        국립공원 관리를 위한 해양생태계 장기 모니터링 프로그램의 발전 방향

        생태계 변화의 모니터링은 효율적 생태계 관리를 위한 기초 정보 수집을 위해 중요한 과정이다. 많은 생태 현상과 그 과정은 오랜 시간에 걸쳐 진행되므로 국립공원의 지속적인 보전 및 효율적 관리를 위해 생태계 모니터링은 장기적으로 이루어질 필요가 있다. 본 연구는 국내외 장기 모니터링 프로그램 사례 분석을 통해 성공적인 장기 모니터링을 위한 방법으로써, 최적화된 모니터링 설계, 표준화된 운용방안 마련, 인적 자원 및 DATA 관리 체계 수립, 안정적인 재정 마련 및 후속지원 등을 제시하였다. 이를 바탕으로, 현재 10년 넘게 수행되고 있는 ‘HS호 유류유출 사고 해역 장기 모니터링 프로그램’의 발전 방향으로 적응형 모니터링 방식이 적용된 표준화된 프로토콜의 정립, QA/QC와 지속적인 자료 관리, 조사 항목의 연계성 강화 등의 구체적 방안을 제안한다. Monitoring ecosystem changes is an important process for collecting basic information for ecosystem management. Since many ecological phenomena and their processes take place over a long period of time, it is necessary to conduct long-term monitoring of ecosystems for the continuous conservation and efficient management of national parks. This study suggested an optimized monitoring design, standardized operation plan, human resource and data management system, stable financial preparation and follow-up support as methods for successful long-term monitoring through case analysis of domestic and foreign long-term monitoring programs. Based on this, it proposes specific measures such as establishing a standardized protocol with adaptive monitoring methods, continuous data management with QA/QC, and strengthening the linkage of investigation items in the development direction of the “HS oil spill accident long-term monitoring program” that has been conducted for more than a decade.

        Sport has taken a position as significant influence all over the world for a long time. These days, sport must exist with us in order to provide pleasure, so people have a tendency to expect it what happens to them. Therefore, a number of sport teams and competitions are able to stimulate people to be interested in sport. Sport event, information, equipment, and sport player management need systems which accompany with security and pleasure because the systematical managements establish security, and help sport players not to make an error that is a fountain of good quality game. Thus, sport players improve their ability, so the audience are willing to pay to see the game. In order to extend the power of sport influence, sport should be organized in terms of managing sport processes well. Unfortunately, the system of management for sport is not still adequate and organized in Korea. In other words, many materials must be sporadic, and newspapers or magazines have treated of sport management with fragmentary knowledge so far. Today, systematical management is essential to all segments of the sport industry, professional or amateur, school or club, private or government at any level community or individual. Hopefully, this research will be instructive as a starting point to derive a special quality of sport management which is suited to korea sport market by comparing to other management systems in order to formulate organized management industry.

      • 중증 재생불량성 빈혈 환자의 동종골수이식 후에 병발한 간정맥폐색질환(Hepatic Veno-Occlusive Disease)의 임상 경과 및 혈액응고 인자들의 변화

        연구배경: 재생불량성 빈혈 경우에는 조혈모세포 이식 후 VOD의 발생에 관한 보고가 거의 없는 실정이다. 저자 등은 cyclophosphamide와 antithymocyte globulin으로 전처치 후에 동종골수이식을 시행한 재생불량성 빈혈 환자들을 대상으로 하여 VOD의 발생과 임상 양상, 혈액응고 인자들의 변화에 관하여 분석하여 보았다. 방법: 재생불량성 빈혈로 동종골수이식을 시행 받은 13예 환자를 대상으로 하였다. 임상적 기준에 따라 VOD를 진단하였으며 임상 경과에 따라서 경중을 분류하였다. Protein C, protein S, antithrombin Ⅲ, tPA, PAI-1 등의 혈액응고 인자들을 전처치 투여전, 골수 주입당일, 이식 후 제7, 14, 21일에 측정하였다. 결과: 7예(53.8%)에서 VOD가 발생하였으며, 경증 5예, 중등도 2예였다. VOD군에서 혈소판치가 20.000/㎕ 이상을 유지하는 데까지 걸리는 시간이 더 길었다(p=0.026). 또한 VOD군에서 적혈구 및 혈소판의 수혈 요구량이 각각 유의하게 많았다(p=0.025, 0.015). 혈액응고 인자들의 변화를 보면, 제 0일에 VOD군에서 PAI-1의 혈중 농도가 유의하게 더 높았다.(p=0.034). 결론: 재생불량성 빈혈환자에서도 조혈모세포 이식 후에 VOD의 높은 발생률을 관찰하였으며, 대부분이 경증이었다. VOD 환자에서의 혈액응고 인자들의 변화는 PAL-1이 VOD의 감별 진단을 위하여 이용될 수 있음을 시사하고 있다. Background: There are few reports about the occurrence of hepatic veno-occlusive disease(VOD) after bone marrow transplantation (BMT) for severe aplastic anemia (SAA). We analyzed the incidence and clinical outcomes of VOD and changes of coagulation factors in patients with SAA who were conditioned with cyclophosphamide plus ATG and underwent allogeneic BMT. Methods: Thirteen patients with SAA who underwent allogeneic BMT were included. A diagnosis of VOD was made according to clinical criteria, and severity of VOD was classified according to clinical course. Plasma levels of protein C, protein S, antithrombin Ⅲ, t-PA, and PAI-1 were determined before preparative chemotherapy, in the day of marrow infusion, and days 7, 14, and 21. Results: VOD occurred in 7/13(53.8%). Five were mild VOD and two were moderate VOD. Time to platelet ≥20,000/L was slower (p=0.026) and red cell and platelet transfusion requirements were higher (p=0.025,0.015) in patients with VOD . The mean plasma level of PAI-1 at 0 were higher in patients with VOD (p=0.034). Conclusion: We observed the high incidence of VOD after allogeneic BMT in patients with SAA. A majority of cases of VOD were mild. The changes of PAI-1 after BMT may be useful marker of hepatic VOD

      • Status of <i>Taylorella equigenitalis</i> Infection in Thoroughbred Horses in the Republic of Korea and the Molecular Characterization of the Korean <i>Taylorella equigenitalis</i> Isolates

        <P><B>Abstract</B></P> <P>Contagious equine metritis is a highly transmissible venereal disease of horses caused by the bacterium, <I>Taylorella equigenitalis</I>. After the first identification of <I>T. equigenitalis</I> in Korea in 2015, targeted surveillance of Thoroughbreds was conducted using real-time polymerase chain reaction. Initially, the prevalence of <I>T. equigenitalis</I> in the Thoroughbred population tested was 13.5% (71/526). All positive animals were placed under quarantine and treated according to previously described protocols (Jeoung et al., J Equine Vet Sci, 2016; 47:42–6). After the initial outbreak identification, annual national surveillance for <I>T. equigenitalis</I> was conducted from 2015 to 2017, and by 2017, prevalence was reduced to 1.0% (22/2,171). Most positive cases were located in Jeju province, the major horse-rearing region in Korea. A total of 12 <I>T. equigenitalis</I> strains were isolated during the surveillance period, and multilocus sequence typing (MLST) and eBURST analysis were applied to determine epidemiological characteristics. All Korean strains showed a unique MLST allelic profile (1-2-1-2-2-2-1; <I>gltA</I>-<I>gyrB</I>-<I>fh</I>-<I>shmt</I>-<I>tyrB</I>-<I>adk</I> in order) and were classified as sequence type 55. In addition, they showed little relationship with <I>T. equigenitalis</I> strains isolated from other countries. Considering the unique biological and genetic characteristics of the Korean isolates, effective quarantine measures and continued surveillance will be required to eradicate the disease and prevent incursions of additional <I>T. equigenitalis</I> strains into Korea.</P> <P><B>Highlights</B></P> <P> <UL> <LI> In Korea, a national <I>T. equigenitalis</I> surveillance program conducted in Thoroughbred horses showed a decrease in prevalence from 13.5% in 2015 to 1.0% in 2017. </LI> <LI> Characterization of 12 Korean <I>T. equigenitalis</I> isolates showed identical bacteriological properties and demonstrated a unique multilocus sequence typing (MLST) profile (1-2-1-2-2-1). </LI> <LI> All Korean <I>T. equigenitalis</I> isolates were classified as MLST sequence type 55 (ST55) and showed little genetic relationship with isolates from other countries. </LI> </UL> </P>

        Flavonoids Inhibit Histamine Release and Expression of Proinflammatory Cytokines in Mast Cells

        Mast cells participate in allergy and inflammation by secreting inflammatory mediators such as histamine and proinflammatory cytokines. Flavonoids are naturally occurring molecules with antioxidant, cytoprotective, and antiinflammatory actions. However, effect of flavonoids on the release of histamine and proinflammatory mediator, and their comparative mechanism of action in mast cells were not well defined. Here, we compared the effect of six flavonoids (astragalin, fisetin, kaempferol, myricetin, quercetin, and rutin) on the mast cell-mediated allergic inflammation. Fisetin, kaempferol, myricetin, quercetin, and rutin inhibited IgE or phorbol-12-myristate 13-acetate and calcium ionophore A23187 (PMACI)-mediated histamine release in RBL-2H3 cells. These five flavonoids also inhibited elevation of intracellular calcium. Gene expressions and secretion of proinflammatory cytokines such as tumor necrosis factor-$\alpha$ (TNF-$\alpha$), interleukin (IL)-$1{\beta}$, IL-6, and IL-8 were assessed in PMACI-stimulated human mast cells (HMC-1). Fisetin, quercetin, and rutin decreased gene expression and production of all the proinflammatory cytokines after PMACI stimulation. Myricetin attenuated TNF-$\alpha$ and IL-6 but not (IL)-$1{\beta}$ and IL-8. Fisetin, myricetin, and rutin suppressed activation of NF-${\kappa}B$ indicated by inhibition of nuclear translocation of NF-${\kappa}B$, NF-${\kappa}B$/DNA binding, and NF-${\kappa}B$-dependent gene reporter assay. The pharmacological actions of these flavonoids suggest their potential activity for treatment of allergic inflammatory diseases through the down-regulation of mast cell activation.

      • Evaluating the short message service alerting system for critical value notification via PDA telephones.

        <P>Physicians need to respond to critical value alerts in a timely and appropriate manner. We evaluated the clinical usefulness of a short message service (SMS) system for notifying physicians of critical values by sending a text message to their personal data assistant (PDA) phones. The clinical response times and clinical response rates to notifications of critical hyperkalemia in inpatients in 2001, when the callback system alone was used to alert physicians, were compared to those in 2005, when the SMS alert system was used in addition to the callback system. The clinical response time to SMS alerts of critical hyperkalemia sent to PDA phones in 2005 was remarkably shorter than the response time to alerts sent using the callback system alone in 2001. With the help of the SMS system for sending critical value alerts to PDA phones, corrective actions could be taken more promptly and patient care was improved.</P>

        Nuclear localization signal domain of HDAC3 is necessary and sufficient for the expression regulation of MDR1

        Histone acetylation/deacetylation has been known to be associated with the transcriptional regulation of various genes. The role of histone deacetylase-3 in the expression regulation of MDR1 was investigated. The expression level of HDAC3 showed an inverse relationship with the expression level of MDR1. Wild-type HDAC3, but not catalytic mutant HDAC3S424A, negatively regulated the expression of MDR1. Wild-type HDAC3, but not catalytic mutant HDAC3S424A, showed binding to the promoter sequences of HDAC3. HDAC3 regulated the expression level, and the binding of Ac-H3K9/14 and Ac-H4K16 around the MDR1 promoter sequences. The nuclear localization signal domain of HDAC3 was necessary, and sufficient for the binding of HDAC3 to the MDR1 promoter sequences and for conferring sensitivity to microtubule-targeting drugs.
