This study examined the association between functional sequence variants (FSVs) of myosin heavy chain 3 (MYH3) genotypes and collagen content in a Landrace and Jeju native pig (JNP) crossbred population. Four muscles (Musculus longissimus dorsi, Musculus semimembranosus, Musculus triceps brachii, and Musculus biceps femoris) were used for the analysis of meat collagen content, and the same animals were genotyped for the FSVs of the MYH3 gene by using PCR-RFLP (polymerase chain reaction-restriction fragment length polymorphism). Three FSVs of MYH3 genotypes were identified and had genotype frequencies of 0.358, 0.551, and 0.091 for QQ, Qq, and qq, respectively. QQ animals for the FSVs of the MYH3 genotypes showed higher collagen content in their M. longissimus dorsi (p < 0.001), M. semimembranosus (p < 0.001), M. triceps brachii (p < 0.001), and M. biceps femoris (p < 0.001) than qq homozygous animals. After the validation of this result in other independent populations, the FSVs of MYH3 genotypes can be a valuable genetic marker for improving collagen content in porcine muscles and can also be applied to increase the amount of collagen for biomedical purposes.

In this study, the nucleation behavior of acicular ferrite in 1 GPa class high strength steel weld metal was investigated. A 1 GPa class as-deposited weld metal specimen was prepared by single-pass welding using a prototype flux-cored wire, and a reheated weld metal specimen was obtained using Gleeble simulation technique. The typical inclusion in both specimens was composed of MnTi2O4 and amorphous. It was found that the inclusion potency for acicular ferrite nucleation was maximized in the as-deposited weld metal because of both the formation of Mn-depleted zone (MDZ) near the MnTi2O4 and the low interfacial energy between the MnTi2O4 and ferrite. In the reheated weld metal specimen, it was observed that the inclusion potency decreased possibly due to the homogenization of Mn in the MDZ that occurred during reheating thermal cycling.

The microstructure and mechanical properties of the high-strength low-alloy steel weld metals with a variation of nickel content were investigated. The weld metals with a variation of nickel content from 2.3 to 3.3 wt% were prepared using Gas Metal Arc Welding process. The amount of acicular ferrite decreased with increasing nickel content; this is accompanied with an increase in the region of bainite and martensite, hence the hardness and tensile strengths were increased with the increase in nickel content, whereas the impact energy was deteriorated.
Characteristics of Cold Cracking in the Heat-Affected Zone of Carbon Steel

The effects of hydrogen and microstructure on the cold cracking susceptibility of the heat-affected zone (HAZ) of carbon steel were investigated. Various carbon steel plates with tensile strengths of 400- and 500-MPa grade were employed, and the cold cracking susceptibilities of the HAZs were evaluated using implant tests. To investigate the effects of hydrogen and microstructure, implant test assemblies were produced with two types of welding electrodes (low-hydrogen and cellulose) and two different welding conditions (low and high heat inputs). The results indicate that the microstructure was more important than hydrogen in determining the cold cracking susceptibility of the HAZ of carbon steel.
