        Purpose: The purpose of this descriptive study is to examine the correlation between nursing students’ perception of nurses and the level of their satisfaction with the major. Method: The survey was done with the total number of 192 nursing students from 10 different universities in S city selected by a convenience samplingmethod. In order to measure the perception of nurses, the measurement which was invented by Lee(1992) and revised by Jang(1998) was used. Also, the measurement designed by Shim, and modified by Kim(2007) was used for measuring of the students’ satisfaction in their major. The collected data was analyzed by SPSS 17.0. Results: 1.The most decisive factor of the conceptualization of the image of nurses for the nursing college students is their experience of meeting nurses as a patient. 2. Students reconsider the role of nurses by experiencing immediate relationships with nurses in field training. 3. Other factors of the survey such as the students’academic year, religion, the field training situation (individual experience, type of the hospital where they experienced the training, and a relationship with the nurse in charge of the training) or the existence of nurses among their family members do not affect the students’ satisfaction. 4. There is a positive correlation between the image of nurses of nursing college students and their satisfaction with the major(0.66). Conclusion: The positive image of nurses is directly related to the students’ satisfaction in the nursing major, which hugely affects on their academic achievements and decisions of career path. Therefore, there should be more efforts to increase the positive perception of nurses among nursing students.

        홍고추의 유전형 및 재배환경 요인에 따른 비타민 성분의 자연변이 연구

        홍고추는 사람의 식이에서 주요 비타민 공급원이며, 우리나라 사람들은 홍고추를 많이 섭취하고 있다. 그러나 홍고추의 비타민 함량에 대한 품종, 재배지역 및 재배연도에 의한 성분의 자연변이 연구는 국내에서 거의 수행되지 않았다. 이를 위하여 본 연구에서는 12개 상업품종을 대상으로 2016년도와 2017년도 2년간, 2개 지역(4개의 재배환경)에서 재배 및 수확하여 홍고추 과육에서 비타민 A, 비타민 B₁, 비타민 B₂, 비타민 B₃, 비타민 B5, 비타민 E(α-, β-, δ-, γ-tocopherol) 함량을 비교 분석하였다. 고추 과육에서 분석한 비타민 함량을 통합하여 4개 재배환경에 대한 자연변이를 비교하기 위해 통계 알고리즘을 개발하고 적용한 결과, 비타민의 성분은 품종별, 지역별, 연도별로 자연변동성에 유의미한 차이를 보였다. PLS-DA를 이용한 주성분 분석과 더불어 R 통계 기법을 이용한 변동성(% variability) 분석을 실시한 결과, 비타민 성분의 자연변동성은 품종 요인보다는 재배지역이나 재배연도 또는 세 가지 통합 요인의 교호작용에 의해 더 많은 영향을 받는 것으로 확인되었다. 특히 비타민 C와 비타민 B₂, 비타민 B₃, γ-tocopherol은 주로 재배연도에 의해 성분변이가 큰 것으로 분석되었다. α-Tocopherol은 품종 요인에 의해 변이가 비교적 큰 것으로 나타났으나, 세 가지 요인의 교호작용에 의한 변이가 가장 큰 것으로 분석되었다. 비타민 B5는 품종별 요인에 의해 성분변이가 가장 크게 나타났으며, β-tocopherol은 지역 요인에 의한 성분변이가 가장 큰 것으로 나타났다. 비타민 분석 결과에 대해 품종별, 지역별, 연도별로 다변량 통계분석(PLS-DA)을 실시한 결과, 재배지역과 재배연도 요인에 의한 데이터 클러스터가 가시적으로는 뚜렷하게 구분되었으나, PC1과 PC2의 합산 분산력이 60% 미만으로 2차원의 주성분으로 전체 데이터의 분산을 설명하기에는 약간 부족한 것으로 확인되었다. 따라서 비타민 성분의 환경 요인에 의한 변동성 또는 분산력을 설명하기 위해서는 정량적 통계분석 방법인 자연변동성(% variability) 분석이 추가로 수행되어야 하는 것을 알 수 있었다. Red peppers are a remarkably rich source of vitamins in the human diet and widely consumed in Korea. However, comprehensive studies have not reported on the effects of genotype, cultivation regions, and year of cultivation on the vitamin composition of the pepper fruits. To address this, twelve commercial pepper varieties were grown at two locations in Korea during 2016 and 2017, and the concentrations of vitamin A, B₁, B₂, B₃, B5, and E (α-, β-, δ-, γ-tocopherols) in pepper pericarps were determined. Statistical analysis of the combined data showed significant differences among varieties, locations, and years for the measured components. The % variability analysis demonstrated that environment (location and year) and genotype-environment interaction contributed more to the nutritional contents than the genotype alone. Particularly, variation in vitamins A, C, B₂, B₃, and γ-tocopherol was attributed to the year of cultivation. α-Tocopherol primarily varied by the genotype-environment interaction. Vitamin B5 was the trait that the most highly affected by genotype. The location effect was significant for β-tocopherol. Partial least squares discriminant analysis (PLS-DA) results showed that the separation of vitamins was distinct by year and location. These results were supported by the % variability analysis. Our study could explain the natural variation in the vitamin compositions of peppers by genotypes and environments.

        On-demand Versus Continuous Maintenance Treatment With a Proton Pump Inhibitor for Mild Gastroesophageal Reflux Disease: A Prospective Randomized Multicenter Study

        Background/AimsIt remains unclear which maintenance treatment modality is most appropriate for mild gastroesophageal reflux disease (GERD). We aimed to compare on-demand treatment with continuous treatment using a proton pump inhibitor (PPI) in the maintenance treatment for patients with non-erosive GERD or mild erosive esophagitis. MethodsPatients whose GERD symptoms improved after 4 weeks of standard dose PPI treatment were prospectively enrolled at 25 hospitals. Subsequently, the enrolled patients were randomly assigned to either an on-demand or a continuous maintenance treatment group, and followed in an 8-week interval for up to 24 weeks. ResultsA total of 304 patients were randomized to maintenance treatment (continuous, n = 151 vs on-demand, n = 153). The primary outcome, the overall proportion of unwillingness to continue the assigned maintenance treatment modality, failed to confirm the non-inferiority of on-demand treatment (45.9%) compared to continuous treatment (36.1%). Compared with the on-demand group, the GERD symptom and health-related quality of life scores significantly more improved and the overall satisfaction score was significantly higher in the continuous treatment group, particularly at week 8 and week 16 of maintenance treatment. Work impairment scores were not different in the 2 groups, but the prescription cost was less in the on-demand group. Serum gastrin levels significantly elevated in the continuous treatment group, but not in the on-demand group. ConclusionsContinuous treatment seems to be more appropriate for the initial maintenance treatment of non-erosive GERD or mild erosive esophagitis than on-demand treatment. Stepping down to on-demand treatment needs to be considered after a sufficient period of continuous treatment.

        Overlooked KCNQ4 variants augment the risk of hearing loss

        Pathogenic variants of KCNQ4 cause symmetrical, late-onset, progressive, high-frequency-affected hearing loss, which eventually involves all frequencies with age. To understand the contribution of KCNQ4 variants to hearing loss, we analyzed whole-exome and genome sequencing data from patients with hearing loss and individuals whose hearing phenotypes were unknown. In KCNQ4, we identified seven missense variants and one deletion variant in 9 hearing loss patients and 14 missense variants in the Korean population with an unknown hearing loss phenotype. The p.R420W and p.R447W variants were found in both cohorts. To investigate the effects of these variants on KCNQ4 function, we performed whole-cell patch clamping and examined their expression levels. Except for p.G435Afs*61, all KCNQ4 variants exhibited normal expression patterns similar to those of wild-type KCNQ4. The p.R331Q, p.R331W, p.G435Afs*61, and p.S691G variants, which were identified in patients with hearing loss, showed a potassium (K+) current density lower than or similar to that of p.L47P, a previously reported pathogenic variant. The p.S185W and p.R216H variants shifted the activation voltage to hyperpolarized voltages. The channel activity of the p.S185W, p.R216H, p.V672M, and p.S691G KCNQ4 proteins was rescued by the KCNQ activators retigabine or zinc pyrithione, whereas p.G435Afs*61 KCNQ4 proteins were partially rescued by sodium butyrate, a chemical chaperone. Additionally, the structure of the variants predicted using AlphaFold2 showed impaired pore configurations, as did the patch-clamp data. Our findings suggest that KCNQ4 variants may be overlooked in hearing loss that starts in adulthood. Some of these variants are medically treatable; hence, genetic screening for KCNQ4 is important.

      • Interacelluar reprogramming of expression, glycosylation, and function of plant-derived anti-virus therapeutic monoclonal antibody

        Plant genetic engineering has led to production of plant-derived mAb (mAbP), which can provides a safe and economically effective alternative to the conventional antibody expression methods. In this study, the expression levels of mAbP SO57 with or without ER-retention peptide extensions signal (Lys-Asp-Glu-Leu; KDEL) in transgenic tobacco plants were analysed in transgenic plant. The expression levels of mAbP SO57 with KDEL were significantly higher than that without KDEL regardless of transcription level. mAbP SO57 with KDEL localized surround to the nucleus suggesting that the mAbP with KDEL is localized in ER. The mAbP without KDEL and mAbH had mainly Golgi type glycans, whereas the ER-localized mAbP with KDEL had glycan profile with both oligomannose type (47.6%) and Golgi type (52.4%). The Fc domains of both purified mAbP (with and without KDEL) and human-derived mAb (mAbH) had similar bingding activity to the Fcγ RI receptor (CD64). Both mAbP (with and without KDEL) had a shorter half-life than mAbH. However, both mAbP with and without KDEL was as effective at neutralizing activity of the rabies virus CVS-11 as the mAbH. These results suggest that ER localization of recombinant mAbP by KDEL reprograms glycosylation and enhances production of the functional antivirus therapeutic antibody in the plant.

        Transition Characteristics and Risk Assessment of Heavy Metal(loid)s in Angelica gigas Nakai and Cnidium officinale Makino

        This study was conducted to identify transition characteristics of heavy metal(loid)s and to assess dietary risk in the Angelica gigas and Cnidium officinale grown at the major medicinal plant producing districts in Korea. Average bio-concentration factor (BCF) values and range were 0.056 (0.006-0.511) in arsenic (As), 2.030 (0.021-15.678) in cadmium (Cd), 0.179 (0.052-0.393) in mercury (Hg), and 0.061 (0.013-0.474) in lead (Pb) in Angelica gigas. Average BCF values and range were 0.044 (0.011-0.264) in As, 0.557 (0.052-4.255) in Cd, 0.174 (0.069-0.286) in Hg, and 0.024 (0.012-0.057) in Pb in Cnidium officinale. The BCF values by heavy metal(loid)s in Angelica gigas and Cnidium officinale were high in Cd, copper (Cu) and zinc (Zn), while As and Pb were found to be less transferable. The BCF values calculated in this study will be useful for predicting the uptake of heavy metal(loid)s. Human exposure to As, Cd, Hg, and Pb through dietary intake of Angelica gigas and Cnidium officinale might not cause adverse health effects although some Angelica gigas were higher than the allowable value for Cd. Further study on uptake and accumulation mechanism of Cd by Angelica gigas is required to assess the human health risk associated with soil contamination.

      • Effect of ER signal peptides (KDEL) on expression and function of anti-virus monoclonal antibody in plant

        Plant genetic engineering has led to the production of plant-derived mAb (mAbP), which provides a safe and economically feasible alternative to the current antibody expression systems. In this study, the expression levels of mAbP SO57 with or without ER-signal peptides (Lys-Asp-Gly-Leu;KDEL) in transgenic tobacco plants were analysed in transgenic plant. PCR and Reverse Transcription-PCR analyses showed existence of heavy and light chain genes of mAb with or without KEDL and their transcription in plant, respectively. Western blot showed that the expression levels of mAbP SO57 with KDEL were significantly higher than that without KDEL. Flow cytometry analysis showed that the Fc domains of both purified mAbP and mammalian-derived mAb have similar binding activity to the FcγRI receptor. High-performance liquid chromatography analysis showed that the mAbP SO57 with KDEL had glycan profile with both oligomannose and golgi type, whereas the mAbP SO57 without KDEL had only golgi type glycans. Neutralizing analysis with rabies virus CVS-11 showed the similar neutralizing activity between mAbP SO57 with and without KDEL. These results suggest that the potential of mAbP SO57 for rabies immunotherapy is regardless of plant specific glycan structures.
