The present study was carried out to investigate the potential acute toxicity of safflower seed by a single oral dose in ICR mice. Twenty animals of each sex were randomly assigned to four groups of 5 rats each and were administered singly by gavage at dose levels of 0, and 2,500 mg/kg bodyweight. Mortalities, clinical findings, and body weight changes were found and complete gross postmortem examinations were performed. Throughout the study period, no treatment-related deaths were observed. There were no adverse effects in clinical signs, body weight, and gross findings at any dose tested. The results showed that the single oral administration of safflower seed did not cause any toxic effect at the dose levels of 2,500 mg/kg in rats and the minimal lethal dose was considered to be over 2,500 mg/kg body weight for both sexes.

This study was performed to evaluation the skin and eye irritation of etofenprox 8% and spinosad 4% combination in male New Zealand White rabbits. In skin irritation test, the combination of etofenprox and spinosad did not showed any change of body weight but caused slight skin irritation with the P.I.I. (primary irritation index) value 0.25. The etofenprox and spinosad combination showed severe eye irritation 1 h, 24 h, 48 h, and 72 h after ocular treatment, having the A.O.I. (acute ocular irritation) value of 37.3. Therefore, these result indicated that the combination of etofenprox and spinosad might be irritant formulation on eye and skin.

저자들은 만성골수성백혈병으로 동종 골수이식과 이식편 부전으로 인하여 추가적인 말초혈액 조혈모세포이식을 시행 받은 환자에서 만성 이식편대숙주질환과 그 폐 합병증인 폐색성 세기관지염에 동반된 자발성 기종격동과 피하 기종이 병발한 1예를 경험하였기에 이를 보고하는 바이다. Obstructive lung disorders following after allogeneic bone marrow transplantation (BMT) in association with graft- versus-host disease (GVHD) contribute significant morbidity and mortality. We report a case of a 28-year-old man who developed spontaneous pneumomediatinum and subcutaneous emphysema complicating bronchiolitis obliterans after allogeneic BMT. He received an allogeneic BMT for chronic phase of chronic myeloid leukemia. Five months after BMT, he was boostered by allogeneic peripheral blood stem cells from the same donor due to graft failure. One month after the boostering, chronic GVHD developed and were treated with cyclosporine and steroid. The patients developed spontaneous pneumomediatinum and subcutaneous emphysema secondary to severe bronchiolitis obliterans 4 months after boostering donor cells. The air-leak syndromes were recovered by conservative management, including high-flow oxygen.
엔로플록사신 액제의 닭 호흡기 및 소화기 감염증에 대한 예방 및 치료효과

We evaluated the efficacy of the liquid form of enrofloxacin against poultry respiratory and digestive infections, the major problems in the poultry industry, from diverse diagnosis viewpoints. Results obtained through this study were as follows: 1. The liquid form of enrofloxacin showed good acceptability to poultry owing to no abhorrence to the drug by chickens, which indicated its suitability for mixing in drinking water. 2. The application of the enrofloxacin formulation improved the productivity of the chicken farm to a great extent by lowering the death toll of chicken raised in the farm. 3. The use of the drug shortened the days to the market, and increased the body weight at the time of marketing. 4. Preventive and therapeutic effects of the liquid enrofloxacin were observed higher than ampicillin, a major antibiotic used in poultry farming. From above results, the liquid form of enrofloxacin would find its use for the prevention and treatment of digestive and respiratory infections, let alone the growth promotion and feed efficiency, especially in the chicken farming practice.

In this study the antimicrobial spectrum and sensitivity of a liquid formulation of enrofloxacin against the major six pathogenic bacteria isolated from the diseased poultry in Korea were evaluated in comparison with ciprfloxacin (CFX), gentamicin (GM) and penicillin (PC). A liquid form of enrofloxacin, a test material and ciprofloxacin showed at least 83% antibacterial activity against all of six test pathogenic microorganisms at the fixed concentration of 10 ㎍/㎖. Gentamicin and penicillin revealed much lower antibacterial activity at the same condition. We evaluated the liquid form of enrofloxacin as having widespread antibacterial spectrum against poultry infections. The liquid form of enrofloxacin showed potent antibacterial activity, with the MIC range being 0.12∼2 ㎍/㎖ against Gram positives, 0.06∼2 ㎍/㎖ against Gram negatives and 0.5∼2 ㎍/㎖ against Mycoplasma spp. These values were equivalent to or somewhat higher than those of ciprofloxacin, but much lower than those of gentamicn and penicillin. This new liquid form of enrofloxacin would find its way in application of mixing with drinking water for the prevention and treatment of diverse infections in chickens.

<P>Hwangryunhaedok-Tang (HR) and berberine-containing single herbs are used to treat bacterial infection and inflammatory diseases in eastern Asia. The combination of berberine-containing herbal medicines and ciprofloxacin can be an excellent antibacterial chemotherapy against multidrug resistance bacteria. To evaluate the pretreatment effect of berberine and HR, vehicle, berberine (25 and 50 mg/kg/day), and HR (1.4 g/kg/day) were daily administered to rats for five consecutive days. On day 6, ciprofloxacin was administered (10 mg/kg, i.v. and 20 mg/kg, p.o.) to rats. To assess cotreatment effect of berberine and ciprofloxacin, berberine (50 mg/kg) and ciprofloxacin (20 mg/kg) were coadministered by single oral gavage. Pharmacokinetic data were estimated by noncompartmental model. Compared with ciprofloxacin alone (control group), coadministration of berberine (50 mg/kg) and ciprofloxacin significantly decreased <I>C</I><SUB>max</SUB> of ciprofloxacin (<I>P</I> < 0.05). In addition, the pretreatment of berberine (50 mg/kg/day) and HR (1.4 g/kg/day) significantly decreased <I>C</I><SUB>max</SUB> and AUC<SUB>0→<I>∞</I></SUB>, compared with control group (<I>P</I> < 0.05). The oral bioavailability of ciprofloxacin was reduced by cotreatment of berberine and pretreatment of berberine and HR. Our results suggest that the expression of P-glycoprotein and organic anion and/or organic cation transporters (OAT/OCT) could take a role in reduced oral bioavailability of ciprofloxacin by berberine and HR.</P>
<i>In Vitro</i> and <i>In Vivo</i> Genotoxicity Assessment of <i>Aristolochia manshuriensis</i> Kom.

<P><I>Arisolochiae species </I>plants containing aristolochic acids I and II (AA I and AA II) are well known to cause aristolochic acid nephropathy (AAN). Recently, there are various approaches to use AAs-containing herbs after the removal of their toxic factors. However, there is little information about genotoxicity of <I>Arisolochiae manshuriensis</I> Kom. (AMK) <I>per se</I>. To obtain safety information for AMK, its genotoxicity was evaluated in accordance with OECD guideline. To evaluate genotoxicity of AMK, we tested bacterial reverse mutation assay, chromosomal aberration test, and micronucleus test. Here, we also determined the amounts of AA I and II in AMK (2.85 ± 0.08 and 0.50 ± 0.02 mg/g extract, resp.). In bacterial reverse mutation assay, AMK dose-dependently increased revertant colony numbers in TA98, TA100 and TA1537 regardless of metabolic activation. AMK increased the incidence of chromosomal aberration in Chinese hamster ovary-K1 cells, but there was no statistically significant difference. The incidences of micronucleus in bone marrow erythrocyte were significantly increased in mice after oral administration of AMK (5000 mg/kg), comparing with those of vehicle group (<I>P</I> < 0.05). The results of three standard tests suggest that the genotoxicity of AMK is directly related to the AAs contents in AMK.</P>
Toxicity of Hematoporphyrin-Coated Magnetic Ferrofluid in Rats

The purpose of this study was to investigate the toxicity of hematoporphyrin-coated magnetic ferrofluid (HP-MF) through intravenous administration in Sprague-Dawley rats. Each group was treated with either saline, or the HP-MF at 0.5, 1, 1.5, 2 and 4 ml/kg body weight (b.w.) for the observation of survival rate, clinical symptoms, laboratory values and histopathological findings. In this study, HP-MF was evaluated for the survival rates, symptoms, laboratory values and histopathological examination after treatments. The result revealed that the animals in the group of HP-MF at 2 and 4 ml/kg b.w. showed some lethality. In serum biochemistry, the levels of AST, ALT and ALP were increased in the MF and HP-MF treated groups. However, histopathological examination for the suspected organs showed no evidence of hepatotoxicity and nephrotoxicity of typical iron poisoning. Though the toxicity of HP-MF was higher than that of HP, long retention of hematoporphyrin via HP-MF provides additional benefit over conventional hematoporphyrin. HP-MF could be utilized as a potential photodynamic agent in cancer therapy. It is suggested to develop an efficient external magnetic device to attract hematoporphyrin in the target site, thereby enabling to administering a small amount of HP-MF.

<P>Surfactin C, produced by <I>acillus subtilis</I> isolated from Korean soybean paste, was given to Sprague-Dawley rats of both sexes at dose of 500, 1000 or 2000 mg/kg for 28 days. There were no surfactin C-related toxicities in survival, clinical signs, and hematological parameters in the experimental period. The highest dose of surfactin C showed the decrease in body weight gain despite normal food and water consumptions and the increase in relative liver weight. alanine aminotransferase (ALT), aspartate aminotransferase (AST) and alkaline phosphatase (ALP) levels were increased in animals administered with surfactin C of 1000 or 2000 mg/kg. Zonal necrosis of hepatocyte around the hepatic vein was observed after administration of the same doses in a dose-dependent manner. In the present study, the no-observed-adverse-effect level (NOAEL) of surfactin C was 500 mg/kg following oral administration in rats.</P>
