과일과 전통발효제로부터 추출한 단백질분해 조효소의 비교 특성

본 연구에서는 과일(무화과, 키위, 파인애플)에서 추출한 단백질분해 조효소와 전통발효제(개량누룩, 전통누룩, 메주, 입국)에서 추출한 단백질분해 조효소의 특징을 비교하였다. 전통발효제에서 추출한 단백질분해 조효소는 과일에서 추출한 단백질분해조효소보다 높은 온도(70°C), 높은 염 내성(1~3%), 그리고 낮은 pH(3~6)에서 활성을 보였다. 같은 조건에서 활성을 비교하였을 때, 바이오 누룩>입국>전통누룩>무화과>파인애플>메주>키위 순으로 활성이 높았다. 이러한 결과는 과일에서 추출한 단백질분해 조효소보다 전통발효제에서 추출한 단백질분해 조효소의 활성이 우수하고 고온, 산성, 그리고 염의 조건에서도 활성이 저하되지 않으므로, 소시지나 치즈 제조 등의 다양한 식품산업에 이용될 수 있는 가능성을 보여준다. In this study, we ascertained the characteristics of crude protease extracted from fruits (fig, kiwifruit and pineapple) as well as traditional Korean fermentation starters (bio nuruk, traditional nuruk, meju and rice koji) to determine their suitability for industrial application. Crude protease extracted from traditional Korean fermentation starters was found to have a higher optimum temperature (70°C) and salt concentration (1~3%) but a lower optimum pH (3~6) value compared to the corresponding values for the protease extracted from fruits. On comparison, the total activities of protease per gram unit follow the order: bio nuruk> rice koji> traditional nuruk> fig> pineapple> meju> kiwifruit. Based on our results, we conclude that protease extracted from traditional Korean fermentation starters has potential for application in food industry, for example, as a meat tenderizer for sausage manufacturing and as a protease for cheese production.

With the recent earthquake measuring 5.4 in Pohang, attention is being raised to the effects of the structures caused by the earthquake. In this study, seismic effect of the tunnel structure was analyzed by the time history analysis method using the measurement results of the nearby structures. Most of the analysis results showed displacement and stress results of one tenth level of the artificial seismic wave using design seismic acceleration, and the actual field survey revealed that the effects of earthquakes were minor.

The seriousness of current environment problems is demanding the drastic change in the view of environment. The purpose of this study was to find out the view of environment shown in oriental thoughts. The oriental thoughts were slightly different in the views of human, nature, and environment, but they had the important point in common. This was that human and nature were one, and human should follow nature. According to the oriental thoughts, human could not be separated from nature. Human were a part of nature, or nature itself. The ideal of human life was the recovery of natural states, and this could be obtained by lessening human artificial doings to nature, and throwing away their desires. The above oriental thoughts could not solve the environment problems completely. But the current views of environment should be changed in order to lessen the pollution of environment and prevent the ecological breakdown. This change could be obtained through education. Based on the above discussion, this study suggests that we should develope the practical new model of environmental movement and environmental education based on the oriental thoughts of human, nature, environment. The final suggestion is that we should study the traditional thoughts of India and Korea deeply in order to elucidate the oriental view of environment.
RAW 264.7 세포에서 발효 울금 추출물의 면역조절 효과

본 연구에서는 울금의 선호도 및 이용율을 높이기 위해 Aspergillus oryzae를 이용하여 발효시킨 후 용매별 추출을 통하여 얻은 발효 울금 추출물들을 사용하여 면역조절 효과를 살펴보았다. MTT assay를 이용하여 실온 추출물 200 μg/mL, 열수 추출물 400 μg/mL, 20% 주정 추출물 200 μg/mL, 80% 주정 추출물 100 μg/mL의 농도가 적정하다고 판단되어 추후 실험에 사용하도록 하였다. 면역조절 효과를 보고자 대식세포의 탐식작용 능력, nitric oxide(NO), TNF-α 생성, NK 세포 활성, SC-1 세포에서 발현되는 LP-BM5 virus의 LP-BM5 eco 유전자 발현을 측정하였다. NK 세포의 활성을 측정한 결과 NK 세포는 YAC-1 세포에 대한 cytotoxity를 관찰했을 때 20% 주정 추출물이 가장 활성을 나타내는 것으로 확인하였다. 발효 울금 추출물 중 열수 추출물과 20% 주정 추출물은 대식세포를 활성화시킴으로써 탐식능력을 증가시켰다. NO 생성을 관찰했을 때 발효 울금처리에 의해 활성화된 대식세포는 효과를 나타내지 않았으나 80% 주정 추출물을 제외한 추출물에서 TNF-α 생성을 정상군에 비해 유의적으로 증가시켰다. SC-1 세포에 있는 LP-BM5 eco 유전자 발현을 비교하여 바이러스 복제 억제능을 측정한 결과 모든 추출물이 유의적으로 억제되었으며, 특히 20% 주정 추출물이 가장 많이 억제하고 있는 것을 확인할 수 있다. 이러한 연구 결과로부터 발효 울금이 면역조절 효과를 가진 천연 기능성 소재로 개발에 있어 기초자료로 활용할 수 있을 것으로 기대할 수 있다. Curcuma longa L. (CL) is a well known traditional medicinal plant that is also used in curries and mustards as a coloring and flavoring agent. However, CL is not usually used as a food source due to its bitter taste. We investigated the immunomodulatory effect of CL fermented by Aspergillus oryzae (FCL) on RAW 264.7 cells. FCL was extracted with cold water (CW), hot water (HW), 20% ethanol (20% EtOH) and 80% ethanol (80% EtOH), after which its effects on phagocytic activity, tumor necrosis factor-alpha (TNF-α), nitric oxide (NO) production, natural killer (NK) cell activity and mRNA expression of LP-BM5 eco were investigated. Phagocytic activity was increased in HW and 20% EtOH when compared to the control. The secretion of nitric oxide (NO) from RAW 264.7 cells did not change significantly relative to the control. However, TNF-α was significantly increased by the addition of FCL extracts. Moreover, FCL 20% ethanol extract showed a four fold increase in NK cell cytotoxity relative to the control group. Finally, we observed suppressed mRNA expression of LP-BM5 eco in FCL extracts, especially in the 20% ethanol extracts group. These results indicate that the FCL extracts can be used as a functional material due to their effective immunomodulating activities.
MMORPG 디아블로 Ⅲ 캐릭터 의상에 나타난 미메시스 특성

The purpose of this study is to analyze the specific mimesis types of common character costumes in MMORPGs, which have stories that are very similar in structure to ancient myths and fantasy novels. As the subject of the research I have selected Diablo 3 created by the American video game developer, Blizzard Entertainment Images of 5 characters, for both genders - Barbarian, Witch Doctor, Monk, Demon hunter, Wizard - were compiled for a total of 10 images, and these images were compared to popular archetypes in mythology, religion and literature. analyzed them separately according to the mimesis type. For this study, Articles and books containing the Aesthetics, Mimesis and Digital culture were researched in order to study the costume features and the Mimesis types and compare them to the costumes in the MMORPG. Firstly, the character Barbarian is a successor of the barbarian character of the previous product. The outfit worn by the game figure is based on the common images expressed in the movie <Conan-The Barbarian>. The makers created an analogous-imitation mimesis by replicating the image in an exaggerated and grand form. Second, the character Witch Doctor derived its motif from the Voodoo cult, and was expressed in analogous-imitation mimesis, using the very shades and patterns used in real life. Third, the character Monk, blending the image of a priest in Eastern Europe and images of its oriental counterparts, was expressed in transposition-diversion mimesis. Fourth, the Demon Hunter took the source of its inspiration from the image of a Witch hunter in the medieval times. The game character has a sharper sleeker figure than its model, and uses more acute shapes and darker gloomier colors expressed in an analogous-imitation mimesis. Finally, the character Magician took a formless character and expressed it in transposition- diversion mimesis taking as its basis some fantasy novels.

본 연구에서는 전통발효식품 13종과 천연물 4종에서 27균주를 분리한 후 생육속도가 빠르고 pH 저하 능력이 우수한 3종의 균주를 1차 선발하였으며, 분자생물학적 방법을 이용하여 동정한 결과 Staphylococcus warneri, Sta. epidermidis, Lactobacillus plantarum과 99% 상동성을 보였다. 선발된 3균주의 발효소시지 스타터로서 이용가능성을 알아보기 위해 발효소시지의 환경과 유사한 model-system에서 배양하며 pH 저하 능력, 총산 생성 능력, 생육 능력, 아질산염 소거 능력을 발효소시지의 스타터로 많이 사용되고 있는 5균주 및 상업용 2균주와 비교하였다. Modelsystem에서 pH 저하능과 총산 생성능, 생육능은 관련성이 있었으며, 상업용 균주보다 분리한 Sta. epidermidis DO 10-1, Lac. plantarum MLK 14-2가 우수한 결과를 나타내었다. 아질산염 소거능의 경우에도 분리한 3균주가 상업용 균주보다 상대적으로 빠른 속도를 보였다. 분리한 3균주는 발효소지지 스타터로서의 발효 소거 능력은 우수할 것으로 보이지만 Staphylococcus는 잠재적인 위험성이 제기되는 균주이므로 Lac. plantarum이 발효소지지 제조에 가장 적합할 것으로 사료된다. The aim of this study was to select the most suitable starter cultures for production of fermented sausages. A total of 27 strains isolated from Korean fermented foods and natural substances were characterized with respect to their physicochemical properties in a fluid (submerged) model system modified according to the special conditions of fermented sausages. Three of these strains were pre-selected for testing as potential cultures based on their ability to grow fast and initiate rapid acidification. The selected strains were identified by API and partial sequence analysis of 16S rRNA. The results exhibited sequence similarity to known sequences of Staphylococcus warneri, Staphylococcus epidermidis and Lactobacillus plantarum. Among them, relatively good growth properties and nitrite reduction activities were detected for S. epidermidis and L. plantarum and low pH values and high total acidities were observed in the model system fermented with these isolates compared with reference strains.

The constant flow control valve is used to control the flow rate of heating water in the large apartment complex and buildings. It is important to have similar heating flow rate in the apartments, even though the apartment is top or bottom floors. To achieve those purposes, the constant flow control valve was developed.<br/> The performance of this control valve is effected by hole area and discharge coefficients of the cartridge holes. The discharge coefficients of orifice hole in the cartridge were testes with various sizes of holes and various flow direction in the holes. The discharge coefficients decreased as the hole size increased due to the collision at the cartridge wall of water jet. The effects of the flow direction at the hole were not significant on the discharge coefficients.
