Increased intracranial pressure after massive blood loss -A case report-

A 4-year old boy with supravalvular ascending aortic stenosis underwent sliding aortoplasty. After cardiopulmonary bypass weaning, aorta suture site was torn accidentally and the patient was in hypovolemic shock. Emergency cardiopulmonary bypass was reinstituted and the aorta was repaired. After removal of the aortic clamp, bradycardia and hypertension were noted. We suspected increased intracranial pressure due to hypoxic brain damage after massive blood loss and the patient was treated to lower the intracranial pressure. Physicians should be aware of the significance of the hemodynamic change associated with increased intracranial pressure to prevent further neurologic damage.
Lenzites betulina에 의한 Tannase 생산 및 성질에 관한 연구

Lenzites betulina(조개껍질버섯균) 등 6종 담자균류의 tannase(tannin acyl hydrolase EC 3.1, 1.20) 생산을 비교하고 Lenzites betulina가 가장 우수하여 이 균주의 배양물로부터 효과적인 tannase 생산조건과 효소의 특성을 검토하였다. Lenzites betulina의 tannase 최적 생산을 위한 배양조건은 25℃, pH6.0에서 21일이었고, tannase 생산을 위한 효과적인 배양기질의 조성은 tannic acid 2g, sucrose 5g, bacto-peptone 2g, KH_PO_4 2g, MgSO_4·7H_2O 0.05g, CuSO_4·5H_2O 2㎎, thiamine·HCl 100㎍, 증류수 1000㎖이었다. 이상의 조건에서 Lenzites betulina가 생산하는 tannase는 223.3unit(gallic acid umole/㎖/min)이었다. Lenzites betulina가 생산하는 tannase의 반응 최적 pH는 6.0, 최적 온도는 40℃이었고, 40℃ 이하에서는 효소활성이 대단히 안정하였으나 60℃ 이상에서는 50% 이상 실활하여 고온에 민감하였다. 또한, pH5.5-6.0의 범위에서는 비교적 안정을 유지하였다. Six species under the basidiomycetes were screened for extracellular tannase(tannin acyl hydrolase EC 3.1, 1.20) production in submerged culture and Lenzites betulina was found to be most effective for the production of tannase. The optimum cultural conditions for tannase production were 25℃, pH 6.0 and 21 days of culture period. The efficient composition of culture medium for the production of tannase was performed in synthetic medium containing tannic acid, 2g; sucrose, 5g; bacto-peptone, 2g; KH_2HO_4, 2g; MgSO·7H_2O, 0.5g; CuSO_4·5H_2O, 2㎎; thiamine HCl, 100 ug and distilled water 100 ㎖. The tannase produced from Lenzites betulina was 223.3 unit(umole of gallic acid/㎖ of broth/min). The tannase had an optimal reaction conditions of pH 6.0 and temperature of 40℃. The enzyme was stable at temperature below 40℃ and lost its activity by 50% above 60℃. And the stable pH range was 5.5 to 6.0.

As a medicinal plant, the roots of Valeriana officinalis have been used as a sedative and tranquilizer. In the present study, we evaluated the neuroprotective effects of valerian root extracts (VE) on the hippocampal CA1 region of gerbils after 5 min of transient cerebral ischemia. Gerbils were administered VE orally once a day for 3 weeks, subjected to ischemia/reperfusion injury, and continued on VE for 3 weeks. The administration of 100 mg/kg VE (VE100 group) significantly reduced the ischemia-induced spontaneous motor hyperactivity 1 day after ischemia/reperfusion. Four days after ischemia/reperfusion, animals treated with VE showed abundant cresyl violet-positive neurons in the hippocampal CA1 region when compared to the vehicle or 25 mg/kg VE-treated groups. In addition, the VE treatment markedly decreased microglial activation in the hippocampal CA1 region 4 days after ischemia. Compared to the other groups, the VE100 group showed the lowest level of lipid peroxidation during the first 24 h after ischemia/reperfusion. In summary, the findings in this study suggest that pretreatment with VE has protective effects against ischemic injury in the hippocampal pyramidal neurons by decreasing microglial activation and lipid peroxidation
