        Molecular Typing of Leuconostoc citreum Strains Isolated from Korean Fermented Foods Using a Random Amplified Polymorphic DNA Marker

        For preliminary molecular typing, PCR-based fingerprinting using random amplified polymorphic DNA (RAPD) is the method of choice. In this study, 14 bacterial strains were isolated from different Korean food sources, identified using 16S rRNA gene sequencing, and characterized through RAPD-PCR. Two PCR primers (239 and KAY3) generated a total of 130 RAPD bands, 14 distinct PCR profiles, 10 polymorphic bands, one monomorphic band, and four unique bands. Dendrogram-based analysis with primer 239 showed that all 14 strains could be divided into seven clades out of which clade VII had the maximum of seven. In contrast, dendrogram analysis with the primer KAY3 divided the 14 L. citreum strains into four clades out of which clade IV consisted of a maximum of 10 strains out of 14. This research identified and characterized bacterial populations associated with different Korean foods. The proposed RAPD-PCR method, based on sequence amplification, could easily identify and discriminate the lactic acid bacteria species at the strain-specific level and could be used as a highly reliable genomic fingerprinting tool.

        영아기 수신증에서 신장 초음파와 MAG3 스캔의 의의

        A prospective study had been performed in infants with hydronephrosis due to ureteropelvic junction stenosis to determine adequate management plan. Forty infants(47 renal units) were selected for the study during the period from January 1995 to July 1997 in the Department of Pediatrics, Asan Medical Center. They were diagnosed prenatally or early postnatally($lt;3 months of age) on the basis of renal ultrasonography and techotium-99m-mercaptoacetyltriglycerine(MAG3) scan. The antero-posterior pelvic diameter(APPD) was measured from serial renal ultrasonography and relative renal function and clearance half time(T1/2) were calculated from serial MAG3 scan. Those who had accom- panying other urogenital anomalies or vesicoureteral reflux were excluded in this study. Follow-up tests(ultrasonography and MAG3 scan) and surgical corrections, if necessary, were performed according to the pre-determined algorithrn. The male to female ratio was 3: 1. Hydronephrosis was involved the left side in 24 renal units, the right side in 9 renal units, and the bilateral sides in 14 renal units. All the hydronephrotic renal units were divided into 2 groups; Group A(19 renal units) those had taken surgical correction, and group B(28 renal units) those revealed spontaneous improvement without surgical correction. The mean follow-up duration was 7.2±6.3 months in group A and 14.1±12.6 months in group B. The mean APPD in the first postnatal renal ultrasonography was 19.4±6.8 mm in group A and 10.0±3.0mm in group B and this was statistically significant(P$lt;0.01). The relative renal function of group A in the first MAG3 scan was under 35% in 7 renal units and over 35M in 12 renal units. In group B, the relative renal function was over 35% in the all 28 renal units. Tl/2 in the first MAG3 scan was longer than 20 minutes in the all 19 renal units in group A. In group B, Tl/2 was shorter than 10 minutes in 15 renal units, between 10 minutes and 20 minutes in 6 renal units, and longer than 20 rninutes in 7 renal units. The mean APPD of the 7 renal units in group B of which Tl/2 was longer than 20 minutes was 12.62.6mm and that of group A was 19.4±6.8mrn and this was statistically significant(P$lt;0.01). In conclusion, the renal ultrasonography and MAG3 scan can be used effectively to determine the degree of the stenosis of the ureteropelvic junction in hydronephosis during infancy. When the babies are diagnosed with hydronephrosis due to ureteropelvic junction stenosis in the early infantile period, serial renal ultrasonography and MAG3 scan play an important role in the decision of the management.

        패턴 최적배치를 위한 유전 알고리즘의 응용에 관한 연구

        A good algorithm for optimum packing of materials is very important in that batch production of small quantities that requires a quirk response of the manufacturing system to the change in the market demand turns out to be an industrial trend. In spite of many researches on optimum packing, performance of pattern packing on the basis of computer aided automation if still inferior to that by manual operation from the material utilization point of view. In this paper we develop a software program based on genetic algorithm that is known to be proper for searching an optimal solution and try to ascertain whether the algorithm adopted is suitable and effective for improving the efficiency of the pattern laying process for apparel manufacturing. The pattern laying problem is divided into two parts: to determine a priority order of pattern and to achieve a high efficiency of materials by the order already given. For determining the order of placing patterns, a genetic algorithm was applied and for a pattern location a path pursuit algorithm. The results show that the pattern packing software, consisting of the genetic alogorithm and the path pursuit strategy used In this study, is one of the ways suitable for making the pattern laying process efficient and consistent, which was assessed indirectly by the random search method. The simulation reveals that the pattern laying efficiency of the method adopted in this study is about 85%, independent of the number of the pattern elements.

        원전 중성미자 검출을 위한 액체섬광검출용액의 감쇄거리 측정

        The antielectron neutrino is emitted during fission in a reactor. Through an inverse beta-decay reaction ($\bar{\nu_e} + p \rightarrow e^{+} + n$, IBD), the positron and the neutron generated in the liquid scintillator will be detected by using the time difference between the prompt signal and the delayed signal. During this process, light will travel a certain distance to reach the photomultiplier tube, so measurements of the optical and the physical properties of the liquid scintillator are very important. For that reason, a precise measurement of the attenuation length in a liquid scintillator is needed. This paper briefly describes a device for measuring the attenuation length in a liquid scintillator and presents the results of those measurements. 원전중성미자 검출은 원자로의 핵분열 과정에서 발생한 반전자 중성미자가 검출기내의 양성자와 충돌한 후, 양전자와 중성자로 변환되는 역베타붕괴 (inverse beta decay, $\bar{\nu_e} + p \rightarrow e^{+} + n$, IBD) 반응을 이용한다. 역베타붕괴에서 생성되는 양전자는 쌍소멸 하며 빛을 방출하고 중성자는 주변의 원자핵에 포획되는 과정에서 빛을 내게 되는데 각각의 신호에 일정 시간 차이를 주어서 구별할 수 있다. 원전 중성미자 표적으로 액체섬광검출용액을 사용할 수 있다. 이때 발생된 빛이 광증폭관에 잘 도달해야 여러 가지 물리적 성질 측정이 가능하기 때문에 액체섬광검출용액의 감쇄거리 측정은 매우 중요하다. 본 논문은 액체섬광검출용액의 감쇄거리를 측정하기 위한 측정 장비 개발 과정 및 결과에 대해 간략히 기술하였다.

        대학생 걷기 및 기립 증진을 위한 앱 기반 프로그램의 금전적 보상 및 푸시알림 효과성 연구

        신체활동 mHealth 앱은 걷기 및 기립 행동을 효과적으로 증진하기 위해 다양한 행동 증진 기술을 사용한다. 그중 금전적 보상과 푸시알림은 상용화된 앱에서 가장 자주 사용되는 기술이며, 일반적으로 조합의 형태로 사용된다. 하지만 이들의 조합 효과성에 대한 연구는 매우 부족하며, 개별 기술의 효과성 연구 결과도 혼재되어 있다. 이에 본 연구에서는 보상과 푸시알림이 걷기와 기립 행동 증진에 효과적인지 파악하기 위해 상호작용과 각각의 효과성을 확인하였다. 이를 위해 대학생 114명을 보상 및 푸시알림 유무에 따라 4개의 집단에 무선 할당하였다. 7일 간의 기저선 기간 동안 걷기와 기립 행동을 측정한 후, 집단별로 21일 간의 개입을 실시하였다. 연구 결과, 보상을 제공한 집단은 제공하지 않은 집단에 비해 걷기와 기립 행동에서 유의미한 증가를 보였다. 한편 푸시알림을 제공한 경우에는 유의미한 신체활동 증가가 나타나지 않았으며, 보상과 푸시알림의 상호작용 효과도 확인되지 않았다. 또한, 개입 만족도 및 유용성 측면에서, 보상 지급 집단의 개입 유용성, 목표 만족도, 목표 유용성 점수가 보상 미지급 집단보다 높았으나, 푸시알림 유무에 따른 차이는 없었다. 이러한 결과를 바탕으로 연구의 의의와 한계에 대해 논의하였다. Physical activity mHealth apps use a variety of behavioral promotion techniques to increase walking and standing behaviors. Financial rewards and push notifications are the most frequently used techniques in commercialized apps. And they are usually used in combination. However, there are few studies on their combination, and mixed results on the effectiveness of individual techniques. In this study, we examined their interaction and individual effectiveness to determine whether rewards and push notifications are effective in promoting walking and standing behaviors. To do so, 114 college students were randomly assigned to four groups with and without rewards and push notifications. Walking and standing behaviors were measured during a 7-day baseline period, followed by a 21-day intervention in each group. The results showed that the reward group showed significant increase in walking and standing behavior compared to the non-reward group. In terms of intervention satisfaction and usefulness, the rewarded group had higher intervention usefulness, goal satisfaction, and goal usefulness scores than the unrewarded group, but there was no difference in the presence of push notifications. Based on these results, the significance and limitations of the study are discussed.

        Bacillus pumilus TX703 유래 Xylanase 유전자(xynK)의 Cloning과 염기서열 분석

        A gene coding for xylanase from thermo-tolerant Bacillus pumilus TX703 was cloned into Escherichia coli DH5 $\alpha$ using pUC19. Among 7,400 transformants, four transformants showed clear zones on the detection agar plates containing oat-spells xylan. One of them which showed highest xylanase activity was selected and its recombinant plasmid, named pXES106, was found to carry 2.24 kb insert DNA fragment. When the nucleotide sequence of the cloned xylanase gene (xynK) was determined, xynK gene was found to consist of 1,227 base-pair open reading frame coding for a polypeptide of 409 amino acids with a deduced molecular weight of 48 kDa. The coding sequence was preceded by a putative ribosome binding site, the transcription initiation signals, and cia-acting catabolite responsive element. The deduced amino acids sequence of xylanase is similar to those of the xylanases from Hordeum vulgare (barley) and Clostridium thermocellum, with 39 and 31% identical residues, respectively. The amino acids sequence of this xylanase was quite different from those of the xylanases from other Bacillus species. Xylanase를 생산하는 내열성 Bacillus pumilus TX703의 chromosomal DNA로부터 xylanase 유전자를 cloning하여 그 염기배열 순서를 결정한 다음 이로부터 유전자 발현에 관련된 구조를 분석하였다. Xylanase 유전자의 cloning을 위해 제한효소 HindIII로 절단한 B. pumilus TX703의 chromosomal DNA와 pUC19을 ligation시켜 E. coli DH5 $\alpha$에 형질전환시킨 후 형질전환체 중에서 xylanase 활성을 나타내는 재조합 plasmid pXES106을 분리하였다. 재조합 plasmid pXES106은 pUC19의 HindIII 부위 내에 2.24 kb의 외래 DNA가 삽입되었고, 이 plasmid DNA를 분리하여 E. coli DH5 $\alpha$에 재형질전환시킨 결과 vector 내에 xylanase 유전자가 cloning되었음을 확인하였다. Cloning된 유전자의 염기배열을 분석한 결과 이 유전자의 총 크기는 2,187 bp였고 이는 409개기 아미노산을 coding 하는 open reading frame 1,227 bp를 포함하고 있었다. 이 염기배열은 ATG개시 codon으로부터 각각 193과 216 base 상류에 TTTAAT의 -10 box와 TCGAAA인 -35 box로 추정되는 염기배열이 존재하였고 -10 box로부터 7 bp하류에 전사개시점인 A가 위치하고 있었다. 또한, 개시 codon으로부터 432 bp 상류에 공통염기배열과 14개의 염기 중 11개의 염기가 일치하는 TGATGGCGTCGGCA의 catabolite responsive element (CRE)가 존재하였다. B. pumilus TX703의 xylanase와 아미노산배열의 유사성이 가장 높은 xylanase는 Hordeum vulgare의 isozyme X-I이었고 본 xylanase는 208번째와 322번째에 glutamic acid 잔기를 가지고 있어 Clostridium thermocellum, Dictyoglomus thermophilum, Thermotoga neapolitana 등에서 밝혀진 바와 같이 glutamic acid 부위가 xylanase의 활성부위라 여겨진다.

        Comparative Analysis of the Achievements and Limitations of Projects that Support Local Businesses in an Area Undergoing Gentrification: A case study on Brixton

        This research has been conducted to find answers to the questions of how projects that support local businesses work in areas undergoing gentrification and what are the achievements and limitations of these projects, based on the case studies of Brixton Pound and Pop Brixton supporting local businesses in Brixton in the London borough of Lambeth. To this end, this study collected data through interviews with local businesses belonging to each project and analysed them based on prior research on the projects, evaluation reports, and articles in the media. It began by investigating the supporting mechanisms of projects that support local businesses in the area, and then analysed and evaluated the projects’ contribution to local businesses and the impact on the local community according to these mechanisms. The two projects in the case study showed contrasts in their support mechanisms, governance, and the role of Lambeth Council, and the local businesses’ evaluations of the projects’ support differed depending not only on the support mechanism of each project but also on business type and perceptions of the impact of gentrification.
