        한국잔디 수집계통들 중에서 우수계통들의 생육특성 비교

        2001년 수집된 한국잔디 133계통들 중에서 우수한 계통들을 선발하기위하여 밀도(품질),<br/> 피복성, 녹색기간, 내병성, 출수유무(종자수) 등의 주요특성과 그 외 생육특성들이 조사되었다. 생육특성이 우수한 6계통이 선발되었으며 그 결과를 요약하면 다음과 같다.<br/> 수집된 133계통들 중에서 가장 녹색기간이긴 계통은 J01067으로 11월초까지 녹색도를 유지하였다. J01106 및 J01129 계통들은 엽폭이 각각 1.5mm 및 2mm 정도로 매우 좁으며, 밀도가 높고, 피복성도 좋은 것으로 나타났으며, 내병성도 강하였다. J01122 계통은 들잔디 계통들 중에서 유일하게 내병성이 강한 계통으로 선발되었다. J01128 계통은 엽폭이 3.2mm로 중엽에 속하였으며, 대비품종인 Sunburst 와 비슷하였다. 이 계통의 경우도J01106, J01129 계통들과 마찬가지로 밀도가 높고 내병성이 강한 것으로 나타났다.<br/> 수집된 133계통들의 형태적 분류를 위하여 5가지의 생육특성을 사용하여 크게 세가지 군집<br/> 으로 분류하였으며, 그 중에서 대비품종으로 사용된 6품종들과 선발된 우수 6계통의 분류는<br/> 다음과 같다. 제 1군집에 속하는 품종 및 계통은 Belare, Meyer, 안양중지(Anyang-jungji), J01067, J01112 등이 포함되었으며, 제 2군집으로는 S-94, J01105가 포함되었고, 제 3군집에는 Sunburst, 건희(Konhee), J01106, J01128, J01129 등이 포함되었다. Growth characteristics such as density(quality), covering speed, green period, disease resistance, heading existence(number of seeds) and other characteristics were examined for selection of superior lines among the 133 zoysiagrass lines collected in 2001. Six superior lines were selected and the results were summarized as follows.<br/> Superior line, J01067 was longest for green period among the 133 zoysiagrasses and stayed green until the beginning of November. Leaf width of J01106 and J01129 lines was very narrow as 1.5㎜ and 2㎜, respectively and superior for density(quality), covering speed and disease resistance. J01122 line was selected for strongest disease resistance among the lines of Zoysia japonica. Leaf width of J01128 was 3.2㎜ showing midium type and similar to standard cultivar, Sunburst. This line also showed higher density and strong disease resistance like J01106 and J01129.<br/> Five growth characteristics for morphological classification of 133 zoysiagrass lines<br/> was used and divided into 3 cluster groups. Of 133 lines, 6 standard cultivars and 6 superior lines were classified as follows. First cluster group contained Belare, Meyer, Anyang-jungji, J01067, J01122, and second group contained S-94 and J01105, and third group contained Sunburst, Konhee, J01106, J01128 and J01129.<br/> <br/>

      • Lipopolysaccharide from <i>Prevotella nigrescens</i> stimulates osteoclastogenesis in cocultures of bone marrow mononuclear cells and primary osteoblasts

        <P>Background and Objective: </P><P>Lipopolysaccharide is thought to be a major virulence factor of pathogens associated with periodontal diseases and is believed to stimulate bone resorption <I>in vivo</I>. Although <I>Prevotella nigrescens</I> has been implicated in periodontitis, its role in osteoclastogenesis has not been reported. In this study, we investigated the effects of lipopolysaccharide from <I>P. nigrescens</I> on the formation of osteoclasts and the production of cytokines related to osteoclast differentiation.</P><P>Material and Methods: </P><P>Mouse bone marrow mononuclear cells were cultured in the presence of macrophage colony-stimulating factor (M-CSF) and receptor activator of nuclear factor &kgr;B ligand (RANKL), with or without lipopolysaccharide. Bone marrow mononuclear cells were also cocultured with calvarial osteoblastic cells in the presence or absence of lipopolysaccharide. Osteoclast formation was determined by tartrate-resistant acid phosphatase cytochemistry. The production of osteoprotegerin (OPG), M-CSF, tumor necrosis factor alpha (TNF-&agr;), transforming growth factor-beta (TGF-&bgr;) and prostaglandin E<SUB>2</SUB> (PGE<SUB>2</SUB>) was determined by enzyme-linked immunosorbent assay (ELISA).</P><P>Results: </P><P><I>P. nigrescens</I> lipopolysaccharide inhibited osteoclast differentiation from bone marrow mononuclear cells cultured in the presence of M-CSF and RANKL. However, in the coculture system, <I>P. nigrescens</I> lipopolysaccharide stimulated osteoclastogenesis. Notably, <I>P. nigrescens</I> lipopolysaccharide decreased OPG production but increased TGF-&bgr; secretion. In addition, treatment with <I>P. nigrescens</I> lipopolysaccharide increased PGE<SUB>2</SUB> production during the late stage of the culture period. There was no difference in M-CSF and TNF-&agr; production.</P><P>Conclusion: </P><P>These results demonstrate that <I>P. nigrescens</I> lipopolysaccharide stimulates osteoclastogenesis in the coculture system by decreasing the production of OPG and increasing the production of TGF-&bgr; and PGE<SUB>2</SUB>. Through the mechanisms involving these factors, <I>P. nigrescens</I> lipopolysaccharide may cause alveolar bone resorption in periodontal diseases.</P>

        New emergence pattern with variant porcine epidemic diarrhea viruses, South Korea, 2012-2015

        Since outbreaks of porcine epidemic diarrhea virus (PEDV) in the United States in 2013, explosive outbreaks of PED in South Korea have infected all age groups of pigs in 2014-2015year. This study analyzed a large collection of the Spike protein coding gene to infer the spatial-temporal diffusion history of PEDV. The studying results suggested that PEDVs in Korea belonged to different genogroups. While classical G1 was continuingly circulating between provinces of Korea, the pandemic G2a were recently introduced from China and USA. By the application of Bayesian phylogeographical analysis, this study demonstrated the spatial-temporal transmission of PEDVs within Korea. Of the recent emerged G2a viruses, J3142 strains showed potential recombination breakpoint (376-2,143nt) of S1 gene between KNU1303_Korea strain_G2a (KJ451046) and 45RWVCF0712_Thailand strain_G2b (KF724935). The pandemic G2a virus was partial neutralized by the antibodies invoked by the G1- based PED vaccine virus.

        The E3 ubiquitin ligase CHIP selectively regulates mutant epidermal growth factor receptor by ubiquitination and degradation

        Somatic mutation in the tyrosine kinase domain of epidermal growth factor receptor (EGFR) is a decisive factor for the therapeutic response to EGFR tyrosine kinase inhibitors (EGFR-TKIs) in lung adenocarcinoma. The stability of mutant EGFR is maintained by various regulators, including heat shock protein 90 (Hsp90). The C terminus of Hsc70-interacting protein (CHIP) is a Hsp70/Hsp90 co-chaperone and exhibits E3 ubiquitin ligase activity. The high-affinity Hsp90-CHIP complex recognizes and selectively regulates their client proteins. CHIP also works with its own E3 ligase activity independently of Hsp70/Hsp90. Here, we investigated the role of CHIP in regulating EGFR in lung adenocarcinoma and also evaluated the specificity of CHIP's effects on mutant EGFR. In HEK 293T cells transfected with either WT EGFR or EGFR mutants, the overexpression of CHIP selectively decreased the expression of certain EGFR mutants (G719S, L747_E749del A750P and L858R) but not WT EGFR. In a pull-down assay, CHIP selectively interacted with EGFR mutants and simultaneously induced their ubiquitination and proteasomal degradation. The expressions of mutant EGFR in PC9 and H1975 were diminished by CHIP, while the expression of WT EGFR in A549 was nearly not affected. In addition, CHIP overexpression inhibited cell proliferation and xenograft's tumor growth of EGFR mutant cell lines, but not WT EGFR cell lines. EGFR mutant specific ubiquitination by CHIP may provide a crucial regulating mechanism for EGFR in lung adenocarcinoma. Our results suggest that CHIP can be novel therapeutic target for overcoming the EGFR TKI resistance.

      • 이동 작동기형 인공심장의 모터부와 작동기부의 효율 분석

        이동 작동기형 인공 심장의 모터부와 모터부를 포함한 작동기부의 효율 측정방법과 측정 결과이다. 이동 작동기형 인공심장은 에너지 변환기로 브러시 없는 직류 모터(S/M 506-26A, Sierracin/Magnedyne, Cafsbad, CA, U.S.A.)를 사용한다. 이 브러시 없는 직류 모터가 기어 열(Gear train)을 따라 원추운동을 하며 혈액주머니에 힘을 가하게 되고, 이 힘에 의해 혈액주머니 안의 혈액이 박출하게 된다. 효율 측정을 위하여 제어기 및 모터부, 기어부 및 작동기부, 혈액주머니 밀 기타부등 크게 세 부분으로 나누었으며, 각각을 다시 소자별로 나누어 입, 출력의 효율을 측정했다. 부하의 토크, 분당 회전수, 각속도, 인가되는 전압 등을 바꾸어 가면서 시스템의 효율을 측정헌고, 이 측정을 통해서 모터 운동시의 전체적인 효율과 부분별 효율을 알 수 있다. 특히 인공심장의 모터부와 작동기부의 효율측정에 중점을 두었다. 측정 결과 4ℓ/min 박출량의 경우, 이동 작동기형 인공심장의 전체 효율은 8 %정도이며, 구체적으로는 모터부에서 50 %정도, 작동기부에서 85 %정도, 그리고 혈액주머니에서 19 %정도의 査율을 보임을 확인했다. This is a test report of motor and actuator efficiency for the moving actuator type total artificial heart. Our moving-actuator type total artificial heart, as an energy converter, utilizes a brushless DC motor(S/M 566-26A, Sierracin/Magnedyne, Carlsbad, CA, U.S.A.) generating a pendulous motion of itself by an epicyclic gear train. It is necessary to know the overall efficiency of our system. We operated the motor at a variable ranee of torque, angular speed and width of voltage pulse. Measurements of power into the drive, from the drive to the motor, and out of the motor were taken in order to derive the efficiency of the drive, efficiency of the motor and overall efficiency for the system. Throughout the test, the power input to the drive, the power output from the drive, and the power output from the motor are measured simultaneously. We focused on efficiency of the motor and actuator parts in the TAH. In case of 4 ℓ/min condition, system efficiency is about 8 %, motor part is about 50%, actuator part is about 85 %. Some experimental results are provided to demonstrate the effectiveness of the presented approach.

      • Effects of E. Coli lipopolysaccharide on the pharmacokinetics of ipriflavone and its metabolites, M1 and M5, after intravenous and oral administration of ipriflavone to rats: Decreased metabolism of ipriflavone due to decreased expression of hepatic CYP1

        <P>It was reported that ipriflavone was primarily metabolized via hepatic CYP1A1/2 and 2C11 in rats. In the present study, the expression of CYP1A2 and 2C11 decreased in the liver, but increased in the intestine in rats pretreated with E. coli lipopolysaccharide (ECLPS; an animal model of inflammation). Thus, pharmacokinetic parameters of ipriflavone and its metabolites, M1 and M5, were evaluated in ECLPS rats. After intravenous administration (20 mg/kg) to ECLPS rats, the AUC of ipriflavone was significantly greater (26.7% increase) and CL<SUB>NR</SUB> of ipriflavone was significantly slower (19.9% decrease) than in the controls. This could have been due to decreased expression of hepatic CYP1A2 and 2C11 compared to the controls. After oral administration (200 mg/kg) to ECLPS rats, the AUC of ipriflavone was also significantly greater (130% increase) than in the controls. Although the expression of intestinal CYP1A2 and 2C11 increased in ECLPS rats, contribution of this increase to the significantly greater AUC of ipriflavone after oral administration of ipriflavone to ECLPS rats was not considerable. This could have also been due to a significantly decreased expression of hepatic CYP1A2 and 2C11 in ECLPS rats.         AFLP Marker를 이용한 한국 재래돼지의 유전적 다양성 및 품종식별

        본 연구는 한국재래돼지의 순수 혈통정립 및 유전자원 보존을 목적으로 AFLP 다형성을 분석하고 이를 유전적 표지인자로 이용하여 재래돼지의 유전적 특성을 분자 수준에서 규명하고 품종 집단내 유전적 변이성과 품종 특이적인 DNA marker를 탐색하며 동시에 타품종들과의 유전적 근연관계를 추정하기 위해 수행하였다. 13종류의 selective primer 조합형을 이용하여 분석한 결과 총 band의 수는 611개로 각 조합형 당 평균 47개의 band가 확인되었으며 이 가운데 다형적 band가 152개로 다형성 수준은 약24.5%였다. 한국 재래돼지의 다형율과 유전적 다양성 값은 각각 29.8%와 2.9로 개량종에 비해서 다소 높은 경향이었다. 한편, 13종류의 primer 조합형 가운데 E35/T38 및 E38/H13 primer 조합형에서 품종간의 차이를 나타내는 DNA marker가 확인되었다. 특히, 재래돼지에서 검출된 E35/T38 primer 조합형의 0.50kb, 0.25kb 및 0.38kb의 DNA band는 개량종과 명확히 구별되어 이들 품종 특이적 DNA band는 한국 재래돼지의 품종식별에 유용한 표지인자로 이용 가능할 것으로 기대된다. 품종간 유전적 근연관계에서 한국 재래돼지는 Hampshire종과 유전적 유사성이 가장 높은 것으로 추정되었다. 본 연구에서 검출한 한국 재래돼지의 AFLP 유전자 지문은 재래돼지 집단의 유전적 변이성 및 타 품종과의 근연관계 분석뿐만 아니라 경제형질과 연관된 marker 개발에 유용한 DNA 표지인자로 활용할 수 있을 것으로 기대된다. For the purpose of genetic conversation and utilization of Korean native pig(KNP) as a valuable animal genetic resource, DNA polymorphisms of AFLP (amplified fragment length polymorphisms) as genetic markers were analyzed in KNP and foreign pig breeds (Landrace, Duroc, Hamphsire and Yorkshire). Using these AFLP markers, the genetic structure and characteristics of KNP population were analyzed at the DNA level and the genetic variability and diversity within and between breed population were evaluated. Breed-specific DNA marker for KNP was screened, and phylogenetic relationship within and among breeds were estimated. A total of 611 AFLP markers were amplified by 13 selective primer combinations, and the average number of bands per primer combination was 47.0. Among them 152 bands were polymorphic (24.5%). The rate of polymorphisms and genetic diversity values of KNP (29.8% and 2.90) was higher than these of other foregin breeds. E35/T38 and E38/H13 primer combinations produced AFLP banding patterns which clearly discriminated between KNP and other foreign pig breeds. Three bands(0.50kb, 0.25kb and 0.20kb) identified in E35/T38 and two bands(0.45kb and 0.38kb) identified in E38/H13 primer combinations were present in all of the KNP examined, but not present in the foreign pig breeds. Therefore, these two primer combinations could be used as breed-specific DNA markers for breed identification of KNP. In comparison of genetic distances among pig breeds, KNP was the most closely related to the Hampshire breed. AFLP fingerprints may be useful for genetic variability and relationships and development of breed-specific DNA markers in pig breeds.

        본 연구에서는 사상관련전위인 수반음성변동(CNV)을 이용하여 쾌/불쾌 향의 영향을 평가하고자 하였다. 즉, 건강한 20대 성인을 대상으로 쾌한 향(레몬)과 불쾌한 향(E3) 자극에 따른 CNV의 전기성분 및 후기성분의 변화를 대뇌부위별, 가산횟수별로 비교 분석하였고, 쾌/불쾌 향의 자극 반복에 따른 주관적 평가도 부가하여 검토하였다. 그 결과, 쾌/불쾌 향은 CNV 후기성분의 중심엽 부위에서 10∼15회 가산평균의 경우 정량적으로 구별될 수 있는 가능성을 보였고, 주관적인 평가에서는 반복 자극횟수가 증가함에 따라 쾌/불쾌감이 저하하는 것을 알 수 있었다.

        EZH2 Generates a Methyl Degron that Is Recognized by the DCAF1/DDB1/CUL4 E3 Ubiquitin Ligase Complex

        Ubiquitination plays a major role in protein degradation. Although phosphorylation-dependent ubiquitination is well known for the regulation of protein stability, methylation-dependent ubiquitination machinery has not been characterized. Here, we provide evidence that methylation-dependent ubiquitination is carried out by damage-specific DNA binding protein 1 (DDB1)/cullin4 (CUL4) E3 ubiquitin ligase complex and a DDB1-CUL4-associated factor 1 (DCAF1) adaptor, which recognizes monomethylated substrates. Molecular modeling and binding affinity studies reveal that the putative chromo domain of DCAF1 directly recognizes monomethylated substrates, whereas critical binding pocket mutations of the DCAF1 chromo domain ablated the binding from the monomethylated substrates. Further, we discovered that enhancer of zeste homolog 2 (EZH2) methyltransferase has distinct substrate specificities for histone H3K27 and nonhistones exemplified by an orphan nuclear receptor, RORα. We propose that EZH2-DCAF1/DDB1/CUL4 represents a previously unrecognized methylation-dependent ubiquitination machinery specifically recognizing ''methyl degron''; through this, nonhistone protein stability can be dynamically regulated in a methylation-dependent manner.

        The purpose of this study was to know relationship between important variables which influence lifestyle in college women`s health promotion, prove adjustable factors and provide basic data which develops nursing intervention health promotion program. The method of this study was descriptive correlational study. The convenience sample was 206 college woman who live alone in lodgings, dormitory, a reading room or off-campus housing. Research instruments were the Multidimensional Health Locus of Control(MHLC) developed by Wallston, Wallston & Devellis(1978), Interpersonal Support Evaluation List developed by Cohern, Hoberman(1983) and The Health Promotion Lifestyle Profile(HPLP) developed by Walker, Pender & Sechrist(1978) and the data was used after Factor Analysis. The result of this study was follows. 1. It can be found that the level of the college woman`s health promotion lifestyle was 2.46±.33 with higher fulfilling level: According to general characteristic, the level of health promotion lifestyle was 5% significant statistical differences by age and period of menstruation. 2. The level of health locus of control was 3.25±.31 with higher fulfilling level. 3. The interpersonal support level was 3.05±.44 with higher fulfilling level. 4. The level of interpersonal support turned out most convincing factor in health promotion lifestyle (9%). In addition to age factor, two factors was 10% power of explanation in whole health promotion.
