권백의 Caspase-3 활성화를 통한 HL-60 세포에서 세포사멸 유도효과

In our previous studies, we reported that Selaginella Tamariscina(ST) induced apoptotic cell death in HL-60 cells selectively. The cell viability after treatment with extract of ST was quantified by MTT assay and trypan bleu exclusion method. The results showed that application with ST in HL-60 induced 40% cell death at the concentration of 400 ㎍/ml. The cancericidic effect of Selaginella Tamariscina was mediated by apoptosis. Thus, HL-60 cells exposed to Selaginella Tamariscina displayed the DNA fragmentation ladder and nucleus chromatin condensation characteristic for apoptosis. The enzyme activity of caspase-3 and actived caspase-3 protein were markedly increased HL-60 cells treated with the extract of Selaginella Tamariscina. In addition, the extract of Selaginella Tamariscina induced cleavage of PARP, a known substrate for caspase-3. The expression of BcI-2, anti-apoptotic protein, was decreased by treatment of the aqueous extract of Selaginella Tamariscina in a dose-dependent manner. And the expression of pro-apoptotic Bax protein was increased. In conclusion, our results suggest that the extract of Selaginella Tamariscina may induce the apoptotic death of HL-60 cells via caspase-3, cleavage of PARP protein, depletion of cellular ATP levels and BcI-2 degradation.

The Sellaginella Tarmariscina is widely used in the traditional oriental herbal medicine for its anti-inflammatory, antioxidant effects. The effects of aqueous extracts of Sellaginella Tarmariscina on the viability and induction of apoptotic cell death were investigated in HL-60 cells. The cell viability after treating with extract of Sellaginella Tarmariscina was quantified by MTT assay. The results showed that could inhibit the proliferation of HL-60 cells and caused a 40% inhibition of HL-60 cells at concentrations of 400 ㎍/ml. The cancericidic effect of Sellaginella Tarmariscina was mediated by apoptosis. Thus, HL-60 cells exposed to Sellaginella Tarmariscina displayed the DNA fragmentation ladder and nucleus chromatin condensation characteristic for apoptosis. In conclusion, our results suggest that the extract of Sellaginella Tarmariscina may induce the apoptotic death in HL-60 cells.

補中益氣湯 및 補肝湯이 白鼠 損傷肝의 回復에 미치는 影響

This study was performed to elucidate the protective effect of several preseriptions on the carbon tetrachloride induced hepatotoxicity in rats. The effect were chemically examined by AST, ALT, ALP, LDH, total cholesterol, protein, triglyceride level in the serum. The experimental group was divided into five groups, normal groups, normal group with no treatment, control group treated with CCl₄and physiological saline, EGI treated with CCl₄and BGT extract, EG II treated with CCl₄and BIT extract, EG III treated with CCl₄and BGT plus BIT extract. The results were as follows; 1. The serum AST and LDH activity after CCl₄treatment was significantly inhibited from 2nd to 7th day after showing the highest level at the 1st day in all experimental groups in comparison with the level of control group. But those findings maintained higher level than normal group. 2. In all experimental groups, AST level showed the highest level at 12 hours after CCl₄administration and then decreased by slow degrees but appeared higher than normal level in all experimental days. Also, the serum AST level of EG III significantly decreased at the 1st, 2nd, 3rd day than that of control group. 3. The serum ALP activity of EG I appeared lower than that of control group, and that of EG II at the significantly decreased in comparison with that of control group. 4. After CCl₄treatment, the serum cholesterol level of all experimental groups slightly increased in comparison with tht of control group, these findings were lower than that of normal group in all experimental days. These results suggested that BGT, BIT and BGT plus BIT have considerable effects to the hepatic injury induced by CCl₄treatment and so these prescriptions are able to reduce the recoveing period of damaged liver.

만성 요통환자에서 성별에 따른 고관절 내회전 각도의 비교

Objectives : This study is planned to classify different biomechanics of men and women, from the comparison of normal range and hip internal rotation angle in chronic low back pain patients. Methods : We measured the hip internal rotation angle of the 30 men and 30 women patients with low back pain that has been over 3 months in BuCheon Jaseng Hospital of Oriental medicine. We set 35-45 degrees as normal range, and found the patients with exceeding normal range, the patients with normal range, and the patients with under normal range. Results : Men appeared to have less hip internal rotation angle than women. Especially, the men's left hip internal rotation angle was less than normal range. On the other hand, women's right hip internal rotation angle was often larger than normal range which was stastically significant. Conclusions : Contraction of hip external rotation muscle including gluteus maxius muscle and piriformis muscle in men, or relaxation of posterior gluteus medius, gluteus maxius and piriformis muscle with contraction of tensor fasciae latae, a part of hip internal rotation muscle, in women seems to be the basis for biomechanics of chronic low back pain.

蘇木의 에틸아세테이트 추출물이 B16/F10 흑색종세포의 멜라닌 합성에 미치는 효과

Caesalpinia sappan L. has been commonly used as analgesic, a cure for contusion and sprain as well as a remedy for thrombosis in the oriental medicine. The main constituent of Caesalpinia sappan L. is brazilin having a flavonoid structure. Melanin is a main pigment found in skin, hair and eyes, and melanin synthesis is controlled by a cascade of enzymatic reactions regulated at the level of tyrosinase. The aim of this study was to examine the effect of Caesalpinia sappan L. on proliferation and melanogenesis of B16/F10 melanoma cells. After 48h treatment of cells with various concentrations of EtOAc extract, the cells exhibited a dose-dependent inhibition in their proliferation without apotosis. Therefore, the growth retardation may be due to the cell arrest, not due to cytotoxicity. And there were estimated total melanin contents as a final product and activity of tyrosinase, a key enzyme, in melanogenesis. EtOAc extract increased the melanin contents and tyrosinase activity in low concentration (0.01-10㎍/ml), whereas it decreased those in high concentration. In conclusion, it was observed that EtOAc extract of Caesalpinia sappan L. differently regulates melanization of cells dependent of its concentrations.

로지스틱 회귀 분석을 이용한 고관절 내회전 각도와 단분절 요추 추간판 탈출증 발생 부위의 상관성 분석

ObjectivesThe purpose of this study is to find out the relationship between the hip in-ternal rotation angle and the site of herniation of single level lumbar intervertebral disc.MethodsThis study was carried out to 418 patients with lumbar disc herniation diag-nosed by M.R.I, who visited Bu-Chun Jaseng Hospital of Korean Medicine with low back pain. Researchers measured their hip internal rotation angle and analyzed the relationship between the hip internal rotation angle and the site of herniation of single level lumbar inter-vertebral disc.Results1. Excessive right internal rotation of hip joint increases risk of L5/S1 HIVD. 2. Limited left internal rotation of hip joint decreases risk of L4/L5 HIVD. 3. In cases of L4/L5 HIVD level, there was evidence that the prevalence of HNP in men was 1.85 times as higher than that in women. 4. In cases of L5/S1 HIVD level, there was the evidence that the preva-lence of HNP in men was 0.64 times as lower than that in women.ConclusionsIn single-segment lumbar HIVD patients, limited left internal rotation of hip joint decreases risk of HIVD of L4/5 and excessive right internal rotation of hip joint mainly leads to L5/S1 HIVD. 전방락킹, 후방락킹 운동 및 MET(Muscle Energy Techniques)를 이용한 Sway-back 자세 환자의 치험 3례

Sway-back 자세를 가진 환자 3례에 전방락킹, 후방락킹 운동 및 MET를 이용하여 치료한 결과 Whole spine X-ray(lateral)상의 척추만곡각도 및 NRS score의 호전을 확인하였다. 이에 저자는 본 증례를 보고하는 바이다. Objectives : The purpose of this study is to investigate the clinical application of Rocking forward, Rocking backward Exercise and MET(Muscle Energy Techniques) to three patients who had sway-back posture. Methods : Three patients diagnosed as sway-back posture, not have discogetic pain, were hospitalized Bu-Chun Jaseng hospital of Oriental Medicine about 20 days and they were treated by Rocking forward, Rocking backward Exercise and MET. To measure the outcome of the patients' improvement, we observed the X-ray of Whole spine and Numerical Rating Scale(NRS). Results : After being treated by Rocking forward, Rocking backward Exercise and MET, the patients' posture was significantly improved and the pain was reduced. The X-ray of Whole spine became almost normal and the improvement of patients' in the NRS score was detected. Conclusions : These results suggest that Rocking forward, Rocking backward Exercise and MET were effective treatment on patients who had sway-back posture.

경근 추나 치료를 적용한 척추유합술 후 슬굴곡근 단축 환자 치험 1례

Objectives : The present study introduces the clinical application of Hamstring Chuna Manual Therapy to a patient who underwent the pain after spinal fusion. Methods : A patient who had the pain after spinal fusion was hospitalized at Bucheon Jaseng hospital of Oriental Medicine for 35 days. During the hospital treatment, the pain was relieved by Hamstring Chuna Manual Therapy. The improvement of the patient was measured by VAS(Visual Analogue Scale) score, SLR(Straight-Leg Raising) test, and walking distance. Results : The values of the patient's VAS score, SLR test, and walking distance measured before and after Hamstring Chuna Manual Therapy presented that the pain was relieved after Hamstring Chuna Manual Therapy. Conclusions : Therefore, this case demonstrates that Hamstring Chuna Manual Therapy is effective treatment method for relieving pain in the spine.

요추부 한 분절 유합술 이후 발생한 추간판 탈출증환자에 대한 보존적 치료 치험 1례

요추부 한 분절 유합술 이후 발생한 추간판 탈출증으로 내원한 외래 환자 1례에 한방치료를 적용하여 요통 및 하지방사통 통증 감소와 요추부 관절가동범위(ROM) 증가에 유의한 효과를 보였기에 보고하는 바이다. Objectives: The purpose of this study is the evaluation of conservative medical treatment on the herniation of intervertebral disc(HIVD) patient after the fusion surgery of a lumbar segment. Methods: We used acupuncture, herbal medication, and manipulation for this patient. And we measured of VNRS score and SLR test result to evaluate effect of conservative medical treatment. Results & Conclusions: Patient's low back and left leg pain VNRS is decreased, and the SLR test result is improved.

고관절 내회전 각도와 단분절 요추 추간판 탈출증 발생 부위의 상관성 분석

Objectives : The purpose of this study is to find out the relationship between hip internal rotation angle and the level of herniation of lumbar intervertebral disc(HIVD) in single-segment. Methods : We investigated 314 patients (158 male, 156 female) who were diagnosed as herniation of lumbar intervertebral disc(HIVD) in single-segment. We measured 314 patients' hip internal rotation angle and analysed the relationship between the hip internal rotation angle and the level of herniation of lumbar intervertebral disc(HIVD). Results : 1. Among 314 cases, the hip internal rotation angle was different between male and female. Hip internal rotation angle of male was mainly limited and that of female was mainly excessive. 2. Among 314 cases, the normal group, defined as patients who have specific range of hip internal rotation angle(male : $35^{\circ}{\pm}10^{\circ}$, female : $45^{\circ}{\pm}10^{\circ}$), tends to be occurred HIVD at L4/5 level. The limited group, defined as patients who have less angle than normal group, the excessive group, defined as patients who have more angle than normal group, and the complex group, defined as patients who have more angle of one leg and less angle of the other leg than normal group, tend to be occurred HIVD at L5/S1 level(p<0.05). Conclusions : In single-segment lumbar HIVD patients, The normal hip internal rotation angle mainly leads to L4/5 HIVD, while the limited and excessive hip internal rotation angle mainly lead to L5/S1 HIVD.
