        갑상선안병증 환자의 일차배양한 안와섬유모세포에 대한 알파-토코트리에놀의 항섬유화 효과

        Purpose: To evaluate the antiproliferative and antifibrotic effects of α-tocotrienols in primary cultured orbital fibroblasts from thyroid-associated ophthalmopathy (TAO) patients. Methods: Orbital fibroblasts were cultured (5 eyes from TAO patients, 5 eyes from normal patients) and classified into a control group, α-tocotrienol group and α-tocopherol group. The cell viability was measured using the 3-(4,5-dimethylthiazol-2-yl)- 2,5-diphenyl tetrazolium bromide (MTT) assay. The proliferation of orbital fibroblasts was measured using the Click-iTTM assay. The collagen production of the control and α-tocotrienol groups was measured using a hydroxyproline assay. Results: The α-tocotrienol and α-tocopherol groups showed no cytotoxicity up to 150 μm in orbital fibroblasts from TAO and normal patients. The proliferation of orbital fibroblasts from TAO and normal patients was significantly inhibited with α -tocotrienol at 80 μm and 120 μm. The collagen production of orbital fibroblasts from TAO patients was significantly inhibited with α-tocotrienol at 120 μm. Conclusions: The results from the present study indicate that non-toxic concentrations of α-tocotrienol have significant antiproliferative and antifibrotic effects on orbital fibroblasts from TAO patients.       • 복강경하 엉치뼈질고정술 후 발생한 Mesh 미란의 수술적 치료

        The sacrocolpopexy has been the gold standard surgery for POP (pelvic organ prolapse). Though the rates of mesh-related complications after sacrocolpopexy are relatively low, the complications should be considered. This case report describes the clinical cases of removal of mesh eroding following hysterectomy, laparoscopic sacrocolpopexy and posterior vaginal repair. The surgery for complete mesh excision was performed at laparotomy or laparoscopy. Although any complications haven't been yet reported in two cases, careful follow-up management should be needed. Successful sacrocolpopexy without complications requires technically experienced pelvic surgeons, the most suitable surgical methods and the best graft material.

        Effects of Bevacizumab on Bcl-2 Expression and Apoptosis in Retinal Pigment Epithelial Cells under Oxidative Stress

        Purpose: To evaluate the effects of bevacizumab on expression of B-cell leukemia/lymphoma (Bcl)-2 and apoptosisin retinal pigment epithelial (RPE) cells under oxidative stress conditions. Methods: RPE cells were treated with H2O2 (0, 100, 200, 300, and 400 μM) and bevacizumab at or above thedoses normally used in clinical practice (0, 0.33, 0.67, 1.33, and 2.67 mg/mL). Cell apoptosis was measuredusing flow cytometry with annexin V-fluorescein isothiocyanate. The expression of Bcl-2 mRNA was determinedusing reverse transcription polymerase chain reaction. Results: Under low oxidative stress conditions (H2O2 100 μM), cell apoptosis was not significantly different atany concentration of bevacizumab, but Bcl-2 mRNA expression decreased with increasing concentration ofbevacizumab (0.33, 0.67, 1.33, and 2.67 mg/mL). Under moderate oxidative stress conditions (H2O2 200 μM),Bcl-2 mRNA expression decreased with increasing concentration of bevacizumab (0.33, 0.67, 1.33, and 2.67mg/mL), but cell apoptosis increased only at 2.67 mg/mL of bevacizumab. Under high oxidative stress (300μM) conditions, cell apoptosis increased at high concentrations of bevacizumab (1.33 and 2.67 mg/mL), but itdid not correlate with Bcl-2 expression. Conclusions: Withdrawal of vascular endothelial growth factor can lead to RPE cell apoptosis and influencesthe expression of anti-apoptotic genes such as Bcl-2 under oxidative stress conditions. Since oxidative stresslevels of each patient are unknown, repeated injections of intravitreal bevacizumab, as in eyes with age-relatedmacular degeneration, might influence RPE cell survival.

        포스트바이오틱스의 암예방 및 치료 효과

        The incidence of cancer has been rising expeditiously, and postbiotics have been receiving great attention from researchers for cancer therapy due to the side effects and complications of conventional cancer treatment. Postbiotics encompass a wide range of complex macromolecules such as inactivated microbial cells, cellular fractions or metabolites and provide various physiological health benefits to the host. Postbiotics exert anticancer effects by apoptosis, anti-proliferative and anti-inflammatory effects, as well as modulating the composition of the gut microbiota and the immune system. This review provides an overview of the conceptual and safety issues surrounding postbiotics, highlighting their biological role in the prevention and treatment of cancer. 암 발병률의 급격한 증가와 기존 암 치료에 따른 부작용과 합병증으로 인해 포스트바이오틱스를 이용한 암 치료에 대한 연구가 증가하고 있다. 포스트바이오틱스는 비활성화된 미생 물 세포, 세포 분획 또는 대사 산물과 같은 광범위한 생리 활성 인자이며 숙주에게 다양한 생리학적 건강상의 이점을 제공한 다. 포스트바이오틱스는 고위험 환자 또는 살아있는 프로바이 오틱 균주를 사용하기 어려운 기저 질환이 있는 환자를 위한 프로바이오틱스의 대안이 될 수 있으며 긴 유통기한, 무독성 및 소화 계통에 대한 안정성 등의 특성을 지닌다. 포스트바이 오틱스는 세포 사멸, 항증식 및 항염증 효과를 통해 항암 효과 를 나타낼 뿐만 아니라 장내 미생물 및 면역 체계의 구성을 조 절한다. 이를 통해 포스트바이오틱스가 부작용 없고 효과적인 새로운 항암제 후보 물질임을 시사한다. 이 논문은 암 예방 및 치료에 대한 포스트바이오틱스의 생물학적 역할을 강조하면 서 포스트바이오틱스를 둘러싼 개념 및 안전성 문제에 대한 개요를 제공한다.

        Anti-adipogenic effect of Lactobacillus fermentum MG431 and MG4244 through AMPK pathway in 3T3-L1 preadipocytes

        This study evaluated the anti-adipogenic effectsand mechanisms underlying the action of Lactobacillusfermentum MG4231 and MG4244 strains on adipogenesisand lipid accumulation in 3T3-L1 preadipocytes. Treatmentwith cell-free extracts (CFEs) from the two strainsreduced lipid accumulation and intracellular triglycerideproduction in 3T3-L1 adipocytes by more than 50%. Theinhibitory effects of L. fermentum on lipid accumulationwere mediated by the downregulation of FAS and aP2resulting from the inhibition of PPARc and C/EBPa geneexpression. Moreover, AMPK and HSL phosphorylationwas upregulated by CFE treatment. These results indicatedthat the anti-adipogenic and lipolysis activities of L. fermentumstrains were caused by increased AMPK and HSLphosphorylation. Both strains displayed high leucine arylamidaseand b-galactosidase enzymatic activity, withexcellent adhesion to epithelial cells. Therefore, we identifiedL. fermentum as potential new probiotics for theprevention of obesity.

        Effects of Continuous Application of Green Manures on Microbial Community in Paddy Soil

        Green manure crops have been well recognized as the alternative for chemical fertilizer, especially N fertilizer, because of its positive effect on soil and the environment. Hairy vetch and green barley are the most popular crops for cultivation of rice in paddy field. This study was conducted to evaluate effects of hairy vetch and green barley on soil microbial community and chemical properties during short-term application (three years). For this study, treatments were composed of hairy vetch (Hv), green barley (Gb), hairy vetch + green barley (Hv+Gb), and chemical fertilizer without green manure crops (Con.). Hv+Gb treatment showed the highest microbial biomass among treatments. Principal component analysis (PCA) showed that PC1 (73.0 %) was affected by microbial biomass and PC2 (21.5 %) was affected by fungi, cy19:0/18:1ω7c (stress indicator). Combined treatment with hairy vetch and green barley could be more efficient than green manure crop treatment as well as chemical fertilizer treatment for improvement of soil microorganisms.
