The study aimed to confirm transformation, morphology, steroid alkaloidal TLC pattern and growth rate of hairy roots. The Solanum nigrum plantlets were inoculated with Agrobacterium rhixogenes strain 15834. Hairy roots were induced by plasmid. Agropine and mannopine were detected in the hairy roots. The organization of hairy roots on the transectional morphology was undifferentiated. Culture on the medium containing hormone(IBA 2, kinetin 0.1 ㎎/ℓ) altered hairy roots into callus. The growth rate of hairy roots on the NN30 liquid medium was 52 times heavier than in the original state and this was higher than on the other media. The results of TLC analysis indicated that the hairy roots produced steroidal alkaloids resembling those of normal roots.
제주도 주목(Taxus cuspidata S. et Z.)의 Taxol 및 Cephalomannine의 함량분석

To seek for a stable supply source of taxol as an anticancer agent urgently needed for the treatment of ovarian cancer, the concentrations of taxol and cephalo mannine in bark and needles of 37 yew (Taxus cuspidata S. et Z.) trees grown in 6 locations of Cheju Island were analyzed by using HPLC. A considerable variation in the content was observed in plant part and the location. The average taxol content in the needles (220.3 ± 56 ㎍ per g dry wt) was ca. 4 times hgher than that in the bark, whereas the content of cephalomannine in the needles (353 ± 141 ㎍) was ca. 5 times higher than that in the bark. Particularly, even within Cheju Island, needles from trees grown near the coast including Sogwipo area contained much higher taxol contents than needles from trees grown at mountain locations, indicating that taxane level is affected by environmental effects. Taxol in the needles was confirmed by electrospray mass spectrometry. These results suggest that the needles of yew trees growing in Cheju Island may provide a renewable source for taxol.

Microsomal △6 desaturase is a key enzyme in the metabolism of polyunsaturated fatty acids, the activity of which is regulated by insulin. To determine if changes in d △6 desaturase activity in experimental diabetes correspond to changes in fatty acid compoistion of microsome and RBC membrane, 18 Sprague-Dawley rats were studied for 6 weeks. The animals were divided into 3 groups with 6 rats in each group. The first group was the contrals, the 2nd 3rd groups were streptozotocin diabetic rats but insulin was injected to the 3rd group. The △ 6 desaturase activities of diabetic rats were 68pg of those of controls (p<0.05) and increased to 119% of controls (p<0.05) with insulin treatment. Linolic (LA), palmitic (PA) and docosahexaenoic acids (DHA) were increased and arachidonic (AA) & oleic acids (OA) decreased in liver microsome of diabetic rats, which were reversed with insulin treatment, respectively. In erythrocyte membrane LA & stearic acids (SA) were increased, and PA, AA, and palmitoleic acids were decreased which were not reversed with insulin treatment, however. To summarize, insulin controls the △ 6 desaturase activities, which in turn affect fatty acid compositions of microsome and RBC membranes. Theee findings suggest that adequate insulin therapy is required for the correction of the polyunsaturated fatty acid metabolism as well as blood glucose control in the insulin dependent diabetics.
