        Three positive regulators of leaf senescence in Arabidopsis, ORE1, ORE3 and ORE9, play roles in crosstalk among multiple hormone-mediated senescence pathways

        Leaf senescence is a developmentally programmed event, but the initiation and progression of leaf senescence are affected by a range of plant hormones including abscisic acid (ABA),ethylene and methyl jasmonate (MeJA). To investigate plant hormone crosstalk during leaf senescence, hormone-induced senescence phenotypes were analyzed in three leaf senescence mutants [ore1 (oresara1), ore3 and ore9] showing delayed senescence phenotypes in age-dependent and dark-induced senescence. The ore mutants exhibited delayed leaf senescence phenotypes following treatment with ABA, ACC (aminocyclopropane-1-carboxylic acid) or MeJA. After each hormone treatment, the photochemical efficiency of photosystem II and chlorophyll content were significantly higher in the ore mutant leaves than in the wild-type leaves. The expression of CAB2and SEN4 in the wild-type was rapidly altered following each hormone treatment. However, the decrease in CAB2 expression and the induction of SEN4 expression in the mutants were less affected by ABA, ACC or MeJA treatment. It is suggested that ORE1, ORE3 and ORE9 are required for the proper progression of leaf senescence mediated by ABA, ethylene and MeJA. This implies that ORE1, ORE3 and ORE9 may be linked to the crosstalk among senescence pathways induced by ABA, ethylene and MeJA, as well as age and darkness.

        17β-Estradiol에 의한 생쥐 자궁 내 Nesfatin-1/NUCB2 발현 조절

        시상하부에서 생성되는 nesfatin-1/NUCB2가 음식물 섭취와 에너지 대사를 조절한다는 것이 보고된 이후로, 최근생쥐의 난소와 자궁에서도 다량의 nesfatin-1/NUCB2가 발현된다는 사실이 새롭게 밝혀졌다. 그러나 생식기관에서nesfatin-1/NUCB2 발현이 어떻게 조절되는지는 알려져 있지 않다. 따라서 본 연구에서는 생쥐의 생식기관 중 자궁에서nesfatin-1/NUCB2 발현이 어떠한 경로를 통하여 조절되는지를 알아보고자 난소를 제거한 암컷 생쥐에 성선자극호르몬과17β-estradiol을 각각 투여한 후 nesfatin-1/NUCB2 발현 정도를 조사하였다. 먼저 자궁 내 NUCB2 mRNA 발현을conventional PCR과 real-time PCR 방법으로 확인하였으며, 아울러 western blot 방법으로 nesfatin-1 단백질의 발현을 관찰하였다. Nesfatin-1 단백질 발현 위치 및 결합 부위를 조사하기 위하여 면역조직화학염색을 수행한 결과, nesfatin-1 단백질은 자궁내막과 자궁샘을 둘러싼 상피세포에서 발현되었으며, nesfatin-1 단백질 결합 부위는 자궁샘을 둘러싼 상피세포와호중구를 포함하는 특정 과립세포에서 관찰되었다. 난소를 제거한 암컷 생쥐에 성선자극호르몬을 투여한 결과, 자궁 내NUCB2 mRNA 발현량은 대조군과 성선자극호르몬 투여군 사이에 차이가 없었으나, 같은 방법으로 난소를 제거한 암컷 생쥐에 17β-estradiol을 투여한 결과, 대조군보다 17β-estradiol 투여군에서 NUCB2 mRNA 발현량이 유의하게 증가하는 것을 확인하였다. 자궁 내 nesfatin-1 단백질의 발현량 또한 NUCB2 mRNA 발현 양상과 유사하게 17β-estradiol 투여군에서 발현량이 유의하게 증가하는 것을 확인할 수 있었다. 본 연구 결과, 자궁 내 nesfatin-1/NUCB2 발현이 뇌하수체에서 분비되는 성선자극호르몬에 의해 조절 받기보다는 난소에서 분비되는 17β-estradiol에 의해 조절 받는다는 사실이 밝혀졌으며, 이러한 결과는 발정 주기에 따른 혈액 내 17β-estradiol 농도의 변화가 자궁 내 nesfatin-1 발현을조절함으로써 nesfatin-1이 자궁내막 발달과 착상을 조절할 수 있는 국부조절인자로 작용할 수 있음을 제시하고 있다.

        Ultrastrucure of Germ Cells during Spermatogenesis and Some Characteristics of Sperm Morphology in Male Mytilus coruscus (Bivalvia: Mytilidae) on the West Coast of Korea

        The ultrastructure of germ cells during spermatogenesis and some characteristics of sperm morphology in male Mytilus coruscus, which was collected on the coastal waters of Gyeokpo in western Korea, were investigated by transmission electron microscope observations. The morphology of the spermatozoon has a primitive type and is similar to those of other bivalves in that it contains a short midpiece with five mitochondria surrounding the centrioles. The morphologies of the sperm nucleus type and the acrosome shape of this species have an oval and modified cone shape, respectively. In particular, the axial rod is observed between the nucleus and acrosome of the sperm. The spermatozoon is approximately 45-50 μm in length including a sperm nucleus (about 1.46 μm in length), an acrosome (about 3.94 μm in length) and tail flagellum (approximately 40-45 μm). The axoneme of the sperm tail flagellum consists of nine pairs of microtubules at the periphery and a pair at the center. The axoneme of the sperm tail shows a 9+2 structure. Some special charateristics of sperm morphology of this species in the genus Mytilus are (1) acrosomal morphology, (2) the number of mitochondria in the midpiece of the sperm, and (3)the existence of a satellite. The axial rod appears in the acrosome and sperm nucleus as one of the characteristics seen in several species of the subclass Pteriomorphia, unlikely the subclass Heterodonta containing axial filament instead of the axial rod. The number of mitochondria in the midpiece of the sperm of this species in the family Mytilidae are five, as one of common characteristics appeared in most species in the family Mytilidae. Most of Mytilus species contain a satellite body which is attached to the proximal centriole in the middle piece of the sperm, as one of common characteristics of sperm morphology in the family Mytilidae.

        Ultrastructure of the Testis and Germ Cell Development During Spermatogenesis in Male Crassostrea gigas (Bivalvia: Ostreidae) in Western Korea

        Ultrastructural characteristics of the testis and spermatogenesis of Crassostrea gigas were investigated by Transmission and Scanning Electron microscope observations. The testis is a diffuse organ consisting of branching acini containing differentiating germ cells in a variety of stages. The acinus is surrounded by an intermitent layer of myoepithelial cells andis divided into subcompartments that are partially separated by pleomorphic accessory cells which remain in close contact with germ cells until late stages of development. these accessory cells contain a large quantity of glycogen particles and lipid droplets in the cytoplasm. Therefore, it is assumed that they are involved in the supplying of the nutrients for germ cell development, while any phenomena associated with phagocytosis of undischarged, residual sperms by lysosomes could be find in the cytoplasm of the accessory cells. The morphology of the spermatozoon has a primitive type and is similar to those of other bivalves. Mature spermatozoa consist of broad, cap-shaped acrosomal vesicle, subacrosomal material (containing axial rod embedded in a granular matrix), a oval nucleus showing deeply invaginated anteriorly, two triplet substructure centrioles surrounded by four spherical mitochondria, and satelite fibres appear to the distal centriole and plasma membrane. Spermatozoa of C. gigas resemble to those of other investigated ostreids. In particular, the anterior region of the acrosomal vesicle is transversely banded. It is assumed that differences in this acrosomal substructure are associated with the inability of fertilization between the genus Crassostrea and other genus species in Ostreidae. Therefore, we can use sperm morphology in the resolution of taxonomic relationships within the Ostreidea. The spermatozoon is approximately 42-47 μm in length including an oval sperm nucleus (about 0.91 μm in length), an acrosome (about 0.42 μm in length) and tail flagellum (40-45 μm). The axoneme of the sperm tail flagellum consists of nine pairs of microtubules at the periphery and a pair at the center. The axoneme of the sperm tail shows a 9 + 2 structure. These morphological charateristics of acrosomal vesicle belong to the family Ostreidae in the subclass Pteriomorphia.

        현대시조의 율격 변이 양상과 그 의미 - 이호우 시조를 중심으로 -

        현대시조의 형식적 면모는 시조장르의 본질 및 존립과 관련하여 논란거리가 되 고 있다. 고시조의 형태를 전제로 창작되어지는 시조 중에서 어느 정도의 일탈을 현대시조로 이해해야 하는가 하는 점이 문제가 되고 있는 것이다. 이와 관련하여 본고에서는 현대시조의 율격적 변이 양상을 살피고 그 의미에 대해 논의하였다. 제2장에서는 시조의 율격이 “반행 내의 앞마디가 뒷마디보다 짧거나 같으며(단, 마지막 반행은 종결부로서 그 반대), 각 마디는 3․4음절로 주로 구성된다.”라는 음수율적 성격을 지닌 것임을 밝혔다. 이어 제3장에서는 그러한 율격의식이 현대 시조에서 어떻게 변화되었는지 이호우 시조의 예를 들어 살펴보았다. 먼저, 3․4 음절을 넘는 過音節로 시조의 마디가 구성된 경우를 검토한 후, 이어 ‘앞마디≦뒷 마디’의 반행 형태가 전도된 경우에 대해 분석하였다. 그 결과, 이러한 변격이 고 시조에 비해 이호우 시조에는 훨씬 잦았음을 보았으며, 이는 시적 주제 및 시의식 과 관련하여 일어난 결과임을 알 수 있었다. 과음절의 다용은 불안정하고 부조화 스러운 정서의 표현과 관련되어 있었으며, 반행 형태의 전도는 심상과 구어에 대 한 중시, 그리고 시상과의 부합을 꾀하는 내재율적 의식 등과 관계가 있었다. 본고의 논의를 요약하면, 첫째, 시조에는 음수율과 관련된 율격 의식이 존재하 며, 둘째, 그러한 율격 의식과 작가의 리듬 충동이 교섭하는 가운데 현대시조의 율격 변이는 이루어진다는 것이다. 만일 음수율과 관련된 시조의 율격 의식을 체 득하지 못하고 그것을 무시한 채 작품을 짓는다면 난삽한 작품이 형성될 것이요, 반대로 예민한 시의식과 내재율의 감각이 부족한 채 시조의 이른바 기준 음수율만 준수하려 한다면 고루한 작 품이 만들어질 것이다. 따라서 현대시조를 짓고 감상하 는 일은 시조의 율격 의식과 시인의 리듬 충동 사이의 팽팽한 긴장감을 느끼는 일 이 되어야 한다. The formal aspects of the modern sijo have been controversial with regard to the essential features of the sijo genre and its survival. The formal deviation of modern sijo poems raised the question such as how much deviation could be allowed. In this regard, this paper investigated the transformation of the sijo metrics in modern sijo and its implication. In chapter2, the metrical features of the sijo genre were discussed because they had still been controversial. This paper suggested that the metrics of the sijo should be understood as syllabic. Although the syllabic number of a colon is not fixed in a sijo, it follows less strict rules; a colon is often composed with three or four syllables, and the latter colon is greater than or equals to the former one in a half line of a sijo poem. In chapter3, the transformation of the sijo metrics in modern sijo was discussed. In Lee Ho-woo's sijo poems, which were selected as examples, the metrical features of the sijo were often transformed with regard to the poetic themes and the poet's literary consciousness. The longer colons that were often used in Lee's works were related to the anxiety and disharmonious feeling of his time. The reversed form of the half-lines also had connection with the modern poet's consciousness, which considers the poetic imagery and colloquialism as important and pursues the rhythm of free verse. In summary, the transformation of the sijo metrics in modern sijo poems occurs while the sijo metrics negotiates with the rhythmic sense of modern sijo poets. Disregarding the existing metrics would result in abstruseness, whereas sticking to the metrical rules would produce antiquated works. Therefore, the writers and the readers of modern sijo poems should be able to feel the tension between the sijo metrics and the poets' rhythmic sense and poetic consciousness.

        Selective adsorption of Ba2+ using chemically modified alginate beads with enhanced Ba2+ affinity and its application to 131Cs production

        The 131Cs radioisotope with a short half-life time and high average radiation energy can treat the cancer effectively in prostate brachytherapy. The typical 131Cs production processes have a separation step of the cesium from 131Ba to obtain a high specific radioactivity. Herein, we suggested a novel 131Cs separation method based on the Ba2þ adsorption of alginate beads. It is necessary to reduce the affinity of alginate beads to cesium ions for a high production yield. The carboxyl group of the alginate beads was replaced by a sulfonate group to reduce the cesium affinity while reinforcing their affinity to barium ions. The modified beads exhibited superior Ba2þ adsorption performances to native beads. In the fixed-bed column tests, the saturation time and adsorption capacity could be estimated with the Yoon-Nelson model in various injection flow rates and initial concentrations. In terms of the Cs elution, the modified alginate showed better performance (i.e., an elution over 88%) than the native alginate (i.e., an elution below 10%), indicating that the functional group modification was effective in reducing the affinity to cesium ions. Therefore, the separation of cesium from the barium using the modified alginate is expected to be an additional option to produce 131Cs.
