      • 구개주름의 형태형성과 Retinoic Acid로 유발된 이상

        Morphogenesis of palatal rugae and retinoic acid-induced anomalies, numerical and pattern disruptions, were studied to know the mechanism of normal rugae formation and the relations between these disruptions and retinoic acid teratogenecity. Sprague-Dawley rats and all trans retinoic acid of Sigma Chemicals were used in this experiment. Retinoic acid was administered at gestational day 8, 9, 10, 11, and 12 by the dose of 60mg/kg 80mg/kg 100mg/kg and 120mg/kg respectively per orally. At the gestational day 17.5, the rats were sac rificed under ether anesthesia and removed entire uterine horns into the saline-filled Petri dish. Congenital anomalies on the head and palatal rugae were observed with stereoscope. and light and transmission electron microscopic observation were performed to investigation histologic changes of the rugal epithelium and mesenchyme after retinoic acid treatment. The results were as follows; 1. Retinoic acid-induced congenital anomalies in this experiment were exencephaly and hydrocephalus in gestational day 8 and 9 groups, and cleft palate in gestational day 9 and 10 groups, and the number of anomalies was increased dose dependently. 2. Severe numerical variation and pattern disruptions such as division, medial and lateral fusion, cross, bifid rugae and rugal papillae were found on the gestational day 9 and 10 groups, and the relation between the retinoic acid teratogenecity and the numerical and pattern disruptions of the rugae were very close. 3. On TEM observation, the basal laminae under the rugal epithelium were absent at many areas and the filopodiae of the epithelial cells protruded into the underlying mesenchyme. According to the above results, it is considered that the mesenchymal cells of palatal shelf migrate into the designated areas of epithelium through the basal lamina, proliferate rapidly and form the rugae. Retinoic acid may inhibit the proliferation and migration into the epithelium of mesenchymal cells and the incompatibility between the growth of palatal shelf and the slowed speed of mesenchymal cells migration into the epithelium will leads the numerical and pattern disruptions of the rugae. And the relation between the teratogenecity of the retinoic acid and the numerical and pattern disruptions of the rugae was considered to be very close.

      • 사람 말초혈액 림프구에서 FudR에 의해 유도되는 Fragile Sites

        Chromosomal abnormalities including fragile sites induced by 10 ㎍/ml and 20 ㎍/ml fluorodeoxyuridine (FudR) were studied in lymphocytes cultures of six healthy individuals. A significant decrease in mitotic indexes in respect to control cultures was observed with two FudR concentrations used. The cells showing chromosome aberrations and the total number of cytogenetic alterations were significantly increased both FudR treated cultures with respect to control cultures. Damaged cells showing chromosome aberrations and total number of chromosomal aberrations were not significantly increased in FudR 20 ㎍/ml treated groups compared to FudR lO㎍/ml treated groups. The most frequent type of chromosomal aberration is a gap. A site showing a gap or break was defind as fragile if it appeated in 1% of cells analyzed and in at least three of six individuals studied with the same culture tre atment. Using these criteria, fragile sites 3p14, 6p25, 3q32, 1Oq22, 12q21 and 13q21 were induced in different proportions by different concentration of FudR.

        해부시신에서 발견된 덧콩팥동맥과 그들의 분지양상

        간추림. 덧콩팥동맥은 콩팥동맥의 발생학적 기형의 일종이다. 그것은 임상의학적 관점에서, 예를 들어 본태성 고혈압, 신혈관성 질환, 아래대정맥 폐쇄, 요관폐쇄, 덧콩팥정맥과 같은 다른 혈관성 기형의 발생, 외과적 중요성, 그리고 신장 이 식 동에 매우 중요한 의미를 가지고 있다. 그러나 최근까지 덧콩팥동맥의 연구는 시체해부에서 매우 희소하다. 22구의 해부시신 중 10구에서 12예의 덧콩팥동맥을 관찰한 내용을 보고하는 바이다. 1. 2예는 양측성 덧콩팥동맥이고, 8예는 편축성 덧콩팥동맥이었다. 2 .7예는 왼쪽 덧콩팥동맥, 6예는 오른쪽 덧콩팥동맥이었다. 3. 2예는 북강동맥과 상장간막동맥 사이에서, 7예는 상창간막동맥과 아래창자간막동맥 사이에서, 그리고 3예는 아래창자간막동맥 아래에서 기시하였다. 4. 덧콩밭동맥 중 7예는 가지를 내지 않았고, 4예는 3가지를 그리고 1예는 2가지를 분지하였다. 5. 덧콩밭동맥 7예는 콩팥문을 통해 실질내로 들어가고, 5예는 상단분절이나 전상분절로 들어가고, 5예는 아래분절로 들어갔다.

        Uitrastructure of Cryptoglena pigra from Korea

        Cryptoglena pigra Ehrenberg from Korea was a photosynthetic euglenoid alga, which had typical characteristics of the Euglenales. The ultrastructure examination of C. pigra revealed certain features which were distinctly photosynthetic euglenoid: one U-shaped chloroplast with thylakoid membranes; two paramylon grains appressed to both sides of the chloroplast; eyespot associated with the chloroplast but not part of it. Three flagellar roots were associated with the two basal bodies. The four-membered dorsal root arose from the dorsal body and extended anteriorly following the reservoir membrane. At the base of the reservoir the dorsal band was nucleated by the dorsal root and it ran anteriorly between the reservoir membrane and eyespot. The dorsal band was continued with the microtubules of the canal and the pellicle. The singlet dorsal microtubules at the transition level arranged into doublets by a successive linkage of the existing adjacent microtubules, and the doublets rearranged into the cytoskeletal microtubules that were continuous with four microtubules in pellicles. Finally, the sixteen ridges gave rise to the pellicular ridges. The five to six-membered ventral root extended anteriorly into a cytoplasmic pocket through the reservoir and lined a cytoplasmic pocket.

        Two Freshwater Cryptomonads New to Korea: Cryptomonas marssonii and C. pyrenoidifera

        We described two brownish freshwater Cryptomonas species, C. marssonii Skuja and C. pyrenoidifera Geitler as first records in Korea. The identification was based on light microscopy, scanning electron microscopy, and nuclear SSU rDNA sequences analysis. Cryptomonas marssonii is characterized by its sigmoid shape with a sharply pointed and dorsally curved antapex, dorso-ventrally flattened cell, two lateral plastids without pyrenoid, and its dimension of 18-25 μm in length and 8-13 μm in width. Cryptomonas pyrenoidifera is characterized by ovoid to elliptical shape with a partially twisted or rounded antapex, dorso-ventrally biconvex cell, lateral plastids with two pyrenoids, and the dimensions of 15-22 μm in length and 10-14 μm in width. Nuclear SSU rDNA sequences between C. marssonii WCK01 from Korea and CCAC0086 from Gernmay, and between C. pyrenoidifera WCK02 from Korea and CCMP152 from Australia were identical, respectively.

        Initial Preliminary Studies in National Long-Term Ecological Research (LTER) Stations of Daechung Reservoir

        Major objective of our study was to introduce initial researches of national long-term ecological monitoring studies on Daechung Reservoir, as one of the representative lentic reservoir ecosystems in Korea. For the long-term ecological research (LTER), we conducted preliminary field monitoring during 2008~2009 and analyzed biological parameters such as phytoplankton, zooplankton, and freshwater fish along with chemical water quality and empirical model analysis. According to phytoplankton surveys, major taxa have varied largely depending on seasons and sites sampled. Overall phytoplankton data showed that cyanophyta dominated in the summer period and diatoms dominated in the winter. In zooplankton analysis, 25 species including 20 rotifers, 3 cladocerans and 2 copepods were collected during the survey. The relative abundance of rotifers (86.5%) was always greater than that of cladocerans (6.3%) or copepods (5.1%). There were distinct spatial and inter-annual changes in the abundance of zooplankton in the reservoir, displaying similar patterns in three sites with the exception of S3 during the study. According to fish surveys, 8 families and 39 species were observed during 2008~2009. The most dominant fish was an exotic species of Lepomis macrochirus (23%), indicating an severe influence of exotic species to the ecosystem. TP averaged $17.9\;{\mu}g\;L^{-1}$ ($6{\sim}80\;{\mu}g\;L^{-1}$), which was judged as a mesotrophy, and showed a distinct longitudinal gradients. TN averaged $1.585\;{\mu}g\;L^{-1}$ during the study and judged as hypereutrophic condition. Unlike TP, TN didn't show any large seasonal and spatial variations. Under the circumstances, nitrogen limitation may not happen in this system, indicating that nitrogen control is not effective in the watershed managements. These data generated in the LTER station will provide key information on long-term biological and water quality changes in relation to global warming and some clues for efficient reservoir ecosystem managements.
