        항균성 동치미액의 첨가에 의한 냉면국물 중의 Listeria monocytogenes 및 Escherichia coli O157:H7 생육억제

        동치미액은 전통적으로 냉면국물로 이용되어왔다. 냉면국물의 미생물 오염문제를 해결하기 위하여 항균활성이 높은 김치 젖산균 Lactobacillus homohiochii B21과 Leuconostoc mesenteroides subsp. mesenteroides C16을 스타터로 혼합사용하여 동치미액을 제조하고, 이를 냉면국물에 100%, 50%, 10% 및 0% 첨가하여 20℃ 및 10℃에서 보관할 때에 동치미액의 항균력으로 인하여 의도적으로 첨가한 Listeria monocytogenes와 Escherichia coli O157:H7의 증식이 억제되는 정도를 조사하였다. 동치미액 100%인 냉면국물은 20℃에서 보관할 때에 Listeria monocytogenes는 8시간만에, Escherichia coli O157:H7는 40시간만이 10^6CFU/ml에서 10^0CFU/ml로 각각 급격히 사멸하였고, 10℃에서 보관할 때에도 시간경과에 따라 생균수가 급격히 감소하였으나 감소속도는 20℃에서 보관할 때보다 느렸다. 동치미액을 50% 첨가한 육수에서도 Listeria monocytogenes와 Escherichia coli O157:H7는 시간경과에 따라 급격히 감소하였으며, 감소속도는 동치미액 100%일 경우보다 느렸다. 동치미액을 10% 첨가한 육수는 20℃에서 보관할 때는 초기에 Listeria monocytogenes 및 Escherichia coli O157:H7의 균수증가가 약간 있었으나 동치미액 무첨가에 비하여 현저히 억제되었으며, 16시간 이후부터는 균수가 서서히 감소하였다. 동치미액을 첨가하지 않은 육수는 20℃에서 보관할 때는 초기부터 Listeria monocytogenes 및 Escherichia coli O157:H7의 균수증가가 급격히 이루어졌으며, 10℃에서 보관할 때는 24시간 이후부터 서서히 증가하였다. Listeria monocytogenes 및 Escherichia coli O157:H7에 대한 본 동치미액의 항균활성은 후자의 미생물보다 전자에 대하여 더욱 강하게 나타났다. 본 연구의 동치미액을 육수에 50% 첨가하여 냉면국물을 제조한다면 보관중의 미생물 오염문제 해결에 큰 도움이 될 것으로 판단된다. Juice of Dongchimi, a Korean traditional vegetable food fermented with lactic acid bacteria has been traditionary used as broth for Naengmyon, a Korean cold noodles with broth. This study was carried out to demonstrate the growth inhibition of two food born pathogens, Listeria monocytogenes and Escherichia coli O157:H7, in Naengmyon-broth containing Dongchimi-juice fermented with high antibacterial lactic acid bacteria, Lactobacillus homohiochii B21 and Leuconostoc mesenteroides C16. Naengmyon-broths were made with beef broth and Dongchimi-juice fermented with lactic acid bacteria, and the changes in viable cell counts of the inoculated pathogens in Naengmyon-broths were investigated during storage at 20℃ and 10℃. In Naengmyon-broth of 100% Dongchimi-juice stored at 20℃, the numbers of Listeria monocytogenes and Escherichia coli O157:H7 were rapidly decreased from 10^6CFU/ml to 10^0CFU/ml in 8 hours and 40 hours, respectively. In Naengmyon-broth containing 50% Dongchimi-juice, their numbers were also rapidly decreased, though the decreasing rates were not so fast as those in 100% Dongchimi-juice. In Naengmyon-broth containing 10% Dongchimi-juice, the growths of the two pathogens were markedly inhibited, compared with those in 100% beef broth, though some growths were occurred in early phase. But in Naengmyon-broth of 100% beef broth, their growths were very fast from early. Antibacterial activity of the Dongchimi-juice was more distinct at 20℃ than at 10℃, and was more active against Listeria monocytogenes than against Escherichia coli O157:H7.

        옥수수주정박의 급여가 산란계 생산성에 미치는 효과

        A layer feeding trial was conducted for 10 weeks to investigate the effects of the addition of corn distillers dried grains with solubles (DDGS) to layer diets on the laying performance, egg qualities, and yolk fatty acid composition. Nine hundred Hyline Brown layers, 24 weeks of age, were randomly allotted to 20 replicate laying cages, 45 birds per replicate. There were four diet treatments (0, 10, 15, and 20% DDGS), and five replicates per treatment. All experimental diets were prepared to contain iso-protein (17%) and iso-calorie (TMEn 2,780 kcal/kg). The use of DDGS up to 20% level in layer diets did not exert any influence on feed intake, laying rate, total egg mass, mean egg weight, and feed conversion ratio. DDGS did not exert any influence in weight of egg, breaking strength, and color of eggshell. The albumen height and Haugh unit was not influenced by DDGS addition. The yolk color was significantly increased by DDGS supplementation. As the DDGS level increased, the oleic acid content decreased, and the linoleic acid increased (P<0.05). The degree of saturation of yolk fatty acids was not affected by dietary DDGS. The inclusion of DDGS up to 20% in layer diets resulted in the decrease of feed cost per kg without any effect in the laying performance. In conclusion, the use of DDGS up to 20% level in layer diets could replace corn and soybean meal without any harmful effect on the laying performances.

      • Bacillus sp. YJ-63이 생산하는 항곰팡이 항생물질의 구조분석

        토양으로부터 분리한 Bacillus sp. YJ-63으로부터 항진균성 항생물질을 분리하여 그 구조를 분석한 결과는 다음과 같다. 항생물질은 7개의 α-amino acid와 1개의 β-amino acid가 결합한 환상구조를 가지는 iturin E 계에 속하며 특히, β-amino acid의 methylene 수가 기존의 iturin E 보다 더 많은 것으로 나타났다. Structural analysis was performed by the ^1H-NMR, ^13C-NMR, amino acid composition analysis and FAB-mass. The instrumental analysis represented that the potential antifungal antibiotic belonged to the iturin E group antibiotic, consisting of 7 α-amino acid residues and a collection of β-amino acid with aliphatic side chain. Compared to the iturin E group, notably, the potent antifungal antibiotic from bacillus sp. YJ-63 carried longer β-amino acid side chain. In conclusion, these findings identified a potential antibiotic, which contained a stable cyclopeptide structure with long β-amino acid side chain.

      • 고위험 유방암환자에서 CPB(Cyclophosphamide, Cisplatin, BCNU) 고용량 화학요법과 자가조혈모세포이식술 : A Phase II Multi-Center Trial

        배경: 림프절 전이가 많은 고위험군 유방암환자에서 CPB(cyclophosphamide, cisplatin, BCNU)를 이용한 고용량화학요법 및 자가조혈모세포이식술이 무병생존율에 미치는 영향과 안전성을 규명하기 위하여 연구를 시행하였다. 방법: 1998년 2000년 까지 한양대학병원, 가천의대병원과 인하대학병원에서 4개 이상 림프절을 침범한 유방암으로 근치적 절제술 시행 후 고용량 화학요법과 자가조혈모세포이식을 시행 받은 13명의 환자를 대상으로 하였고 대조군은 4개 이상 림프절을 침범한 유방암으로 보조화학요법을 받은 환자들 중 39명을 무작위로 선택하여 비교분석하였다. 고용량 화학요법은 제-6일에 BCNU 450 mg/m², 제-5일부터 3일까지 cyclophosphamide 2.0 g/m²와 cisplatin 55 mg/m²를 각각 투여하였다. 결과: 채집된 단핵구수의 중앙값은 8.5 x 10^(8)/kg (범위 2.44-17.90)이었고 CD34양성세포수의 중앙값은 11.0 x 10^(6)/kg (범위 0.8-78.35)이었다. 모든 환자가 이식 후 중앙치 10.0일(범위 8-15)에 말초혈액 과립구가 500 /mm³에 도달하였고 중앙치 9.0일(범위 0-18)에 혈소판이 20,000 /mm³에 도달하였다. 고용량 화학요법을 시행 받은 환자군의 추적기간의 중앙값은 24.6 개월 (범위 15.5-40.5)이었으며 총 13명의 환자 중 1명이 뇌전이로 인하여 사망하였으며 3명(23.1%)에서 재발이 관찰되었다. 대조군은 재발율이 25.6%이었고 무병생존율은 고용량 화학요법을 시행 받은 군(70.5%)과 대조군(78.8%) 사이에서 통계학적으로 의미 있는 차이를 나타내지 않았다 (P=0.744). 고용량 화학요법을 시행 받은 환자 중 10명 76.9%에서 발열이 관찰되었고 WHO 기준 3도 및 4도의 부작용은 혈액학적 부작용이 6예, 소화기적 부작용이 6예에서 관찰되었다. 결론: 고위험군 유방암 환자에서 CPB를 이용한 고용량화학요법 및 자가조혈모세포이식술은 안전하게 적용할 수 있는 치료의 한 방법이나 고용량 화학요법의 효과에 대해서는 앞으로 발표될 대규모 3상 연구의 결과를 주시해야 할 것으로 생각된다. Background: Despite the improvement of prognosis by multimodality therapy for the treatment of breast cancer, primary breast cancer patients with multiple involved axillary lymph node still have a high risk of recurrence. Methods: Between January 1998 and May 2000, we treated 13 women with histologically proven stage II or III breast cancer with 4 or more axillary lymph nodes at the time of primary mastectomy with high-dose chemotherapy and autologous stem cell transplantation. Results: The median age was 45 years (range: 32-57) and the median number of involved lymph node was 13 (range: 6-31). Ten patients were premenopausal state and 40 percent were positive hormone receptor. Seven patients received radiation therapy. Patients were treated with high-dose CPB (cyclophosphamide 2 g/m² D-5--3, cisplatin 55 mg/m² D-5--3, BCNU 450 mg/m² D-6) with autologous peripheral blood stem cell transplantation after 2-4 cycles of adjuvant chemotherapy (CAF, CEF or AC). We compared those patients with age and other clinical factors-matched 39 patients who received conventional adjuvant chemotherapy. The median follow-up for surviving patients who received high-dose chemotherapy was 25.6 months. The relapse rate and 2-year relapse-free survival was 23.1%, 25.6% and 70.5%, 78.8% for high-dose and conventional dose chemotherapy, respectively, but there was no significant difference between two groups (P>0.05). Conclusion: Further follow-up and additional studies are required to resolve the role of high-dose chemotherapy with autologous stem cell transplantation in high-risk breast cancer.

        Performance Evaluation of VITEK MS for the Identification of a Wide Spectrum of Clinically Relevant Filamentous Fungi Using a Korean Collection

        The correct identification of filamentous fungi is challenging. We evaluated the performance of the VITEK MS v3.0 system (bioMérieux, Marcy-l’Étoile, France) for the identification of a wide spectrum of clinically relevant filamentous fungi using a Korean collection. Strains that were added to the upgraded v3.2 database were additionally identified by the VITEK MS v3.2 system. Of the 105 tested isolates, including 37 Aspergillus (nine species), 41 dermatophytes (seven species), and 27 other molds (17 species), 43 (41.0%) showed “no identification” or “multiple species identification” results at the initial VITEK MS testing; these isolates were retested using the same method. Compared with sequence-based identification, the correct identification rate using VITEK MS for Aspergillus, dermatophytes, other molds, and total mold isolates was 67.6%, 56.1%, 48.1%, and 58.1% at the initial testing and 94.6%, 78.0%, 55.6%, and 78.1% with retesting, respectively. Following retesting, 19 (18.1%) and two (1.9%) isolates showed “no identification” and “misidentification” results, respectively. VITEK MS reliably identified various filamentous fungi recovered in Korea, with a very low rate of misidentification.

      • Identification of Tumor Suppressor Loci on the Long Arm of Chromosome 5 in Pulmonary Large Cell Neuroendocrine Carcinoma

        <P>BACKGROUND: A recent cytogenetic analysis of non-small cell lung cancer revealed hot-spot regions for deletion on the long arm of chromosome 5 and suggested the existence of putative tumor suppressor genes in that region. However, similar studies on genetic alterations in large cell neuroendocrine carcinoma (LCNEC) have been very limited. To our knowledge, this is the first report to screen for the loss of heterozygosity (LOH) and to examine the location of putative tumor suppressor genes on chromosome 5q in LCNEC. OBJECTIVES: To identify tumor suppressor loci on chromosome 5q in LCNEC by microsatellite analysis. PATIENTS AND METHODS: Microsatellite instability and LOH in tumor and normal tissue samples from 13 patients with LCNEC, who had undergone surgical resections, were analyzed by polymerase chain reaction using a panel of 19 microsatellite DNA markers spanning chromosome 5q. RESULTS: LOH was found in all of the 13 tumors (100%) in at least one informative marker tested. The following four common minimally deleted regions were noticed on chromosome 5q: 5q14.3-q21.1; 5q22.2-q23.1; 5q23.3-q33.2; and 5q35.1-q35.2. Three of 13 individual tumors (23.1%) exhibited shifted bands for at least one of the tested microsatellite markers. Shifted bands occurred in 6 of 224 loci (2.7%) tested. CONCLUSION: These data suggest the presence of at least four tumor suppressor loci on chromosome 5q in LCNEC, and further investigations into cloning candidate tumor suppressor genes are warranted.</P>

        The MAKE Biomarker Discovery for Enhancing anTidepressant Treatment Effect and Response (MAKE BETTER) Study: Design and Methodology

        Objective Depression is associated with a major disease burden, and many individuals suffer from depressive symptoms due to an insufficient response to ostensibly adequate antidepressant treatment. Therefore, it is important to identify reliable treatment response predictors for use in developing personalized treatment strategies. Methods The MAKE Biomarker discovery for Enhancing anTidepressant Treatment Effect and Response (MAKE BETTER) study was performed to identify predictors of antidepressant response using a 2-year naturalistic prospective design. Participants in the MAKE BETTER study were consecutively recruited from patients who visited the Psychiatry Department of Chonnam National University Hospital, Gwangju, South Korea for treatment of a depressive disorder. Data on demographic and clinical characteristics, genetic markers measured by whole-exome sequencing, and blood markers were obtained. The types and doses of antidepressants were determined based on the clinical judgment of the psychiatrist, and the treatment outcomes (e.g., depressive and other psychiatric symptoms and issues related to safety) were assessed. Results We will be able to use the data collected in this study to develop a treatment-response prediction index composed of biomarkers. Conclusion The MAKE BETTER study will provide an empirical basis for a personalized medicine approach to depression by enabling the prediction of antidepressant treatment response according the characteristics of each patient. It will thereby support evidencebased decision-making that decreases the use of a trial-and-error approach to the treatment of depressive disorders.
