        영지버섯(Ganoderma lucidum)의 폴리페놀화합물과 Pancreatic Lipase 저해 활성

        In this study, a Ganoderma lucidum methanol extract was fractionated according to the polarity of the solvent, Diaion HP 20 gel column chromatography was used to isolate the polyphenol compounds in the ethyl acetate fraction that showed the highest antioxidant capacity. The anti-obesity effects of each fraction were examined by measuring, the pancreatic lipase inhibitory activities. The polyphenol compounds were identified by HPLC. The antioxidant capacity and pancreatic lipase inhibitory activities in the ethyl acetate fraction were the highest among them (P<0.05). To isolate the polyphenol compounds, the ethyl acetate fraction was obtained with 30%, 60%, and 90% ethanol fractions by Diaion HP 20 gel chromatography. The DPPH (1.93 mM gallic acid equivalent) and ABTS (5.36 mM Trolox equivalent (TE)) radical scavenging activity, FRAP activity (5.92 mM TE), total phenolic content (121.46 mg%), and total flavonoid content (24.91 mg%) in 60% ethanol fraction were the highest (P<0.05). The pancreatic lipase inhibitory activity in the 60% ethanol fraction (1,000 μg/mL) also showed the highest activity as 93.30% (P<0.05). HPLC was used to identify the polyphenol compounds (gallic acid, quercetin, naringenin, kaempferol, and hesperetin) from the G. lucidum methanol extract, ethyl acetate fraction, and 60% ethanol fraction. Among them, the 60% ethanol fraction showed the highest content of total polyphenol compounds. Hesperetin (186.19±5.76 μg/mL) and quercetin (178.60±6.41 μg/mL) were obtained at high levels, followed by naringenin, kaempferol, and gallic acid (P<0.05). 본 연구에서는 항비만 기능성 소재로서 영지버섯의 활용성을 증대하기 위하여 영지버섯을 메탄올로 추출하고 이를 용매의 극성에 따라 분획하였다. 그중 활성이 가장 우수한 ethyl acetate 분획물을 Diaion HP 20 gel chromatography에 의해 폴리페놀화합물을 분리하였다. 폴리페놀 분획물의 항산화능 및 pancreatic lipase 저해 활성을 조사하였으며, HPLC를 이용하여 폴리페놀화합물을 분석하였다. 용매 분획물 중 ethyl acetate 분획물에서 항산화능이 가장 높게 나타났으며, 활성이 가장 우수한 ethyl acetate 분획물을 Diaion HP 20 gel chromatography에 의하여 30%, 60%, 90% ethanol 획분을 얻었다. 60% ethanol 획분에서 DPPH(1.93 mM GAE) 및 ABTS(5.36 mM TE) 라디칼 소거 활성, FRAP 활성(5.92 mM TE)과 총 페놀성 화합물 함량(121.46 mg%) 및 총 플라보노이드 화합물 함량(24.91 mg%)이 가장 높았다(P<0.05). Pancreatic lipase 저해 활성 역시 60% ethanol 획분(1,000 μg/mL)에서 93.30%로 가장 높은 활성을 나타내었다(P<0.05). HPLC를 이용하여 메탄올 추출물, ethyl acetate 분획물, 60% ethanol 획분(20 mg/mL)에서 폴리페놀화합물인 gallic acid, quercetin, naringenin, kaempferol, hesperetin을 확인하였다. 그중 60% ethanol 획분에서 hesperetin과 quercetin의 함량이 각각 186.19±5.76 μg/mL, 178.60±6.41 μg/mL로 가장 높았으며, naringenin, kaempferol, gallic acid 순으로 높았다(P<0.05). 따라서 영지버섯의 60% ethanol 획분의 강한 항산화능 및 pancreatic lipase 저해 활성을 나타내는 주요 활성 물질이 hesperetin 및 quercetin일 것으로 사료된다.

        제주 동백나무 겨우살이를 첨가한 막걸리의 항산화 및 항비만 활성

        The objective of the present study was to evaluate in vitro antioxidant activity, antioxidant content and pancreatic lipase inhibitory activities of Makgeolli supplemented with 0, 1, 2, and 4% (w/v) Jeju camellia mistletoe during fermentation. Total phenolic and flavonoid contents tended to increase as content of Jeju camellia mistletoe increased. Supplementation with Jeju camellia mistletoe resulted in a significant increase in the scavenging capacities of 1,1-diphenyl-2-picrylhydrazyl, hydrogen peroxide, nitric oxide and superoxide anion radicals, and reducing power activity. Moreover, pancreatic lipase inhibitory activity was significantly elevated by Jeju camellia mistletoe addition. These results suggest that Jeju camellia mistletoe is considered to be a good material to improve antioxidant and pancreatic lipase inhibitory activities of makgeolli.

        In Vitro a-Amylase, a-Glucosidase, Pancreatic Lipase, Xanthine Oxidase Inhibiting Activity of Agaricus bisporus Extracts

        In this study, the a-amylase inhibitory activity, a-glucosidase inhibitory activity, pancreatic lipase inhibitory activity, and Xanthine Oxidase inhibitory activity of the fruiting body extracts of 5 varieties of Agaricus bisporus (AB) were confirmed. First, the a-amylase inhibi- tory activity of AB12, AB13, AB18, AB34, and AB40 methanol extracts was lower than that of acarbose, a positive control, in all concentration ranges. The a-glucosidase inhibitory activity of the AB40, AB13, and AB12 methanol extracts at the extract concentration of 1.0 mg/mL was 80.5%, 81.3%, and 78.5%, respectively, similar to that of acarbose, a positive control. The pancreatic lipase inhibitory activity of the methanol extract of Agaricus bisporus fruiting body was significantly lower than that of the positive control orlistat in the concentration range of 50 1.000 (mg/mL). The Xanthine Oxidase inhibitory activity was 0.5 8.0 mg/mL of each extract, which was significantly lower than that of the positive control allopurinol in the same concentration range. However, the Xanthine Oxidase inhibitory activity of AB13 and AB40 at 8.0 mg/mL was about 70%, which was higher than that of other mush- rooms. In conclusion, five kinds of Agaricus bisporus fruiting bodies seem to have inhibitory effects on enzymes such as a-amylase, a-glucosidase, pancreatic lipase, and Xanthine Oxidase that degrade starch and protein. In particular, it has an inhibitory effect and a reduction effect on xanthine oxidase that causes gout, so it is expected that it can be developed and used as a food or health supplement with health functional properties through future research.

        <P>Twenty-three phenolic compounds were isolated from <I>Dioscorea opposita</I> by bioactivity-guided method and their inhibitory effect against pancreatic lipase was evaluated. A total of 15 isolates reduced lipase activity at IC<SUB>50</SUB> values of less than 50 µM and 3,3′,5-trihydroxy-2′-methoxybibenzyl showed the highest inhibition with an IC<SUB>50</SUB> value of 8.8 µM. This study is a first to reveal the pancreatic lipase inhibitory activity by both <I>D. opposita</I> and its isolated compounds.</P>

The rf value of the compound was 0.51 at TLC with butanol : methanol :water (4: 1: 2) solvent system. The molecular weight of the compound was determined to be 293 by EI-MS.

        In vitro α-Glucosidase and Pancreatic Lipase Inhibitory Activities and Antioxidants of Samnamul (Aruncus dioicus) during Rehydration and Cooking

        In vitro α-glucosidase and pancreatic lipaseinhibitory activities of ethanol extracts of samnamul (driedyoung shoots of Aruncus dioicus) during rehydration(soaking, 30 min boiling, and re-soaking in water) andsubsequent cooking at 180oC with/without perilla oil weredetermined for evaluation of its anti-diabetic and antiobesitypotential. Polyphenol and flavonoid contents weremonitored by spectrophotometry. Rehydration and boilingof samnamul reduced α-glucosidase and pancreatic lipaseinhibitory activities as well as polyphenol and flavonoidcontents, whereas soaking time did not have a significanteffect. Of the rehydration and cooking processes, 30 minboiling was the most critical in reducing anti-diabetic andanti-obesity activities as well as antioxidant contents. Perillaoil addition improved α-glucosidase inhibitory activity,possibly through polyphenol addition. These results suggestthat dietary intake of samnamul can reduce the risks ofdiabetes and obesity, and perilla oil can additionallyimprove its health benefits.

        토마토 발효액을 이용한 고추장의 이화학적 및 기능적 특성

        Bacillus subtilis SRCM100333로 발효한 토마토 발효액을 제조하고, 이를 사용하여 제조한 고추장의 이화학적 및 기능성 특성을 비교하였다. 대조구로는 토마토 발효액 대신 발효하지 않은 토마토 액을 첨가하였다. 2종의 토마토 고추장 시료는 수분함량 44-45%, 염도 6.2%, pH 4.7, 적정 산도 2.8 mL/g, 색도(L 값) 22-23을 나타냈으며 아미노산성 질소를 제외한 모든 항목에서 유의적인 차이가 없는 경향을 보였다. 토마토 착즙액과 발효액 및 이를 첨가한 고추장의 기능성 분석결과, α-glucosidase 저해(AGI) 활성, pancreatic lipase 저해(PLI) 활성, 항산화, 지방세포분화 억제활성을 보였으며 모든 시료에서 세포독성은 나타나지 않았다. 토마토 발효액은 토마토 착즙액에 비하여 더 높은 PLI 활성과 SO소거능을 보였다. 토마토 발효액을 사용하여 고추장 제조 시 52%의 NO 생성 억제능을 보였으나, 대조구인 토마토 착즙액 고추장에는 NO 생성 억제능이 확인되지 않았다. 발효하지 않은 토마토착즙액을 이용하여 고추장을 제조한 경우와 비교 하였을 때, 토마토 발효액을 첨가하여 제조한 고추장은 고유의 붉은색은 유지하면서 강한 매운맛은 감소하였으며 다양한 기능성이 향상된 것을 확인할 수 있었다. 이러한 연구결과를 토대로 다양한 소비자들의 기호를 충족시킬 수 있는 고추장 개발과 같은 추가적인 연구가 지속되기를 바란다. This study was conducted to investigate the physicochemical and functional properties of Gochujang in the presence of fermented tomato products. To accomplish this aim, tomatoes were fermented using Bacillus subtilis SRCM100333, and the products were used with Gochujang. As a control, non-fermented tomato liquid was used with Gochujang. There were no significant differences in quality characteristics except for amino nitrogen in Gochujang. The functional properties of tomato liquid (TL), fermented tomato product (FTP), Gochujang made with tomato liquid (TLG), and Gochujang made with fermented tomato products (FTG) were investigated. There were significant differences in the pancreatic lipase inhibition (PLI) and superoxide scavenging activity between FTP (87% and 28%, respectively) and TL (77% and 14%, respectively). Nitric oxide scavenging activity of FTG (52%) was higher than that of other groups (TL, FTP, and TLG). Overall, the results indicate that the fermented tomato can be used to develop functional Gochujang.

        추출용매에 따른 영지버섯(Ganoderma lucidum)의 항산화 및 소화효소 저해활성

        This study investigated the nutritional properties and biological activities of Ganoderma lucidum (GL). The round type of GL contained higher carbohydrate content, while the Nokgak type of GL contained higher crude ash, crude fat, and crude protein content. The most abundant amino acid, fatty acid, mineral, and soluble vitamin observed were valine (round type: 11.90 mg/g and Nokgak type: 17.18 mg/g), linoleic acid (round type: 47.56% and Nokgak type: 75.68%), potassium (round type: 116.50 mg/100 g and Nokgak type: 184.36 mg/100 g), and vitamin B3 (round type: 1.78 mg/100 g and Nokgak type: 1.81 mg/100 g), respectively. In addition, the β-glucan content were 34.15 g/100 g (round type) and 30.07 g/100 g (Nokgak type). The GL 70% ethanol extract at 40℃ showed higher radical scavenging as well as carbohydrate and lipid enzyme inhibition than other conditions. At 1 mg/mL of treatment with the 70% ethanol extract at 40℃ of round type GL, the DPPH, ABTS, hydroxyl radical scavenging, and α-glucosidase, α-amylase, and pancreatic lipase inhibition activities obtained were approximately 92.85, 99.74, 58.09, 89.68, 44.68, and 67.56%, respectively.
