspot Panel v2. Results: The median age of patients was 64 years (range, 32 to 83 years) and the majority had stage IV NSCLC at the time of cancer diagnosis (90%). Among the 162 patients, 161 patients (99.4%) had novel or hotspot mutations (range, 1 to 21 mutated genes). Mutations were found in 41 genes. Three of the most frequently mutated genes were TP53 (151, 93.2%), KDR (104, 64.2%), and epidermal growth factor receptor (EGFR; 69, 42.6%). We also observed coexistence of EGFR and other oncogene (such as KRAS, PIC3CA, PTEN, and STK11) mutations. Given that 69.6% (48/69) of EGFR mutant patients were treated with EGFR tyrosine kinase inhibitors, EGFR mutant status had higher prognostic ability in this study. Conclusions: These results suggest that targeted NGS using small biopsy samples is feasible and allows for the detection of both common and rare mutations in NSCLC.

        PKCη Regulates the TGFβ3-induced Chondrogenic Differentiation of Human Mesenchymal Stem Cell

        Transforming growth factor (TGF) family is well known to induce the chondrogenic differentiation of mesenchymal stem cells (MSC). However, the precise signal transduction pathways and underlying factors are not well known. Thus the present study aims to evaluate the possible role of C2 domain in the chondrogenic differentiation of human mesenchymal stem cells. To this end, 145 C2 domains in the adenovirus were individually transfected to hMSC, and morphological changes were examined. Among 145 C2 domains, C2 domain of protein kinase C eta (PKCη) was selected as a possible chondrogenic differentiation factor for hMSC. To confirm this possibility, we treated TGFβ3, a well known chondrogenic differentiation factor of hMSC, and examined the increased-expression of glycosaminoglycan (GAG), collagen type II (COL II) as well as PKCη using PT-PCR, immunocytochemistry and Western blot analysis. To further evaluation of C2 domain of PKCη, we examined morphological changes, expressions of GAG and COL II after transfection of PKCη-C2 domain in hMSC. Overexpression of PKCη-C2 domain induced morphological change and increased GAG and COL II expressions. The present results demonstrate that PKCη involves in the TGF-β3-induced chondrogenic differentiation of hMSC, and C2 domain of PKCη has important role in this process.

        컨템퍼러리 댄스의 상호매개성(Intermediality) : 몸-아카이브의 형성을 중심으로

        본 연구는 관습적인 춤이나 무용예술의 매체특정성을 벗어나고 있는 컨템퍼러리 댄스가 제기하는 매체에 대한 비판성을 탐구하는 데 목적을 두었다. 이를 위해 컨템퍼러리 댄스에서 나타난 주요 변화들의 맥락과 이에 대한 논의, 그리고 ‘안무(choreography)’ 개념이 제기하는 문제들을 검토했고, 컨템퍼러리 댄스의 모호하고 경계적인 속성을 상호매개성(intermediality)의 개념을 통해 논했다. 컨템퍼러리 댄스에 이르기까지 발전되어 온, 일반적인 의미의 안무 개념은 춤을 사라지는 것으로 이해하며 이에 대한 포획을 시도하는 장치로서 매체적 습관을 나타내었다. 이런 특성은 소통을 위한 사이 공간이나 소통을 가능하게 하는 것으로서의 아카이브가 등장하면서 불안정하게 된다. 아카이브는 단지 안무적 작동에 의해 포획되어 고정된 상태를 지칭하는 것을 넘어, 아직 소진되지 않은 잠재적 가능성을 찾고 새롭게 발현하는 것으로 이해된다. 즉, 컨템퍼러리 댄스에서 아카이브는 퍼포먼스에서 남겨진, 포획된 것과 새로이 진행되는 것의 동시 존재를 가능하게 하는 확장된 시간성을 나타낸다. 그리고 이런 아카이브는 퍼포먼스를 이론적, 교육적, 혹은 공동체적인 논의의 대상으로 확장시킬 수 있다는 점에서 생활공간으로 확장되며, 예술과 일상, 가상과 현실 등 이질적인 층위들이 만나는 상호매개적 장소로 작동한다. 이런 작동 하에서 퍼포먼스에 참여하는 몸들은 그 순간의 물질적 현존을 넘어 생활공간 속의 살아 있는, 그리고 살아가는 몸으로 이해된다. 이런 변화 속에서 안무는 무용의 자율적인 형식을 주장하는 대신 여러 매체와의 교류와 협력을 나타내며, 담론과 이론 등 지식생산과도 긴밀한 관계를 나타낸다. 이에 본고에서는 컨템퍼러리 안무에 이르러 확장된 매체적 특성을 기존 매체적 관습에 대한 비판으로서 이해하고, 퍼포머가 본인으로서 자신의 이야기를 하는 자전적 퍼포먼스 작품들을 소개함으로써 이런 실천을 구체적으로 고찰하고자 한다. 이런 작품들은 언어와 다큐멘트의 도입을 통해 양식적인 구분의 모호함을 나타냄은 물론, 기억과 경험의 아카이브이자 현실과 예술의 모호한 경계에 있는 존재, 즉 살아있는 몸-아카이브로서의 존재와의 소통 속에 참여자들을 불러온다. 이에 따라 자전적 퍼포먼스는 몸-아카이브의 잠재적인 정체성을 파편적으로 경험하고 구성하도록 하는 상호매개적 작동을 일으킨다. This study aims to explore the critical approach toward media in contemporary dance, which avoids conventional understanding of dance and artistic media. After reviewing important contexts and debates concerning contemporary dance, this study discuss the matter of its ambiguous boundaries and expansion through the concept of intermediality. The idea of performance and dance with its ephemeral nature and media being used to preserve it, has constituted a conventional understanding of its temporality. However in contemporary dance, the archive expresses the request for extended temporality that enables the simultaneous existence of what is remaining, what is captured, and what is progressing in the performance. In addition, these archives expand into living spaces so that performances can lead to theoretical, educational, or political discourses, and function as a mediating space where heterogeneous entities such as art, virtual reality, and real life meet. Moreover through this study, I wish to illuminate the expanded understanding of media in contemporary choreography as a criticism on previous media conventions, by introducing certain examples of autobiographical performance pieces in which the performer tells his or her own story. These works show the blurred boundaries among verbal languages, documents, and corporeal performance. They also bring participants in communication with performers, who can be understood as a physical archive of memories and experiences, roaming the ambiguous space inbetween reality and arts. Accordingly, an autobiographical performance creates an intermediality that allows the body-archive to experience and construct the virtual identity of the body in a fragmented manner.

        재조합 대장균으로부터 추출‧정제된 베타-카로틴의 주름개선 활성

        This paper described the extraction/purification of β-carotene from recombinant E.coli and evaluation of anti-wrinkle activity of purified β-carotene. No significant differences in extraction yields were observed when hexane or isobutyl acetate was used. However, extraction from wet-cell cake resulted in 2-fold higher amount of β-carotene than that from dry cells. Disruption of 5 g-wet cells by ultrasonic homogenizer, acetone dehydration, extraction with isobutyl acetate resulted in 36 mg of β-carotene corresponding to 61.2% of recovery. The formation and separation of β-carotene crystal improved the purity. 633 mg of β-carotene crystal with 93% purity was obtained from 223 g/L of wet-cell cake harvested from 2.5-L fed-batch culture broth. The cultures of normal human primary fibroblast were performed to investigate the effect of β-carotene on cytotoxicity as MTT assay and anti-wrinkle activity as collagen synthesis assays. 1.7 μM of β-carotene was found to be optimal concentration at which 1.4-fold higher amount of collagen was synthesized than that in absence of β-carotene. This indicates that highly purified β-carotene can be obtained from recombinant E.coli by applying simple method with less toxic solvent and can be used in functional cosmetics as anti-wrinkle agent.

        Genetic Alterations and Their Clinical Implications in High-Recurrence Risk Papillary Thyroid Cancer

        Purpose Papillary thyroid carcinomas (PTCs) frequently involve genetic alterations. The objective of this study was to investigate genetic alterations and further explore the relationships between these genetic alterations and clinicopathological characteristics in a high-recurrence risk (node positive, N1) PTC group. Materials and Methods Tumor tissue blocks were obtained from 240 surgically resected patients with histologically confirmed stage III/IV (pT3/4 or N1) PTCs. We screened gene fusions using NanoString’s nCounter technology and mutational analysis was performed by direct DNA sequencing. Data describing the clinicopathological characteristics and clinical courses were retrospectively collected. Results Of the 240 PTC patients, 207 (86.3%) had at least one genetic alteration, including BRAF mutation in 190 patients (79.2%), PIK3CAmutation in 25 patients (10.4%), NTRK1/3 fusion in six patients (2.5%), and RET fusion in 24 patients (10.0%). Concomitant presence of more than two genetic alterations was seen in 36 patients (15%). PTCs harboring BRAF mutation were associated with RET wild-type expression (p=0.001). RET fusion genes have been found to occur with significantly higher frequency in N1b stage patients (p=0.003) or groups of patients aged 45 years or older (p=0.031); however, no significant correlation was found between other genetic alterations. There was no trend toward favorable recurrence- free survival or overall survival among patients lacking genetic alterations. Conclusion In the selected high-recurrence risk PTC group, most patients had more than one genetic alteration. However, these known alterations could not entirely account for clinicopathological features of high-recurrence risk PTC.

        EGFR C797S as a Resistance Mechanism of Lazertinib in Non-small Cell Lung Cancer with EGFR T790M Mutation

        The non-small cell lung cancer with activating epidermal growth factor receptor (EGFR) mutation eventually acquires resistant to either first or second-generation EGFR tyrosine kinase inhibitor (TKI). As the following option, targeting EGFR T790M with third-generation EGFR TKI is now established as a standard treatment option. In this study, we are reporting the first case of resistance mechanism to the novel third-generation EGFR TKI, lazertinib, which showed promising clinical efficacy in phase 1-2 study. The patients showed resistance to the treatment by acquiring the additional EGFR C797S mutation in cis which is also confirmed from the patient-derived cell lines.
