        검은콩 첨가 비율에 따른 앙금의 품질 특성

        This study investigated the quality characteristics of black soybean sediments to diversify the availability of soybean. The cooking method selected for black soybean sediment preparation was a pressure cooking process without soaking, considering the isoflavone content. The black soybean sediments were prepared by the addition of 0, 10, 30, 50 and 100% (w/w) black soybean. When 0% to 100% black soybean was added to the black soybean sediments, the moisture and crude protein contents increased from 53.17% to 54.41% and from 12.07% to 21.68%, respectively. The total isoflavone content of the black soybean sediments was increased from 2.69 ㎍/g to 696.09 ㎍/g, respectively, by the addition of black soybean. The anthocyanin content of the black soybean sediments ranged from 279.29 ㎍/g to 387.8 ㎍/g by the addition of black soybean. The total polyphenol content and the total flavonoid content of the black soybean sediments range from 1.72 mg/g to 2.00 mg/g and 0.89 mg/g to 0.92 mg/g, respectively, by the addition of black soybean. Given the isoflavones, total polyphenol, and anthocyanin content of the black soybean sediments, it is appropriate that the ratio of added black soybeans is at least 50% after the pressure-cooking process, regardless of soaking.

        검정콩 된장의 사이토카인 생성 및 종양전이 억제에 미치는 영향

        본 연구에서는 노란콩과 검은콩의 메탄올 추출물과 각각의 콩으로부터 제조된 된장 메탄올 추출물이 IL-2, IL-6 및 TNF-α를 포함하는 사이토카인의 생성과 종양전이 억제에 미치는 영향에 대해 검토하였다. IL-2의 경우, LPS를 처리하지 않았을 때 대조군과 2개월 발효 검은콩 된장 메탄올 추출물(10 μg/ml)에서 가장 높은 IL-2 생성을 나타내었고 LPS (1 ㎍/ml)를 처리한 후에는 검은콩 메탄올 추출물에서 가장 높은 IL-2 생성을 보였고 대조군에서 가장 낮은 생성을 나타내었다(p<0.05). 또한 IL-6 생성의 경우 LPS를 처리하지 않았을 때 2개월 발효 검은콩 된장 메탄올 추출물(10 ㎍/ml)에서 가장 높은 수치를 나타내었고 대조군과 2개월 발효된 노란콩 된장 메탄올 추출물(10 ㎍/ml)에서 가장 낮았다(p<0.05). LPS (1 ㎍/ml)를 처리한 경우, 대조군, 노란콩, 2개월 및 7개월 발효된 검은콩 메탄올 추출물(10 ㎍/ml)에서 검은콩 및 2개월 발효된 노란콩 된장 메탄올 추출물(10 ㎍/ml)에서 보다 유의적으로 높은 수치를 나타내었다(p<0.05). TNF-α의 생성에서 2개월 발효된 검은콩 메탄올 추출물(10 ㎍/ml)은 LPS를 처리하지 않을 때와 LPS (1 ㎍/ml)를 처리한 후 둘다의 경우 기타 실험군들에 비해 유의적으로 높은 TNF-α의 생성을 나타내었다(p<0.05). 한편 종양전이의 예방적 효과에 대한 실험결과에서 1 ㎎/mouse 농도로 투여하였을 때 2개월 발효된 노란콩 된장, 검은콩, 2개월 검은콩 된장, 7개월 검은콩 된장에서 각각 18%, 26%, 21%, 50%로 7개월 발효된 검은콩 된장에서 종양전이를 크게 억제하는 것으로 나타났다. 따라서 검은콩으로 제조한 된장은 사이토카인 생성을 증가시켜 면역증진 활성을 나타내었으며 암전이의 예방효과도 우수하였음을 확인할 수 있었다. We investigated the effect of black soybean and doenjang with black soybean on production of cytokines including interleukin-2 (IL-2), interleukin-6 (IL-6) and tumor necrosis factor-α (TNF-α) compared with yellow soybean and doenjang with yellow soybean. We also determined inhibitory effect of two types of soybeans and doenjang on tumor metastasis produced by colon 26-M3.1 carcinoma cells. The cytokine productions of mouse splenocytes increased by the exposure of lipopolysaccharide (LPS, 1 ㎍/ml) compared to without treatment of LPS. The LPS-induced IL-2 production was highest in the black soybean methanol extract, while the methanol extract from black soybean doenjang fermented for 2 mo showed the higher levels of IL-2 without LPS (p<0.05). In case of LPS-induced IL-6 production, the methanol extracts from control, yellow soybean, black soybean doenjang fermented for 2 and 7 mo showed higher levels of IL-6 compared to those of black soybean, and yellow soybean doenjang fermented for 2 mo (p<0.05). The methanol extract of black soybean doenjang fermented for 2 mo showed significantly higher levels of TNF-α in both with and without LPS (p<0.05). In experiment of tumor metastasis, the treatment (1 mg/mouse) of methanol extract from black soybean doenjang fermented for 7 mo inhibited tumor metastasis by 50% and had the highest inhibitory effect among other samples (p<0.05). From these results, doenjang manufactured with black soybean modulated the production of cytokine and showed anticancer effect, suggesting that this effect was increased with increased periods of fermentation.

        검은콩과 발효검은콩 추출물이 인간 모유두 세포 성장에 미치는 효과

        본 연구 결과 검은콩 물・에탄올 추출물과 Lactobacillus rhamnosus GG(LGG)와 Bifidobacterium animals subsp. lactis BB-12(BB-12)를 이용하여 발효시킨 발효검은콩 물・에탄올 추출물의 발효 전과 후의 성분변화를 분석하고, 검은콩과 검은콩 발효 추출물이 모유두 세포 성장에 미치는 효과를 알아보기 위해 primary 인간 모유두 세포(HFDPC)에 검은콩 추출물(BWE, BEE)과 발효검은콩 추출물(BWEF, BEE-F)을 다양한 농도로 처리하여 세포독성을 확인하였으며, 이를 바탕으로 적정 처치 농도를 결정하여 모발 성장촉진(VEGF와 KGF/FGF7)과 억제(TGF-β1과 AR)에 관여하는 유전자 발현을 mRNA 수준에서 확인하였다. 나아가 검은콩 추출물이 HFDPC의 생존 촉진에 미치는 영향을 Akt와 Erk의 인산화 활성을 통해 비교 분석하였다. LGG와 BB-12를 이용하여 발효시킨 검은콩은 발효가 진행됨에 따라 pH, 총폴리페놀, 총당・환원당의 함량이 감소하였으며, 검은콩 4종 추출물(BWE, BEE, BWE-F, BEE-F) 중 BWE, BEE, BWE-F가 VEGF의 mRNA 발현을 증가시키고, 모든 처리군에서 KGF/FGF7의 mRNA 발현을 유의적으로 증가시킴을 확인할 수 있었다. 나아가 BWE, BEE, BWE-F가 Erk의 활성을 증가시켰음을 확인할 수 있었다. 따라서 검은콩 물 추출물과 검은콩 에탄올 추출물, 그리고 발효검은콩 물 추출물이 인간 모유두 세포의 세포성장에 모발 성장 촉진인자의 활성과 Erk의 활성화 등을 통한 기전으로 긍정적인 영향을 미침으로써 모발 성장 및 모발 건강을 위한 기능성 원료로서의 가능성을 확인할 수 있었으며, 유산균 2종(LGG, BB-12)을 이용한 발효검은콩이 검은콩과 비교하여 모발 성장 촉진 관련 단백질 발현에서는 유의적인 우월성을 가지지는 않음을 확인하였다. 추후 다른 유산균 균 총을 이용한 발효검은콩의 연구와 더불어 보다 정밀한 발효를 통한 검은콩 추출물의 성분과 조성의 변화를 바탕으로 한 모발 성장 및 모주기 관련 연구가 이루어져야 할 것이라 생각된다. This study was conducted to examine the effects and potential mechanisms of action of black soybean extracts and fermented black soybean extracts by Lactobacillus rhamnosus GG (LGG) and Bifidobacterium animals subsp. lactis BB-12 (BB-12) on proliferation of human follicle dermal papilla cells (HFDPC). We examined changes in pH, total polyphenol, sugar, and reducing sugar contents according to fermentation period of black soybean extracts. Assay using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide was performed to determine cell toxicity levels of the four black soybean extracts [black soybean water extract (BWE), black soybean ethanol extract (BEE), fermented BWE (F-BEW), and fermented BEE (F-BEE)]. Changes in mRNA expression levels of hair growth promoting factors and hair growth inhibiting factors by the four black soybean extracts were measured by real-time PCR. In addition, phosphorylation levels of mitogen-activated protein kinase family proteins were measured by western blot analysis. As a result, fermentation of black soybeans significantly reduced pH, total polyphenols, and sugar/reducing sugar contents. All four black soybean extracts showed no cellular toxicity in HFDPC. In fact, BEE significantly enhanced cell viability of HFDPC at 100 μg/mL compared to control. BWE, BEE, and BWE-F significantly increased mRNA expression of vascular endothelial growth factor, and all four extracts increased mRNA expression of fibroblast growth factor. However, mRNA expression levels of apoptosis-related genes were not affected by black soybean extracts in HFDPC. Furthermore, BWE, BEE, and BWE-F significantly increased phosphorylation levels of extracellular signal-regulated kinase compared to control. Taken together, we demonstrated that black soybean extracts enhanced proliferation of human follicle dermal papilla cells partially via activation of hair growth promoting factors, although no particular significant effects on proliferation were observed by fermentation of black soybeans.

      • 자연치유 관점에서 대두콩과 검정콩 섭취가 인체에 미치는 효과에 대한 사례연구

        본 연구는 자연치유 관점에서 대두콩과 검정콩 섭취를 실천함으로 콜레스테롤을 줄이고 건강에 미치는 효과에 대한 사례연구이다. 연구방법은 대두콩과 검정콩을 섭취하기 전의 총 콜레스테롤 수치와 LDL, HDL, Triglyceride를 참여자 3명에게 병원에서 혈액검사를 실시하였고, 참여자가 섭취한 후에 총 콜레스테롤 수치와 LDL, HDL, Triglyceride를 다시 검사하여서 결과를 알아보았다. 또 하나의 연구방법으로는 실천 전과 실천 후에 혈압계를 측정해서 결과를 보았고 건강 척도도 비교하였다. 식단은 아침에 검은콩 20g과 잡곡밥 1/3 공기, 점심은 대두콩 30g과 검은콩 20g과 잡곡밥 1/3 공기, 저녁은 대두콩 30g과 검은콩 20g, 잡곡밥 1/4 공기를 먹고 간식은 과자나 빵을 먹지 않도록 하였다. 자연치유 효과를 확인하기 위하여 대두콩과 검정콩을 섭취한 결과 건강의 척도는 얼굴 화끈거림도 줄어들었고 머리부터 땀나는 것도 줄어들었으며, 깊은 잠을 자지 못하였는데 수면을 잘 취하게 되었다. 총 콜레스테롤 수치가 줄어들었으며 , LDL 콜레스테롤도 감소되었고, Triglyceride의 수치도 줄어들었다. HDL 콜레스테롤의 수치는 향상되었으며, 혈압의 수치도 정상적인 결과를 나타내었다. 대두콩과 검정콩의 섭취 후에 건강 척도와 혈압측정도 개선시키는 결과를 가져왔다. 현대에는 고콜레스테롤 환자가 점점 많아지고 있는데 병원에서는 약물로 콜레스테롤 수치를 줄이는 사람들이 늘고 있는 현실이다. 고콜레스테롤을 줄이기 위하여 약물을 섭취하면 우리 인체에 미치는 영향으로 신체적 건강이 해로울 수 있지만, 자연치유를 실천하면 치유력이 향상되어 건강을 회복하고 질병을 예방할 수 있어 행복한 삶, 조화로움, 건강한 삶, 활기찬 삶을 살아가는데 도움이 된다고 본다. This study is a case study on the effects on health by reducing cholesterol by practicing intake of soybean and black soybean from the viewpoint of natural healing. For the study method, blood tests were conducted on three participants at the hospital for total cholesterol levels, LDL, HDL, and triglycerides before ingestion of soybeans and black soybeans, and total cholesterol levels, LDL, HDL, and triglycerides were tested again after the participants consumed So, I found out the result. As another research method, blood pressure gauges were measured before and after practice, the results were seen, and health measures were compared. The diet consisted of 20g of black beans and 1/3 bowl of multigrain rice for breakfast, 30g of soybean beans, 20g of black beans and 1/3 bowl of multigrain rice for lunch, 30g of soybean beans, 20g of black beans, and 1/4 bowl of multigrain rice for lunch, and snacks for snacks. forbade me to eat bread. As a result of taking soybean and black soybean to confirm the natural healing effect, the measure of health was reduced face flushing, sweating from the head, and sleep well even though it was difficult to sleep deeply. Total cholesterol levels decreased, LDL cholesterol levels decreased, and triglyceride levels also decreased. The level of HDL cholesterol was improved, and the level of blood pressure also showed normal results. Health measures and blood pressure measurements were also improved after consumption of soybean and black soybean. In modern times, there are more and more patients with high cholesterol, and in hospitals, more and more people are reducing cholesterol levels with drugs. If you take drugs to reduce high cholesterol, your physical health can be detrimental due to the effect on the human body, but if you practice natural healing, you can restore your health and prevent disease by improving your healing power, so you can live a happy, harmonious, healthy life, lively I think it helps in life.

        재배지역에 따른 검정콩 부위별 추출물의 항산화 활성

        With the aim of developing region specialized crops, this study was conducted to clarify effects of variant and cultivation region on antioxidative activities in various black soybean (Glycine max L.) seed tissues. Three black soybean varieties (SCEL-1, Wonheug, and Cheongja 3) were each cultivated in 4 different regions (Jeonju, Pyeongchang, Paju, and Cheonan). Harvested seeds were used to assess DPPH and ABTS radical scavenging activity, and total polyphenol, flavonoid and anthocyanin content. SCEL-1 soybean hull contained higher DPPH and ABTS radical scavenging activity (61% and 85% respectively) compared to Wonheug (40% and 50% respectively). SCEL-1 cultivated in Pyeongchang displayed the highest total polyphenol and flavonoid content (1,189 mg GAE/100g sample and 951 mg CTE/ 100g sample, respectively). Total anthocyanin content was ranked in the following order: SCEL-1>Wonheug>Cheongja 3. All black soybeans showed much higher antioxidant activity in the soybean hull than in the dehulled soybean. The antioxidant activity of black soybeans cultivated at high latitudes was high. These results suggest that the best black soybean variant for high beneficial biological activities is the SCEL-1 variant. For a complete understanding of the potential of black soybean as functional foods, we plan to further analyze their antioxidant activities in future studies.

        검정콩 종피 함유 안토시아닌의 최적 추출조건

        Soybeans (Glycine max) with a black seed coat have been widely utilized as food source and as a medicinal herbs in Korea. The pigmentation in the seed coat of black soybean is due to accumulate anthocyanins in the epidermis palisade layer. The anthocyanin content and composition of the black soybean seed coat are considered as a standard of quality evaluation of black soybean. The main objective of this study was to investigate the optimal condition for an extraction method of anthocyanins and compare anthocyanin quantity and composition within black soybean varieties and germplasms. In the test of extraction solvent, absorbance at 530 nm and Hunter’s a value were increased as increasing the concentration of MeOH, but Hunter’s L and b values were the exact opposite of absorbance and Hunter’s a values. There was no significant difference for anthocyanin contents from 1% HCl - H₂O to 1% HCl - 80% MeOH. In the aspects of anthocyanin contents and HPLC peak resolution, 1% HCl - 20% MeOH extraction solution was the most suitable solvent. Among the 5 kinds of extraction method using 1% HCl - 20% MeOH solution, the anthocyanin contents of room temperature extraction at 72 h was the highest among the methods. High extraction temperature, sonication and reflux method influenced on the decrease of anthocyanin contents because of breakdown of anthocyanins. There was no significant difference for extraction time between 12 h and 24 h. However, the optimal extraction condition were at room temperature for 12 h. The anthocyanin contents in seed coats of black soybean were determined on the basis of HPLC peak area at 530 nm. Ten black soybean varieties and germplasms were tested with optimal conditions founded in this study. On the basis of antocyanin component, these can be classified into three groups; C3G, C3G + D3G and C3G + D3G + Pt3G. The total anthocyanin content in seed coats ranged from 1.58 to 10.62 mg/g of seed. The total anthocyanin content of the variety “Geomjeongol” was about 7 times higher than that of variety “Heugchong”. Information for extraction method and diversity in antocyanin of soybean seed coats can be used for future research for germplasm evaluation and development of high quality black soybean varieties.

      • 검정콩의 일반성분 함량 및 종실 경도

        Black colored soybean has been cultivated for a long time from ancient in Korea, and its uses such a for cooking with rice are peculiar as compared with ordinary soybean. This study was conducted to investigate the chemical component content and hardness of black colored soybean seed. Soybean seeds of 13 selected lines were analyzed for protein, oil, total sugar, fatty acid and amino acid content. The ranges in values for protein, oil and total sugar were 37.3 to 42.0, 17.2 to 21.3,and 8.3 to 13.0 % respectively. And unsaturated fatty acid contents were 84.6 to 87.0 %. In case amino acid, serine, threonine and thyrosine content in black colored soybean seeds were higher than yellow seed coat soybean, especially, methionine was detected a little amounts in black colored soybean while on the other hand that was not analyzed in ordinary soybean. The results show that black colored soybean seed components may be different in comparison with yellow seed coat soybean's. Dried soybean seed absorbed water fast after the first 2hrs followed by a show rate of uptake, which is reached completely hydration after 24hrs at 25℃. The hardness of soybean measured by texturemeter ranged from 5.04 to 7.58 ㎏, which was not shown a certain tendency to alter according to seed size, cotyledon color and 100 seeds weight.

        대두와 쥐눈이콩 추출물의 항산화 활성에 관한 연구

        This study was carries out to analyzed the antioxidant activities of extracts extrated from soybean and small black bean for the development of functional materials. Yields of hot-water and ethanol extracts were higher in the soybean than small black bea, and yields of hot-water extracts were higher than ethanol extracts. Total phenol contents were 26.22 mg/g in the ethanol extracts from small black bean, and were higher than those in the soybean extracts. Total flavonoid contents were 30.52 mg/g of hot-water extracts from small black bean. The EDA values were increased by the increase of extract concentrations and were high in the hot-water and ethanol extracts from bean and small black bean The EDA values was high in the extracts of small black bean from low concentrations. The SOD-like activity of hot-water and ethanol extracts from small black bean was higher than those of soybean extracts. The SOD-like activity was 73%, 62% of the hot-water and ethanol extracts from small black bean in 10 ml/mL extract concentration, respectably. The nitrite scavenging ability was high extracts from soybean was 47.60% in pH 1.2, Those of hot-water extracts were higher than in ethanol extracts in 10 ml/mL extract concentration. The xanthine oxidase inhibitory activities were increased by the increase of extract concentrations and were high in ethanol extracts from small black bean of low concentration. The xanthine oxidase inhibitory activities was hot-water extracts from soybean were higher than 5 ml/mL extract concentration than those of other extracts. The inhibitory activities of tyrosinase were increased by increase of extract concentrations and were high each other 42.05%, 45.67% of hot-water and ethanol extracts from small black bean in 10 ml/mL extract concentration. Reducing power was increased by increase of extract concentrations in extracts of small black bean, and were high more than double in hot-water and ethanol from small black bean than extracts of soybean.

Ames test를 이용한 항돌연변이 실험에서 간접돌연변이원인 AFB1에 대해 모든 종류의 콩 메탄올 추출물들은 농도 의존적으로 돌연변이 억제 효과가 증가하였다(p<0.05). 대두 메탄올 추출물보다는 검정콩들인 흑태, 서목태 및 서리태 메탄올 추출물에 의한 효과가 높은 경향을 나타내었다. 검정콩 중에서 약콩이라고 불리는 서목태에 의한 항돌연변이 효과가 높아 첨가농도 5 ㎎/plate일 때 82%의 효과를 나타내었다. 직접 돌연변이원인 MNNG에 대한 각 종 콩 메탄올 추출물들의 항돌연변이성 실험을 한 결과, AFB1에 대한 효과와 유사하게 검정콩 중 서목태에 의한 효과가 높았으며 첨가농도 2.5 및 5 ㎎/plate일 때부터 각각 58% 및 61%로 돌연변이를 억제시켰다. 인체 암세포를 이용하여 대두 및 검정콩 메탄올 추출물들에 의한 암세포 증식 억제 효과를 검토한 결과, 모든 종류의 콩 메탄올 추출물(첨가농도 1 ㎎/㎖)을 인체 위암세포(AGS)에 처리했을 때 50% 이상의 암세포 증식 억제 효과를 나타내었고 대두보다는 검정콩에 의한 효과가 높았으며 흑태, 서목태 및 서리태 메탄올 추출물은 각각 61%, 70% 및 65%의 암세포 증식 억제효과를 나타내었다. 인체 결장암세포(HT-29)의 경우, 검정콩 메탄올 추출물에 의한 효과가 대두보다 높은 경향을 나타내었으나 서목태 메탄올 추출물을 제외하고는 유의적 차이는 살펴 볼 수가 없었다. 인체 간암세포(Hep 3B)에 의한 증식 억제효과는 이상의 암세포에 대한 효과보다 다소 낮았으나 흑태, 서목태 및 서리태 메탄올 추출물은 첨가농도 1 ㎎/㎖에서 각각 51%, 59% 및 52%의 저해효과를 나타내어 여기서도 서목태에 의한 억제효과가 높았다. 따라서 본 연구의 결과로부터 Ames test를 이용한 항돌연변이 및 인체 암세포 증식 억제 실험에서 검정콩 메탄올 추출물에 의한 억제 효과가 높았고 특히 소립 검정콩에 해당하는 서목태에 의한 생리활성이 우수하였으므로 검정콩에 많이 함유된 색소에 의한 효과라고 추정되며 여기에 대한 향후 연구가 필요하다. This study compared the inhibitory effects of methanol extracts from yellow and black soybeans (black soybean, Seomoktae and Seoritae) on mutagenicity using the Ames test and growth of human cancer cells (AGS human gastric adenocarcinoma, HT-29 human colon cancer, Hep 3B hepatocellular carcinoma cells). In the Ames test system using Salmonella typhimurium TA100, aflatoxin B₁ (AFB₁)-induced mutagenicity was significantly inhibited by treatments with the methanol extracts from either yellow or black soybeans in a dose dependent manner (p<0.05). The methanol extracts from various black soybeans tended to have a greater inhibitory effect compared to those from yellow soybeans. As for N-methyl-N'-nitro-N-nitrosoguamidine (MNNG)-induced mutagenicity, the methanol extracts (5 ㎎/assay) from black soybean, Seomoktae and Seoritae showed 51%, 61% and 53% inhibitory rates, respectively, indicating that Seomoktae, a type of black soybean, had a stronger antimutagenic activity against mutagens (both AFB₁ and MNNG). Methanol extracts from black soybeans showed an inhibitory rate of greater than 50% on the growth of human cancer cells (AGS, HT-29 and Hep 3B) and the inhibition was more effective in the methanol extract from Seomoktae. Our results suggested that the methanol extracts from black soybeans showed stronger inhibitory effects on mutagenicity and growth of cancer cells than those from yellow soybean. It is concluded that intake of black soybean can be recommended for improving health.

        검정콩 종피 함유 안토시아닌의 최적 추출조건

        검정콩 품질평가의 한 기준이 되는 안토시아닌의 함량평가를 위한 분석기술 개발의 일환으로 검정콩 종피 함유 안토시아닌의 최적의 추출용매 및 추출방법을 확립하고, 주요재배 검정콩 종피 함유 안토시아닌의 함량을 정량적으로 평가한 결과를 요약하면 다음과 같다. 1. 검정콩 종피 함유 안토시아닌의 최적 추출용매를 검토한 결과 1% HCl - H2O 로부터 1% HCl - 80% MeOH용액까지 통계적 차이가 인정되지 않았으나, 산술적으로 1% HCl - 60% MeOH 용액으로 추출하였을 때 총 안토시아닌의 함량이 7.72 mg/g으로 가장 높은 양상을 나타내었으나, 추출용매에 메탄을 함량이 높아질수록 안토시아닌 피크의 분리도가 극히 불량한 결과를 나타내므로 종합적 견지에서 볼 때 1% HCl - 20% MeOH를 이용하여 안토시아닌을 추출하는 것이 가장 우수한 것으로 평가되었다. 2. 검정콩 종피 함유 안토시아닌의 최적 추출방법 및 추출시간을 검토한 결과 상온 12시간 추출이 가장 우수한 결과를 나타내었으며, 추출시간이 24시간 이상 지속될 경우 추출시간이 증대됨에 따라 안토시아닌의 함량이 소량 감소되는 양상을 확인할 수 있었다. 3. 국내외 주요 재배 검정콩 품종별 종피 함유 안토시아닌 색소의 함량을 비교한 결과 총 안토시아닌의 함량 범위는 1.58-10.62 mg/g으로 조사되었고, 조사된 품종 중 검정 올콩이 10.62 mg으로 가장 높은 양상을 나타내었으며, 가장 낮은 함량을 나타낸 흑청콩 대비 약 7배 높은 안토시아닌 함량을 나타내었다. Soybeans (Glycine max) with a black seed coat have been widely utilized as food source and as a medicinal herbs in Korea. The pigmentation in the seed coat of black soybean is due to accumulate anthocyanins in the epidermis palisade layer. The anthocyanin content and composition of the black soybean seed coat are considered as a standard. of quality evaluation of black soybean. The main objective of this study was to investigate the optimal condition for an extraction method of anthocyanins and compare anthocyanin quantity and composition within black soybean varieties and germplasms. In the test of extraction solvent, absorbance at 530 nm and Hunter's a value were increased as increasing the concentration of MeOH, but Hunter's Land b values were the exact opposite of absorbance and Hunter's a values. There was no significant difference for anthocyanin contents from 1% HCl - H2O to 1% HCl - 80% MeOH. In the aspects of anthocyanin contents and HPLC peak resolution, 1% HCl - 20% MeOH extraction solution was the most suitable solvent. Among the 5 kinds of extraction method using 1% HCl - 20% MeOH solution, the anthocyanin contents of room temperature extraction at 72 h was the highest among the methods. High extraction temperature, sonication and reflux method influenced on the decrease of anthocyanin contents because of breakdown of anthocyanins. There was no significant difference for extraction time between 12 h and 24 h. However, the optimal extraction condition were at room temperature for 12 h. The anthocyanin contents in seed coats of black soybean were determined on the basis of HPLC peak area at 530 nm. Ten black soybean varieties and germplasms were tested with optimal conditions founded in this study. On the basis of antocyanin component, these can be classified into three groups; C3G, C3G + D3G and C3G + D3G + Pt3G. The total anthocyanin content in seed coats ranged from 1.58 to 10.62 mg/g of seed. The total anthocyanin content of the variety "Geomjeongol" was about 7 times higher than that of variety "Heugchong". Information for extraction method and diversity in antocyanin of soybean seed coats can be used for future research for germplasm evaluation and development of high quality black soybean varieties.
