The pure-separation of calcium chloride-treated fibroin hydrolysates could be carried out using gel filtration chromatography. Also, the effect of its enzymatic hydrolysis was investigated in order to find out the enhancement of their functionality. The average molecular weight(Mw), solubility and free amino acid compositions of three hydrolysates samples (calcium chloride, calcium chloride-flavo-urzyme and calcium chloride-thermoase) were measured to compare their characteristics. The molecular weight of calcium chloride hydrolysate was about Mw 46,800 and it can be reduced to Mw12,500 and 1,070 upon the enzymatic hydrolysis by flavourzyme and thermoase, respectively. A solubility of calcium chloride-treated samples shows about 60% while calcium chloride/enzyme-treated samples are perfectly soluble (100% solubility). The total amino acid composition of calcium chloride enzymatic hydrolysates are much higher than that of calcium chloride hydrolysate.

The preparation of skin care gauze could be make to mixing separated cellulose from waste milk park and degummed silk fibroin fiber. Also, its wound covering and anti-barterial activity were investigated in order to find cut the enhancement of their functionality. By the 30% silk fibroin fiber including skin care gauze. the anti-bacterial activity values of Sraphylococcus stran are much 4 times higher than of0~10% silk fibroin fiber including skin care gauze. The average yield of cellulose from waste milk pack was obtalned 50-6O%, and their morphologie,, physical properties, modulus and biodegradation ratios are studies, respectively.

The bond direction between fiber axis and N-H bond direction of $^15N$-Gly labeled silk fibroin fiber and poly-p-phenyleneterephthalamide(PPTA) oriented were obtained from the dipolar-bipolar interaction information, respectively. Theoretical method for the determination of molecular bond directions relative to the axis of fiber was derived on the basis of solid state nuclear magnetic resonance dipolar-dipolar interaction tensor. The oriented samples were placed parallel to the applied magnetic field, and the $^15N-^1H$ hetero-nuclear dipolar-dipolar splitting values were obtained. Substituting this values into the derived equation, the N-H bond angles ($$\theta$_NH$=83$^{\circ}$ and 97$^{\circ}$) are calculated. Also, the N-Hbond direcyions to fiber axis of PPTA are 44$^{\circ}$ and 67- These obtained values compared with the values from the angular dependation spectra simulation of oriented PPTA sample.

The collagen growth effects in rats using B. mori fibroin sponge sheet, pure fibroin solution and chitosan sponge sheet were investigated. Histopathological inspection of the wound 12 days later showed the increase of a vascular ingrowth and the absence of inflammatory cells. Wound healing effects were accelerated in the order of Fibroin sponge sheet ≥Fibroin dressing solution > Chitosan sponge sheet > Control. The mean percentage of collagen production in rats after 12 days was 46.12±4.58% and 42.11±5.67% in fibroin sponge sheet and dressing solution-treated rats, respectively. These values were higher than the relative content in the chitosan sponge sheet and control groups, which was 23.87±3.24 and 2.42±0.32%, respectively.

The behavior of tyrosine(Tyr.) residue of Bombyx mori silk fiber from yellow fluorescent cocoon has been examined for the dependence of pH in aqueous silk solution under the presence of orange II salt. Through the peak separation of angular dependence of spectral pattern of 15N-Tyr. and (1-13C)-Tyr. between the fiber axis and the molecular bond direction, N-H bond in fiber as well as the orientation distribution around the fiber axis were analyzed. Also, and sericin component was obtained from these angular dependence of oriented spectral pattern. The pH dependence of the 13C NMR chemical shift of B. mori silk fibroin was examined in aqueous solution in the presence of orange II hydrophobic interaction such as stacking between the Tyr.-ring and the orange ll. All peaks of orange II are broad at pH ≥ 7.0. However, these become sharper at pH ≥8.0 and remain sharp at higher pH. In these higher pH range, a chemical shift change occurs due to the deprotonation of the Tyr. side group of fibroin. At higher pH, such a hydrophobic cluster is destroyed because of the electrostatic interaction according to the deprotonation of the Tyr.-OH group.

실크단백질 식이공급이 동물대장암에 미치는 효과를 알아본 결과 다음과 같은 결과를 얻을 수 있었다. 1. 동물 대장암에 투여한 식이공급용 선발 실크단백질의 피브로인 및 세리신 단백질은 각각 Mw 65,000 내외의 피브로인 및 Mw 100,000만 내외의 세리신 단백질이며, 동물사료의 경우, 섭식 등 물성 등이 작년대비 개선된 3% 첨가된 전용 사료를 조제하여 섭식하도록 하였다. 2. 1,2-DMH에 의한 동물 대장암 유발(1회/1주, 4회 주사) 후 6주 시료 투여에 있어서 대장암 유발군(CA)의 경우, 체중변화 및 식이 섭취량의 관계인 식이효율은 세리신 섭취군(S)의 경우 정상군(C)의 85% 수준을 나타내었다. 3. 동물대장암 조직의 조직관찰에 있어서는 대장암 조직의 COX-2 발현을 염색한 결과, 대장암 유도군(CA)의 경우 확실한 COX-2에 의한 발현을 나타내었으며, 세리신 섭취군(S)의 경우는 그 정도가 미비함을 나타내어 대장암에 효과가 있음을 나타내었다. 4. 대장암 인자변화를 정량적으로 확인한 결과, 정상군(C)에 비하여 대장암 유발군(CA)은 확실한 7배 정도의 COX-2량이 확인되었으며, 세리신 섭취군(S)의 경우는 유발군(CA) 대비 1/2 이하 수준으로 줄어듬을 확인할 수 있어서 금후 대장 질환 관련 소재 개발 가능성을 나타내었다. This study was done for the dietary effect of silk protein, molecular controlled silk sericin and silk fibroin, on colon cancer of animal. The ICR mice were given weekly injections of 1,2-dimethylhydrazine (DMZ) for the initial 10 mg/kg with four weeks, and fed a diets supplemented with 3% (v/v) molecular controlled sericin and fibroin for 6 weeks, respectively. Then, the body weight, efficiency of dietary, typical histologic appearance of animal colon tissue and COX-2 effect were studied, too. Molecular controlled sericin protein are shown more higher dietary effect on animal colon cancer than fibroin supplemental case. The results suggest a potential usefulness of sericin as a chemopreventive for colon carcinogenesis and the development functional food.
