      • Lactobacillus acidophilus 88과 Lactobacillus bulgaricus IFO 13953간의 세포융합주의 특성에 관한 연구

        유산균의 균주개량방법의 일환으로 protoplast fusion 기법과 electrofusion법을 이용하여 protease 활성, lipase 활성, 내열성, 내산성 등이 우수한 L. bulgaricus 와 bacteriocin을 생산하는 L. acidophilus를 융합시켜 얻은 융합주들의 생리학적 성질을 검토하였다. 산 생성능, 내열성, 내산성, protease, lipase 활성 등은 L. bulgaricus가 L. acidophilus 보다 우수하였다. L. bulgaricus는 lactose와 sorbose를 이용하였으나 maltose와 sorbitol을 이용하지 못하는 반면, L. acidophilus는 maltose를 이용하고 lactose와 sorbose를 이용하지 못하였다. 융합주 가운데서는 3, 6, 7, 8, 10번이 모균주의 발효능 특성을 함께 지님으로서 재조합체임을 확인 할 수 있었으며, sorbitol의 경우 모균주에서는 발효능이 전혀 나타나지 않았음에도 불구하고 융합주 4, 7번이 발효능을 나타내어 융합과정에서 새로운 형질을 획득하기도 한다는 사실을 알 수 있었다. Lactase 활성은 모균주 모두 높은 β-galactosidase 활성을 보였으나, phospho-β-galactosidase 활성은 거의 없었으며 융합주들도 다소 차이는 있었지만 모균주와 유사한 효소 활성을 나타내었다. 발효에 있어서 key enzyme으로 작용하는 protease, lipase 등의 효소 활성도 모균주의 활성 보다 우수한 융합주도 존재하였다. Aninterspecific fusant was made from the protoplasts of two strains of lactobacillus genus (streptomycin and lincomycin resistant L. bulgaricus and kanamycin resistant L. acidophilus 88). The functional properties of the fusant were examined by determining bacteriocin productivity, acid producing activity, ability of carbohydrates fermentation and three important enzyme activities. The recombinant strain revealed bacteriocin productivity. Acid production and β galactosidase, phospho-β-galactosidase, lipase and protease activity of L. bulgaricus were better than those of L. acidophilus 88. Among fusants, β-galactosidase activity of two strains were better than that of the parent strains but phospho-β-galactosidase activity remarkably lower. One fusant revealed the improved proteolysis compared to that of the parent strains- Lipase activity of L. bulgaricus was better than that of L. acidophilus but another fusant exhibited the highest lipase activity.

        Rheumatoid arthritis frequently involves the cervical spine and may lead to neurologic impairement. However, direct involvement of CNS structures by inflammatory cells has been reported infrequently. The prevalence of this complication of rheumatoid arthritis is unknown. Inflammatory CNS involvement in rheumatoid arthritis reportedly occurs in the setting of longstanding, active, erosive articular disease and is accompanied by extracranial and extraspinal nodules and vasculitis. This is diagnosed by radiologic finding of CNS nodules or meningeal thickening and by biopsy or autopsy. Treatment with corticosteroid, cytotoxic agent or surgical decompression is helpful. But the majority of patients die within several months of onset of neurologic symptom. Recently, we experienced a case of pachymeningitis caused by rheumatoid arthritis, which resolved repeatedly with azathioprine treatment.

        초등학교 아동의 치과치료장면 그림의 주조색과 불안도의 연관성에 관한 연구

        The purpose of the study was to observe the association between the main colors of school chidlren's drawings of dental treatment situation and their dental anxiety. The subjects of study were 688 school children from 2nd to 4th grade. Caries experience was investigated by oral examination. Questionnaire survey was done regarding children's attitude toward the dentist and dental treatment and their experience of pain and fear during the dental treatment. Colors with high frequency of use were green, blue, yellow, orange, pink, purple, gray, brown, yellow ochre, red, white and black in order. Boys used green and blue, and girls used yellow and orange, more than each other. As the grade increased, white, yellow, and blue increased and brown decreased. Colors with high dental anxiety level were black, brown, red, white, blus, orange, yellow and green, yellow ochre, gray, pink, and purple in order, and the upper four represented 10.5% of the subjects. Children with high decayed teeth rate had a tendency to use colors of high dental anxiety level. Black color had the most obvious association with the pain and fear of dental treatment.

      • 항암화학요법을 받는 암 환자에 대한 Nucare^R의 영양지지효과

        Purpose : The purpose of this study was to evaluate the effect of nutritional support with enteral liquid supplement in cancer patients receving chemotherapy for possible benefit in nutritional, immunologic and golbal function of patients. Patients and Methods : From October 1995 to February 1997, 30 advanced cancer patients receving chemotherapy were divided two roups. The Nycare group, in addition to normal diet, Nucare^R enteral supplement was given for 1week right after chemotherapy for the duration of 2 chemotherapy cycles. Control group received only normal diet without parenteral fluid supplement for 2 chemotherapy cycles. Results : Median ages were 53 end 56 years for Nucare group and Contrl group respectively. Performance scores was less than 2 by ECOG scale. All patients were stage Ⅲ or Ⅳ. The physical parameters, such as weight, arm muscle circumference(AMC) and triceps skin fold(TSF) were decreased in both groups after 2 cycles of anticancer therapy. but it was less severe in Nucare group(p<0.05). Serum transferrin was maintained in mild deficit state in Nucare group, whereas it aggravated form mild to moderate deficit in Control group(p<0.05). Serum albumin level increased in Nucare group without statistical significance. but it decreased from normal to mild deficit in Control group. Serum total protein did not change significantly in Nucare group. but in Control group, serum total protein was decreased from 7.24±0.9 to 6.52±0.5(P<0.05). Total lymphocyte count did not change significantly in both groups. Conclusion : This study shows that the nutritional support with Nucare^R was effective in the prevention of nutritional deficit status in patients receving a nticancer chemotherapy.

      • 김치 유산균인 Lactobacillus sp. JJ-2001이 생산하는 Bacteriocin에 관한 연구

        The microorganisms producing bacteriocin from kimch were screened, and among them,a strain showing the highest bacteriocin activity and stability was isolated. Based on the taxonomic characteristics of its morphological, cultural and physiological properties, the isolatef strain was characterized as Lactobacilius sp..To identify optimal conditions for the growth and the production of bacteriocin from the isolated strain,nutrient source, initial pH, and culture temperature were evaluated. Physiochemical properties of bacteriocin was also investigated. The potimal condition for the production of bacteriocin and the cell growth was obtained at 2% glucose, 1% beef extract as organic nitrogen source,1%(NH_4)_2SO_4 as inorganic nitrogen source,and 0.1% Tween 80. The isolated strain showed the maximum growth at 25℃. The inhibitory range of its antimicrobial substance aginst gram positive bacteria was marrow and its inhibitory activity was maintained between pH3.0 and pH8.0. The antimicrobial activity of bacteriocin from the isolated strain was stable at 50℃.

      • Citrobacter sp.에 의한 Triphenylmethane계 색소의 분해

        염색공장 폐수처리장의 폭기조로부터 분리한 Citrobacter sp. SK-3균주의 생육 특성을 검토하고, triphenylmethane계 색소중 염기성 염료로 사용빈도가 높은 crystal violet을 비롯한 pararosaniline, brilliant green등에 대한 분해능을 조사하였으며, crystal violet의 분해산물을 HPLC로 추정하였다. Citrobacter sp. SK-3균주의 triphenylmethane계 색소를 분해하기 위한 최적배지 조성은 최소 무기염배지에 탄소원으로서는 galactose 0.5%, 질소원으로서는 beef extract 0.1%, 초발 pH는 8.0-9.0 였으며, 최적배지에 crystal violet 200ppm을 첨가하여 40℃ 부근에서 배양하였을 때 24시간 이내에 100%의 분해율을 나타내었다. Citrobacter sp. SK-3균주는 triphenylmethane계 색소중 crystal violet뿐만 아니라 malachite green, pararosaniline, brilliant green, methyl violet, basic fuchsin 등과, Azo계 색소인 methyl red를 분해하였다. Citrobacter sp. SK-3균주는 무기염 배지상에서 crystal violet, brilliant green을 유일한 탄소원, 질소원으로 이용하여 생육 가능하였다. Citrobacter sp. SK-3균주에 의한 crystal violet 분해산물을 TLC 및 HPLC로서 분석한 결과, crystal violet의 기본골격에 결합된 methyl기가 점차 demethylation되어 pentamethyl-, tetramethyl-, trimethyl- pararosaniline등의 물질들로 분해되어짐을 확인할 수 있었다. The optimal condition for degradation of crystal violet and other triphenylmethane dyes by Citrobacter sp. SK-3 isolated from the activated sludhe of dye manufacturing factory was investigated. The optimal culture medium for the degradation of triphenylmethane dye was composed of minimum inorganic salt medium supplemented with 0.5% galactose, 0.1% beef extract, with the initial pH of 8.0 to 9.0. Under this condition, Citrobacter sp. SK-3 degraded 200ppm of crystal violet completely within 24 hours. Citrobacter sp. SK-3 also degraded efficiently malachite green, pararosaniline, brilliant green, methyl violet, basic fuchsin and methyl red. Analysis of the degradation products of crystal violet through thin layer chromatography and high performance liquid chromatography indicated that the methyl groups bound to crystal violet backborn were gradually demethylated to pentamethyl-, tetramethyl- and trimethylpararosaniline.

        광중합형 수복용 복합레진의 인장강도에 미치는 thermal cycling과 시효 처리의 영향

        The improvement in the mechanical properties of restorative composites has contributed to their increased use in recent years. In spite of their growing popularity, composites still have low resistance to fracture and wear in oral environments. This study was performed to examine whether the tensile strengths of light-cured restorative composites were affected by thermal cycling and aging in water. Five commerically available light-activated composites(Heliomolar : HM, Aelitefil : AF, Amelogen : AG, Clearfil AP-X : CF, Z100 : ZH) were used. Rectangular tension test specimens were fabricated in a teflon mold giving 5 mm in gauge length and 2mm in thickness. All samples were divided into 3 groups. Group 1 was stored in a desiccator for 10 days. Group 2 was immersed in a 37℃ distilled water for 10 days. Group 3 was subjected to 10,000 thermal cycles between 5℃ and 55℃, and the immersion time in each bath was 15 seconds per cycle. Tensile testing was carried out at a crosshead speed of 0.5 mm/min and fracture surfaces were observed with scanning electron microscope. The results obtained were summarized as follows; 1. The strength data fit to single-mode Weibull distribution(r²> 0.86) and Weibull parameters decreased after thermal cycling test. 2. The highest characteristics strength was observed in the CF group but the highest Weibull modulus was obtained in the AF group after thermal cycling test. 3. The highest tensile strength value of 83.92 MPa was obtained in the CF group and the lowest value of 13.91 MPa was obtained in the AG group. Tensile strengths showed no significant difference between HM and AG groups but other groups showed the significant differences in each group(p<0.05). 4. According to the microscopic observation of the fracture surfaces, fractures of large fillers were observed in the hybrid composites of the AF and CF groups.

        남북한 산업 규격의 비교 연구

        The technological and industrial traits of North Korean today are very different with those of South Korea as politics, economy, culture of both Koreas are, which have been developed in respective ways during the last fifty years. Facing with today's many real activities for the interchange and the cooperation like the construction of railroads and industrial complexes, lack of the general understanding of dissimilarities in characteristics of both countries are making it difficult to accomplish joint objects. However, any examination about those problems has never been tried because of the lack of practical materials and data about the technology and the industry in the North. This paper addresses the analysis procedure and method of the difference of the national industrial standard between South and North Korea in order to understand the difference. We already investigated the hierarchical classifications, organization, regulative items before and the systematic analysis procedure including the comparative study of the national standard based on the characteristics of each standard and the guide table which can be used in the comparison of the standards are accomplished in this paper. Based on the suggested comparison procedure, we reported the difference of the standard in the area of mechanical industry as a result of the comparison of the standards of South and North Korea over 360.

      • 단기배양을 통한 말초혈액 CD34 양성세포의 체외증폭

        Background: It is suggested that clinical practice in the areas of bone marrow transplantation and gene therapy might rely on the ex vivo expansion of hematopoietic stem/progenitor cells. However, the condition for ex vivo expansion of hematopoietic progenitor cells is not well established. The authors pursued a series of experiments to define the proper conditions for the expansion of hematopoietic cells in the short-term liquid suspension culture of mobilized peripheral blood CD34+ cells. Methods: 1.0ml cultures were initiated with 9×10^3 PB CD34+ cells, which were isolated from PB mononuclear cells (MNCs) by high-gradient cell sorting, in 12 well plates with the various combinations of hematopoietic growth factors(HGF). The following recombinant human HGFs were used: stem cell factor(SCF) 100ng/ml, granulocyte colony-stimulating factor(G-CSF) 100ng/ml, GM-CSF(granulocyte, macrophage colony-stimulating factor) 100ng/ml, interleukin-1 beta(IL-1B) 1ng/ml, interleukin-3(IL-3) 20ng/ml, interleukin-6 (IL-6) 100ng/ml. At the end of culture, colony-forming cells were evaluated by semisolid clonogenic assay. Results: 1) Using the high-gradient magnetic sorting system, CD34^+ cells were isolated with a yield of 40 3% 2) In 7 day culture of PB CD34^+ cells(9×10^3 cells), nucleated cells expanded mean 10×10^3(range, 9 to 20×10^3) with the addition of SCF alone, 35×10^3(range, 10 to 60×10^3) with SCF plus G-CSF plus GM-CSF, and 130×10^3(range, 40 to 300×10^3) with the combination of SCF, G-CSF, IL-1, IL-3, IL-6, GM-CSF. In 14 day culture, nucleated cells expanded 10×10^3 to 1,860×10^3 with combination of human hematopoietic growth factors. 3) In 10 day culture without medium change of PB CD34^+ cells, CFU-GM numbers expanded 16. 5 fold(range, 7 to 59 fold) with the addition of SCF plus G-CSF plus Il-1 plus IL-3, 31.3 fold(range, 20.5 to 101.1 fold) with the combination of SCF, G-CSF, IL-1, IL-3, GM-CSF. In 14 day culture with or without medium change of PB CD34^+ cells was inferior to 10 day culture for CFU-GM expansion. 4) There was no significant difference for CFU-GM expansion between five growth factors(SCF,G-CSF,IL-1,IL-3,GM-CSF) and six growth factors(five growth factors plus IL-6). Conclusion: The authors could confirm that short-term suspension culture of peripheral blood CD34+ cells could expand hematopoietic progenitor cells. Ten-day culture with medium change of CD34+ cells with the addition of five growth factors, i.e. SCF, G-CSF, IL-1B, IL-3, and GM-CSF, might be the most efficient in this system.

      • 전자석 작동기를 이용한 드릴의 선회진동제어 : Control of the Whirling Vibration of the Drill by Electromagnetic Actuator

        The drill is the most widely used tool in the world to make holes on the workpiece. But the performance of the drill is affected by many factors. One of them is to get the position of the drill at the beginning of the penetrating stage. The drill vibrates laterally while it rotates. Especially in case of a thin and long drill, the magnitude of the whirling vibration of the drill becomes very large and makes critical problem. Therefore, several methods have been used to get a correct position during drilling process. As an example, center drills or guides are used to make the drill get correct position. But these methods are passive. In this paper, studies for an active control are carried out to get the correct position of the drill by reducing the whirling vibration of the drill which occurs before the drill contacts the workpiece. The active phase lead algorithm is applied in order to control the vibration of a twist drill, which has dimension as follows: diameter φ3 and length 150mm.
