몽골 유래 Brucella melitensis 동정 및 특이 SNP를 이용한 real-time PCR법에 의한 진단 평가

A real-time PCR assay using hybridization probe (HybProbe) has been developed to detect Brucella (B.) melitensis strains. The primer and HybProbe sets were designed based on the gap gene of chromosome I with a specific single nucleotide polymorphism of B. melitensis. Specificity of the assay was confirmed by comparison to reference Brucella species and other related strains. In the melting curve analysis, B. melitensis generated a peak at $67^{\circ}C$ unlike those for other Brucella species observed at $61^{\circ}C$. Sensitivity of the assay for B. melitensis ranged from 20 ng to 200 fg of genomic DNA. The ability to identify 94 Mongolian B. melitensis isolates using the real-time PCR assay was identical to that of classical biotyping methods and differential multiplex PCR. These data showed that this new molecular technique is a simple and quick method for detecting B. melitensis, which will be important for the control and prevention of brucellosis.

The aim of this paper is to explore the profile of food tourists whose destination is Jeju. Fieldwork was carried out among a serises of restaurant establishments in Jeju. A demand survey was conducted in 20 particular restaurants that were selected among those offering local dishes and regularly visited by tourists. This study contributes regional balanced local food and restaurant industry development by proposing differentiated food tourists in Jeju region and contributes to promote related studies by suggesting advanced skills in establishing policies on Jeju tourist food industry and food tourism.

The IEEE-1394 High Performance Serial Bus provides the real-time isochronous transport with capability of bandwidth reservation, however, there is a problem that isochronous bandwidth is used inefficiently on transmitting multimedia data with variable rate via the isochronous transport. To improve utilization of isochronous bandwidth, this paper proposes a bandwidth stealing technique to be able to recognize available isochronous bandwidth without any effects on current isochronous transmissions with reserved bandwidths, standard nodes and extended nodes with the bandwidth stealing technique can be connected together on the same bus. The ABR-1394 transport enables applications to utilize available isochronous bandwidth efficiently. In the respect of the utilization of available bandwidth, ABR-1394 is similar to the ABR service of ATM, however, it is different in that an ABR-1394 node itself can do flow control independently without any help from the network.
Alkaline protease inhibitor를 생산하는 해양유래 방선균의 탐색 및 동정

Alkaline protease는 식품 가공 공정에 있어 가공 원료의 선도 저하를 초래하거나 맛살류 제품 원료인 어류의 근육조직을 파괴하여 gel 구조를 파괴시키는 것으로 알려져 있다. 본 연구는 해양으로부터 alkaline protease에 대한 저해력을 가지는 방선균들을 분리하여 그 중 저해력이 가장 높은 C12 균주를 최종 선정하였다. 본 균주의 형태학적, 배양적 및 생리학적 특성을 조사한 결과, 포자의 크기는 2.0 ㎛로 외형은 원통형이고, 편모가 없으며, 포자형태는 smooth하였다. ISP 9 배지를 제외한 대부분의 배지에서 잘 성장하였다. 또한 15~50oC에서 잘 성장하였으며, 9% (w/v) NaCl이 포함된 배지에서도 성장하는 것으로 확인되었다. Gram 양성, citrate 음성, catalase 양성이었으며, melanin 색소를 생성하지 않았다. Starch, casein 및 gelatin 분해능이 있었으며, glucose, galactose, maltose, lactose, fructose 및 mannse 등은 잘 이용하였지만, sorbitol과 sucrose는 이용하지 않았다. 이러한 특성을 토대로 본 균주는 Streptomyces sp.로 확인되었다. 보다 정확한 균주 동정을 위하여 16S rDNA 염기서열 분석을 수행하였으며, 그 결과 C12 균주는 S. thermocarboxydus와 계통진화학적으로 가장 유연관계가 높았다. In this study, we screened and identified the bacterial strain showing high alkaline protease inhibitor activity from marine environment. Nine bacterial strains with alkaline protease inhibitor activity were isolated from marine sediments. Among them, strain C12 had the highest alkaline protease inhibitor activity and was selected for further taxonomical study. On the basis of morphological and physiological characteristics, strain C12 was identified as the genus Streptomyces. A phylogenetic analysis of the 16S rDNA showed that the isolated strain was actually a member of the genus Streptomyces, because the determined sequence exhibited a higher homology with Streptomyces thermocarboxydus. Morphological characteristics showed cylindrical spore chain and smooth spore surface by scanning electron microscope. Strain C12 was grown on all media except for ISP 9 agar. This strain could be grown in the medium containing up to 9% NaCl.

This study was carried out to develop a vine crusher for harvesting sweet potato. The experimental two-row vine crusher attachable to agricultural tractor composed of vine crushing part with frail type vine crushing blades and vine lifting blades, power transmission part with chain and gear transmission mechanism, crushing height control part with two control wheels and manual levers, and implement frames, was designed and fabricated. And this vine crushing performance was also analyzed. From vine crushing tests, backward travel direction (i.e., rotational direction of the vine crushing blades) showed better vine crushing performance than forward travel direction. Crushing ratio of remained vine was increased, and length of remained vine and length of crushed vine were decreased as working speed was decreased and rotational speed of vine crushing blades was increased. At a working speed of 0.27 m/s and rotational speed of vine crushing blades of 800 rpm, crushing ratio of remained vine was 98%, length of remained vine was 104 mm, and length of crushed vine was 327 mm. But, when crushing vine on irregular ridges, vines and mulching vinyl were wound in the vine crushing part. Therefore, change of location of power transmission chain mechanism, and an automatic control device for controlling crushing height were needed.
VAL의 FORALL 배열구문에 대한 효율적인 파이프라인 구성법에 관한 연구

본 논문은 정적 데이타 플로우 머신상에서 산술계산용 데이타 플로우 언어인 VAL의 FORALL 배열구문에 대한 파이프라인 구성방법에 관한 연구이다. VAL로 작성된 데이타 플로우 프로그램인 데이타 플로우 머신의 기계어에 해당하는 데이타 플로우 그래프로 컴파일될 때 컴파일하는 동안 비용면에서 우수한 소프트웨어 파이프라인을 구성하면서 파이프라인 구성으로 인한 처리량의 저하를 막기 위하여 불필요한 응답신호를 제거하고 두가지 수준에서 각기 다른 파이프라인 구성법을 사용하여 이 저하를 보충할 수 있는 방법을 고안하였다.
