In order to enumerate the T lymphocytes in bovine peripheral blood lymphocytes(PBL) by E rosette assay, KGRBC were treated with various concentration of 2-aminoethylisothiouronium bromide hydrobromide(AET) and dextran(Dex), singte or combination. The PBL from 10 clinically normal Korean native cattle were formed as low percentage of spontaneous E rosette(6.7±2.4%) in control group, whereas KGRBC treated with 0.1M AET for 20 minutes and 8%Dex were formed as 37.3±2.7% and 45.1±2.1%, respectively. And the synergistic effects were noted no less than 66.5±5.6% when the KGRBC treated with 0.1M AET and 8% Dex subsequently. These results suggest that the KGRBC would be a useful agent for the enumeration of T tymphocytes by E rosette assay in bovine PBI.
Eimeriosis in Korean indigenous calves with bloody diarrhea from March, 2006 to March, 2007

The aim of this study was to examine the epidemiological features of Eimeria in calves with acute diarrhea. Samples were collected from between 15 days and 90 days old calves (n=83) in Gimje area from March 2006 to March 2007. Feces of bloody diarrhea were examined for the presence of Eimeria oocysts using a sucrose flotation method. Out of 83 calves, 62 (74.6%) had Eimeria oocysts. In the results of monthly analysis, the highest prevalence(12.0%) of Eimeria oocysts was found on June. In the seasonal infection rate, spring was the highest prevalence(30.1%), followed by summer(24.0%). Furthermore, the highest prevalence(44.5%) was found in calves from between 31 - 60 days old in the analysis by ages. However, there was no significant differences between female and male sex even though the prevalence was slightly bigger in female than in male. The prevalence of the present study to detect Eimria oocysts for infection may have been affected by weather-conditions in the spring. Young calves should be separated to minimize the infection from cattle as much as possible. Additional studies are necessary to find other factors for infection and combining molecular methods with a highly sensitive system for Eimeria detection could be a reliable and economic way of Eimeria eradication.

This study was undertaken to develop the monoclonal antibody(MAb) for lymphocytes of Korean native cattle by the cell hybridization of myeloma P3/NS-1/ 1-Ag-4-1 and spleen cells from BALB/c mice hyperimmunized with nylon wool column eluted peripheral T lymphocytes of Korean native cattle. The isotype of MAb KCT-14 against T lymphocyte was mouse $IgG_1$. KCT-14 positivity of mononuclear cells(MNC) from peripheral blood lymphocytes, nylon wool nonadherent and adherent-lymphocytes was 41.7%, 58.4% and 22.6%, respectively. And that of mesenteric lymph node-, spleen and thymus-MNC was 43.3%, 40.2% and 33.6%, respectively. Immunoperoxidase staining of frozen tissue sections showed that the MAb positive cells were located in the medulla of the thymus and in the paracortical area and the mantle zone of the germinal center in the lymph nodes. These results indicated that KCT-14 was one of the MAb for investigate of T lymphocyte subpopulations in the Korean native cattle.
Detection of rotavirus and coronavirus from suckling Korean indigenous calves with acute diarrhea

In order to evaluation of prevalence of rotavirus and/or coronavirus, forty suckling Korean indigenous calves(between 2 and 98 days old) with acute diarrhea were investigated by the immunochromatographic rapid test in the field(veterinary practice) on October and on December 2006. Rotavirus and coronavirus were detected in 13 (32.5%) and 8(20.0%) of the fecal samples from diarrheal calves, respectively. The highest mortality rate in diarrheal calves occurred in the neonatal period from 2 to 7 days old. Totally, 40.0% of diarrheal calves showing acute enteritis were not detected with rotavirus and/or coronavirus. The rotavirus infection rate was significantly difference(p<0.05) between October and December, and the detection rate of rotavirus was bigger than that of coronavirus. These results suggested that rotavirus can be frequently associated with acute diarrhea of suckling calves and affected with changes of temperature.

A monoclonal antibody(MAb), KCT-23ER, with specificity for E-rosetted T cells of Korean native cattle was prepared by cell hybridization of myeloma P3/NS-1/1-Ag-4-1 and spleen cells from BALB/c mice hyperimmunized with E-rosetted lymphocytes. The isotype of KCT-23ER to T lymphocytes was mouse $IgG_{2b}$. KCT-23ER was reacted with 53.6% to peripheral blood lymphocytes and with 67.8% to nylon wool nonadherent blood lymphocytes. And it was reacted with 72.2%, 59.2% and 35.3% to thymocytes, prescapular lymph node cells and splenocytes, respectively. Immunocytological reactive rates to E-rosetted and non-E-rosetted cells were 72.5% and 22.4%, respectively. These results indicated that KCT-23ER reacted to E-rosetted cells was one of the MAb for investigate of $CD_2$ receptor positive cell subset in the Korean native cattle.
유소아의 조절호흡에 사용한 Mapleson B System 의 임상적 고찰

This study was undertaken to observe whether an adult ventilator with a preset volume could be used as a controlled ventrolled ventilator far pediatric anesthesia. 35 patients ranging in age from 3months to 7 years were divided into two groups based on body weight(Group 1: 5∼10kg, 14 cases, Group 2: 11∼15kg, 21 cases) and anesthetized with halothane-N_2O/O_2 - pancuronium using the Mapleson B system. Immediately after induction, the reservoir bay of the Mapleson B system was replaced by the reservoir tube of the adult ventilator (MCM 801). Arterial blood gas studies 30 and 60 minutes after induction were performed, and the data from group 1 was compared with that of group 2. The magnitude of PCO_2 increase 30 minutes after induction was not significantly different from that at 60 minutes(p$gt;0.05), and alterations of PCO_2 in group 1 were not statistically significant with group 2 (P$gt;0.05). In conclusion, it is suggested that the Mapleson B system attached to adult ventilator is an useful and convenient device f controlled ventilation in pediatric patients.

Experiments were performed on mice to investigate the effects of a polycycic aromatic hydrocatbon, 3-methylcholanthrene (MCA) on Arthus reaction, delayed-type hypersensitivity (DTH) and antibody production to sheep red blood cells (SRBC). Mice were sensitized iv with 0.1㎖ of 1% SRBC suspension were treated with a single ip injection of olive oil alone or with different doses of MCA in oil (0.5∼50㎎/㎏0 at various time before (-) or after (+) sensitization (day 0) and were challenged at 4 days after SRBC. Atrhus reaction was measured at 3 hours after challenge and other responses at 24 hours. Treatment with MCA inhibited Arthus reaction and DTH to SRBC, measured by footpad swelling reaction, and this immunosuppressing effect was dependent on the dose and time of MCA treatment in relation to SRBC sensitization. Humoral immune responses as measured by serum hemagglutinin-and hemolysin-titers to SRBC were significantly depressed when MCA was injected before or at the same time of sensitization. However, the response was slightly depressed when injected after SRBC. These result indicate that MCA suppress the function of the cells involved in immune responses.
