        생강 저장 중의 향기성분 변화

        This study was carried out to investigate the changes in volatile constituents concerning with the flavor of the green ginger (Zingiber officinale Roscoe) during storage in underground pit (15℃, RH 95%). And the constituents of essential oil of etiolated shoots formed on the mother rhizomes during the five months storage in the dark under same conditions were compared with those of mother rhizomes. The essential oils of Korean domestic ginger (Bong-dong cultivar) were isolated by simultaneous steam distillation and extraction method (SDE). Then the compositions of the essential oils were analysed by GC and GC-MS spectrometry. The major compounds of essential oil from the fresh rhizomes were zingiberene, citronellol+β-sesquiphellandrene, β-phellandrene, camphene, geranial, γ-bisabolene, ar-curcumene+geranyl acetate, α-pinene, β-gurjunene, limonene and neral. The content of monoterpene hydrocarbons increased with a concomitant lowering in the amounts of sesquiterpene hrdrocarbons and oxygenated sesquiterpenes during storage of rhizomes although contents of the oxygenated monoterpens changed little or slightly during the storage. During the storage the content of such monoterpenes as camphene, β-phellandrene and citral (neral and geranial) increased w hereas the content of such sesquiterpenes as zingiberene and citronellol+β-sesquiphellandrene decreased. The composition of shoot oil differed from that of mother rhizome oil in having higher content of terpene hydrocarbons and also in the higher content of bornyl acetate, β-gurjunene and ar-curcumene+geranyl acetate and lower in citral (neral and geranial).

        소동물 발광영상 측정을 위한 광학분자영상기기의 개발

        목적: 광학영상기술은 소동물이나 임상연구에서 분자영상법으로 알려진 첨단연구 분야이다. 광학영상기기는 소동물영상연구 및 추적연구에 중요한 역할을 수행하고 있다. 발광영상에서 소동물을 영상화 하기 위해서는 피부조직을 뚫고 나오는 광자를 검출하기 위한 고민감도 CCD카메라가 필요하다. 이 연구에서는 소동물에서 발생하는 발광신호를 검출하기 위해 개발한 광학영상기기를 소개하고자 한다. 대상 및 방법: 냉각형 CCD카메라와 집광렌즈, 8개의 백색광 LED 광원을 암실상자 안에 장치하였다. 팬텀 및 튜브를 이용한 영상을 얻은 후 발광 박테리아를 이용하여 CT26 암모델 누드마우스에서 영상을 획득하였다. 결과: 발광영상을 얻기 위한 광학영상기기를 설계하고 개발하였다. 영상획득이 성공적으로 수행되었고, 시스템을 완성하였다. 개발된 장비는 분자영상연구에 사용되고 있다. 결론: 개발된 광학영상장비는 다양한 실험적 조건을 만족하는 연구에 최적화하여 유용한 도구로 자리잡을 것으로 기대한다. Purpose: Optical imaging is providing great advance and improvement in genetic and molecular imaging of animals and humans. Optical imaging system consists of optical imaging devices, which carry out major function for monitoring, tracing, and imaging in most of molecular in-vivo researches. In bio-luminescent imaging, small animals containing luciferase gene locally irradiate light, and emitted photons transmitted through skin of the small animals are imaged by using a high sensitive charged coupled device (CCD) camera. In this paper, we introduced optical imaging system for the image acquisition of bio-luminescent signals emitted from small animals. Materials and Methods: In the system, Nikon lens and four LED light sources were mounted at the inside of a dark box. A cooled CCD camera equipped with a control module was used. Results: We tested the performance of the optical imaging system using effendorf tube and light emitting bacteria which injected intravenously into CT26 tumor bearing nude mouse. The performance of implemented optical imaging system for bio-luminescence imaging was demonstrated and the feasibility of the system in small animal imaging application was proved. Conclusion: We anticipate this system could be a useful tool for the molecular imaging of small animals adaptable for various experimental conditions in future.         Based on the establishment of a design contest for the H industrial complex dormitory establishment project, this study proposes various unit households and public facilities through legal review, case analysis, and prospective residents interviews, and aims to provide an optimal living environment for users. For the proper planning of the dormitory, it is necessary to consider the types and characteristics of single-person households according to various situations in the future, and to study the harmony of various classes and generations.

        젖소의 번식장애 및 비임신진단을 위한 Milk Progesterone 측정 EIA-kit의 이용에 관한 연구

        This study was carried out to investigate the effect, range of practice, and propriety for diagnosis of early non-pregnancies and reproductive disorders by dairy cows' milk progesterone analysis used EIA-kit of home products. The results were summarized as follows : 1. During 2 to 6 months after artificial insemination, the results of milk progesterone measurement by Home-kit and Auto ELISA reader-kit with pregnant dairy cows(152 heads) certified by rectal palpation were revealed, in Home-kit, 145 heads(95.4%) of positive reaction, 7 heads(4.6%) of quasi-positive, and 0 heads(0%) of negative among 152 heads and, in Auto ELISA reader-kit, 152 heads(100%) of positive reaction among 152 heads. 2. During 19 to 22 days after artificial insemination, the results of milk progesterone measurement by Home-kit, and thereafrer during 50 to 90 days after that, the results of pregnant test by rectal palpation were summarized as follows : 147 heads(82.1%) among 179 heads of positive reaction by Home-kit and 5 heads(31.3%) among 16 heads of quasi-positive were revealed pregnant cows by rectal palpation, and 42 heads(100%) among 42 heads of negative were non-pregnant. 3. During 19 to 22 days after artificial insemination, the results of milk progesterone measurement by Auto ELISA reader-kit, and thereafrer during 50 to 90 days after that, the results of pregnant test by rectal palpation were summarized as follows : 146 heads(86.9%) among 168 heads of positive reaction by Auto ELISA reader-kit and 6 heads(28.6%) among 21 heads of quasi-positive were revealed pregnant cows by rectal palpation, and 48 heads (100%) among 48 heads of negative were non-pregnant. 4. For the accuracy of the rectal palpation, Home-kit and Auto ELISA reader-kit were used in the cows of ovarian diseases. The results were following : in the cows of reproductive disorders expected negative milk progesterone, the accuracies of rectal palpation were the same 75.5%(40 heads among 53 heads) by Home-kit and Auto ELISA reader-kit, and in the cows of reproductive disorders expected positive milk progesterone, the accuracies of rectal palpation were 82.6%(19 heads among 23 heads) and 91.3%(21 heads among 23 heads) by Home-kit and Auto ELISA reader-kit, respectively, and the general accuracies of rectal palpation were 77.6%(59 heads among 76 heads) and 80.3%(61 heads among 76 heads) by Home-kit and Auto ELISA reader-kit, respectively.

        갈근에서 분리한 화합물의 $\alpha$-glucosidase와 $\alpha$-amylase 활성 저해 효과

        Compounds of isolated from roots extract of Pueraria thunbergiana were tested their inhibitory effects on $\alpha$-glucosidase and $\alpha$-amylase. Inhibitory activity of methylene chloride (MC) fraction and ethyl acetate (EA) fraction against $\alpha$-glucosidase showed more than 60% at a concentration of $500{\mu}g/m{\ell}$. Among the nine compounds tested on $\alpha$-glucosidase, biochanin A, (-)-tuberosin and calycosin from MC fraction and daidzein from EA fraction were stronger inhibitors than acarbose ($IC_{50}=530{\mu}g/m{\ell}$), and their $IC_{50}$ were 9, 144, 328 and $20{\mu}g/m{\ell}$, respectively. Biochanin A and (-)-tuberosin also inhibited $\alpha$-amylase activity as like as acarbose $IC_{50}=20.5{\mu}g/m{\ell}$), and their $IC_{50}$ were 22 and $348{\mu}g/m{\ell}$, respectively. Although daidzein was already known $\alpha$-glucosidase inhibitory effects, it was newly evaluated that biochanin A and (-)-tuberosin inhibited $\alpha$-glucosidase as well as $\alpha$-amylase, and that calycosin did $\alpha$-glucosidase.

        발광영상에 대한 정량화 방법 개발

        목적: 광학적 분자 발광영상은 발광효소를 이용하여 발광하는 빛의 신호를 영상화하는 기법이다. 발광하는 광량이 분자 변화 또는 세포 수와 비례하고 신호 대 잡음비가 좋아서 영상을 얻고, 정략분석이 가능하다. 이 연구에서는 정량적 분석을 위해 비례적 측정 정량화기법을 개발하였다. 대상 및 방법: 개발 중인 ALIS (animal light imaging system) 광학발광영상 카메라에서 박테리아 수를 달리한 박테리아 광원 3가지와 또 다른 3가지 광원을 이용하여 발광영상을 측정하였다. 일정한 세기의 광원에 대해서 측정 방법을 수학적으로 표현하기 위해 ㏅와 광속의 개념을 이용하여 간단한 수식을 유도하였다. 실험을 통해 측정시간 1초를 기준으로 얻어진 값으로 표준 정량화 함수를 얻었다. 얻어진 정량화 함수를 이용하여 박테리아를 이용한 실험에 필요한 함수의 상수 값을 구했으며, 세 가지 세기가 다른 광원을 이용하여 측정한 값을 측정시간과 함께 정량화 식에 대입하여 측정하였다. 결과: 표준측정함수를 이용하여 측정시간에 비례하는 정량적 값을 얻을 수 있었다. 정량화결과를 측정시간으로 나눠준 값은 일정하였으며, 측정시간에 대비한 비례적 값을 얻을 수 있었다. 결론: 측정한 결과를 정량화 함수에 대입하여 정량화시킨 값은 표준정량화 하기에 적합하였다. 이 정량화 방법은 다른 광학적 분자영상 장비에 적용하여도 빛의 세기를 표준화 시킬 수 있을 뿐 만 아니라 성격이 다른 각각의 광원에 대해서도 보다 정량적인 분석을 시행할 수 있으므로, 새로운 표준 정량화 방법으로 발전시킬 수 있을 것으로 기대한다. Purpose: Optical molecular luminescence imaging is widely used for detection and imaging of bio-photons emitted by luminescent luciferase activation. The measured photons in this method provide the degree of molecular alteration or cell numbers with the advantage of high signal-to-noise ratio. To extract useful information from the measured results, the analysis based on a proper quantification method is necessary. In this research, we propose a quantification method presenting linear response of measured light signal to measurement time. Materials and Methods: We detected the luminescence signal by using lab-made optical imaging equipment of animal light imaging system (ALIS) and different two kinds of light sources. One is three bacterial light-emitting sources containing different number of bacteria. The other is three different non-bacterial light sources emitting very weak light. By using the concept of the candela and the flux, we could derive simplified linear quantification formula. After experimentally measuring light intensity, the data was processed with the proposed quantification function. Results: We could obtain linear response of photon counts to measurement time by applying the pre-determined quantification function. The ratio of the re-calculated photon counts and measurement time present a constant value although different light source was applied. Conclusion: The quantification function for linear response could be applicable to the standard quantification process. The proposed method could be used for the exact quantitative analysis in various light imaging equipments with presenting linear response behavior of constant light emitting sources to measurement time.
