        Response Regulator RssB의 활성 조절

        많은 세균들은 환경적 스트레스에 대항하기 위해 세균 생존에 유용한 특정 유전자들의 전사를 유도하는 대체시그마 인자 RpoS를 활용한다. 세포 내 RpoS 단백질의 농도는 주로 ClpXP 단백질 분해효소의 조절을 통해 결정된다. RpoS를 ClpXP로 전달하기 위해서는 adaptor 단백질 RssB가 반드시 필요하다. Two-component-type response regulator RssB는 RpoS와 지속적으로 상호작용을 하지만, 다양한 환경변화에 의해 RssB-RpoS 상호작용이 억제되어 세균에서 RpoS 양을 증가시킨다. 본 총설에서는 최근까지 연구 된 RssB-RpoS 상호작용에 관여하는 RssB의 anti-adaptor 단백질 IraD, IraM, IraP 등의 조절인자들과 RssB의 N-terminal 수용체 도메인의 인산화에 대해 설명하고 요약하였다. 이러한 RssB의 정교한 활성을 통한 RpoS 분해조절 과정은 외부환경 스트레스로부터 보다 효율적으로 세균을 보호할 수 있다. Against environmental stresses, many bacteria utilize the alternate sigma factor RpoS that induces transcription of the specific set of genes helpful in promoting bacterial survival. Intracellular levels of RpoS are determined mainly by its turnover through proteolysis of ClpXP protease. Delivery of RpoS to ClpXP strictly requires the adaptor protein RssB. The two-component-type response regulator RssB constantly interacts with RpoS, but diverse environmental changes inhibit this interaction through modification of RssB activity, which increases RpoS levels in bacteria. This review discusses and summarizes recent findings on regulatory factors in RssB-RpoS interactions, including IraD, IraM, IraP anti-adaptor proteins of RssB and phosphorylation of N-terminal receiver domain of RssB. New information shows that the coordinated regulation of RssB activity in controlling RpoS turnover confers efficient bacterial defense against stresses.

        전기영동적층법을 통한 판상 알루미나 입자와 전기영동 수지의 배향 유무기 복합체 제조 및 물성평가

        Plate-shaped inorganic particles are coated onto a stainless steel substrate by the electrophoretic deposition of a precursor slurry which includes the inorganic particles of $Al_2O_3$ and polymer resin in mixed solvents to mimic the abalone shell structure, which is a composite of plate-shaped inorganic particles and organic interlayer binding materials with a layered orientation. The process parameters of the electrophoretic deposition include the voltage, coating time, and conductivity of the substrate. In addition, the suspension parameters are the particle size, concentration, viscosity, conductivity, and stability. We prepared an organic-inorganic composite coating with a high inorganic solid content by arraying the plate-shaped $Al_2O_3$ particles and electrophoretic resin via an electrophoretic deposition method. We analyzed the effect of the slurry composition and the electrophoretic deposition process parameters on the physical, mechanical and thermal properties of the coating layer, i.e., the thickness, density, particle orientation, Young's modulus and thermogravimetric analysis results.

        추출조건에 따른 귀리 추출물의 면역활성

        추출조건에 따른 귀리의 crude ${\beta}$-glucan에 대한 면역증진효과을 살펴보기 위해 추출온도, 에탄올 농도 및 pH를 달리하면서 얻은 ${\beta}$-glucan 분획 중 in vitro 실험에서 항암성이 우수하게 나타난 5개 분획인 A분획 ($55^{\circ}C,\;5%,\;pH\;6$) B분획($45^{\circ}C,\;15%,\;pH\;6$), C분획 ($50^{\circ}C,\;20%,\;pH\;7$), D분획 ($50^{\circ}C,\;0%,\;pH\;7$) 및 E 분획($50^{\circ}C,\;10%,\;pH\;9$)를 대상으로 복강 큰 포식세포의 nitric oxide 생성능과 경구투여 후 혈액중의 carbon 탐식효율을 조사하였다. In vitro 실험에서 ${\beta}$-glucan을 $10{\sim}1,000{\mu}g/mL$ 범위에서 단독으로 또는 $IFN-{\gamma}$와 병행 처리했을 때 모두 대식 세포의 nitric oxide 생성량은 처리 농도에 비례하여 증가하였다. 단독 처리시에는 고용량일 때($1,000{\mu}g/mL$) 모든 분획에서 nitric oxide 생성량이 유의적으로 증가하였다. $IFN-{\gamma}$와 병행 처리시에는 C분획을 제외한 모든 분획의 고농도 처리구에서 유의적으로 증가하였는데, 특히 E분획은 $100{\mu}g/mL$의 낮은 농도에서도 효율적인 증가를 보였다. In vivo 실험에서 100mg/kg의 ${\beta}$-glucan 분획을 7일간 경구투여한 후 carbon 탐식효율을 조사한 결과 B, D 및 E 분획에서 유의적인 효과를 나타내었다. 이상의 결과를 종합해볼 때, ${\beta}$-glucan은 고용량일 때 직접적으로 또는 $IFN-{\gamma}$ 존재시에는 저용량에서도 복강 큰 포식세로를 활성화시킬 뿐 아니라, 탐식효율도 높임으로써 면역기능을 증진 시키는 것으로 나타났고, 그 효과는 crude ${\beta}$-glucan의 추출조건에 따라 달라지는 것을 알 수 있었다. Immunomodulatory activities of crude ${\beta}$-glucans extracted from oat bran under different conditions, fractions A ($55^{\circ}C,\;5%,\;pH\;6$), B ($45^{\circ}C,\;15%,\;pH\;6$), C ($50^{\circ}C,\;20%,\;pH\;7$), D ($50^{\circ}C,\;0%,\;pH\;7$), and E ($50^{\circ}C,\;10%,\;pH\;9$) were investigated. All crude ${\beta}$-glucan fractions stimulated macrophages, producing nitric oxide dose-dependently, and, efficiently promoted nitric oxide production in presence of IFN-${\gamma}$. Except for fraction C, in vivo test indicated fractions B, D, and E (100 mg/kg) substantially enhanced carbon-phagocytic indices of blood macrophages by oral administration of crude ${\beta}$glucan for 7 days prior to carbon injection. These immunomodulatory effects could be determined with extraction conditions of crude ${\beta}$-glucan.

        지방자치단체 자체평가모형에 관한 연구

        This article is about to search for the appropriate self-evaluation model for Korea's local governments. For this purpose, comparative analysis among three kinds of self-evaluation model is carried out on the basis of some selection criteria, which covers relevance, user-friendliness, cost requirement, inclusiveness, process, comparative ability, and objectivity. Furthermore, two lines of empirical inquiry has been performed to find out the perception and preference of local government officers.The findings are as follows:1) Local government officers prefer the self-evaluation tool which is objective is measurement, relevant to local circumstances, comprehensive in contents, and ease to use.2) As a results, it is identified that performance evaluation model was more appropriate for self-evaluation than program analysis or agency evaluation.But the local evaluation capacity is too weak to perform the performance evaluation, because of the lack of the personnel who takes charge of evaluation job, and the insufficiency of practitioner's knowledge and skill. Therefore, evaluation capacity of local governments has to be built up to operate the performance evaluation system.

        자체감사기구의 평가기준 개발

          This study intends to develop desirable criteria for evaluating comprehensive performance of an audit unit in different levels of government and other public authorities. These criteria can help improve the effectiveness of performance evaluation system of an audit unit in public organizations.   For this purpose, a normative approach was used to extract evaluation criteria from existing government audit standards of domestic and foreign countries. Based on 44 audit standards, the survey for audit staffs was conducted to determine the importance and feasibility of those standards. According to the result of the survey, 36 standards were selected as important evaluation criteria.   Furthermore, a simple validity test for the criteria was conducted using 2006 survey data gathered by the Board of Audit and Inspection of Korea(BAI). Due to the limited data, however, only 19 criteria were tested to check the relation to the effectiveness of self-audit and 10 criteria were found be relevant to the effectiveness of self-audit. recognized to be relevant to the effectiveness of self-audit.

        본 논문은 LMS와 함께 e-러닝 시스템을 구성하는 중요한 요소 중 하나인 학습 콘텐츠 관리 시스템을 본래의 목적인 콘텐츠 정보, 자원의 재사용이 가능하도록 하는 모형을 제시하였다. 콘텐츠 분류체계, 콘텐츠 구성 및 등록, 메타 데이터 조회 기능, 자원 업로드 및 연결 기능, 재조합 생성 기능 등을 기본 구성요소로서 설계한 것이다. 이 설계를 바탕으로 LCMS의 구현이 가능할 것이며, 한국전력의 요구사항에 알맞은 설계내용으로 전력산업분야 e-러닝을 체계적으로 지원하고, 저비용 고효율 콘텐츠를 만드는데 도움이 될 것이다.

        지방자치단체 평가의 운영실태분석 -메타평가 모형 활용을 중심으로-

        This study set out to examine the perceptions of citizens and experts regarding local government evaluations, to diagnose the problems with the evaluations, and to suggest future directions for improvement. The meta evaluation model was employed to analyze the status quo of local government evaluations. As a result, the respondents had negative cognitions of the evaluations from every aspect of meta evaluation including the conditions, activities and outcomes. The current evaluation system of local governments, in particular, failed to provide the citizens with necessary information. The evaluation results did not provide feedback to the improvement of local governments. It means that the effectiveness of local government evaluations performed by the central government is very low, and the current evaluation system should be reviewed. Given that the mission of local governments are to vitalize the local community and enhance the life quality of the citizens, local government evaluations should be based on a model that allows for effective assessment of the performance of these two strategic goals.

        회계비리 근절을 위한 내부통제 개선 연구: 여수시 횡령사건을 중심으로

        The purpose of this study is to evaluate internal controls in local governments and suggest how to improve the accounting control system, especially focused on an embezzlement case involving a low-ranking public official in Yeosu City Government. Through evaluating internal controls in Yeosu City Government, based on five elements of internal control, it was found that internal control system did not work at all in Yeosu City Government. The findings are as follows: (ⅰ) Despite the importance of a strong internal control environment, the government`s internal controls became weak because local high-ranking officials took the lead in committing an irregularity. (ⅱ) Before opening the Expo 2012, the government did not evaluate and manage internal and external risks associated with accounting operations. (ⅲ) Due to the lack of segregation of duties, one public official was in complete control of the situation regarding the entire accounting transaction, resulted in significant financial loss. Accounting Information System, namely e-Hojo, was circumvented by hand work. Besides, control activities including reconciliation was performed as a mere formality. (ⅳ) Formal and informal information and communication system was not successful. (v) Top management did not periodically review and evaluate internal control system. As a result, such overall weak system of internal control incurred the embezzlement case of Yeosu City Government. Therefore it could be urgent for local governments to effectively establish and maintain internal control mechanisms that are necessary to prevent accounting fraud.
