      • Vibrio fluvialis에 의한 위장관염 1 예

        V. fluvialis는 해양 및 담수 서식 세균으로서 드물게 인체에서 위장감염을 일으킨다. 현재까지 외국에서 보고된 예는 있으나 국내에 보고된 예는 없었다. 저자들은 여름철에 조개(고막)를 먹은 후 심한 복통과 설사 및 탈수증 등의 증상이 1주일간 지속된 55세의 간경변 남자 환자에서 대변배양 검사상 확진된 V. fluvialis에 의한 위장관염 1예를 적극적인 항생제 치료를 하였으나, 간경변 합병증으로 사망한 증례를 경험하였기에 이에 보고하는 바이다. V. fluvialis is a gram-negative, oxidase-producing, halophilic bacterium. It is normally found in coastal waters and seafoods. There have been a few reports on Vibrio fluvialis gastroenteritis in other countries, whereas as there has been no previous report of V. fluvialis infections in Korea. Reports from other countries showed that V. fluvialis was isolated mostly from infants and children. We experienced a race case of gastroenteritis due to V. fluvialis in a 55-year-old man with liver cirrhosis who ate an ark shell. He was admitted due to mild abdominal pain and severe watery diarrhea followed by rapidly progressive dehydration, electrolyte imbalance, and hepatorenal syndrome for a week. This patient was not improved by intensive care and antibiotic therapy.

        Vibrio fluvialis에 의한 균혈증 1예

        Vibrio fluvialis는 바닷물 또는 해산물에서 발견되는 호염성 그람음성 간균이며 주로 설사 및 위장관염을 일으킨다. 저자들은 71세 남자가 2주 전 해산물을 먹은 후 V. fluvialis에 의한 균혈증으로 내원한 드문 증례를 경험하였기에 보고하는 바이다. 그는 발열을 주소로 내원하여 혈액배양 검사에서 V. fluvialis가 확인되었고 수액 및 항생제 치료로 호전되어 내원 11일째 퇴원하였다. Vibrio fluvialis is a halophilic gram-negative bacilli usually found in sea water or seafood, and mainly cause gastroenteritis and diarrhea illness. We present a rare case of bacteremia caused by V. fluvialis in 71-years old man who has a history of sea food ingestion 2 weeks ago. He was admitted because of fever and V. fluvialis was isolated from blood. The patient was cured with fluid and antibiotics therapy and discharged after 11 days.

        Vibrio fluvialis에 의한 위장관염 1예

        V. fluvialis는 해수 및 담수에 서식하는 세균으로서 우리나라나 일본같이 해산물을 많이 섭취하는 나라에서 잘 발생함에도 불구하고 국내에서는 한 예만 보고된 바 있고, 그 환자는 간신증후군으로 사망하였다. 저자들은 지난 8월 해산물 섭취력이 없는 39세 알콜성 간질환 남자 환자의 대변 배양검사에서 V. fluvialis에 의한 위장관염을 확진하였고 급성신부전으로 악화되었으나 치료된 증례를 경험하였기에 이에 보고하는 바이다. Vibrio fluvialis is one of the non-cholera, halophilic, gram-negative bacterium that causes gastroenteritis. There have been a few reports of V. fluvialis infections worldwide. In the present report, one case of gastroenteritis caused by V. fluvialis which occurred in chronic liver disease patient in Korea is presented. This was a rare case of gastroenteritis caused by V. fluvialis in 39-years old man who had no history of seafood ingestion. He was admitted because of severe watery diarrhea and abdominal pain, and developed acute renal failure. The patient was improved after fluid and antibiotic therapy for 2 weeks.

        16S-3S rRNA Intergenic Spacer Region을 이용한 Vibrio fluvialis의 검출

        We have examined the 16S rRNA intergenic spacer regions(ISR) of Vibrio Fluvialis. ISRs were PCR amplified, cloned into a plasmid vector and then sequenced. As results of ISR nucleotide sequence analysis, total of 6 clones were isolated depending on the size. The clones were different in both the number and the composition of the tRNA genes, and were designated ISR-A, ISR-E, ISR-E1, ISR-IA, ISR-EKV, ISR-EKAV. ISR-A contains tRNA^(Ala) ; ISR-IA, tRNA^(Ile)-tRNA^(Ala) ; ISR-EKV, tRNA^(Glu)-tRNA^(Lys)-tRNA^(Val) ; ISE-EKAV, tRNA^(Glu)-tRNA^(Lys)-tRNA^(Ala)-tRNA^(Val) ; ISR-E and E1, tRNA^(Glu) clusters. ISR-EKV was shown to be a minor type out of the six ISR tyoes and showed a very limited homology between ISR-EKV from V. Fluvialis and ISRs from other Vibrio species. Therefore ISR-EKV sequence was used to design species-specific primers to detect V. fluvialis from other Vibrio species by PCR reaction . The specificity of the primers was examined using genomic DNA of other Vibrios as Templates for PCR reaction. The result showed that PCR can be a useful method to detect V. fluvialis among Vibrio species in a single PCR reaction.

        Vibrio fluvialis의 Oligopeptide Permease Gene 결손에 의한 생육과 Biofilm 생산의 비교

        Various ${\Delta}opp$ mutants of Vibrio fluvialis were constructed by allelic exchange method. The mutants occurred in target genes were confirmed by PCR and Southern hybridization analyses. After the exact mutants were identified, cell growth and biofilm production were examined using the respective mutants. The growth of wild strain was more rapid than mutants within 4hr incubation. Thereafter, the growth of wild strain and mutants reached to same level. When the productivities of wild strain and mutants were examined, ${\Delta}oppA$ mutant showed the highest productivity. Though ${\Delta}oppC,D$ and F mutants produced the lower production than that of ${\Delta}oppA$ mutant, the productivities of those mutants were much higher than that of wild strain.

        Vibrio fluvialis is a haplophilic gram-negative bacterium normally found in coastal water and seafood and causes gastroenteritis. There have been a few reports on V. fluvialis gastroenteritis in Korea, but no previous report of isolation from blood. We isolated V. fluvialis from the blood of two patients undergoing cancer chemotherapy.

        Vibrio fluvialis 유래의 hemolysin 정제와 생화학적 특성

        Hemolysin (VFH) of V. fluvialis, which is a pathogenic bacteria, causing watery diarrhea with vomiting, abdominal croup, was purified. V. fluvialis was cultivated in BHI medium and the culture supematant was precipitated by ammonium sulfate. The protein was purified by chromatographies on columns of DEAE-cellulose and Mono-Q. Molecular weight of the purified VFH was estimated as 79kDa by SDS-PAGE. The optimal temperature for a maximum hemolytic activity was at around 35$^{\circ}C$ and the activity was decreased at 4$0^{\circ}C$ Cytotoxicity of VFH was also investigated using RTG-2 cell line. LDH assay study showed that 50$\mu\textrm{g}$/m1 of VFH release 80% of total cellular LDH (lactate dehydrogenase) from RTG-2 cell and microscopic observation also showed the morphological change of cell.

        Biosynthetic Pathway of Indole-3-Acetic Acid in Basidiomycetous Yeast Rhodosporidiobolus fluvialis

        IAA biosynthetic pathways in a basidiomycetous yeast, Rhodosporidiobolus fluvialis DMKUCP293, were investigated. The yeast strain showed tryptophan (Trp)-dependent IAA biosynthesis when grown in tryptophan supplemented mineral salt medium. Gas chromatography-mass spectrometry was used to further identify the pathway intermediates of Trpdependent IAA biosynthesis. The results indicated that the main intermediates produced by R. fluvialis DMKU-CP293 were tryptamine (TAM), indole-3-acetic acid (IAA), and tryptophol (TOL), whereas indole-3-pyruvic acid (IPA) was not found. However, supplementation of IPA to the culture medium resulted in IAA peak detection by high-performance liquid chromatography analysis of the culture supernatant. Key enzymes of three IAA biosynthetic routes, i.e., IPA, IAM and TAM were investigated to clarify the IAA biosynthetic pathways of R. fluvialis DMKU-CP293. Results indicated that the activities of tryptophan aminotransferase, tryptophan 2-monooxygenase, and tryptophan decarboxylase were observed in cell crude extract. Overall results suggested that IAA biosynthetic in this yeast strain mainly occurred via the IPA route. Nevertheless, IAM and TAM pathway might be involved in R. fluvialis DMKU-CP293.

        Vibrio fluvialis oligopeptide permease (oppA) 유전자 deletion에 의한 생리적 특성

        Oligopeptide is known to be an essential nitrogen nutrient for bacterial growth. Oligopeptide can be transported into cytoplasm by a specific transport system, Opp system. Opp system is composed of five proteins, which are transcribed by an operon. These are responsible for oligopeptide binding protein (OppA), permease (OppB and OppC) and energy generation system (OppD and OppF), respectively. Previously, we isolated the opp operon from Vibrio fluvialis and constructed the oppA mutant by allelic exchange method. In this study, we investigated the growth pattern and biofilm production under the different growth condition. When the cells were cultivated using brain heart infusion(BHI) medium, the wild type was faster than the mutant in growth during the exponential phase. However, it showed that the growth pattern of two strains in M9 medium is very similar. The growth of wild type showed better than that of the mutant grown at pH 8. At pH 7, there was no an obvious difference in growth. After 5 mM $H_2O_2$ was treated to the cells $(OD_{600}=1.2)$, the cell survival was examined. The oppA mutation did not affect in survivability. In the presence of $10{\mu}g/ml$ polymyxin B, the biofilm production of the oppA mutant was higher than that of the wild type.
