      • Fluorescence in situ Hybridizaton에 의한 염색체 이상의 절단점의 규명

        Fluorescence in situ hybrididzation(FISH) is a novel molecular genetic technique. It used for gene mapping in researches and in clinical cytogenetic fields. We perfomed FISH using various kinds of probes for the determination of breakpoints in chromosomal abnormalities. Following results were obtained: 1. Determination of breakpoint in complex translocations. FISH results with chromosome 14/22 centromere probe showed that the breakpoint was 14q11 in the patient with corpus callosum agenesis and complex transocation. 2. Determination of breakpoint of inv(16) in acute myelomonocytic leukemia. FISH results with inv(16) probe showed that the breakpoint was between 16pll and p13 in an abnormal inv(16) chromosome. 3. Determination of breakpoint of dup(17q) in acute myelocytic leukemia. FISH results with iso 17q probe showed that the breakpoint was 17q21 and the duplicated segment from q21-q25. 4. Determination of breakpoint of isodicentric X chromosome. FISH results of Turner syndrome with two iso Xq chromosome using a-satellite X centomere probe showed that the breakpoint was in proxim portion of centromere. From the above results, we could easily find out the breakpoints of chromosomal abnormalities by FISH. It was helpful for the tracing of pathogenesis of genetic disease and cancers.

      • 대장직장암 환자의 술전 및 술후 혈청 CEA측정의 의의

        This report is a clinical study of the significance of preoperative and postoperative serial check of plasma CEA levels in 36 cases of colorectal cancer who were admitted and treated at the Chungnam National University Hospital from Mar. 1987 to Oct. 1988 and the results were as follows: 1. The positive rate of preoperative CEA level(over 5ng/ml) in colorectal cancer was 58.3% In resection group, positive rate and mean CEA level were 53.6% and 7.6ng/ml respective and in unresection group they were 75% and 36.7ng/ml respectively (p<0.05). 2. The proportion of patients with abnormal CEA level(over 5ng/ml)was 33.3% in Dukes B, 46.2% in Dukes' C and 69.2% in Dukes'D. 3. The proportion of patients with abnormal CEA level(over 5ng/ml) was 25% in wel1.diffe rentiated type, 60.9%in moderately differentiated type and 37.5 % in poorly differentiated type. 4. The recurrence rate after curative resection in preoperative normal CEA(0~2.5ng/ml) group was presumed 0% in borderline(2.6~ 5ng/ml) group was 33.3%, in high(over 5ng/ml) group was 16.7% 5. In 3 patients with postoperative liver metastasis had the highest frequency of elevated level(over 60ng/ml), whereas those with local recurrence had a lower frequency of elevated level. 6, A patient had second look procedure include segmental resection of local recurrence, buthe was dead of peritoneal caricinomatosis several months later.

      • 메탄올-아세톤 혼합용매계에서 염화벤질술포닐과 파라치환 아닐린과의 친핵성 치환반응에 대한 속도론적 연구

        The second order rate constants have been determined for the nucleophilic substitution reaction of benzylsulfonyl chloride with anilines in MeOH-(Me)_2CO mixtures at 25℃ and 35℃. Results were shown that (ⅰ)the rate constant of the reaction is the largest in 100-90% MeOH solvent and increase with the electron donating substituent anilines, (ⅰ) the magnitudes of ρ and β associated with a change of substituent in the nucleophile are large and indicate a relatively advanced bond-formation in the transition state, (ⅲ) the reaction procceds via S_n2 mechanism with product-like transition state from solvent effect and quantum mechanical approach.

        The rates of reaction of 4-substituted-2, 6-dinitrochlorobenzenes with para-substituted anilines in methanol-acetonitrile mixtures were measured by conductometry. It was observed that the rate constant increase in the order of Y=α-CH3>β-CH3>Py-H>α-CH3, where Y is a substituent in the pyridine ring. The electrophilic catalysis by methanol may be ascribed to the formation of hydrogen bonds between alcoholic hydrogen and nitrogen of amines in the ground state.

      • 테녹시캄과 β-시콜로멕스트린 간의 고체분산체 형성에 의한 용출 증가

        The purpose of this study is to investigate the interaction of tenoxicam with various cyclodextrins (CDs) in the aqueous solution and in solid state, and finally to formulate a tablet with enhanced dissolution rate. CDs used in solubility studies were α-, β- and γ -CD, 2-hydroxypropyl-β-CD (HPCD), sulfobutyl ether-β-CD (SBCD), dimethyl-β-CD (DMCD) and trimethyl-β-CD (TMCD). The effect of CDs on the solubility of tenoxicam was observed. Solid dispersions were prepared with β-CD using solvent evaporation and freeze-drying process and physical mixtures were prepared by trituration in a mortar. The ratios of drug to carrier were 1:1 and 1:2 molar ratio. These solid dispersions were evaluated by differential scanning calorimetry (DSC), Fourier-transform infrared spectroscopy (FT-IR) and dissolution studies. Solid dispersions were also formulated to tablets and then dissolution rates were compared with that of a commercial product. In aqueous solution, complex formation with α- and β-CD gives rise to Bs type solubility phase diagram, and DMCD, TMCD, HPCD and SBCD showed AL type diagrams. The solubility increase of tenoxicam was in the order of SBCD>γ-CD>β-CD>HPCD>DMCD>α-CD>TMCD. The presence of water soluble polymers such as povidone and hydroxypropyl methylcellulose in β-CD solution did not increase the solubility of tenoxicam. DSC and FT-IR studies confirmed the formation of an solid complex of tenoxicam with β-CD. Dissolution of solid dispersion was about 4-fold higher than that of drug alone and about 2-fold higher than that of physical mixture within 10 min at pH 1.2. As the ratio of carrier to drug increased, dissolution rate became faster. The dissolution rate of tenoxicam from the tablets prepared using freeze-dried solid dispersion at 1:2 molar raio was fast; more than 80% was dissolved within 15 min at pH 1.2. All tenoxicam tablets prepared using tenoxicam-β-CD solid dispersions showed more rapid dissolution rates than commercial tablets. Therefore, the solid dispersion technique is an effective method for increasing the dissolution rate of poorly water soluble tenoxicam. β-CD was thought to be a candidate carrier for preparing solid dispersion with tenoxicam.

        To overcome poor dissolution property of nifedipine (NFD), which is used in the treatment of ischemic heart disease and hypertension as a calcium antagonist, effects of β-cyclodextrin (β-CyD) derivatives on the solubility of NFD in aqueous buffered solutions were investigated. The aqueous solubility of NFD increased linearly with increase in the concentration of CyDs, showing an A???-type phase solubility diagram. The stability constant of NFD-CyD complexes was in the order of 2-hydroxypropyl-β-cyclodextrin (HP-β-CyD) <β-CyD ≪diemthyl-β-cyclodextrin (DM-β-CyD), and the solubilizing ability of the β-CyDs was also in the same order. The dissolution rate of NFD-DM-β-CyD (1 :2 molar ratio) or NFD-β-CyD (1 :2 molar ratio) solid dispersions was much faster than those of NFD-β-CyD physical mixtures and the drug alone. Therefore, solid dispersion system of NFD with β-CyD (1 : 2 molar ratio) was prepared by solvent evaporation and encapsulated with Eudragit RS as the coat at core to wall ratios of 1 : 0.1 and 1:0.3 (w/w) using phase separation method by non-solvent addition. All the Eudragit RS microcapsules of NFD-β-CyD solid dispersion at core to wall ratio of 1:0.1 or 1:0.3 (w/w) were small and uniform. Although dissolution of NFD from Eudragit RS microcapsules was retarded with increasing the ratio of core wall from 1:0.1 to 1:0.3 (w/w), the release rate was sustained in a much higher concentration than those of drug alone and commercial sustained release NFD tablets.

      • 기계적 합금화에 의한 Al-Fe 및 Al-Fe-Ce 합금의 고온 변형거동

        The deformation behavior of a mechanical alloyed Al-Fe and Al-Fe-Ce alloy was studied in the temperature range of 300℃ to 500℃. The steady state for Al-8wt.%Fe powder was obtained after 1000 minutes of mechanical alloying at the process condition of 2wt.% stearic acid, 46:1 of grinding media/powder weight ratio, and 300 rpm of impeller revolving velocity. The relative densities of Al-8wt.%Fe alloy powders reached almost 100% after 1 hour sintering at 470℃ under vacuum state. With increasing the amount of PCA, hardness of the sintered alloys increased but the relative densities were not affected. It was also found that decrease in hardness and tensile strength after high temperature aging treatment was reduced with increasing the amount of Ce addition.

        민간 산후조리원 공공성 제고를 위한 산후조리서비스 체계 고찰 및 개선 방안

        The study aimed to review the current postpartum care service system and suggest measures of improvement for the public reinforcement of the private-postpartum care center (PCC). We conducted a review of literature, including an internet search, and we analyzed the in-depth interviews using questionnaires answered by workers from the 6 private-PCCs operated in Goyang city. Using these data, we derived possible measures which could improve the publicity of private-PCCs. It was found that the 11 nationwide public-PCCs had serious operating deficits, and its utilization by the vulnerable was insufficient. In areas where private-PCCs are concentrated, we can propose the following policies to strengthen publicity, rather than establishing another public-PCC. First, it is important to provide support for private-PCCs to apply cost reductions for vulnerable groups, similar to the maternal and newborn medical care of public- PCCs. Second, publicity would be strengthened by reorganizing the pregnancy and childbirth education program, while also offering additional services for mothers with stillbirths and abortions. For the integrated management of postpartum care services, this should be shifted towards reinforcing the publicity of private-PCCs, rather than operating a public-PCC in relative ly concentrated private-PCC areas. Through these policies, it is expected that financial and administrative waste will be reduced, and an integrated management system for postpartum care services will be established.

      • 탐구 학습과 STS 관점에서 본 5,6차 과학 교과서의 비교 분석 : 중학(Ⅰ) 교과서의 화학 영역을 중심으로 Reference to the Chemistry Field of Science Textbooks(Ⅰ) in the Middle School

        This study treats of the comparative analysis of the science textbooks in relation to the inquiry learning ability and STS viewpoint in order to offer some aids that help teaching/learning in class. The major findings of the study are summarized below. 1. The textbooks on the 6th curriculum contain more variety of inquiry process than those on the 5th, which makes students get interested in their learning and get the result of experiments without making experiments. 2. The pictures on the 6th rose 27 percent compared with 5th. It can help students reduce the burden of learning and understand science more easily. 3. The textbooks on the 6th are intended to develop a balanced and varied inquiry ability, while the textbooks on the 5th are rather biased to several items. 4. The textbooks on the 6th have more contents and pictures connected with everyday science than those on the 5th.
